Verfahren zur Herstellung von Hydrazinderivaten
Die vorliegende Erfindung bezieht sich auf ein Verfahren zur Herstellung von Hydrazinderivaten, welche für die Änderung des endokrinen Zustandes bei Mensch und Tier wertvoll sind.
Nach dem Verfahren gemäss der Erfindung werden Hydrazinderivate der Formel: R1-NH-CS-NH-NH-CS-NHR2 worin R1 und R je einen Alkenylrest mit mindestens 3 Kohlenstoffatomen bedeuten, wobei diese Alkenylreste gleich oder verschieden sein können und zusammen mehr als 6 Kohlenstoffatome enthalten, hergestellt, indem man eine Verbindung der Formel:
R,-NCS oder R1-NH-CSSR5 worin R1 die obige Bedeutung besitzt und R für einen Methyl- oder Äthylrest steht, mit einem Thiosemicarbazid der Formel: H2N-NH-CS-NHR2 worin R die obige Bedeutung besitzt, umsetzt.
Der Alkenylrest Rl bzw. R2 ist vorzugsweise ein Allylrest, welcher durch einen bis 3 Kohlenstoffatome enthaltenden Alkylrest substituiert sein kann; insbesondere bedeutet der Alkenylrest einen durch einen z-Me- thylrest substituierten Allylrest.
Geeignete Alkenylreste sind z.B. der Allyl-, a-Me thylallyl-"8-Methylallyl-, r-Methylallyl-. But-3-enyl-, - -Äthylallyl-, y-Äthylallyl, a-n-Propylallyl-, y-n-Propylallyl-, asr-Dimethylallyl-lz-Äthyl-,3-methylallyl- und Pent -$enylrest.
Wenn das Verfahren gemäss der Erfindung mit einer Verbindung der Formel R1-NCS durchgeführt wird, kann man in einem Verdünnungsmittel oder Lösungsmittel, beispielsweise in Äthanol, arbeiten; vorzugsweise arbeitet man in siedendem Äthanol.
Wenn das Verfahren gemäss der Erfindung mit einer Verbindung der Formel R1-NHCSSR5 durchgeführt wird, bildet sich aus dieser unter den Reaktionsbedingungen eine Verbindung der Formel Rl-NCS. Bei dieser Verfahrensvariante arbeitet man mit Vorteil in siedendem Wasser.
Wenn R1 und R2 denselben Alkenylrest bedeuten, kann man auch ein Isothiocyanat der Formel R1-NCS mit Hydrazin umsetzen, wobei sich zunächst das Thiosemicarbazid der Formel H2N-NH-CS-NHR1 bildet, welches dann nach dem Verfahren gemäss der Erfindung mit einem zweiten Molekül Isothiocyanat unter Bildung eines symmetrischen Hydrazinderivats reagiert.
Wie bereits erwähnt, besitzen die erfindungsgemäss erhältlichen Hydrazinderivate wertvolle Eigenschaften zur Änderung des endokrinen Zustandes bei Mensch und Tier. Insbesondere hemmen sie die gonadotrope Hypophysenfunktion und eignen sich daher zur Behandlung des Sexualzyklus und dessen Abweichungen; unter anderem können sie auch zur Regulierung des Östrus (Brunst) bei Haustieren verwendet werden. Für diese Zwecke können sie in Form von pharmazeutischen oder veterinären Präparaten verwendet werden.
Die pharmazeutischen und veterinären Präparate können in einer für orale oder für parenterale Verwendung geeigneten Form vorliegen. Die Präparate können beispielsweise auch in Form von Mischungen mit Tierfutter oder in Form von als Beigabe zu Tierfutter geeigneten Mischungen vorliegen.
In den folgenden Beispielen sind die Teile Gewichtsteile.
Beispiel I
Eine Lösung von 5,65 Teilen cc-Methylallyl-isothio- cyanat in 10 Teilen Äthanol wird tropfenweise in eine ge kühlte Lösung von 1,25 Teilen Hydrazinhydrat (100%in) in 10 Teilen Äthanol eingerührt. Das Gemisch wird während 30 Minuten unter Rückfluss erhitzt, hierauf mit Kohle versetzt und das Gemisch filtriert. Das Filtrat wird gekühlt und filtriert und der Rückstand mit Äthanol gewaschen und getrocknet. Auf diese Weise erhält man nach dem Umkristallisieren aus Äthanol Nl,N2-Bis-(oc- -methylallyl-thiocarbamoyl)-hydrazin, Smp. 186 - 1870 C.
