Verfahren zur Herstellung von 2-Chlor-5-aryl-1,3,4-thiadiazolen
Die Erfindung liegt in einem neuen Verfahren zur Herstellung von 2-Chloro-5-aryl-1,3,4-thiadiazolen.
Nach dem bisherigen Stand der Technik bedurfte es zur Herstellung dieser Verbindungen eines mehrstufigen Verfahrens, wobei Benzaldehyd-thiosemicarbazon mit Eisenchlorid zum 2-Amino-1, ,3,4-thiadiazol cyclisiert (I1 Farmaco, ed sci. 13, 187 [1958] und dieses nach Diazotierung (J. Sci. Ind. Res. [NewDelhi] 21C, 444 [1962]) durch die Sandmeyer-Reaktion in das 2-Chlor-5-phenyl 1,3 ,4-thiadiazol übergeführt wurde.
Es wurde nun gefunden, dass gleiche Endprodukte auch durch eine Eintopfreaktion erhalten werden, wenn man ein Arylaldazin mit Thiophosgen reagieren lässt.
Die so erhaltenen Verbindungen stellen infolge ihres leicht beweglichen Chloratoms insbesondere begehrte Zwischenprodukte für die Herstellung von in 2-Stellung substituierten 5-Phenyl-1,3,4-thiadiazolen dar. Die aus den Verfahrensprodukten z. B. durch Umsetzung insbesondere mit Ammoniak, primären oder sekundären Aminen, aber auch mit Hydroxy-, Alkoxy- oder Aryloxy Verbindungen, Merkaptanen, Alkylthio- und Arylthioverbindungen erhaltenen Produkte weisen pharmazeutisch interessante Eigenschaften auf (I1 Farmaco, 1 .c., sowie Brit. J. Pharmacol. 13, 357 [1958]).
Das Ringsystem des als Ausgangsmaterial dienenden Arylaldazins kann auch substituiert sein und beispielsweise ein oder mehrere Halogenatome oder gegebenenfalls halogenierte Alkyl-, Alkoxy-, Alkylthio-, Dialkylamino-, Dialkylaminoalkyl-, Dialkylsulfonamido-, Cyano-, Carboxamido-, Sulfonsäure-, Alkoxycarbonylgruppen aufweisen.
Die Reaktion wird zweckmässig bei erhöhter Temperatur von vorzugsweise mindestens 1000 C und in Anwesenheit eines in bezug auf beide Reaktionspartner indifferenten Lösungsmittels durchgeführt; mit Vorteil wird das Thiophosgen im Überschuss eingesetzt.
Beispiel (Alle angegebenen Schmelzpunkte sind unkorrigiert)
Eine Mischung von 65 ml Thiophosgen (853 mM) mit 87,8 g Benzaldazin (421 mM) und 300 ml abs.
Xylol wurden in einem 2-Liter-Glasautoklaven mit Ma gnetrührung im Ölbad unter dem Abzug und unter Verwendung eines Schutzschildes schnell auf 135 C Badtemperatur erwärmt und 1 Stunde auf dieser Temperatur gehalten. Bei 112" C entstand im Gasraum ein karminroter Blitz, und die Gefässwand erhielt einen feinen schwarzen Belag. Nach Abkühlung wurde das Reaktionsgemisch im Rotationsevaporator auf dem Wasserbad eingedampft und der Rückstand fraktioniert. Zwischen 1130 C/0,8 mm Hg und 1800 C/2 mm Hg gingen 27,78 g Rohprodukt über. Dieses lieferte nach Umkristallisieren aus Benzin 12,635 g 2-Chlor 5-phenyl-1,3,4-thiadiazol in Form hellgelber Kristalle mit Smp. 83,5-86" C (Smp. gemäss J. Sci. Ind.
Res., l. c. : 88" C/Athanol). Ausbeute: lS,30/o d. Th. Das Produkt konnte auch aus viel Wasser oder aus verdünnter Salzsäure umkristallisiert werden.

Mit dem in diesem Beispiel nach dem erfindungsgemässen Verfahren erhaltenen Produkt wurden folgende Reaktionen durchgeführt: Reaktion n mit Methylamin
Eine Mischung von 8,54 g (43,4 mM) der erhaltenen 2-Chlor-Verbindung mit 25 ml wasserfreiem Methylamin und 250 ml absolutem Äthanol wurde 10 Stunden in einem l-Liter-Glasautoklaven auf 100" C erhitzt, worauf das Reaktionsgemisch im Vakuum auf dem 400 ml Benzol erhitzt und heiss filtriert wurde. Das Filtrat wurde nach dem Abkühlen mit wenig Benzin versetzt und in den Eisschrank gestellt.
Durch Filtration erhielt man dann 5,76 g feine Kristalle vom Smp.
170-178,5 C und durch Aufarbeiten der Mutterlaugen weitere 1,18 g desselben Materials, welches nach Umkristallisieren aus Benzol 2-Methylamino-5-phenyl1,3,4-thiadiazol vom Smp. 176,5-179 C lieferte. Ausbeute: 83,7% d. Th.

