Verfahren zur Herstellung von am Kohlenstoff des Heterocyclus alkylierten N,N'-disubstltuierten Diazacycloalkanen
Aufgabe der vorliegenden Erfindung ist die Herstellung von neuen, am Kohlenstoff des Heterocyclus alkylierten N,N'-disutbstituierten Diazacycloalkanen mit verbesserter coronardilatatorischer Wirksamkeit und ihren pharmakologisch unbedenklichen Salzen.
Es sind schon offenkettige und ringförmige N,N'-disubstituierte Diamine mit coronardilatorischer Wirksamkeit bekannt. In der deutschen Auslegeschrift 1 182 239 sind beispielsweise Verbindungen der allgemeinen Formel I

beschrieben. Keine dieser Verbindungen führte jedoch zu einem Handelsprodukt. Als Handelsprodukt bekannt geworden ist das z. B. in der österreichischen Patentschrift Nr. 231 432 beschriebene Produkt N,N'-Dimethyl-N,N'-bis- [3-(3',4',5' trimethoxybenzoyloxy)-propyl] äthylendiamindihydrochl'oA d.
Man ist jedoch stets bestrebt, neue Verbindungen zu schaffen, die in bezug auf ihre chemotherapeutischen, pharmakologischen und sonstigen Eigenschaften weiter verbessert sind.
Die erfindungsgemäss erhältlichen neuen, am Kohlenstoff des Heterocyclus alkylierten N,N'-disubstituierten Diazacycloaikane entsprechen der Formel II

worin R1, , R3 und R4, die gleich oder verschieden sein können, Wasserstoffatome oder niedermolekulare Alkylreste mit 1-4 Kohlenstoffatomen sind und mindestens eine der Gruppen Rot 4 ein solcher Alkylrest ist, m 0 oder die Zahl 1 und n 2 oder 3 bedeutet.
Die neuen Produkte zeichnen sich durch eine starke coronargefäss-erweitern,de Wirkung aus. Sie sind zu den wirksamsten Coronardilatatoren zu zählen.
Wegen ihrer Eigenschaft sind diejenigen Verbindungen der Formel II und ihre pharmakologisch unbe denklichen Säureadditionssalze bevorzugt, worin Rl, R2, R3 oder R4 Wasserstoff oder die Methyl- oder eine Äthylgruppe ist, m O oder die Zahl 1 und n die Zahl 2 oder 3 bedeuten.
Besonders bevorzugt sind dabei diejenigen Verbindungen der Formel II und ihre pharmakologisch unbedenklichen Säureadditionssalze, worin n die Zahl 3, Rt und R2 Wasserstoff und m O bedeutet und dabei R5 die Methyl- oder Äthylgruppe und R4 Wasserstoff oder die Methyl- oder Äthylgruppe ist oder n die Zahl 3, Rt und R2 Wasserstoff und m die Zahl 1 bedeutet und dabei R8 die Methylgruppe und R4 Wasserstoff oder die Methylgruppe ist.
Als pharmakologisch unbedenkliche Salze kommen insbesondere die Hydrochloride, Sulfate, Maleinate, Embonate, Citrate und Tartrate in Frage.
Die ganz besonders bevorzugte Verbindung ist diejenige der Formel III:

und ihre pharmakologisch unbedenklichen Säureadditionssalze.
In den pharmakologischen Versuchen hat sich gezeigt, dass insbesondere die Verbindung der Formel III in Form des Dihydrochlorids mehr als 2,5 mal so wirksam bezüglich der Steigerung der Myocarddurchblu- tung von Hunden ist, als das im Handel befindliche N,N'-Dimethyl-N,N'-bis- [3-(3',4',5'- trimethoxybenzoyloxy)-propyl] -äthylendiamindihydrochlorid (Hexobendin).
Die Myocarddurchblutung wurde an Hunden in Nforphin-Chlornlose-Urethan-Narkose mit Wärmeleitsonden bestimmt (Methode s. BETZ, BRAASCH und HENSEL, Arzneimittelforschung 11, S. 333-336 (1961)).
