Verfahren zur Herstellung von Verbindungen mit beruhigender Wirkung
Die Erfindung betrifft ein Verfahren zur Herstellung von neuen Verbindungen, welche gegen psychische und/ oder organische Störungen wirksam sind und eine spezifische Wirkung gegen Überreiztheit aufweisen.
Die neuen Verbindungen haben insbesondere wegen ihrer Anti-Tremor-Wirkung beträchtliches therapeutisches Interesse. Sie wirken beruhigend z.B. in Fällen nervöser Spannungen, Angstgefühlen, Furcht, Hemmungen, Phobien, Übermüdung, psychomotorisch bedingten Störungen, infantilen Störungen, Dunkelheitsfurcht, Stottem, nervösem Zucken, vasomotorische Gemütsstörungen und Zittern emotionellen, alkoholischen, medizinischen sowie geistigen Ursprungs.
Die neuen Verbindungen haben die Formel I

worin R1 und R2, die gleich oder verschieden sind, je ein Wasserstoffatom oder einen aliphatischen, araliphatischen oder aromatischen Rest, z.B. einen Alkyl- oder Alkenylrest, und R3 und R4 gleiche oder verschiedene, gegebenenfalls mit Alkoxyresten substituierte aliphatische Gruppen bedeuten, die mindestens einen Carbamoylrest tragen.
R1 und/oder R2 können z.B. Allyl-, Phenylreste oder Alkylreste von 1-6 Kohlenstoffatomen sein, z.B. Äthyloder Isopentylreste.
R3 oder R4 sind vorzugsweise mit n-Butoxyresten substituierte Propylreste.
Verbindungen der Formel I, worin jedoch R4 ein Wasserstoffatom ist, sind bekannt. Ihre Synthese und manche ihrer Eigenschaften sind in Helvetica Chimica Acta, XLIV, Seiten 105-113 (1960) beschrieben. Sie sind gute Sedative ohne hypnotische Eigenschaften.
Die neuen Verbindungen haben abweichend von diesen bekannten eine ausgesprochene Anti-Tremor-Wirkung und sind krampflösend.
So wurden z.B. Verbindungen der Formel I, worin R3 und R4 gleich sind und beide die Gruppe

darstellen, in vielen Fällen von Zittern verschiedensten Ursprungs mit Erfolg angewendet, ebenso zur Behebung von Trunkenheit und psychischer und seniler Störungen.
Es wurde auch festgestellt, dass die neuen Verbindungen überraschenderweise in Kombination mit solchen Verbindungen der gleichen Formel, worin jedoch R3=R4=H ist, also mit Malonylharnstoffen gleichfalls besondere beruhigende Wirkungen ausüben. Diese Verbindungen haben die Formeln

Man verwendet also zweckmässig Gemische aus 30 bis 45 Gew.-% einer Verbindung der Formel I, 40 bis 60 Gew.-% einer Verbindung der Formel II und 10 bis 20 Gew-% einer Verbindung der Formel III.
Die neuen Verbindungen der Formel I werden erfindungsgemäss hergestellt durch Einführung der Gruppe R4 an das unsubstituierte Stickstoffatom einer Verbindung der Formeln II und/oder III, indem man ein Alkalimetallderivat einer Verbindung der Formel II mit einer Verbindung der Formel Hal R4, worin Hal ein Halogenatom, vorzugsweise Chlor oder Brom bedeutet, umsetzt. Das Alkalimetall ist zweckmässig Natrium.
Wünscht man eine Verbindung der Formel I herzustellen, worin R3 und R4 gleich sind, kann man ein Dialkalimetallderivat einer Verbindung der Formel III mit einer Verbindung der Formel III mit dem Hal R4 umsetzen.
Wünscht man ein Gemisch der Verbindungen der Formeln I und II herzustellen, kann man ein Gemisch der
Die Eigenschaften sowie der therapeutische Index des als Schlafmittel und Sedativum bekannten Phenobarbitals sind genau beschrieben. Hingegen sind die Eigenschaften der Verbindungen der Formeln I und II sowie des oben angegebenen Gemisches völlig unerwartet und sehr verschieden. Nachfolgend werden die festgestellten Eigenschaften im Vergleich miteinander und mit einem der besten Tranquilizer, nämlich mit Meprobamat, angegeben.
