Diese Verbindungen weisen eine ausge prägte analgetische oder schmerzlindernde Wirkung zu sammen mit der hochgradig erwünschten Eigenschaft, dass sie weder gewohnheitsbildend noch einschläfernd sind, auf.
Die nach dem erfindungsgemässen Verfahren erzeugten 5,9-Diäthyl-benzomorphane zeigen beim üblichen Ratten schwanztip- oder Rattenschwanzklemmversuch geringe Wirksamkeit, woraus hervorgeht, dass sie entweder keine od. nur eine geringe Süchtigkeit bewirken, da bei den oben angegebenen Versuchen eine unmittelbare Beziehung zwi schen erhöhter Süchtigkeit und erhöhter Wirksamkeit be steht. Andererseits zeigen die nach dem erfindungsgemäs- bei sen Verfahren hergestellten Benzomorphan- einem geänderten Versuch nach Randall-Selitto eine gute Wirksamkeit auf.
Eine gute Wirksamkeit bei diesem Ver such deutet auf eine nützliche analgetische Wirkung hin, da Cyclazocin und Pentazocin, zwei bekannte Analgetika, bei diesem Versuch eine Wirkung zeigen und zusätzlich als wenig gewohnheitsbildend bekannt sind.
Die nach dem erfindungsgemässen Verfahren herge stellten Benzomorphan-Derivate haben minimale Neben wirkungen und eine minimale Toxizität, und viele von ihnen weisen eine geringe oder überhaupt keine narko tische antagonistische Wirkung auf. Der Mangel an nar kotischer antagonistischer Wirkung kann von Vorteil sein, da beobachtet wurde, dass eine hohe antagonistische Wir kung im allgemeinen mit unerwünschten Nebenwirkungen verbunden ist.
Es wurde festgestellt, dass die nach dem erfindungs- gemässen Verfahren erzeugten Verbindungen zweckmäs- siger als Glieder der Morphinfamilie natürlich vorkom mender alkaloidartiger Analgetika synthetisch hergestellt werden können.
<B>0,001</B> Die nach dem erfindungsgemässen Verfahren erzeug ten Verbindungen werden vorzugsweise in eine: wässrigen Lösung des salzsauren Salzes und in einer Menge von mg/kg his etwa 18 mg/kg oral oder subkutan etwa verabreicht. Die Verbindungen haben eine günstige phar makologisch kleinste, therapeutische Dosis.
Das erfindungsgemässe Verfahren zur Herstellung oben definierter Verbindungen ist dadurch. gekennzeich net, dass man 5,9-Diäthyl-2'-hydroxy-6,7-benzomorphan mit einem Säurehalogenid der Formel RCOX, worin R die obige Definition hat und X Chlor oder Brom ist, um setzt, wobei ein 5,9-Diäthyl-2'-acyloxy-2-acy1-6,7-benzo- morphan gebildet wird, das man dann zur Bildung des gewünschten Produktes mit einem Metallhydrid reduziert.
Dieses erfindungsgemässe Verfahren wird gewöhnlich so durchgeführt, dass man das Ausgangs-5,9-diäthyl-2'- -hydroxy-6,7-benzomorphan mit zwei Äquivalenten eines Säurehalogenids der Formel RCOX, in der X Chlorid oder Bromid ist und R die oben bei der Struktur I ange gebene Definition hat und insbesondere Spiroalkyl ist, in einem inerten Lösungsmittel, wie z.B. Chloroform oder Benzol, und vorzugsweise in Gegenwart einer tertiären Aminbase, wie z.B. Pyridin oder Triäthylamin, zur Bil dung des entsprechenden 5,9-Diäthyl-2'-acyloxy-2-acyl-

hat] umsetzt.
Durch Reduktion dieser Derivate, z.B. mit LithiumaIuminiumhydrid in Diäthyläther oder Tetrahy- drofuran, erhält man die gewünschten, biologisch aktiven Verbindungen.
Das gewünschte Produkt kann nach dem Fachmann bekannten Verfahren aus ihren jeweiligen Reaktionslö sungen oder -gemischen isoliert werden.
