Verfahren zur Herstellung von 5-Phenyl-2,3-dihydro-lH-benzodiazepinen
Die vorliegende Erfindung bezieht sich auf ein Verfahren zur Herstellung von 2,3-Dihydro-lH-1,4-benzo- diazepinen der folgenden allgemeinen Formel:

worin X und Y jeweils ein Wasserstoffatom, ein Halogenatom oder einen Trifluormethylrest bedeuten.
Die 2,3-Dihydro- lH-l,4-benzodiazepinderivate der allgemeinen Formel I und deren Salze sind wertvolle Tranquillizer, Hypnotika, Muskelrelaxantien undkrampflösende Mittel und eignen sich ferner als Zwischenprodukte für die Herstellung von anderen Benzodiazepinderivaten, die als Tranquillizer, Muskelrelaxantien und krampflösende Mittel wirksam sind.
Man kann auch Salze dieser Benzodiazepinderivate durch Behandeln der Verbindungen der Formel I mit einer Mineralsäure, wie z.B. Salzsäure, Schwefelsäure, Salpetersäure oder Phosphorsäure, oder mit einer organischen Säure, wie z.B. Maleinsäure, Fumarsäure, Bernsteinsäure, Ameisensäure, Essigsäure oder Weinsäure, herstellen.
Einige Methoden für den Herstellung der Benzodiazepinderivate der Formel I sind bereits beschrieben worden. So werden Benzodiazepinderivate dadurch erhalten, dass man ein 2-Glycylamido-benzophenonderivat cyclisiert und das so erhaltene 2,3-Dihydro-lH-l,4-benzodia- zepin-2-onderivat (L.H. Sternbach, E. Reeder und G.A.
Archer, J. Org. Chem. 28 2456) reduziert.
Gemäss vorliegender Erfindung werden Benzodiazepinderivate der Formel I sowie deren Salze in vorteilhafter Weise erhalten. Dieses neue und wertvolle Verfahren unterscheidet sich von den bekannten Methoden und kann daher als eine Verbesserung angesehen werden. Gemäss vorliegendem Verfahren lassen sich die 2,3-Dihy dro-lH-1,4-benzodiazepinderivate der Formel I so erhalten, dass man ein 2-(Dioxopiperazino)-benzophenonderivat der Formel:

Die 2-(Dioxo-piperazino)-benzophenonderivate der allgemeinen Formel IV sind neue Verbindungen und können dadurch hergestellt werden, dass man neue Piperazinoindolderivate der Formel:

Alle diese Methoden verlaufen glatt und ergeben die gewünschten Produkte in hohen Ausbeuten. Es handelt sich somit um für die Praxis vorteilhafte Methoden.
Das Verfahren wird vorteilhafterweise so ausgeführt, dass man die Piperazinobenzophenonderivate der Formel IV mit einem Hydrolysiermittel in einem geeigneten Lösungsmittel, z.B. Wasser, einem Alkohol, wie z.B. Methanol oder Äthanol, oder Pyridin, erhitzt. Als Hydrolysiermittel kommen beispielsweise Alkalimetallhydroxyde, wie z.B. Natriumhydroxyd und Kaliumhydroxyd, Alkalimetallcarbonate, wie z.B. Kaliumcarbonat, Erdalkalimetallhydroxyde, wie z.B. Bariumhydroxyd und Calciumhydroxyd, sowie Ammoniumverbindungen, wie z.B. Ammoniumhydroxyd, in Frage. Hydroxyde von Alkalimetallen oder Erdalkalimetallen sind besonders bevorzugt.
Die Umsetzung erfolgt im allgemeinen bei erhöhter Temperatur und vorzugsweise bei der Siedetemperatur des verwendeten Lösungsmittels.
Milde Hydrolysierbedingungen für die 2-(Dioxopiperazino)-benzophenonderivate der Formel IV führen zu Zwischenprodukten.
Die der Formel V entsprechenden Piperazinoindolderivate können aus l-Cyanomethylindol-2-carbonsäurederivaten der Formel VI durch Umsetzung mit einem Reduktionsmittel erhalten werden.
Die Umsetzung erfolgt vorzugsweise durch katalytische Hydrierung. Verwendete Katalysatoren sind beispielsweise Platinoxyd, Palladium, Raneynickel, Nickelborid, Raneykobalt usw. Wasserstoff kann bei einem geeigneten Druck, vorzugsweise bei Atmosphärendruck, verwendet werden. Die Umsetzung erfolgt im allgemeinen bei Zimmertemperatur, doch kann man auch erhöhte Temperaturen anwenden.