Zur quantitativen Auswertung der Dosis-Wirkungsbeziehungen wurde die Wirkung der Präparate mit dem Effekt einer festen Dosis (1 mg/kg) des bekannten Coronarmittels Prenylamin (LINDNER, Arzneimittelforschung 10, S. 569-573, (1960), verglichen und als DE 1,00 aus den Dosis-Wirkungskurven die Dosis extrapoliert, die 1 mg/kg Prenylamin (N-[3'-Phenyl-propyl-(2' > ] -1,1- diphenyl-propyl-(3)-amin) wirkungsgleich war. Zum Vergleich diente das nächstverwandte Handelspräparat Hexobendin (N,N'-Dimethyl-N,N'-bis-[3-(3',4',5'trimethoxybenzoyloxy)-propyl] -äthylendiamin).
Die Ergebnisse sind in der folgenden Tabelle 1 wiedergegeben

Die Umsetzungen werden vorzugsweise in Gegenwart eines geeigneten inerten organischen Lösungsmitteil, wie z. B. einem Alkohol, einem Äther wie Dioxan, einem aromatischen Kohlenwasserstoff wie Benzol, Toluol oder Xylol oder insbesondere Dimethylformamid und bei erhöhter Temperatur, insbesondere im Bereich von etwa 100"C bis etwa 1800 C bzw. dem Siedepunkt des verwendeten Lösungsmittels durchtge- führt.
Als Verbindung der Formel IV, in der X nicht die Gruppe -OMe ist, haben sich als besonders günstig die Säurehalogenide, insbesondere des Säurebromids oder -chlorids erwiesen, wobei in bevorzugter Ausfüh rungsform in Gegenwart eines inerten organischen Lösungsmittels und in Gegenwart eines säurebindenden Mittels, insbesondere eines tertiären Amins wie Tri äthylamin umgesetzt wird. Als inerte organische Lösungsmittel können hierbei insbesondere die Äther wie Dioxan oder aromatische Kohlenwasserstoffe wie Benzol, Toluol oder Xylol verwendet werden. Bevorzugt wird auch bei dieser Verfahrensvariante bei erhöhter Temperatur, vorzugsweise bei der Siedetemperatur des Reaktionsgemisches gearbeitet.
Anstelle der basischen Verbindungen VI und VIII können auch de ren Salze eingesetzt werden, wobei dann in Anwesenheit eines säurebindenden Mittels, wie z. B. Pottasche oder einem tertiären Amin gearbeitet wird.
Vorzugsweise werden die Reaktionskomponenten in äquivalenten Mengen eingesetzt. Basische Verbindungen können auch im Uberschuss angewandt werden, wobei der Überschuss als säurebindendes Mittel dient.
Die erhaltenen Basen der Formel II können gewünschtenfalls anschliessend in Säureadditionssalze umgewandelt oder aus entstandenen Salzen die Basen freigemacht werden.
Die Herstellung von Verbindungen der Formel VI ist z. B. in L. J. Kitchen u. E. S. Hanson, AM. Soc. 73, S. 1838 (1951). T. Ishiguro et al., J. pharm. Soc. Japan [Yakugakuzasschi] 78, S. 229 (1958), ref. C. A. 52, 11862 a (1958) sowie F. Poppelsdorf u. R. Myerly J. org. Chem. 26, 131 (1961) beschrieben.
Beispiel 1
N,N'-Bis-[y-(3', 4', 5'
Trimethoxybenzoyl-oxy) -propyl] methyl-piperazin-dihydrochiorid.
23 g (0,1 Mol) 3,4,5-Trimethoxybenzoylchlorid und 9,5 g (0,1 Mol) 1,3-Propylenchlorhydrin wurden 3 Std. auf 1000 C erwärmt und nach Zugabe von 50 ml Dimethylformamid, 5,0 g (0,05 Mol) 2-Methylpiperazin und 6,9 g (0,05 Mol) Kaliumkarbonat 20 Stunden bei 1200 C unter Rühren erhitzt.
