Verfahren zur Herstellung von 2 --Cyan-4u,5a-epoxy-3-oxo-steroiden Die Erfindung betrifft ein Verfahren zur Herstellung von U-Cyan-4a,5a-epoxy-3-oxo-steroiden mit 17 bis 23 C-Atomen, C-Atome von allfälligen Esterresten ausge schlossen.
In der deutschen Patentschrift Nr. 1 148 544 sind 2 -Cyan-3-oxo-steroide beschrieben, deren Steroidrest 17 bis etwa 23 C-Atome aufweist, ausgeschlossen Esterreste oder ein Salz davon, wobei diese Verbindungen durch Be handlung des entsprechenden Steroido-[2,3-d]-isoxazols mit einer starken Base, und gegebenenfalls durch An säuern des erhaltenen Basensalzes, wobei das freie Pro dukt erhalten wird, hergestellt werden können.
Die Verfahrensprodukte stellen eine neue Klasse von Verbindungen mit besonderer pharmakologischer Aktivi tät dar und können sich von jedem Steroid des allgemei nen Typs ableiten, der dafür bekannt ist, hormonale oder andere pharmakologische oder endokrinologische Eigen schaften zu zeigen.
Derartige Steroidreste weisen 17 bis 23 C-Atome auf, ohne die Kohlenstoffe von allfälligen Esterresten. Veresterte Hydroxysteroide fallen in den Rahmen des erfindungsgemässen Verfahrens, jedoch wird, wie gesagt, die Anzahl der C-Atome des Säure restes des Esters nicht als Teil der Anzahl der C-Atome des Steroids betrachtet.
Der Steroidrest kann irgendein Rest aus der östran-, 18-Nor-östran-, Androstan-, Äthiocholan-, Pre2nan- oder Allopregnanreihe sein. Der Steroidrest kann verschiedene Grade an Ungesättigtheit und eine Vielzahl von Substi- tuenten in Form von Kohlenwasserstoffresten oder von funktionellen Gruppen enthalten, wie sie auf dem Ste roidgebiet üblich sind.
Repräsentative Steroidreste. aus denen die erfindungsgemäss erhältlichen Verbindungen hergestellt werden, weisen vorzugsweise in 17-Stellung einen Hydroxy-, Acyloxy-, Oxo- oder sowohl einen Hy- droxyl- als auch einen Niedrigalkylrest auf; diese sind für androgene und anabolische Steroide charakteristisch.
Das Vorliegen eines Niedrigalkenyl-, Niedriaalkinyl-, Acetyl-, Hydroxyacetyl-, 1,2-Dihydroxyäthyl-, 1-Hydroxy- iithylrestes und von ähnlichen Resten ist für progestatio- nelle und adrenal-cortikale Steroide charakteristisch.
Der Steroidrest kann auch einen oder mehrere Substituenten in anderen Stellungen des Steroidgerüsts enthalten, bei- spielsweise Hydroxy-, Acyloxy- oder Oxoreste in den Stellungen 6, 7, 11, 12.
14 oder 16, Epoxygruppen in be nachbarten Stellungen, beispielsweise in 9, 11-, 11, 12- oder 15, 16-Stellung, Halogenatome, vorzugsweise Fluor, Chlor oder Brom, beispielsweise in 6-. 7-, 9-, 12-, 16-, 17- oder 21-Stellung, und Niedrigalkylgruppen, beispielsweise in 5-, 6-, 7-, 11- oder 16-Stellung. Der Steroidrest kann ebenfalls eine oder mehrere Doppelbindungen enthalten, beispielsweise in 6, 7-, 9, 11- oder 16,
17-Stellung. Der Steroidrest weist gewöhnlich angulare Methylgruppen am C-10 und C-13 auf, wenn auch 18- und 19-Norsteroide und 18, 19-Bisnorsteroide, bei denen eine oder beide der angularen Methylgruppen am C-13 bzw. am C-10 fehlen, ebenfalls repräsentative Steroide darstellen.
Die 18.19-Bis-norsteroid-, 18,19-Nor-steroid- und die normalen Steroidreste in den erfindungsgemäss erhält lichen Verbindungen können 17, 18 bzw. 19 Kohlenstoff atome und zusätzlich einen beliebieen Gehalt an Kohlen stoffatomen, der von einem oder mehreren kohlenstoff- halti,-en Kernsubstituenten stammt, bis zu einschliesslich insgesamt 23 Kohlenstoffatomen, C-Atome von allfälli- Ren Esterresten ausgeschlossen, enthalten.