Reaktion mit Kaliumäthylat bei Raumtemperatur
Einer Lösung von 4,16 g (21,15 mM) der 2-Chlor Verbindung in 25 ml abs. Äthanol wurde eine Lösung von 0,83 g (21,2 mg-Atom) Kaliummetall in 50 ml abs.
Äthanol zugegeben, worauf man das Gemisch drei Tage gut verschlossen bei Raumtemperatur stehen liess und anschliessend filtrierte. Der durch Eindampfen des Filtrats im Vakuum erhaltene Rückstand (4,08 g) wurde in einem Gemisch von 100 ml Äthanol und 5 ml Wasser gelöst und nach Schichtentrennung mit zweimal 5 ml Wasser neutralgewaschen. Die organische Schicht lieferte nach Trocknung und Eindampfen 3,965 Rückstand in Form eines rasch erstarrenden Öls mit Smp.
65-67,5 C (Ausbeute: 90,9% d. Th.) Durch Umkristallisieren aus viel Wasser erhielt man daraus das 2 Äthoxy-5-phenyl-1,3,4-thiadiazol in Form von feinen, farblosen, verfilzten Nadeln mit Smp. 67-68 C (H.
Ohta und M. Ohta, J. chem. Soc. Japan, pure chem. section 78, 700 (1957) Zentalblatt 1958, 5663 geben demgegenüber an: Smp. 26-27 C, Sdp. 140-145 C/ 4 mm Hg).

Reaktion mit Kaliumäthylat in der Hitze
Eine Lösung von 4,45 g (22,6 mM) der 2-Chlor Verbindung in 25 ml abs. Äthanol wurde mit einer Lösung von 1,30 g (33,3 mg-Atom) Kaliummetall in 50 ml abs. Äthanol vereinigt und im bombenrohr 10 Stunden auf 125-130 C erhitzt. Das Raktionsgemisch wurde anschliessend im Vakuum eingedampft, in 100 ml Äther aufgenommen und sechsmal mit je 5 ml Wasser gewaschen, bis der pH-Wert auf 8 gesunken war.
Die vereinigten wässrigen Phasen wurden angesäuert und mit Äther extrahiert. Der ätherische Extrakt lieferte nach trocknung und Eindampfen 2,37 g rückstand mit Smp. 141-144 C (Ausbeute: 58,8% d. Th.), aus welchem man nach Umkristallisieren aus Tetrachlorkohlenstoff das 2-Hydroxy-5-phenyl- 1,3 ,4-thiadia- zol in Form von leicht bräunlichen, gefiederten Nadeln vom Smp. 145-146 C erhielt (Smp. gemäss J. chem.
Soc. 1961, 1617: 146-148 C/Chloroform-Petroläther).