Nach dem Verdünnen mit 300 mol Eiswasser wird auf pH 10 eingestellt und mit Methylenchlorid extrahiert. Die organische Phase wird mit verdünnter Salzsäure ausgezogen und die salzsaure Phase nach dem Alkalisiereu mit Äther extrahiert und getrocknet. Nach dem Abziehen des Lösungsmittels hinterblieben 6,1 g Öl. Das Öl wird in Benzol, dem etwa 20 /o Äthanol und eine Base wie Ammoniak oder Triäthylamin zugesetzt waren, gelöst und auf einer Kieselgelsäure chromatografiert. Das eingeengte Eluat wird mit einem Gemisch von Aceton und Ather aufgenommen und mit salzsaurem Äther gefällt. Aus Isopropanol umkristallisiert, erhält man ein Produkt vom F:185-1900 C.
Ausbeute: 3,8 g (d. s. 11 /o d. Th.)
Beispiel 2 N,N'-Bis-[y-(3',4',5'-Trimethoxybenzoyl-oxy)- propyl]-2-methyl- piperazin-dihydrochlorid
40,4 g (0,14 Mol) 3,4,5-Trimethoxybenzoesäure-(3'-chlorpropyl)-ester, 7 g (0,07 Mol) Methylpiperazin und 7 g (0,07 Mol) K2COa werden in 70 ml Dimethylformamid 25 Std. unter Rühren auf 120"C erhitzt. Das Reaktionsgemisch nimmt man in Wasser auf und extrahiert zweimal mit Methylenchlorid. Die vereinigten Auszüge wäscht man mit Wasser und trocknet die organische Phase über Natriumsuifat. Nach Abziehen des Lösungsmittels erhält man die Rohbase in einer Ausbeute von 34 g (800/o d. Th.). 25 g (0,04 Mol) der Rohbase werden in 100 ml Aceton gelöst, mit dem gleichen Volumen abs.
Ather versetzt und unter Rühren mit abs. salzsaurem Äther das Dihydrochlorid ausgefällt. Es wurde aus Isopropanol unter Zusatz von Tierkohle umkristallisiert.
F: 185-190 C.
Das Produkt zeigt gemischt mit dem Produkt des Beispiels 1 keine Schmelzpunktsdepression.
Beispiel 3 N,N'-Bis- [y-(3',4', 5'- trimethoxybenzoyl-oxy)-propyl] -cis
2,5-dimethyl-piperazin-dihydrochlorid
Eine Mischung von 40,4 g (0,14 Mol) 3 ,4,5-Trimethoxybenzoesäure-(3'-chlorpropyl)-ester, 8,0 g (0,07 Mol) cis-2,5-Dimethylpiperazin und 9,7 g (0,07 Mol) K2COS in 70 ml Dimethylformamid werden 25 Stunden unter Rühren auf 1200 C erhitzt. Das erkaltete Reaktionsgemisch wird in Wasser gegeben und das abgeschiedene Öl mit Äther extrahiert. Die vereinigten Atherauszüge wäscht man zweimal mit Wasser aus und schüttelt die ätherische Phase dann gut mit 2n Salzsäure durch. Die salzsaure Phase wird sofort abgetrennt, nach kurzer Zeit kristallisiert aus dieser das Dihydrochlorid aus, das man absaugt und aus Äthanol umkristallisiert.
F: 207-2090 C.
Beispiel 4 N,N'-Bis- [y-(3',4',5'- trimethoxybenzoyl-oxy)-propyl] -trans
2,5-dimethylpiperazin-dihydrochlorid
Eine Mischung von 40,4 g (0,14 Mol) 3,4,5-Trimethoxybenzoesäure (3'-chlorpropyl)-ester, 8,0 g (0,07 Mol) trans-2,5-Dimethylpiperazin und 9,7 g (0,07 Mol) K2CO3 in 70 ml Dimethylformamid werden 25 Stunden unter Rühren auf 1200 C erhitzt. Das erkaltete Reaktionsgemisch wird in Wasser gegeben und das abgeschiedene Produkt einige Male mit Methylenchlorid extrahiert. Die vereinigten Auszüge wäscht man zweimal mit Wasser aus. Beim anschliessenden Durchschütteln der Methylenchlorid-Phase mit 2 n Salzsäure kristallisiert das Dihydrochlorid aus. Man saugt ab und kristallisiert aus Dimethylformamid um.
