Verfahren zur Herstellung neuer Polypeptide
Aus dem französischen Patent Nr. 1 329 840 ist bekannt, dass das Heptapeptid
L-Asparagyl-L-alanyl-L-phenylalanyl-L-isoleucyl glycyl-L-leucyl-L-methioninamid, dessen freie Carboxylgruppe am Asparagylrest auch durch Amidbildung abgewandelt sein kann, eine starke, insbesondere erweiternde Wirkung auf das Blutgefässsystem besitzt.
Im Hauptpatent Nr. 488 662 wird ein Verfahren zur Herstellung neuer Polypeptide der allgemeinen Formel
R-L-Alanyl-L-phenylalanyl-L-a-aminoacyl glycyl-L-leucyl-L-methioninamid, in der R ein Wasserstoffatom oder den Rest der natürlichen Aminosäuren, Glycin, Alanin, Valin, Leucin, Isoleucin, Asparagin, Glutamin, Glutaminsäure, Serin, Threonin, Cystein, Methionin, Phenylalanin, Tyrosin, Prolin, Oxyprolin, Lysin, Histidin, Arginin, Ornithin, Norleucin, Norvalin, a-Aminobuttersäure, a,r-Diamino- buttersäure, Pyroglutaminsäure, Tryptophan oder einer natürlichen N - Methylammsäure bedeutet und L Aminoacyl für den L-Isoleucyl-, L-Leucyl- oder I-Valylrest steht, beschrieben.
Die nach diesem Verfahren hergestellten Verbindungen zeigen eine dem Eledoisin, einem vor allem blutdrucksenkenden Undekapeptid, vergleichbare biologische Aktivität.
Das Verfahren gemäss dem Hauptpatent ist dadurch gekennzeichnet, dass man die erforderlichen Aminosäuren und L-Methioninamid in der genannten strukturellen Reihenfolge unter Bildung der Peptidbindungen durch Kondensation vereinigt.
Es wurde nun gefunden, dass nicht nur die Verbindungen, in denen R für einen natürlichen, nicht acylierten Aminosäurerest steht, eine eledoisinartige Wirkung besitzen, sondern überraschenderweise in verstärktem Masse jene Verbindungen, in denen R den Formylrest oder den Acylrest einer mit funktionellen Gruppen substituierten Carbonsäure bedeutet, wie z. B.
Acylreste mit einer zusätzlichen Carbonyl-, Carboxyl-, Halogen-, Hydroxyl- oder nicht a-ständigen Aminogruppe.
Gegenstand der vorliegenden Erfindung ist daher ein Verfahren zur Herstellung neuer Polypeptide der allgemeinen Formel I R-L-Alanyl-L-phenylalanyl-L-a-aminoacyl- glycyl-L-leucyl-L-methioninamid, in der R den Formylrest oder den Acylrest einer mit funktionellen Gruppen substituierten Carbonsäure, ausgenommen den Acylrest einer natürlichen, am Stickstoffatom nicht acylierten Aminosäure bedeutet, und L-a-Aminoacyl für den L-Isoleucyl- oder L-Valylrest steht. Dieses Verfahren ist dadurch gekennzeichnet, dass man die erforderlichen Säuren und Methioninamid in der genannten strukturellen Reihenfolge unter Bildung der Peptidbindungen durch Kondensation vereinigt.
Bei der Durchführung des erfindungsgemässen Verfahrens kann man zunächst kleinere Peptid-Einheiten herstellen, die dann ihrerseits zu den Polypeptiden der Formel I vereinigt werden.
Bei den erfindungsgemäss hergestellten Polypeptiden kann R auch neben den genannten funktionellen Substituenten zusätzlich in nStellung zur Carboxylgruppe eine acylierte, beispielsweise acetylierte, Aminogruppe aufweisen, z. B. R = Säurerest des a-Monoacetyllysins.
Die folgende Tabelle I gibt eine Übersicht über die biologische Aktivität der Verbindungen R-Ala-Phe-Ileu Gly-Leu-Met-NH:

Die höhere Wirkung der erfindungsgemäss hergestellten Verbindungen ist insofern unerwartet, da sie sich nicht durch den verhinderten enzymatischen Abbau mit Aminopeptidasen erklären lässt. Dies beweist die etwa nur dem Eledoisin entsprechende Wirkung des Diacetyl-lysyl-Derivates und die niedrige Aktivität der Capronyl- und Caprinoyl-Verbindungen (30 % bzw.
10 % des Eledoisins), die ebenfalls nicht mit Aminopeptidasen abbaubar sind.
Die erfindungsgemässen Verbindungen zeichnen sich auch in vielen Fällen gegenüber den bisher bekannten Produkten durch ihre leichtere Herstellbarkeit aus, beispielsweise weil beliebig substituierte organische Säuren häufig leichter zugängig sind als viele Aminosäuren.
Die Synthese der neuen Verbindungen erfolgt nach an sich bekannten Methoden.
Beispiel 1
Formyl-Ala-Phe-Ileu-Gly-Leu-Met-NH2
Formyl-Ala-Phe-OMe wurde in der üblichen Weise nach dem Anhydrid-Verfahren erhalten [Fp. 115-117 (aus Äthanol-Pctroläther), [aln = -10,8 (c = 0,8; DMF)] und mit der vierfachen Menge Hydrazinhydrat in das Hydrazid übergeführt. Fp. 207 bis 2080 C (aus DMF/Petroläther), [a]D = -49,90 (c = 0,5; Eisessig).
Umsetzung von 0,3 g des Hydrazides (Azidbildung mittels tert.-Butylnitrit in Chlorwasserstoff/Tetrahydrofuran) mit 0,6 g H-Ileu-Gly-Leu-Met-NHa HCl führte zu 0,5 g der gewünschten Verbindung, Fp. 267-268 C, [a]i = -42,0 (c = 0,5; DMF).
Beispiel 2 Nicotinoyl-Ala-Phe-Ileu-Gly-Leu-Met-NHg
Aus 3,2 g Nicotinsäurechlorid und 5,7 g H-Ala Phe-OMe HCl wurden in Chloroform nach Zugabe von 8,4 cm Triäthylamin, 5,6 g Nicotinoyl-Ala-Phe OMe erhalten, Fp. 161 bis 1620 C (aus Äthanol/Petrol- äther), [ai = + 32,05 (c = 0,5; Eisessig).
Hydrazinolyse mit der vierfachen Menge Hydrazinhydrat lieferte das Hydrazid [Fp. 209-211 , [α]D = -16,6± (c = 0,5; Eisessig)], das wie in Beispiel 1 mit H-Ileu-Gly-Leu-Met-NH2-HCl umgesetzt wurde, Fp.
272 bis 274@ C, [α]D = -27,5C (c = 0,5; DMF).
Beispiel 3 α-Hydroxyisovalery-Ala-Phe-Ileu-Gly-Leu
Met-NHa
Aus 0,3 g L - a - Hydroxyisovaleriansäurehydrazid wurden mit 0,3 cm3 tert.-Butylnitrit in 2,9 cm3 einer 1 ,5n-Lösung von Chlorwasserstoff in Tetrahydrofuran das Azid hergestellt, das nach dem Neutralisieren mit Triäthylamin mit 1,5 g H-Ala-Phe-Ileu-Gly-Leu-Met NH2.HCl und 0,3 cm Triäthylamin umgesetzt wurde.
Ausbeute 1,3 g, Fp. 267 bis 270 C, [α]D - -38,7 (c = 0,5; DMF).
Beispiel 4
Chloracetyl-Ala-Phe-Ileu-gly-Leu-Met-NH2
0,2 g Monochloressigsäure wurden mit 0,3 cm Triäthylamin und 0,2 cm Chlorameisensäureäthylester in das gemischte Anhydrid übergeführt und dieses mit 1,4 g H-Ala-Phe-Ileu-Gly-Leu-Met-NH gekuppelt.
Ausbeute 1,2 g, Fp. 238 bis 242 C, [α]D = -36,4' (c = 0,5; DMF).
Beispiel 5 a) Ac-L-Lys(BOC)-NHNH2
4,0 g (15,4 mMol) H-L-Lys(BOC)-OMe wurden mit 2,8 g (15,4 mMol) Essigsäure-p-nitrophenylester acetyliert. Aus dem nur als Öl erhaltenen Methylester [3,9 g (84,1 %); [aj @D = -18,73 (c = 1,5; Pyridin)j wurden durch Hydrazinolyse mit der 2,5fachen Menge Hydrazinhydrat das Hydrazid hergestellt. Ausbeute: 2,1 g (55 %), Fp. 123 bis 125 C (aus Essigester); b) Ac-L-Lys(BOC)-L-Ala-L-Phe-L-lleu-Gly
L-Leu-L-Met-NH2
1,5 g (5 mMol) Ac-L-Lys(BOC)NHNH2 wurden mit (),5 cm (5,2 mMol) tert.-Butylnitrit in das Azid übergeführt und dieses mit 3,2 g (5 mMol) H-L-Ala L-Phe-L-Ileu-Gly-L-Leu-L-Met-NH2 gekuppelt. Nach dem üblichen Waschen der festen Substanz und Umfällen aus DMF/wässr.
Citronensäure betrug die Ausbeute 2,8 g (61,5%); Fp. 249 bis 254 C, [α]22D = -40,80 (c = 0,5; Eisessig); c) Ac-L-Lys-L-Ala-L-Phe-L-Ileu-Gly
L-Leu-L-Met-NH2 wurde in quantitativer Ausbeute durch Schutzgruppenabspaltung mit HC1/Eisessig aus der in 5 b) dargestellten Verbindung erhalten.
Fp. 225 bis 245 C, [α]22D= -42,5 (c = 0,5; Eis- essig).
Beispiel 6 a) BOC-L-Lys(Ac)-OH
2,3 g (12 mMol) H-L-Lys(Ac)-OH wurden in Dioxan/Wasser 1:1 gelöst und in Gegenwart von 1,5 g (36 mMol) Magnesiumoxyd mit 3,4 g (14,2 mMol) tert. - Butyloxycarbonyl - p -mitrophenylester umgesetzt.
Ausbeute: 1,31 g (38%); Fp. 137 bis 1380 C (aus Essigester/Petroläther); [α]22D -10,lc' (c = 0,5; Eisessig).
b) BOC-L-Lys(Ac)-OPhNO2
1,0 g (3,4 mMol) BOC-L-Lys(Ac)-OH wurde mit 0,6 g (4,5 mMol) p-Nitrophenol nach der Carbodiimid Methode verestert. Ausbeute: 1,3 g (92%); Fp. 96 bis 98 C (aus Essigester/Petroläther, [α]22D = -24,3 (c = 0,5; Äthanol).
c) BOC-L-Lys(Ac)-L-Ala-L-Phe-L-Ileu-Gly
L-Leu-L-Met-NH2
1,2 g (3 mMol) BOC-L-Lys(Ac)-OPhNO2 wurden mit 2,0 g (3 mMol) H-L-Ala-L-Phe-L-Ileu-Gly-L-Leu L-Met-NH in DMF 3 Tage bei 403 C umgesetzt. Nach der üblichen Aufarbeitung wurden 1,3 g (48 %) erhalten. Fp. 239 bis 2600 C, [α]22D = -44,0 (c = 0,5; Eisessig).
d) H-L-Lys(Ac)-L-Ala-L-Phe-L-Ileu-Gly
L-Leu-L-Met-NH2 wurde aus der geschützten Verbindung durch Schutzgruppenabspaltung mit HCl/Eisessig erhalten. Fp. etwa 2300 C (Zers.); [a]D = 27,00 (c = 0,5; Eisessig).