Das vorliegende erfindungsgemässe Verfahren umfasst auch die Herstellung der Isomeren der oben angegebenen Strukturen, bei denen die beiden Äthylgruppen in einer cis-Beziehung zueinander stehen; diese Isomere werden hierin als normale oder α-Isomere bezeichnet.
Der oben erwähnte cis-Isomer kann in seine optische Isomere [dextro (+) und laevo (-)] gespalten werden, indem mit optisch aktiven Säuren, entweder d (+) oder 1 (-), die diastereomeren Salze erzeugt werden, die dann durch fraktionierte Kristallisation getrennt werden kön nen.
Vor dieser Erfindung war allgemein bekannt, dass linksdrehende (-) Isomere von Benzomorphanverbin- dungen im wesentlichen die ganze gewünschte, analgeti sche Wirkung aufwiesen, die das Gemisch von Isomeren zeigt, während das rechtsdrehende (+) Isomer eine gerin ge oder überhaupt keine analgetische Wirkung hatte.
Es wurde nun überraschenderweise gefunden, dass im Gegensatz zu dem, was von diesen Isomeren hinsichtlich ihrer Wirkungslosigkeit im allgemeinen angenommen wird, die rechtsdrehenden (+) Isomere vieler der nach dem erfindungsgemässen Verfahren erzeugten neuartigen 5,9-Diäthyl-2'-hydroxy-2-substituierten-6,7-benzomorpha- ne analgetische und andere oben angegebene wünschens werte Eigenschaften aufweisen.
Deshalb umfasst das vorliegende erfindungsgemässe Verfahren nicht nur die Herstellung neuartiger 5,9-Diäthyl-2'-hydroxy-2-substi- tuierter-6,7-benzomorphan-Derivate, sondern auch die Herstellung einzelner optischer Isomere, d.h. sowohl der rechtsdrehenden (+) als auch der linksdrehenden (-) Isomere der nach dem erfindungsgemässen Verfahren hergestellten neuartigen Benzomorphanverbindungen.
Obwohl alle oben beschriebenen, erfindungsgemäss erzeugten Verbindungen eine analgetische Wirkung auf weisen, sind die besonders wirksamen, bevorzugt erfin dungsgemäss erzeugten Verbindungen jene, bei denen R ein Spiroalkyl mit 5-7 C-Atomen ist, wie z.B. Spirohex- -5-yl, Spiro-3,3-hept-2-yl-Radikale und ähnl.
Die nach dem erfindungsgemässen Verfahren herge stellten 5,9-Diäthyl-2'-hydroxy-2-substituierten-6,7-benzo- morphane können als solche oder in Form ihrer nicht toxischen pharmazeutisch annehmbaren Säureadditions salze verwendet werden. Solche Salze werden aus ge eigneten Säuren, wie z.B. anorganischen Säuren wie Salz säure oder Schwefelsäure, oder organischen Säuren wie Essigsäure oder Maleinsäure und dergleichen, erzeugt. Die Säureadditionssalze werden durch Umsetzung der Base mit ungefähr einem Äquivalent der ausgewählten Säure in einem organischen Lösungsmittel, wie z.B. Di- äthyläther oder Alkohol, hergestellt.
Das Ausgangsmaterial zur Herstellung der neuartigen Verbindungen der vorliegenden Erfindung, nämlich 5,9- -Diäthyl-2'-hydroxy-6,7-benzomorphan, wird aus 5,9-Di- äthyl-2'-hydroxy-2-methyl-6,7-benzomorphan, einer be kannten Verbindung, hergestellt. Die Herstellung des Ausgangsmaterials erfolgt vorzugsweise dadurch, dass 5,9 - Diäthyl - 2' - hydroxy-2-methyl-6,7-benzomorphan in Stellung 2' mit Essigsäureanhydrid acyliert wird, die acy- lierte Verbindung mit Bromcyan in einem organischen Lösungsmittel, vorzugsweise einem halogenierten Kohlen wasserstoff, wie z.B.
Chloroform, auf eine erhöhte Tem- peratur erhitzt wird, wobei die 2-Methylgruppe durch eine Cyangruppe ersetzt wird, und die Cyan- und Acetyl- gruppen dann durch Hydrolyse in einer verdünnten Säu relösung entfernt werden.