F: 220-221 C.
Beispiel 5 N,N'-Bis-[y-(3',4',5'- trimethoxy-benzoyl-oxy)-propyl]
2-äthylpiperazin-dihydrochlorid
40,4 g (0,14 Mol) 3,4,5-Trimethoxybenzoesäure- (3'-chlorpropyl)-ester und 8,0 g (0,07Mol) 2-Äthylpiperazin mit 9,7 g (0,07 Mol) K2CO3 in 70 ml Dimethylformamid werden unter Rühren 22 Stunden bei 120"C erhitzt. Das erkaltete Reaktionsgemisch wird in Wasser gegeben, zweimal mit Äther extrahiert und die vereinigten Auszüge dann mehrhmals mit Wasser ausgewaschen. Die ätherische Lösung schüttelt man gut mit 2 n Salzsäure durch und trennt die salzsaure Phase danach schnell ab. Nach kurzer Zeit kristallisiert daraus das Dihydrochlorid aus, das abgesaugt und aus Isopropanol umkristallisiert wird.
F: 202-2070 C.
Beispiel 6
N,N'-Bis-[ss-(3',4',5' trimethoxybenzoyl-oxy)-äthyl]
2,5-dimethyl-piperazin-dihydrochlorid
Zu einer warmen Lösung von 10,1 g (0,05 Mol) N,N'-Bis-(ss-hydroxy-äthyl)-2,5-dimethylpiperazin und 12,1 g (0,12 Mol) Triäthylamin in 150 ml abs.
Benzol werden unter Rühren 27,7 g (0,12 Mol) 3,4,5 Trimethoxybenzoesäurechiorid in 50ml abs. Benzol getropft. Anschliessend erhitzt man 11/2 Stunden unter Rückfluss. Nach dem Erkalten wird vom Triäthylaminhydrochlorid abfiltriert und das Filtrat nach 3-maligem Waschen mit Wasser über Na-Sulfat getrocknet. Die nach dem Abziehen des Benzols erhaltene Base wird in Aceton aufgenommen und mit dem gleichen Volumen Ather verdünnt. Diese Lösung versetzt man unter Kühlung bis zur schwach sauren Reaktion mit salzsaurem Ather. Das dabei ausgefallene Dihydrochlorid wird abgesaugt, mit Äther nachgewaschen und getrocknet.
F: 223-226 C, Ausbeute: 33 g = 99,4 /o d. Th.
Beispiel 7
N,N'-Bis-ty-(3',4',5'- trimethoxybenzoyl-oxy)-propyl]-
2,5-dimethyl-piperazin-dihydrochlorid
Zu einer warmen Lösung von 11,5 g (0,05 Mol) N,N'-Bis-(A-hydroxy-propyl)-2,5-dimethylpiperazin und 12,1 g (0,12 Mol) Triäthylamin in 150 ml abs. Benzol werden unter Rühren 27,7 g (0,12 Mol) 3,4,5-Trimethoxybenzoesäurechlorid in 50 ml abs. Benzol getropft. Anschliessend erhitzt man 1 1/2 Stunden unter Rückfluss. Nach dem Erkalten wird vom Triäthylaminhydrochlorid abfiltriert und das Filtrat nach 3-maligem Waschen mit Wasser über Na-Sulfat getrocknet. Die nach dem Abziehen des Benzols erhaltene Base wird in Aceton aufgenommen und mit dem gleichen Volumen Äther verdünnt.
Diese Lösung versetzt man unter Kühlung bis zur schwach sauren Reaktion mit salzsaurem Äther. Das dabei ausgefallene Dihydrochlorid wird abgesaugt, mit Äther nachgewaschen und getrocknet.
F: 207-215 C, Ausbeute: 28 g = 80,90/o d. Th.
