Verfahren zur Herstellung von Hefe, vorzugsweise von Futterhefe
Die vorliegende Erfindung bezieht sich auf Verfahren zur Herstellung von Hefe, insbesondere von Futterhefe auf wässerigen Nährmedien, die als Quelle der Kohlenstoffnahrung der Hefe Hydrolysate von pflanzlichen Geweben oder Kohlenwasserstoffe der n-Paraffinreihe bei der Belüftung des Mediums enthalten.
Es sind bekannt Verfahren zur Herstellung von Hefe auf wässerigen als Kohlenstoffquelle Hydrolysate von pflanzlichen Geweben enthaltenden Nährmedien, in denen die Wachstumshemmstoffe der Hefe (Furfurol, Oxymethylfurfurol (sowie die pflanzlichen Gerbstoffe, phy siologisch wirksame Kolloide und andere einen negativen Einfluss auf die Prozesse deren Atmung und Vermehrung ausüben.
Dadurch sinkt die Ausbeute an Hefe und wird deren Vermehrung verlangsamt. Zur Steigerung der Ausbeute müssen die Nährmedien stark verdünnt werden (bis zu einer Konzentration der reduzierenden Stoffe von etwa in,). was die Verwendung der Behälter der Apparatur negativ beeinflusst.
Bei der Anwendung der bekannten Verfahren zur Züchtung von Hefe auf wässerigen Nährmedien, die als Kohlenstoffquelle Kohlenwasserstoffe, z.B. n-Paraffine enthalten, wird die Belüftung entweder allein mit Luft oder mit Luft unter Zugabe von Sauerstoff durchgeführt.
Die genannten Bedingungen gewährleisten jedoch eine genügende Intensität des Prozesses der Züchtung von Hefe nicht, da die Kohlenwasserstoffe schwer oxydierbare Verbindungen sind und von den Mikroorganismen mit geringer Geschwindigkeit verwertet werden.
Das Ziel der vorliegenden Erfindung ist die Entwicklung eines Verfahrens zur Züchtung von Hefe, vorzugsweise von Futterhefe, auf wässerigen Nährmedien, das es möglich macht, den Prozess der Züchtung von Hefe zu intensivieren und die Ausbeute an Hefe zu steigern.
Die genannten und weitere Ziele werden dadurch erreicht, das das wässerige Nährmedium, das als Kohlenstoffquelle eine Verbindung enthält, die aus der Gruppe von Stoffen gewählt ist, die aus den Hydrolysaten der pflanzlichen Gewebe und den Kohlenwasserstoffen der n Paraffinreihe besteht, mit Luft belüftet wird, in die das Ozon eingeführt ist.
Das Ozon kann in einer Konzentration von 1066 - 1,5 Vol.-% hinzugegeben werden.
Man führt zweckmässig das Ozon in die Luft, die für die Behandlung des wässerigen Nährmediums verwendet wird, im Prozess dessen Bereitung ein.
Darüber hinaus beim Züchten der Hefe kann das Ozon in die Luft eingeführt werden, die für die Belüftung wässeriger Nährmedien verwendet wird.
Ausserdem kann das Ozon in die Luft eingeführt werden, die für die Behandlung von Wasser vor dessen Zuführung zur Bereitung des wässerigen Nährmediums verwendet wird.
Zur Bereitung des wässerigen Nährmediums wird als die einzige Kohlenstoffquelle eine Verbindung genommen, die aus der Gruppe von Stoffen gewählt wird, die aus den Hydrolysaten der pflanzlichen Gewebe und den Kohlenwasserstoffen der n-Paraffinreihe besteht.
Als Kohlenwasserstoff verwendet man die Paraffinfraktion des Erdöls mit 11 bis 30 Kohlenstoffatomen, die bei einer Temperatur von 240-3600C siedet.
Als Hydrolysate der pflanzlichen Gewebe verwendet man die Abfälle der Landwirtschaft, z.B. Hydrolysate der Baumwollschalen, der Maiskolben u.a.m., die Abfälle der Holzbearbeitungsindustrie sowie die Hydrolysate von Holz.