Neben der razemischen Form des Ausgangsmaterials kann auch eine seiner isomeren Formen verwendet wer den, die nach bekannten Verfahren unter Verwendung eines Aufspaltungsmittels, wie z.B. Kampfersulfosäure, Weinsteinsäure, Dibenzoylweinsteinsäure und dergleichen, zur Trennung der linksdrehenden (-) und rechtsdrehen den (+) Isomere erzeugt werden. Jedes optische Isomer von 5,9-Diäthyl-2'-hydroxy-6,7-benzomorphan kann dann als Ausgangsmaterial in den verschiedenen Beispielen verwendet werden, wobei man dann rechtsdrehende (+) oder linksdrehende (-) 5,9-Diäthyl-2'-hydroxy-2-substi- tuierte-6,7-benzomorphane erhält.
Andererseits können die razemischen 5,9-Diäthyl-2'-hydroxy-2-substituierten- -6,7-benzomorphan-Endprodukte selbst gemäss einem dem obigen Verfahren ähnlichen Verfahren aufgespalten werden, wobei man die rechtsdrehenden (+) und links drehenden (-) Isomere von 5,9-Diäthyl-2'-hydroxy-2- -substituierten-6,7-benzomorphanen erhält.
Folgendes Beispiel veranschaulicht das oben beschrie bene bevorzugte Verfahren zur Herstellung der neuarti gen 5,9-Diäthyl - 2' - hydroxy - 2 - substituierten-6,7-benzo- morphane.
In diesem Beispiel wird als Ausgangsmaterial die cis- -(normale oder α)-Form der 5,9-Diäthyl-2'-hydroxy-6,7- -benzomorphane verwendet, wobei das entsprechende cis- Endprodukt erzielt wird. Es kann aber auch das rechts drehende (+) oder das linksdrehende (-) Isomer von cis-5,9-Diäthyl - 2' - hydroxy-6,7-benzomorphan als Aus gangsmaterial verwendet werden, wobei man das entspre chende rechtsdrehende (+) oder linksdrehende (-) End produkt erhält.
<I>Beispiel</I> 5,9 -Diäthyl - 2'- hydroxy-2-(spirohex-5-yl-methyl)-6,7- -benzomorphan, 5,9 - Diäthyl - 2' - hydroxy-2-(spiropentyl- methyl)-6,7-benzomorphan oder 5,9-Diäthyl-2'-hydroxy- -2[spiro (3,3) - hept-2-yl-methyl] - 6,7 - benzomorphan wird erzeugt, indem eine Lösung von 0,0125 Mol Spiro(3,2- -hexan-5-carbonylchlorid, Spiropentylcarbonylchlorid od.
Spiro(3,3)-hept-2-yl-carbonylchlorid in 10 ml Chloroform langsam in eine abgekühlte Lösung von 0,006 Mol 5,9- -Diäthyl-2'-hydroxy-6,7-benzomorphan in 35 ml trocke nes Chloroform und 5,5 ml trockenes Triäthylamin ge geben wird. Das Reaktionsgemisch wird dann etwa 12 Stunden bei Rückfluss erhitzt und dann abgekühlt, mit verdünnter Salzsäure und verdünntem Natriumcarbonat gewaschen und über Natriumsulfat getrocknet. Der Chlo roformextrakt wird unter vermindertem Druck verdampft, wobei man das entsprechende 5,9-Diäthyl-2'-acyloxy-2- -acyl-6,7-benzomorphan erhält.
Die 2'-Acyloxy-2-acyl- -Zwischenverbindung wird dann zum gewünschten Pro dukt reduziert, indem sie in etwa 6,2 ml Tetrahydrofuran verdünnt und die Tetrahydrofuranlösung in eine gerührte Suspension von 0,9 g Lithiumaluminiumhydrid in 10 ml Tetrahydrofuran gegeben wird. Das Reaktionsgemisch wird 4 Stunden bei Rückfluss erhitzt; 4 ml Wasser wer den dann tropfenweise beigefügt und die gefällten anorga nischen Salze durch Filtrieren isoliert und mit Äthyläther gewaschen.