In der folgenden Vergleichstabelle sind die Dosierungen in mg/kg Körpergewicht angegeben. Das erhaltene Gemisch der Verbindungen der Formeln I und II kann ohne deren Trennung als Arzneimittel dienen. Die Trennung der beiden Verbindungen wäre schwierig, kann jedoch mittels Chromatographie und/ oder fraktionierte Fällung erfolgen.
Die Barbiturate der Formel III sind bekannt; eine wichtige Verbindung dieser Klasse ist Phenyl-äthylmalonylharnstoff oder Phenobarbital.
Versuche haben gezeigt, dass die kombinative Wirkung der drei Verbindungen der Formeln I, II und III nicht nur von der bekannten Wirkung der Verbindungen der Formel III sondern auch von derjenigen der gegen Tremor viel weniger wirksamen Verbindungen der Formel II deutlich verschieden ist, und auch von der Wirkung der Verbindungen der Formel I, welche wohl sehr wertvolle Anti-Tremor-Wirkung besitzen aber nicht einschläfernd wirken.
Nachfolgend wird ein Versuch mit einem erfindungsgemäss erhältlichen Produkt ausführlich beschrieben. Bei diesem Versuch war der Barbiturat-Kern der Kern von Phenobarbital und die Gruppen R3 und R4 an den Verbindungen der Formeln I und II hatten die Struktur

Beim Versuch wurden die Wirkungen von Verbindungen der Formeln I und II miteinander sowie mit der Wirkung eines Gemisches dieser drei Verbindungen mit der Verbindung der Formel III verglichen.
Das Gemisch hatte die folgende Zusammensetzung in Gewichtsprozenten: Verbindung der Formel I 35% Verbindung der Formel II 50% Verbindung der Formel III (Phenobarbital) 15%
Aus diesem Versuch ist ersichtlich: a) Die Kombination der Verbindungen der Formeln I, II und III ist qualitativ in gleicher Weise wirksam wie das Meprobamat, aber in signifikant geringerer Dosierung; b) Der Einfluss der geringen Phenobarbitalmenge auf die Eigenschaften des Gemisches der drei Verbindungen, insbesondere was die Toxizität betrifft, ist sehr bemerkenswert. Es sei in dieser Hinsicht bemerkt, dass die Verbindung der Formel I an sich praktisch keine akute Toxizität aufweist.
Diese pharmakologischen Untersuchungen wurden ergänzt und bekräftigt mittels klinischer Versuche an mehreren Dutzend Patienten mit psychischen und organischen Störungen, insbesondere mit Tremor.
Die Versuche haben gezeigt, dass bei gleicher Dosierung a) die Verbindungen der Formel II an sich wirksame Sedative sind, aber gegen Tremor emotionellen sowie organischen Ursprungs schwächer wirken; b) die Verbindungen der Formel I an sich eine Anti Tremor-Wirkung aufweisen und sogar krampflösend wirken, aber bei der Behandlung emotioneller Störungen schwächer wirksam sind und keine hypnotischen Wirkungen haben; c) demgegenüber das Gemisch der Verbindungen der Formel I und II, sogar bei Zutat kleiner Mengen von Phenobarbital, unerwartete und aufsehenerregende Wirkungen zeigt, da die individuellen Eigenschaften der einzelnen Verbindungen stark erhöht werden.
Gemische entsprechender Mengen aller drei Verbindungen zeigen sämtliche Wirkungen der Tranquilizer (z.B. der Benzodiazepine oder des Meprobamats). Die Gemische sind besonders brauchbar bei allen eingangs angeführten Störungen. Sekundäre Wirkungen, zum Beispiel ständige Schlafsucht, Verdauungsstörungen und die bei vielen bekannten Sedativen auftretenden Persönlichkeitswechsel treten praktisch nicht auf. Ausserdem er möglicht bei alkoholischem Tremor das N-(2-Carbamoyloxy-3-n-butoxypropyl)-phenobarbital den Bruch der Kreiswirkung der ständigen Intoxikation und des Rückgreifens auf Alkohol zur Verhinderung des Tremors.
Beispiel 1 N,N'-Di-(2-carbumyloxy-3-butoxy-propyl)- phenolbarbital
Eine Lösung von 2,3 g metallischem Natrium, das in 50 ml Methanol gelöst ist, wird unter Rühren zu 25,4 g trockenem handelsüblichen Mononatriumphenobarbital zugegeben.
