Verfahren zur Herstellung neuer basischer Phenyläther
Die vorliegende Erfindung betrifft ein Verfahren zur Herstellung neuer 1-tert.-Butylamino-3-phenoxy-2-pro- panole mit einer wertvollen pharmakodynamischen Wirksamkeit. Die erfindungsgemäss erhältLichen Verbin- dungen haben insbesondere die Eigenschaft, ss-adrener gische Rezeptoren zu blockieren ; sie wirken daher gegen Angina und Arrhythmie.
Die/ ?-RezeptorenbIocker-Wirkung der neuen Verbindungen lässt sich dadurch zeigen, dass sie der stimu- lierenden Wirkung von Adrenalin auf das Herz entgegen- wirken, was sich in der Verminderung des Anstiegs der Sohlagzahl bei Einwirkung von Adrenalin auf isolierte rechte Aurikula von Ratten zeigt. Diese Verbindungen haben in hohem Ausmass blockierende Wirksamkeit ohne die unerwünschte direkte Stimulierung oder cardiale Depression.
Die erfindungsgemäss erhältlichen Verbindungen haben die folgende allgemeine Formel I

worin R, Chlor, Brom oder eine niedere Alkylgruppe mit 1-4 Kohlenstoffatomen, R Wasserstoff, Chlor oder Brom, R3 Wasserstoff oder niedere Alkylgruppen mit 1-4. Kohlenstoffatomen darstellen, wobei allfällige Alkylsubstituenten in 2-und 5-Stellung sowie R2 und Rs voneinander verschieden sind. Vorzugsweise ist R1 Chlor, Ra Wasserstoff und R3 Methyl.
Weiterhin umfasst die vorliegende Erfindung auch ein Verfahren zur Herstellung der Salze der Aminover- bindungen der Former I mit ungiftigen pharmakologisch unbedenklichen organischen und anorganischen Säuren.
Geeignete Säuren sind Maleinsäure, Fumarsäure, Benzoe- säure, Ascorbinsäure, Pamoinsäure, Bernsteinsäure, Methansulfonsäure, AthandisulfonsauTe, Cyclohexyl- sulfamsäure, Essigsäure und Weinsäure. Geeignete anorganische Säuren sind Chlorwasserstoffsäure, Bromwasserstoffsäure, Schwefe) säure und Phosphorsäure.
Die Verbindungen der Formel I sowie deren Salze mit untgiftigen pharmakologisch unbedenklichen Säuren können dadurch hergestellt werden, da# man eine Verbindung der allgemcinen Formel 11

mit einem tert.-Butylhalogenid umsetzt, wobei in der Formel II RI, R2 und R : t die gleiche Bedeutung wie in Formel I haben, und die so erhaltenen Basen gege benenfalls in Salze mit ungiftigen, pharmakologisoh unbedenklichen Säuren überführt.
Die folgenden Beispiele erläutern das erfindungs- gemässe Verfahren.
Beispiel
32, 5 g 1 (2, 4-Dibromphenoxy)-2-hydroxy-3-amino- propan werden mit 20, 5 g tert.-Butylbromid und 100 ml Athanol auf dem Wasserbad 10 Stunden zum Sieden erhitzt. Anschliessend wird unter vermindertem Druck zur Trockne destilliert und der Rückstand unter Zugabe von Salzsäure in Wasser gelöst. Die wässrige Lösung wird mittels NaOH alkalisiert und die sich abscheidende Base in Benzol aufgenommen. Nach dem Trocknen wird in die Benzol-Lösung der Base trockenes Hcl-Gas eingeleitet. Das sich hierbei abscheidende 142, 4-Dibrom phenoxy)-2-hydroxy-3 tert.- : butylaminopropanhydro- chlorid wird aus Athanol umkristallisiert (Fp. 204 bis 206 C).

Process for the production of new basic phenyl ethers
The present invention relates to a process for the production of new 1-tert-butylamino-3-phenoxy-2-propanols with valuable pharmacodynamic activity. The compounds obtainable according to the invention have the particular property of blocking β-adrenergic receptors; therefore they work against angina and arrhythmia.
The /? -Receptor-blocking effect of the new compounds can be shown by the fact that they counteract the stimulating effect of adrenaline on the heart, which results in the reduction of the increase in the bottom number when adrenaline acts on isolated right auricles of rats shows. These compounds have high blocking effectiveness without the undesirable direct stimulation or cardiac depression.

Furthermore, the present invention also comprises a process for the preparation of the salts of the amino compounds of Former I with non-toxic, pharmacologically acceptable organic and inorganic acids.
Suitable acids are maleic acid, fumaric acid, benzoic acid, ascorbic acid, pamoic acid, succinic acid, methanesulfonic acid, AthandisulfonsauTe, cyclohexylsulfamic acid, acetic acid and tartaric acid. Suitable inorganic acids are hydrochloric acid, hydrobromic acid, sulfuric acid and phosphoric acid.
The compounds of the formula I and their salts with non-toxic pharmacologically acceptable acids can be prepared by using a compound of the general formula 11

with a tert-butyl halide, where in formula II RI, R2 and R: t have the same meaning as in formula I, and the bases thus obtained are optionally converted into salts with non-toxic, pharmacologically harmless acids.
The following examples explain the process according to the invention.

32.5 g of 1 (2,4-dibromophenoxy) -2-hydroxy-3-aminopropane are heated to boiling on a water bath for 10 hours with 20.5 g of tert-butyl bromide and 100 ml of ethanol. It is then distilled to dryness under reduced pressure and the residue is dissolved in water with the addition of hydrochloric acid. The aqueous solution is made alkaline using NaOH and the base which separates out is taken up in benzene. After drying, dry HCl gas is passed into the benzene solution of the base. The 142,4-dibromophenoxy) -2-hydroxy-3-tert.-: butylaminopropane hydrochloride which separates out is recrystallized from ethanol (melting point 204-206 ° C.).