Wenn Esterverknüpfungen in dem Steroidmolekül vorliegen, leiten sich die Acylreste vorzugsweise von Carbonsäuren mit 1 bis 10 Kohlenstoffatomen ab, wie sie üblicherweise auf dem Steroidgebiet verwendet wer den, und mit einem Molekulargewicht unter etwa 200 ab. Repräsentative Acylreste, die anwesend sein können.
sind Alkanoylreste mit 1-8 C-Atomen, beispielsweise Formyl, Acetyl, Propionyl-Butyryl, Isobutyryl, Caproyl, Heptanoyl, Octanoyl, Trimethylacetyl und dgl., Carb- oxyniedrigalkanoylreste, beispielsweise Succinyl (-Carb- oxypropionyl),
Cycloalkyl-niederigalkanoylreste. bei spielsweise g-Cyclopentylpropionyl,@tss-Cyclohexylpropio- nyl und dgl., monocarbocyclische Aroylreste, beispiels weise Benzoyl, p-Toluyl, p-Nitrobenzoyl, 3,4,5-Trime- thoxybenzoyl und dgl., monocarbocyclische Aryl-niedrig- alkanoyl- oder -alkenoylreste, wie Phenylacetyl,
@-Phe- nylpropionyl, Cinnamoyl und d-1., und monocarbocycli- sche Aryloxy-niedrigalkanoylreste, wie p-Chlorphenoxy- acetyl und dgl.
Ester anorganischer Säuren, wie der Phos phorsäure, sind Teichfalls vor;esehen. Wenn monocarbo- cyclische Arylgruppen in den Esterresten vorliegen, so umfasst monocarbocyclisches Aryl Phenyl und mit 1 bis 3 Niedrigalkyl-, Niedrigalkoxy-, Halogen- oder Nitro- gruppen substituiertes Phenyl.
Die Ester können nach herkömmlichen Verfahrens weisen erhalten werden, beispielsweise durch Behandlung des entsprechenden Steroidalkohols mit dem geeigneten Säurehalogenid oder -anhydrid.
Die genannten 2a-Cyan-4.z,52-epoxy-3-oxo-steroide werden erfindungsgemäss durch Spaltung eines entspre chenden Steroido-[2,3-d]-isoxazols mit einer starken Base Uemäss dem folgenden Schema

hergestellt. Zur Umwandlung des Isoxazols in Cyanketon kann jede starke Base verwendet werden, jedoch sind die Al kalimetallalkylate bevorzugt und die Reaktion wird am besten in einem wasserfreien Medium durchgeführt.
Die 2a-Cyan-4a,5a-epoxy-3-oxo-steroide sind von Na tur aus sauer, wobei sie ein aktives Wasserstoffatom in der 2-Stellung aufweisen. und sie bilden deshalb Salze mit starken Basen, wie Alkalimetallhydroxyden oder -alkyl- aten. So wird bei der Spaltung des Isoxazols ein Salz des 2x-Cyan-44,5,j,-epoxy-3-oxo-steroids gebildet und dieses Salz kann durch Ansäuern in das freie 2a-Cyait-4x,5x- -epoxy-3-oxo-steroid umgewandelt werden.
Die Salze stellen volle Äquivalente der freien Säuren (d.h. der freien 2x-Cyan-4x,5x-epoxy-3-oxo-steroide) dar.
Die 4x.5a-Epoxy-steroido-[2,3-d]-isoxazole können durch Umsetzung eines _\\,'-Steroido-[2,3-d]-isoxazols mit einem Oxydationsmittel, das fähig ist, eine olefinische Doppelbindung in ein Epoxyd umzuwandeln, gemäss der folgenden Gleichung hergestellt werden:

Zu derartigen Oxydationsmitteln gehören insbeson dere Wasserstoffperoxyd unter alkalischen Bedingungen oder eine Percarbonsäure, beispielsweise Perbenzoesäure. Monoperphthalsäure, Peressigsäure und dol.
Die endokrinologische Untersuchung der erfindungs- gemäss erhältlichen 2a-Cyan-4a,5a-epoxy-3-oxo-steroide hat gezeigt, dass sie andreno-cortikal inhibierende Eigen schaften besitzen.
Beispielsweise wird bei der Untersu chung an Ratten gefunden, dass das 9-x-Cyan-4a,5a- -epoxy-androstan-17ss-ol-3-on die ACTH-indtizierte Zu nahme an Plasmacorticosteroiden Lind adrenalen Cor- ticosteroiden, ACTH-induzierten Stickstoffverlust und Thymolyse und die von ACTH und Kaltstrom induzierte niedere Tryptophan-Pyrrolase-Aktivität verhindert.