Die Fermentation wird nach kontinuierlichem Verfahren auf wässerigem Nährmedium durchgeführt, das eine Standartzusammensetzung der Mikroelemente und Salze Kalium-, Magnesium-, Phosphor- und Stickstoffsalze enthält.
Der pH-Wert des Nährbodens soll in einem Bereich von 3 - 6, vorzugsweise von 4 - 5,5 aufrechterhalten werden (will be maintained). Die optimale Temperatur des Fermentationsprozesses liegt im allgemeinen in einem Bereich von 25-400C.
Das Ozon ist eines der stärksten Oxydationsmittel, das verschiedenartige chemische Verbindungen zu oxydieren vermag.
Wenn die Hydrolysate durch das Ozon behandelt werden, sollen der pH-Wert des Mediums, die Temperatur und die Konzentration des Ozon-Luft-Gemisches in einem Bereich aufrechterhalten werden, der eine selektive Oxydation der Hemmstoffe ohne Oxydation der Kohlenhy drate gewährleistet, die für die Nahrung der Hefe erforderlich sind.
So senkt z.B. die Behandlung des Hydrolysats, das durch ein Ozon-Luft-Gemisch mit einer Konzentration des Ozons von 10-- Vol.-% auf einen pH-Wert von 4,5-5,5 neutralisiert wird, bei einer Temperatur von 50-60 C die Konzentration des Furfurols von 0,043 Gew.-% auf 0,023 Gew.-%. Es vermindert sich entsprechend auch die Konzentration des Oxymethylfurfurols. Die physiologisch aktiven Fraktionen der Kolloide bringen zur Ausflockung.
Die Konzentration der Kohlenhydrate verändert sich praktisch nicht.
Bei der Züchtung von Hefe auf Nährmedien, die aus ozonierten Hydrolysaten bereitet werden, steigt die Ausbeute an Hefe um 12-25% in Abhängigkeit von der Zusammensetzung des Mediums.
Die Behandlung der Kohlenwasserstoffe der n-Paraffinreihe durch Ozon-Luft-Gemische mit einer Konzentration des Ozons von 1,0-1,5 Vol.-% bei einer Temperatur von etwa 1000C führt zur Oxydation der entständigen Gruppen der n-Kohlenwasserstoffe und der Beimengungen von zyklischen Kohlenwasserstoffen, falls solche vorliegen. Im Ergebnis entstehen oberflächenaktive und or spanische Säuren, die die Emulsionen der Kohlenwasserstoff-Nährmedien stabilisieren. Eine mehrfache Vergrösserung der Trennfläche der Phasen, auf der sich die Hefe vermehrt, beschleunigt deren Vermehrung und erhöht die Ausbeute an Biomasse.
Durch die Behandlung des Wassers mit dem Ozon vor dessen Zuführung für die Verdünnung der Hydrolysate und die Bereitung von Nährmedien aus den Kohlenwasserstoffen der n-Paraffinreihe steigt dessen Redoxpotential bis zu einem Stand. bei dem die fremden Mikroorganismen und deren Sporen sowie die Viren vollständig absterben. Das azonierte Wasser ist praktisch steril.
Ausserdem oxydiert das ozonierte Wasser, wenn es unmittelbar nach der Behandlung mit Ozon verwendet wird, die Wachstumshemmstoffe der Hefe in den Hydrolysaten und erhöht die Stabilität der Emulsionen der Koh lenwasserstoff-Nährmedien.
Im Prozess der Hefezüchtung hängt die Geschwindigkeit der Speicherung der Biomasse und die Ausbeute an Produkt von der Intensität der Oxydations-Reduktions-Prozesse ab. Die Verwendung von Ozon, gemischt mit der Luft, die zur Belüftung zugeführt wird, erhöht die Redoxpotentiale der Kulturmedien (d.h. wässeriges Nährmedium mit der darauf gezüchteten Hefe) an der Trennfläche der Phasen in den Bläschen des dispergierten Ozon-Luft-Gemisches.
Dadurch entstehen günstige Bedingungen für die selektive Oxydation der oberflächenaktiven Hemmstoffe u.
der Metabolismusprodukte und werden die fermentativen Systeme der Hefe aktiviert.