Beispiel 8
N,N'-Bis-[y-(3',4',5' trimethoxybenzoyloxy)-propyl] -cis
2,3-dimethyl-piperazin-dihydrochlorid
40,4 g (0,14 Mol) 3,4,5-Trimethoxybenzoesäure-(3'-chlorpropyl)-ester und 8,0 g (0,07 Mol) cis-2,3-Dimethylpiperazin werden mit 9,7 g (0,07 Mol) K2COS in 70 ml Dimethylformamid 20 Stunden unter Rühren auf 1400 C erhitzt. Das erkaltete Reaktionsgemisch wird in Wasser gegeben und dreimal mit Äther extrahiert. Die vereinigten ätherischen Auszüge werden zweimal mit verdünnter Salz säure ausgeschüttelt. Danach macht man die salzsauren Auszüge mit Natronlauge alkalisch und extrahiert wieder dreimal mit Äther. Die vereinigten Ätherauszüge werden über Na2So4 getrocknet. Die so erhaltenen 11 g (25,44/o d. Th.) Rohbase werden unter Kühlung in abs.
Äther mit der entsprechenden Menge salzsaurem Äther versetzt. Das ausgefallene Salz wird einmal aus Wasser und dann einmal aus Isopropanol umkristallisiert.
F: 186-187 C.
Beispiel 9
N,N'-Bis-[y-(3',4',5' trimethoxybenzoyloxy) -propyl]-5,7- dimethyl-homopiperazin-dihydrochlorid
25 g (0,075 Mol) 3 ,4,5-Trimethoxybenzoesäure-(3 '-brompropyl)-ester und 10 g (0,075 Mol) 5,7-Dimethylhomopiperazin wurden in 75 ml Dimethylformamid 24 Stunden auf 1000 C erhitzt. Nach dem Abdestillieren des Lösungsmittels wurde in Wasser aufgenommen und mit Essigester ausgeschüttelt. Die organische Phase wurde mit Wasser gewaschen und zweimal mit verdünnter Salzsäure extrahiert. Die saure Lösung wurde mit Kaliumkarbonat gesättigt und das ausgeschiedene Öl mit Äther extrahiert. Der nach dem Trocknen und Eindampfen verbliebene Rückstand wurde in Alkohol gelöst und mit ätherischer Salzsäure in das Dihydrochlorid überführt.
Ausbeute: 8 g, 29 /o d. Th. Farblose Kristalle
F: 195-200 C.
Beispiel 10
N,N'-Bis-[ss-(3',4',5'- trimethoxybenzoyloxy)-äthyl]-5,7- dimethyl-homopiperazin-maleat
25 g (0,10 Mol) 3,4,5-Trimethoxybenzoesäure-(2-chloräthyl)-ester und 12,8 g (0,10 Mol) 5,7-Dimethylhomopiperazin wurden in 100 ml Alkohol abs. 55 Stunden unter Rückfluss erhitzt. Nach dem Abdestillieren des Lösungsmittels wurde bis zur Isolierung der Blase, wie im Beispiel 9 angegeben verfahren. Die Base wurde in ätherisch-alkoholischer Lösung mittels Maleinsäure in das Maleat überführt. Die farblosen Kristalle haben einen Schmelzpunkt von F: 121-124 C.
Beispiel 11 N,N'-Bis-[7o-(3',4',5'- trimethoxybenzoyloxy)-propylj-5- methyl-homopiperazin-dihydrochlorid
In 120 ml Benzol werden 27,6 g (0,12 Mol) 3,4,5 Trimethoxybenzoylchlorid, 11,5 g (0,05 Mol) N,N'-(y-Hydroxypropyl)-5-methyl-homopiperazin und 10,1 g (0,10 Mol) Triäthylamin gelöst, wobei Erwärmung auftrat. Es wird 2 Stunden unter Rückfluss erhitzt und nach dem Abkühlen Triäthylamin-Hydrochlorid durch Filtration entfernt. Die Benzollösung wird mit verdünnter Salzsäure extrahiert und anschliessend die Base nach dem Alkalisieren der wässrigen Schicht ausgeäthert. Die äthanolische Lösung des Atherrückstandes wird mit ätherischer Salzsäure in das Dihydrochlorid überführt. Nach zweimaligem Umkristallisieren aus Alkohol werden 17g (d. s. 55 /o d.