Bei der Untersuchung an Hunden ist gefunden worden, dass die genannte Verbindung ACTH-stimulierte adrenale Hy peraktivität umkehrt und adrenale Steroidgenese inhi- biert, wobei die adrenale 3ss-Hydroxy-steroid-Dehydro- genase blockiert wird.
Aus diesen Ergebnissen geht her vor, dass die erfindungsgemäss erhältlichen 2x-Cyan-4a. 5x-epoxy-3-oxo-steroide bei der Behandlung von patho logischen Zuständen, wie beim Cushing-Syndrom, bei adrenaler Neoplasie, physischem Trauma, Hirsutismus, psychischen Leiden und primärem und sekundärem Aldo- steronismus, verwendbar sind.
Die erfindungsgemäss erhältlichen Verbindungen kön nen für die Anwendung durch Dispeigieren in einer wäss- rigen Suspension oder durch Auflösen in einem pharma kologisch verträglichem Öl oder einer pharmakologisch verträglichen öl/Wasser-Emulsion zur parenteralen Ver abreichung oder durch Vermischen mit Bindemitteln zur oralen Verabreichung präpariert werden. Die Struktur der erfindungsgemäss erhältlichen Verbindungen wird durch ihre Herstellungsweise, durch Elementaranalyse und durch Ultraviolett- und Infrarotspektraldaten bestimmt.
<I>Beispiel 1</I> a) Zu einer Lösung von 6,16 g 17ss-Hydroxy-4-andro- steno-[2,3-d]-isoxazol, 0,5g Natriumacetat und 50 ml Essigsäure in 500 ml Benzol wird eine Lösung von 3,8 ml 42,2%iger Peressigsäure in 10 ml Essigsäure gegeben.
Die Reaktionsmischung wird bei Raumtemperatur einen Tag stehengelassen. Das Lösungsmittel wird von der Mi schung abgedampft Lind der Rückstand wird zuerst aus Aceton (Ausbeute 4,42 g) und dann aus Äthanol umkri stallisiert, wobei sich das 17ss-Hydroxy-4a,5x-epoxy-an- drostano-[2,3-d]-isoxazol mit einem F = 205 bis 207 C ergibt. [a]=sn = +107,8 (1% in Chloroform).
b) Eine Mischung von 0,40 g 17ss-Hydroxy-4x,5x- -epoxy-androstano-[2,3-d]-isoxazol, 0,1 g Natriumme- thylat und 10 ml Tetrahydrofuran wird 15 Minuten ge rührt. Es werden 5 ml Methanol zugegeben und die Ai- schung wird 10 Minuten gerührt. Dann wird allmählich Wasser zugegeben, bis sich eine trübe Lösung ergibt. Die Mischung wird mit Bcnzol und Äther extrahiert und die wässrige Schicht wird mit verdünnter Schwefelsäure an gesäuert.
Der gebildete Niederschlag wird gesammelt, ge trocknet und aus Pyridin/Dioxan umkristallisiert, wo bei sich das 2x-Cyan-4x,5a-epoxy-androstan-17ss-ol-3-on mit einem F = 258 bis 270 C (Zersetzung) ergibt.
@_ 13T40 (107, in Pyridin). <I>Beispiel 2</I> a) Zu einer Lösung von 3,00 g lvlaleinsätircanhydrid in 150 ml iViethylendichlorid, die einige mg Natriumacetat enthält, werden 0,97 ml 88%iges Wasserstoffperoxyd und anschliessend 7,1 g 17ss-Acetoxy-4-androsteno-[2,3-d]- -isoxazol und 10 Tropfen Pyridin gegeben. Die Reak tionsmischung wird über Nacht im Eisschrank belassen.
Die überschüssige Persäure wird durch Zugabe von Na- triumbisLilfitlöstin2 zerstört und die Mischung wird mit Wasser Lind mit Z#atriumbicarbonatlösungg gewaschen. Die organische Lösung wird getrocknet und im Vakuum kon zentriert.
Der Rückstand wird nacheinander aus Aceton, Äthylacetat (Ausbeute 6,63 g) und Benzol/Methanol um kristallisiert, wobei sich das 17P-Acetoxy-4a,5a-epoxy- -androstano-[2,3-d]-isoxazol mit einem F = 228,5 bis 2300C ergibt. [a]=n = + 76,50 (1a/, in Chloroform).

The compounds obtainable according to the invention can be prepared for use by dispersing them in an aqueous suspension or by dissolving them in a pharmaceutically acceptable oil or a pharmacologically acceptable oil / water emulsion for parenteral administration or by mixing them with binders for oral administration. The structure of the compounds obtainable according to the invention is determined by their method of preparation, by elemental analysis and by ultraviolet and infrared spectral data.