Bei der Belüftung durch Ozon-Luft-Gemische, die von 1,5. 10 bis 1 10-6 Vol.-% Ozon enthalten, des Nährmediums, das aus Hydrolysaten hergestellt wurde, die reduzierende Stoffe in einer Menge von 1,5 bis 4,0% enthalten, steigt die Ausbeute an Biomasse der Hefe unter dem Einfluss des Ozons um 10-30% und mehr.
Bei der Anwendung der bekannten Verfahren zur Herstellung von Hefe beobachtet man häufig im Kulturmedium unter den Bedingungen des kontinuierlichen technologischen Prozesses eine Zunahme der Menge der ungewünschten Mikroorganismen über die zulässigen Normen hinaus. Zur Vermeidung dieser Erscheinung senkt man allgemein den pH-Wert des Mediums auf 4,03,5, was sich auf die Ausbeute an Biomasse und auf andere technisch-wirtschaftliche Kennwerte ungünstig auswirkt.
Durch die Ausnutzung der bakteriziden Eigenschaften des Ozons wird es möglich, die Sterilität aller in die Hefebottiche gelangenen Materialien, der Kulturmedien, des Wassers und der Luft zu gewährleisten. Ausserdem ist es beim Vorliegen von Ozon in die Luft, die zur Belüftung verwendet wird, möglich. den Stand des Redoxpotentials des Kulturmediums derart zu regeln, dass günstige Bedingungen für die Hefe herbeigeführt und die Vermehrung einer Reihe ungewünschter Mikroorganismen erschwert werden.
Die bakteriziden Eigenschaften des Ozons werden ausgenutzt für die Bekämpfung der Vermehrung ungewünschter Mikroorganismen durch Sterilisierung der Hydrolysate, Kohlenwasserstoffe, des Wassers, der Luft und der Kulturmedien durch Ozon-Luft-Gemische.
Darüber hinaus wird es durch das Hinzufügen von Ozon in die Luft, die für die Behandlung der Hydrolysate der pflanzlichen Gewebe verwendet wird, möglich, sogar solche von diesen wirksam zu verwenden, die über 1,5% reduzierende Stoffe und bedeutende Wachstumshemmstoffmenge der Hefe (Furfurol, Oxymethylfurfurol) enthalten.

Beispiel I
Die Hefe des Stammes Candida tropikalis (CD-10) züchtete man nach dem kontinuierlichen Verfahren innerhalb 60 Stunden in einem 1,5 1 -Hefebottich (Arbeitsvolumen des Substrats 0,5 1) auf dem Hydrolysat der Baumwollschalen, die 2,5% reduzierende Stoffe und bedeutende Mengen von Hemmstoffen enthalten, bei einer Temperatur von 32,0 + 0,5 C, einem pH-Wert von 4,2 - 4,5 mit 12 stündigem Austausch der Kulturflüssigkeit.
Die Belüftung der Kulturflüssigkeit erfolgte durch die Luft, die 1,5-10-4 Vol.-ic Ozon enthält. Die Ausbeute an absolut trockener Hefe betrug 55,8 Gew.-%.
Beispiel 2 Die Hefe des Stammes Candida guillermondii (Hll-4) züchtete man nach dem kontinuierlichen Verfahren innerhalb von 18 Stunden in einem 1,5 1 - Hefebottich (Arbeitsvolumen des Substrats 0,5 1) auf wässerigem Nährmedium, das eine Standardzusammensetzung der Mikroelemente und Salze von K, Mg, P und N enthält, bei einer Konzentration der gereinigten Paraffine von 1 Vol.-e,Zc, bei einer Temperatur von 32,0 i 0,50C und einem pH Wert von 4,2-4,5 mit 10stündigem Austausch der Kulturflüssigkeit. Die Belüftung der Kulturflüssigkeit erfolgt durch Luft. die 10-4 Vol.- c Ozon enthält. Die Ausbeute an absolut trockener Hefe, bezogen auf die verbrauchten Paraffine betrug 130 Gew.-%.

The ozone can be added in a concentration of 1066 - 1.5% by volume.