42 g 1-Chlor-2-carbamyloxy-3-butoxy-propan werden eingeführt und das Gemisch auf dem Wasserbad erhitzt, bis das Methanol destilliert. Es wird nach und nach, zum Schluss im Vakuum, abgedampft.
Wenn die Reaktion beendet ist, wird die Masse zur Entfernung des gebildeten Natriumchlorids mit heissem Wasser gewaschen. Der Rückstand wird in 500 ml Äther aufgenommen und die ätherische Lösung mit normalem Ätznatron gewaschen, bis ein aliquoter Teil der Waschflüssigkeit nach Zugabe von verdünnter Salzsäure keinen Niederschlag mehr bildet. Die ätherische Lösung wird schliesslich mit reinem Wasser bis zur Neutralität gewaschen, die Lösung über wasserfreiem Natriumsulfat getrocknet und der Äther verdampft. Die so erhaltene gewünschte disubstituierte Verbindung ist ein transparentes Harz. Ausbeute -42,5%.
Beispiel 2
N,N' -Di-(2-carbamyloxy-3-butoxy- -propyl)-phenobarbital
25,4 g Natriumphenobarbital werden in 100 ml Methanol suspendiert und 21 g 1-Chlor-2-carbamyloxy-3-butoxypropan zugegeben. Das Gemisch wird auf dem Wasserbad erhitzt und das Lösungsmittel nach und nach abgezogen, bis ein trockener Rückstand erhalten wird.
Der Rückstand wird dann acht Stunden auf 1000C erhitzt. Der erhaltene ölige Rückstand wird mit einer Mischung von 500 ml Äther und 100 ml normalem Ätznatron aufgenommen. Das Gemisch wird gründlich gerührt, die zwei Schichten werden getrennt und die ätherische Lösung nochmals extrahiert, bis ein aliquoter Teil der alkalischen Waschflüssigkeit mit Salzsäure keinen weiteren Niederschlag mehr gibt. Die ätherische Lösung wird darauf mit Wasser gewaschen, über Natriumsulfat getrocknet und der Äther verdampft, worauf man das gewünschte Produkt erhält. Ausbeute -35%.
Beispiel 3 N,N'-Di-(2-carbamyloxy-3-butoxy-propyl)-veronal
Natriummethoxyd wird wie in den vorhergehenden
Beispielen aus 23 g Natrium in 1 Liter absolutem Metha nol hergestellt und 184 g Diäthylbarbitursäure zugesetzt.
Damit sich das Reaktionsprodukt löst, ist Erhitzen auf Rückflusstemperatur erforderlich. Nach 30 Minuten wer den 300 ml Methanol und dann 209 g 1 -Chlor-2-carb- amyloxy-3-butoxy-propan, die zuvor geschmolzen worden sind, unter Rühren zugegeben. Das Methanol wird dann vollständig entfernt, zum Schluss im Vakuum bei einer
Temperatur von 920C, und zwar im Wasserbad. Wenn die Reaktion beendet ist, enthält das von Verunreinigun gen befreite Rohprodukt 36,4% der Verbindung I,
45,4% der Verbindung II und 18,2% der Verbindung III (nicht umgewandelt).
Beispiel 4
N,N' -Di-(2-carbatnyloxy-3-äthoxy-propyl)-amoharhital
In der gleichen Weise, wie in den vorhergehenden Beispielen beschrieben, werden 5,75 g Natrium in 150 ml Methanol gelöst und 56,7 g Amobarbital in 250 ml Methanol zugesetzt. Nach kräftigem Rühren werden 50 g 1 -Chlor-2-carbamyloxy-3 -äthoxy-propan eingeführt und die Lösung mit Methanol auf 400 ml gebracht. Das Methanol wird wie in den vorhergehenden Beispielen verdampft und 30% N,N'-Di-(2-carbamyloxy-3-äthoxypropyl)-amobarbital erhalten.
Beispiel 5
N,N' -Di-(2-carbamyloxy-3-butoxypropyl)-5,5- -diallyl-barbitursäure
63,7 g 1-Chlor-2-carbamyloxy-3-butoxy-propan werden in 250 ml Äthanol gelöst und das Gemisch gerührt und erhitzt, um Lösung zu bewirken. 70 g Natriumdiallylbarbiturat werden zu der Lösung zugegeben und das gesamte Äthanol nach und nach abdestilliert, wie in den vorhergehenden Beispielen. Die Reaktion wird durch Erhitzen im Vakuum bei 90-930C während 4 Stunden beendet, worauf ein Aufarbeiten wie in den vorhergehenden Beispielen erfolgt. Ausbeute 36,6% N,N'-Di-(2-carbamyloxy-3 -butoxy-propyl)-5,5-diallyl-barbitursäure.