Die kombinierten Filtrate werden unter ver mindertem Druck zur Trockene verdampft, worauf der kristalline Rückstand aus heissem Äthanol kristallisiert wird. Das erhaltene 5,9-Diäthyl-2'-hydroxy-2-(spirohex-5- -yl-methyl)-6,7-benzomorphan weist einen Schmelzpunkt von 201 - 203 C auf.

in which R is cycloalkenyl, alkyl-substituted cycloalkenyl, alkenyloxyalkyl, alkoxyalkyl or spiroalkyl with up to 7 carbon atoms. These compounds have a pronounced analgesic or pain relieving effect along with the highly desirable property that they are neither habit-forming nor soporific.
The 5,9-diethylbenzomorphans produced by the process according to the invention show little effectiveness in the usual rat tail tip or rat tail clamping test, from which it can be seen that they either cause no or only a low addiction, since there is a direct relationship between the above tests increased addiction and increased effectiveness. On the other hand, the benzomorphans produced by the method according to the invention show a good effectiveness in a modified test according to Randall-Selitto.
A good effectiveness in this attempt indicates a useful analgesic effect, since cyclazocine and pentazocine, two known analgesics, show an effect in this attempt and are also known to be less habit-forming.
The benzomorphan derivatives prepared by the process of the invention have minimal side effects and toxicity, and many of them have little or no narcotic antagonistic effect. The lack of narcotic antagonistic effect can be an advantage, since it has been observed that a high antagonistic effect is generally associated with undesirable side effects.
It has been found that the compounds produced by the process according to the invention can be produced synthetically more expediently as members of the morphine family of naturally occurring alkaloid-like analgesics.
<B> 0.001 </B> The compounds produced by the process according to the invention are preferably administered orally or subcutaneously in an aqueous solution of the hydrochloric acid salt and in an amount from mg / kg to about 18 mg / kg. The desired product can be isolated from their respective reaction solutions or mixtures by methods known to the person skilled in the art.
The present inventive method also comprises the preparation of the isomers of the structures indicated above in which the two ethyl groups are in a cis relationship to one another; these isomers are referred to herein as normal or alpha isomers.
The above-mentioned cis isomer can be split into its optical isomers [dextro (+) and laevo (-)] by using optically active acids, either d (+) or 1 (-), to generate the diastereomeric salts, which then can be separated by fractional crystallization.
Prior to this invention, it was well known that levorotatory (-) isomers of benzomorphane compounds exhibited essentially all of the desired analgesic effect exhibited by the mixture of isomers, while the dextrorotatory (+) isomer had little or no analgesic effect would have.
It has now surprisingly been found that, contrary to what is generally assumed of these isomers with regard to their ineffectiveness, the dextrorotatory (+) isomers of many of the novel 5,9-diethyl-2'-hydroxy-2 produced by the process according to the invention -substituted-6,7-benzomorphane analgesic and other desirable properties indicated above.
Therefore, the present inventive method comprises not only the production of novel 5,9-diethyl-2'-hydroxy-2-substituted-6,7-benzomorphane derivatives, but also the production of individual optical isomers, i. both the right-handed (+) and the left-handed (-) isomers of the novel benzomorphane compounds produced by the process according to the invention.
Although all of the above-described compounds produced according to the invention have an analgesic effect, the particularly effective compounds preferably produced according to the invention are those in which R is a spiroalkyl with 5-7 carbon atoms, e.g. Spirohex--5-yl, spiro-3,3-hept-2-yl radicals and the like.
The 5,9-diethyl-2'-hydroxy-2-substituted-6,7-benzomorphans prepared by the process according to the invention can be used as such or in the form of their non-toxic, pharmaceutically acceptable acid addition salts. Such salts are prepared from suitable acids, e.g. inorganic acids such as hydrochloric acid or sulfuric acid, or organic acids such as acetic acid or maleic acid and the like generated. The acid addition salts are prepared by reacting the base with approximately one equivalent of the selected acid in an organic solvent such as e.g. Diethyl ether or alcohol.