Die l-Cyanomethylindol-2-carbonsäurederivate der Formel VI können dadurch erhalten werden, dass man Indolderivate der Formel VII mit einem reaktionsfähigen Ester von Cyanomethylalkoholen in Gegenwart eines basischen Kondensationsmittels behandelt, oder nach der Bildung der entsprechenden basischen Metallsalze, indem man sie mit basischen Kondensationsmitteln behandelt.
Reaktionsfähige Ester von Cyanomethylalkohol, welche Verwendung finden können, sind Halogenide und Sulfonsäureester. Die Halogenide sind beispielsweise Chloride, Bromide oder Jodide, während als Sulfonsäureester beispielsweise Methylsulfonsäureester, p-Toluolsulfonsäureester oder !,8-Naphthalinsulfonsäureester in Frage kommen.
Basische Kondensationsmittel, welche man beispielsweise verwenden kann, sind Alkalimetalle, Erdalkalimetalle, Alkalimetallhydride, Erdalkalimetallhydride, Alkalimetallhydroxyde, Erdalkalimetallhydroxyde, Alkalimetallamide, Erdalkalimetallamide, Alkalimetallalkoxyde, Erdalkalimetallalkoxyde, Alkylalkalimetalle u. Arylalkalimetalle usw. Natriumhydrid, Lithiumhydrid, Natriumamid, Kaliumamid und Lithiumamid werden bevorzugt.
Die Umsetzung erfolgt im allgemeinen in einem Lösungsmittel, wie z.B. Benzol, Toluol, Xylol, Dimethylformamid, Dioxan oder flüssigem Ammoniak usw.
Herstellung der Ausgangsprodukte a) 1 -Cyanomethyl-2-äthoxycarbonyl-3-phenyl-5-chlor- indol
Zu einer Mischung von 1,1 g 50 obigem Natriumhy- drid und 10 cm3 Dimethylformamid gibt man eine Lösung von 6 g 2-Äthoxycarbonyl-3-phenyl-5-chlorindol in 30 cm3 bei 30 bis 350C hinzu.
Nach dem Rühren während 15 Minuten bei 300C versetzt man tropfenweise mit einer Lösung von 1,7 g Chloracetonitril in 10 cm3 Dimethylformamid bei 30 bis 350C und rührt das Gemisch während 1 Stunde bei 25 bis 300C. Dann gibt man Wasser hinzu. Das Gemisch wird hierauf mit Äther extrahiert. Die ätherische Schicht wird mit Wasser gewaschen und über Natriumsulfat getrocknet. Das Lösungsmittel wird durch Destillation entfernt, wobei man 7 g einer festen Substanz erhält. Zu dieser festen Substanz gibt man 100 cm3 Hexan hinzu und filtriert das Gemisch. Auf diese Weise erhält man 5,5 g 1 -Cyanomethyl-2-äthoxycarbonyl-3-phenyl-5 chlor-indol.
Die Kristalle werden aus einem Lösungsmittelgemisch von Benzol und Hexan umkristallisiert, wobei man blassgelbe Nadeln vom Schmelzpunkt 126,5 bis 1 270C erhält.
b) 7-Chlor-9-phenyl-10-oxo- piperazino-(1,2-a)-indol
Zu einer Lösung von 10 g 1-Cyanomethyl-2-äthoxycarbonyl-3-phenyl-5-chlor-indol in 100 cm3 Tetrahydrofuran gibt man einen Katalysator hinzu, welcher aus 50% Raneynickellegierung unter Behandeln mit einer wässrigen Natriumhydroxydlösung bei 1000C während 1 Stunde hergestellt wurde. Zu dieser Mischung gibt man 2 Mol Wasserstoff unter Atmosphärendruck bei 180C während 12 Stunden solange hinzu, bis die Reduktion durch Absorption beendet ist.
Nach dem Entfernen des Katalysators durch Filtrieren verdampft man das Tetrahydrofuran unter vermindertem Druck und löst den Rückstand in heissem Äthanol.
Das Gemisch wird über Nacht in einem Kühlschrank stehen gelassen. Die auf diese Weise erzeugten Kristalle werden durch Filtrieren gesammelt. Durch Umkristallisieren aus einer Mischung von Äthanol und einer kleinen Menge Benzol erhält man 3,6 g hellgelbe Nadeln, welche aus 7-Chlor-9-phenyl- 1 0-oxo-piperazino 1,2--a)-indol bestehen und bei 244,5 bis 2450C schmelzen.
c) 2-(2",3"-Dioxo-piperuzino)-5-chlor-benzophenon
Zu einer Mischung von 1,6 g 7-Chlor-9-phenyl-10- -oxo-piperazino-(1,2-a)-indol und 35 cm3 Essigsäure gibt man eine Lösung von 1,6 g Chromsäureanhydrid in 2 cm3 Wasser bei 100C hinzu. Das Gemisch wird während 16 Stunden bei 200C gerührt und hierauf in 500 cm3 Wasser gegossen.