L-Leu-L-Met-NH2
800 mg (0,9 mMol) a-Acetyl-Lysin-Heptapeptidamid-Hydrochlorid wurden nach dem Freisetzen mit 0,13 cm Triäthylamin in DMF mit 360 mg (2 mMol) Essigsäure-p-nitrophenylester in üblicher Weise umge- setzt. Ausbeute: 640 mg (80%), Fp. 260 bis 266 C, [α]22D= -44,8 (c = 0,5; Eisessig).
Beispiel 7 a) #-BOC-Aminocapronsäure(BOC-#-Acap-OH)
Aus der #-Aminocapronsäure mit tert.-Butyloxycarbonyl-p-nitrophenylester werden in der üblichen Weise 77 % eines kristallisierenden Öls erhalten. Fp. 30 bis 31 C.
b) BOC-E-Acap-L-Ala-L-Phe-L-Ileu-Gly
L-Leu-L-Met-NH2
Nach der Anhydrid-Methode wurden aus 1,4 g (6 mMol) BOC-E-Aminocapronsäure und 3,9 g (6 mMol) H-L-Ala- L-Phe-L-Ileu-Gly-L-Leu-L-Met-NHl, 4,1 g (78%) erhalten. Fp. 247 bis 2480 C; [c'iD = -33,60 (c = 0,6; DMF).
c) H-E-Acap-L-Ala-L-Phe-L-Ileu-Gly
L-Leu-L-Met-NH2.HCl
Durch Schutzgruppenabspaltung mit HC1/Eisessig wurde das Hydrochlorid in quantitativer Ausbeute erhalten. Fp. 221 bis 2300 C, [a] 2D- = -35,00 (c = 0,5; DMF).
Process for the production of new polypeptides

The compounds prepared by this process show a biological activity comparable to eledoisin, an undekapeptide that lowers blood pressure in particular.
The process according to the main patent is characterized in that the required amino acids and L-methionine amide are combined in the structural order mentioned with formation of the peptide bonds by condensation.
It has now been found that not only the compounds in which R stands for a natural, non-acylated amino acid residue have an eledoisin-like effect, but surprisingly to a greater extent those compounds in which R is the formyl residue or the acyl residue of a carboxylic acid substituted with functional groups means such as B.

The greater effect of the compounds prepared according to the invention is unexpected in that it cannot be explained by the prevented enzymatic degradation with aminopeptidases. This proves the effect of the diacetyl-lysyl derivative, which corresponds roughly only to the eledoisin, and the low activity of the capronyl and caprinoyl compounds (30% and 30% respectively).

Formyl-Ala-Phe-OMe was obtained in the usual manner by the anhydride method [mp. 115-117 (from ethanol-pctrolether), [aln = -10.8 (c = 0.8; DMF)] and converted into the hydrazide with four times the amount of hydrazine hydrate. 4.0 g (15.4 mmol) of H-L-Lys (BOC) -OMe were acetylated with 2.8 g (15.4 mmol) of p-nitrophenyl acetic acid.