In the experiment the effects of compounds of the formulas I and II were compared with one another and with the effect of a mixture of these three compounds with the compound of the formula III.
The mixture had the following composition in percent by weight: Compound of Formula I 35% Compound of Formula II 50% Compound of Formula III (phenobarbital) 15%
From this experiment it can be seen: a) The combination of the compounds of the formulas I, II and III is qualitatively in the same way as the meprobamate, but in a significantly lower dosage; b) The influence of the small amount of phenobarbital on the properties of the mixture of the three compounds, particularly with regard to toxicity, is very remarkable. It should be noted in this regard that the compound of the formula I per se has practically no acute toxicity.
These pharmacological investigations were supplemented and confirmed by means of clinical trials on several dozen patients with mental and organic disorders, especially tremors.
The tests have shown that with the same dosage a) the compounds of the formula II are effective sedatives per se, but have a weaker effect against tremors of emotional and organic origin; b) the compounds of the formula I per se have an anti-tremor effect and even have an antispasmodic effect, but are less effective in the treatment of emotional disorders and have no hypnotic effects; c) on the other hand, the mixture of the compounds of the formula I and II, even when adding small amounts of phenobarbital, shows unexpected and sensational effects, since the individual properties of the individual compounds are greatly increased.
Mixtures of corresponding amounts of all three compounds show all the effects of tranquilizers (e.g. benzodiazepines or meprobamate). The mixtures are particularly useful for all of the disorders mentioned at the outset. Secondary effects, such as constant sleep addiction, digestive disorders and the personality changes that occur with many known sedatives, practically do not occur. In addition, in alcoholic tremor, N- (2-carbamoyloxy-3-n-butoxypropyl) -phenobarbital breaks the circular effect of constant intoxication and recourse to alcohol to prevent the tremor.
Example 1 N, N'-Di- (2-carbumyloxy-3-butoxy-propyl) -phenol barbital
A solution of 2.3 g of metallic sodium dissolved in 50 ml of methanol is added to 25.4 g of dry, commercially available monosodium phenobarbital with stirring.
42 g of 1-chloro-2-carbamyloxy-3-butoxy-propane are introduced and the mixture is heated on the water bath until the methanol distills. It is gradually evaporated, finally in a vacuum.
When the reaction has ended, the mass is washed with hot water to remove the sodium chloride formed. The residue is taken up in 500 ml of ether and the ethereal solution is washed with normal caustic soda until an aliquot of the washing liquid no longer forms a precipitate after adding dilute hydrochloric acid. The ethereal solution is finally washed with pure water until neutral, the solution is dried over anhydrous sodium sulfate and the ether is evaporated. The desired disubstituted compound thus obtained is a transparent resin. Yield -42.5%.
Example 2
N, N '-di (2-carbamyloxy-3-butoxy-propyl) -phenobarbital
25.4 g of sodium phenobarbital are suspended in 100 ml of methanol and 21 g of 1-chloro-2-carbamyloxy-3-butoxypropane are added. The mixture is heated on the water bath and the solvent is gradually stripped off until a dry residue is obtained.
The residue is then heated to 100 ° C. for eight hours. The oily residue obtained is taken up with a mixture of 500 ml of ether and 100 ml of normal caustic soda. The mixture is stirred thoroughly, the two layers are separated and the ethereal solution is extracted again until an aliquot of the alkaline washing liquid with hydrochloric acid gives no further precipitate. The ethereal solution is then washed with water, dried over sodium sulfate and the ether evaporated, whereupon the desired product is obtained. Yield -35%.
Example 3 N, N'-di- (2-carbamyloxy-3-butoxy-propyl) -veronal
Sodium methoxide is used as in the previous ones

63.7 g of 1-chloro-2-carbamyloxy-3-butoxy-propane are dissolved in 250 ml of ethanol and the mixture is stirred and heated to effect dissolution. 70 g of sodium diallyl barbiturate are added to the solution and all of the ethanol is gradually distilled off, as in the previous examples. The reaction is terminated by heating in vacuo at 90 ° -930 ° C. for 4 hours, followed by working up as in the previous examples.