The starting material for the preparation of the novel compounds of the present invention, namely 5,9-diethyl-2'-hydroxy-6,7-benzomorphane, is obtained from 5,9-diethyl-2'-hydroxy-2-methyl-6 , 7-benzomorphan, a known compound produced. The starting material is preferably prepared by acylating 5,9-diethyl-2'-hydroxy-2-methyl-6,7-benzomorphane in the 2 'position with acetic anhydride, the acylated compound with cyanogen bromide in an organic solvent, preferably a halogenated carbon, such as
Chloroform, is heated to an elevated temperature, the 2-methyl group being replaced by a cyano group, and the cyano and acetyl groups are then removed by hydrolysis in a dilute acid solution.
In addition to the racemic form of the starting material, one of its isomeric forms can also be used, which can be obtained by known methods using a resolving agent such as e.g. Camphor sulfonic acid, tartaric acid, dibenzoyltartinic acid and the like, for separating the left-handed (-) and right-handed (+) isomers. Any optical isomer of 5,9-diethyl-2'-hydroxy-6,7-benzomorphane can then be used as starting material in the various examples, in which case dextrorotatory (+) or levorotatory (-) 5,9-diethyl-2 '-hydroxy-2-substituted-6,7-benzomorphane is obtained.
On the other hand, the racemic 5,9-diethyl-2'-hydroxy-2-substituted-6,7-benzomorphane end products themselves can be split up according to a method similar to the above, using the right-handed (+) and left-handed (- ) Isomers of 5,9-diethyl-2'-hydroxy-2-substituted-6,7-benzomorphanes are obtained.
The following example illustrates the preferred process described above for the preparation of the novel 5,9-diethyl-2'-hydroxy-2-substituted-6,7-benzomorphanes.
In this example the starting material used is the cis- (normal or α) -form of the 5,9-diethyl-2'-hydroxy-6,7- -benzomorphans, the corresponding cis end product being obtained. But it can also be the right-turning (+) or the left-turning (-) isomer of cis-5,9-diethyl - 2 '- hydroxy-6,7-benzomorphane used as the starting material, with the corresponding right-turning (+ ) or counter-clockwise (-) end product.
<I> Example </I> 5.9 - diethyl - 2'-hydroxy-2- (spirohex-5-yl-methyl) -6.7- -benzomorphan, 5.9 - diethyl - 2 '- hydroxy-2 - (spiropentyl-methyl) -6,7-benzomorphan or 5,9-diethyl-2'-hydroxy- -2 [spiro (3,3) -hept-2-yl-methyl] -6,7-benzomorphan is produced by adding a solution of 0.0125 mol of spiro (3,2- hexane-5-carbonyl chloride, spiropentyl carbonyl chloride or.
Spiro (3,3) -hept-2-yl-carbonyl chloride in 10 ml of chloroform slowly into a cooled solution of 0.006 mol of 5,9-diethyl-2'-hydroxy-6,7-benzomorphan in 35 ml of dry chloroform and 5.5 ml of dry triethylamine will be given. The reaction mixture is then refluxed for about 12 hours and then cooled, washed with dilute hydrochloric acid and dilute sodium carbonate, and dried over sodium sulfate. The chloroform extract is evaporated under reduced pressure to give the corresponding 5,9-diethyl-2'-acyloxy-2-acyl-6,7-benzomorphane.
The 2'-acyloxy-2-acyl intermediate is then reduced to the desired product by diluting it in about 6.2 ml of tetrahydrofuran and adding the tetrahydrofuran solution to a stirred suspension of 0.9 g of lithium aluminum hydride in 10 ml of tetrahydrofuran. The reaction mixture is refluxed for 4 hours; 4 ml of water who then added dropwise and the precipitated inorganic salts isolated by filtration and washed with ethyl ether.
The combined filtrates are evaporated to dryness under reduced pressure, whereupon the crystalline residue is crystallized from hot ethanol. The 5,9-diethyl-2'-hydroxy-2- (spirohex-5- -yl-methyl) -6,7-benzomorphane obtained has a melting point of 201-203.degree.