Das Methylallyl-isothiocyanat, welches als Ausgangsmaterial verwendet wird, kann man dadurch erhalten, dass man -Methylallylbromid und Ammoniumthiocyanat miteinander umsetzt, wobei bei dieser Umsetzung eine Umlagerung unter Bildung von os-Methylallyl-iso- thiocyanat stattfindet.
Beispiel 2
Man wiederholt das Verfahren gemäss Beispiel 1 wobei aus ,3-Methylallyl-isothiocyanat Nl,N2-Bis-(p-me- thylallyl-thiocarbamoyl)-hydrazin, Smp. 200 bis 2010 C (unter Zers.) erhalten wird.
Beispiel 3
Eine Mischung von 6,5 Teilen 4-Allylthiosemicarbazid, 5,52 Teilen tx-Methylallylisothiocyanat und 25 Teilen Äthanol wird am Rückfluss während 1 Stunde erhitzt. Die Mischung wird gekühlt und filtriert und der Rückstand mit Äthanol gewaschen, getrocknet und aus Äthanol kristallisiert. Auf diese Weise erhält man N1 -Allylthiocarbamoyl - N2'K- methylallylthiocarbamoyl-hydrazin, Smp. 196-198 C (unter Zers.).
Beispiel 4
Die bereits beschriebene Verfahrensweise wird unter Einsatz der geeigneten Thiosemicarbazide und der geeigneten Isothiocyanate durchgeführt, wobei man die in folgender Tabelle angeführten Verbindungen erhält: R2 R: Smp. (OC) Kristallisiert aus p-Methylallyl Allyl 195-196 (Zers.) Äthanol But-3-enyl Allyl 194-196 Äthanol y-Methylallyl Allyl 184-186 50% wässr. Process for the preparation of hydrazine derivatives
The present invention relates to a process for the preparation of hydrazine derivatives which are valuable for changing the endocrine state in humans and animals.

If the process according to the invention is carried out with a compound of the formula R1-NCS, one can work in a diluent or solvent, for example in ethanol; preferably one works in boiling ethanol.
If the process according to the invention is carried out with a compound of the formula R1-NHCSSR5, a compound of the formula R1-NCS is formed from this under the reaction conditions. In this process variant, it is advantageous to work in boiling water.
If R1 and R2 are the same alkenyl radical, an isothiocyanate of the formula R1-NCS can also be reacted with hydrazine, whereby the thiosemicarbazide of the formula H2N-NH-CS-NHR1 is formed first, which is then formed with a second molecule by the method according to the invention Isothiocyanate reacts to form a symmetrical hydrazine derivative.
As already mentioned, the hydrazine derivatives obtainable according to the invention have valuable properties for changing the endocrine state in humans and animals. In particular, they inhibit the gonadotropic pituitary function and are therefore suitable for treating the sexual cycle and its deviations; Among other things, they can also be used to regulate oestrus (heat) in pets. For these purposes they can be used in the form of pharmaceutical or veterinary preparations.
The pharmaceutical and veterinary preparations may be in a form suitable for oral or parenteral use. The preparations can, for example, also be in the form of mixtures with animal feed or in the form of mixtures suitable for addition to animal feed.

Example I.
A solution of 5.65 parts of cc-methylallyl isothiocyanate in 10 parts of ethanol is stirred dropwise into a cooled solution of 1.25 parts of hydrazine hydrate (100% in) in 10 parts of ethanol. The methylallyl isothiocyanate, which is used as the starting material, can be obtained by reacting methylallyl bromide and ammonium thiocyanate with one another, a rearrangement taking place with the formation of os-methylallyl isothiocyanate in this reaction.
Example 2
The process according to Example 1 is repeated, Nl, N2-bis (p-methylallyl-thiocarbamoyl) hydrazine, melting point 200-20 ° C. (with decomposition), being obtained from 3-methylallyl isothiocyanate.
Example 3
A mixture of 6.5 parts of 4-allylthiosemicarbazide, 5.52 parts of tx-methylallyl isothiocyanate and 25 parts of ethanol is refluxed for 1 hour. The mixture is cooled and filtered and the residue is washed with ethanol, dried and crystallized from ethanol. In this way, N1 -allylthiocarbamoyl-N2'K-methylallylthiocarbamoylhydrazine, melting point 196-198 ° C. (with decomposition) is obtained.
Example 4