Theorie) der gewünschten Verbindung vom Schmelzpunkt 195-200 C erhalten.

The new products are characterized by a strong coronary vessel-expanding effect. They are among the most effective coronary dilators.
Because of their properties, those compounds of the formula II and their pharmacologically harmless acid addition salts are preferred in which R1, R2, R3 or R4 is hydrogen or the methyl or ethyl group, m is O or the number 1 and n is the number 2 or 3.
Particularly preferred are those compounds of formula II and their pharmacologically acceptable acid addition salts in which n is 3, Rt and R2 is hydrogen and m is O and R5 is the methyl or ethyl group and R4 is hydrogen or the methyl or ethyl group or n is the Number 3, Rt and R2 is hydrogen and m is the number 1 and R8 is the methyl group and R4 is hydrogen or the methyl group.

The pharmacological tests have shown that the compound of the formula III in particular in the form of the dihydrochloride is more than 2.5 times more effective in increasing the myocardial blood flow in dogs than the commercially available N, N'-dimethyl N, N'-bis [3- (3 ', 4', 5'-trimethoxybenzoyloxy) propyl] ethylenediamine dihydrochloride (hexobendine).
The myocardial blood flow was determined in dogs under Nforphin-Chlornlose-Urethane anesthesia with heat conduction probes (method see BETZ, BRAASCH and HENSEL, Arzneimittelforschung 11, pp. 333-336 (1961)).
For the quantitative evaluation of the dose-effect relationships, the effect of the preparations was compared with the effect of a fixed dose (1 mg / kg) of the known coronary drug prenylamine (LINDNER, Arzneimittelforschung 10, pp. 569-573, (1960), and recorded as DE 1 00 the dose was extrapolated from the dose-response curves, which 1 mg / kg prenylamine (N- [3'-phenyl-propyl- (2 '>] -1,1-diphenyl-propyl- (3) -amine) had the same action. 23 g (0.1 mol) of 3,4,5-trimethoxybenzoyl chloride and 9.5 g (0.1 mol) of 1,3-propylene chlorohydrin were heated to 1000 ° C. for 3 hours and, after addition of 50 ml of dimethylformamide, 5.0 g (0.05 mol) of 2-methylpiperazine and 6.9 g (0.05 mol) of potassium carbonate were heated at 1200 ° C. for 20 hours with stirring.
After dilution with 300 mol of ice water, the pH is adjusted to 10 and the mixture is extracted with methylene chloride. The organic phase is extracted with dilute hydrochloric acid and the hydrochloric acid phase is extracted with ether and dried after the alkaline solution. Example 2 N, N'-Bis- [y- (3 ', 4', 5'-trimethoxybenzoyl-oxy) propyl] -2-methyl-piperazine dihydrochloride
40.4 g (0.14 mol) of 3,4,5-trimethoxybenzoic acid (3'-chloropropyl) ester, 7 g (0.07 mol) of methylpiperazine and 7 g (0.07 mol) of K2COa are dissolved in 70 ml Dimethylformamide heated to 120 ° C. for 25 hours with stirring. The reaction mixture is taken up in water and extracted twice with methylene chloride. The combined extracts are washed with water and the organic phase is dried over sodium sulfate. To a warm solution of 11.5 g (0.05 mol) of N, N'-bis (A-hydroxypropyl) -2,5-dimethylpiperazine and 12.1 g (0.12 mol) of triethylamine in 150 ml Section. 40.4 g (0.14 mol) of 3,4,5-trimethoxybenzoic acid (3'-chloropropyl) ester and 8.0 g (0.07 mol) of cis-2,3-dimethylpiperazine are added with 9.7 g (0.07 mol) K2COS in 70 ml of dimethylformamide heated to 1400 ° C. for 20 hours with stirring. 25 g (0.10 mol) of 3,4,5-trimethoxybenzoic acid (2-chloroethyl) ester and 12.8 g (0.10 mol) of 5,7-dimethylhomopiperazine were dissolved in 100 ml of alcohol abs. Heated under reflux for 55 hours.