Nach dem Anpassen des pH-Wertes auf 7 bis 8 unter Zugabe von wässrigem Ammoniak wird das Reaktionsgemisch mit Chloroform extrahiert. Die Chloroformschicht wird mit Wasser gewaschen und über Natriumsulfat getrocknet. Das Lösungsmittel wird hierauf durch Destillation entfernt, wobei man 1,5 g einer hellgelben, festen Substanz erhält. Durch Umkristallisation aus wässrigem Äthanol erhält man farblose Kristalle von 3"-Dioxopiperazino)-5-chlor-benzophenon vom Schmelzpunkt 1980C.
Beispiel I 5-Phenyl-7-chlor-2,3-dihydro-IH- H-1,4-benzodiazepin
Zu einer Lösung von 2 g Natriumhydroxyd in 5 cm3 Wasser und 50 cm3 Äthanol gibt man 3,3 g 2-(2",3" -Dioxo-piperazino)-5-chlor-benzophenon hinzu. Das Gemisch wird während 3 Stunden unter Rückfluss zum Sieden erhitzt. Nach der Zugabe von 50 cm3 Wasser wird das Reaktionsgemisch unter Rückfluss während 17 Stunden weiter erhitzt und der grösste Teil Äthanol abdestilliert. Zum Rückstand gibt man 200 cm3 Wasser hinzu und erhitzt das Gemisch auf 800C, um ein Pulver zu erhalten, welches durch Filtrieren gesammelt wird. Auf diese Weise erhält man 2 g 5-Phenyl-7-chlor-2,3-dihydro -1H- 1,4- benzodiazepin. Durch Umkristallisieren aus Äthanol gelangt man zu gelben Kristallen vom Schmelz punkt 1730C.
Beispiel 2 5-Phenyl-2,3-dihydro-1H-1,4-benzodiazepi 5-Phenyl-2,3 -dihydro- 1 H- 1,4-benzodiazepin, Schmelzpunkt 144 bis 1460C, wird aus 2-(2",3"-Dioxo-piperazino)-benzophenon nach einer ähnlichen Methode wie in Beispiel 1 erhalten. Das Produkt wird aus Petroläther umkristallisiert.
Beispiel 3
5-(o-Chlor-phenyl)-7-chlor-2'3-dihydro-1H-1 H-1,4-benzo- diazepin
5-(o-Chlor-phenyl)-7-chlor-2,3-dihydro- 1 H- 1 ,4-benzo- diazepin, Schmelzpunkt 175 bis 1770C, wird aus 2 -(2'',3 "-Dioxo-piperazino)-2' ,5-dichlor-benzophenon nach einer ähnlichen Methode wie in Beispiel 1 erhalten.
Beispiel 4 5-(o-Fluor-phenyl)-7-chlor-2'3-dihydro-1H-1'4- benzo diazepin 5-(o-Fluor-phenyl)-7-chlor-23-dihydro- 1H- 1,4-benzo- diazepin, Schmelzpunkt 161 bis 1630C, wird aus 2 -(2",3"-Dioxo-piperazino)-5-chlor- 2'- fluor-benzophenon nach einer ähnlichen Methode wie in Beispiel 1 erhalten.
Beispiel 5 S-Phenyl-7-trifluormethyl-2,3-dihydro-lH-1,4-benzo- diazepin und sein Hydrochlorid
5-Phenyl-7-trifluormethyl-2,3-dihydro- 1H- 1,4-benzodiazepin, Schmelzpunkt 110 bis 111 C, wird aus 2 -(2",3"-Dioxo-piperazino) -5- trifluormethylbenzophenon nach einer ähnlichen Methode wie in Beispiel 1 erhalten.
Beispiel 6
Methanolische Salzsäure wird zu einer Lösung von 5-Phenyl-7-trifluormethyl-2,3-dihydro- lH- 1,4-benzodiaze- pin in Methanol bei 100C hinzugegeben und das Gemisch wird während 1 Stunde bei Zimmertemperatur gerührt. Das Lösungsmittel wird unter vermindertem Druck entfernt und der Rückstand mit Benzol gewaschen, wobei man 5-Phenyl-7-trifluormethyl-2,3-dihydro-lH-1,4- -benzodiazepin-hydrochlond erhält. Durch Umkristallisieren aus einer Mischung von Methanol und Äther gelangt man zu gelben Kristallen vom Schmelzpunkt 283 bis 285 C.