Reaktion mit Ammoniak
Eine Lösung von 7,00 g (35,6 mM) der 2-Chlor Verbindung in 50 ml abs. Äthanol wurden mit 150 ml einer bei 0 C gesättigten äthanolischen Ammoniaklösung vereinigt und in einem Autoklaven mit Glaseinsatz während 10 Stunden auf 1000 C erhitzt. Das Re aktiongsgemisch wurde dann im Vakuum auf dem Wasserbad zur Trockene eingedampft, worauf man den Rückstand (8,2 g) mit einem Gemisch aus 100 mol äther und 10 ml Wasser und anschliessend zweimal mit je einem Gemisch aus 100 ml Benzol und 10 ml Wasser schüttelt und das nicht gelöste Material abfiltrierte.
Nach Eindampfen der getrockneten organischen Phase blieb 5,66 g nicht umgesetztes Ausgangsmaterial zurück (Smp. 79-83 C, identifiziert durch Misch-Schmelzpunkte; 80,90/o des Einsatzes). Der in den organischen Lösungsmitteln unlösliche Rückstand (1,07 g; Ausbeute: 17% d. Th.) bestand aus einem farblose@ Pulver vom Smp 200-204 C, aus welchem man durch Umkristallisieren aus Essigester das 2-Amino-5-phenyl1,3,4-thiadiazol in Form feiner Nadeln mit Smp. 212213 C (Smp. gemäss Chem. Ber. 96, 1059 (1963): 223 C) erhielt.
Process for the preparation of 2-chloro-5-aryl-1,3,4-thiadiazoles
The invention resides in a new process for the preparation of 2-chloro-5-aryl-1,3,4-thiadiazoles.
According to the prior art to date, a multistage process was required for the preparation of these compounds, benzaldehyde thiosemicarbazone cyclizing with iron chloride to form 2-amino-1,3,4-thiadiazole (I1 Farmaco, ed. Sci. 13, 187 [1958] and this after diazotization (J. Sci. Ind. Res. [NewDelhi] 21C, 444 [1962]) was converted into the 2-chloro-5-phenyl 1,3,4-thiadiazole by the Sandmeyer reaction.
It has now been found that the same end products are also obtained by a one-pot reaction if an arylaldazine is allowed to react with thiophosgene.
The compounds obtained in this way are, owing to their easily mobile chlorine atom, particularly sought-after intermediates for the preparation of 5-phenyl-1,3,4-thiadiazoles substituted in the 2-position. B. products obtained by reaction in particular with ammonia, primary or secondary amines, but also with hydroxy, alkoxy or aryloxy compounds, mercaptans, alkylthio and arylthio compounds have pharmaceutically interesting properties (I1 Farmaco, 1. J. Pharmacol. 13, 357 [1958]).
The ring system of the arylaldazine used as starting material can also be substituted and, for example, have one or more halogen atoms or optionally halogenated alkyl, alkoxy, alkylthio, dialkylamino, dialkylaminoalkyl, dialkylsulfonamido, cyano, carboxamido, sulfonic acid, alkoxycarbonyl groups.
The reaction is expediently carried out at an elevated temperature of preferably at least 1000 ° C. and in the presence of a solvent which is indifferent to both reactants; the thiophosgene is advantageously used in excess.
Example (all specified melting points are uncorrected)
A mixture of 65 ml of thiophosgene (853 mM) with 87.8 g of benzaldazine (421 mM) and 300 ml of abs.
Xylene was quickly heated to 135 ° C. bath temperature in a 2 liter glass autoclave with magnetic stirring in an oil bath under the fume cupboard and using a protective shield and kept at this temperature for 1 hour. At 112 "C a carmine-red flash appeared in the gas space, and the vessel wall had a fine black coating. After cooling, the reaction mixture was evaporated in a rotary evaporator on a water bath and the residue was fractionated. Between 1130 C / 0.8 mm Hg and 1800 C / 2 mm Hg, 27.78 g of crude product passed over. After recrystallization from gasoline, this yielded 12.635 g of 2-chloro 5-phenyl-1,3,4-thiadiazole in the form of light yellow crystals with a melting point of 83.5-86 "C (melting point according to J. Sci. Ind.
Res., L. c. : 88 "C / ethanol). Yield: 15, 30 / o of theory. The product could also be recrystallized from a lot of water or from dilute hydrochloric acid.

The following reactions were carried out with the product obtained in this example by the process according to the invention: Reaction n with methylamine
A mixture of 8.54 g (43.4 mM) of the 2-chloro compound obtained with 25 ml of anhydrous methylamine and 250 ml of absolute ethanol was heated to 100 ° C. for 10 hours in a 1 liter glass autoclave, whereupon the reaction mixture was im Vacuum on which 400 ml of benzene were heated and filtered while hot. Filtration then gave 5.76 g of fine crystals of mp.
170-178.5 ° C. and, by working up the mother liquors, a further 1.18 g of the same material which, after recrystallization from benzene, gave 2-methylamino-5-phenyl1,3,4-thiadiazole with a melting point of 176.5-179 ° C. Yield: 83.7% of theory Th.

Reaction with potassium ethylate at room temperature
A solution of 4.16 g (21.15 mM) of the 2-chloro compound in 25 ml of abs. Ethanol was a solution of 0.83 g (21.2 mg atom) of potassium metal in 50 ml of abs.
Ethanol was added, whereupon the mixture was left to stand well sealed for three days at room temperature and then filtered. The residue (4.08 g) obtained by evaporating the filtrate in vacuo was dissolved in a mixture of 100 ml of ethanol and 5 ml of water and, after separation of the layers, washed neutral with twice 5 ml of water. After drying and evaporation, the organic layer gave 3.965 residue in the form of a rapidly solidifying oil with mp.
65-67.5 ° C. (yield: 90.9% of theory). Recrystallization from a lot of water gave 2 ethoxy-5-phenyl-1,3,4-thiadiazole in the form of fine, colorless, matted needles with m.p. 67-68 C (H.
Ohta and M. Ohta, J. chem. Soc. Japan, pure chem. Section 78, 700 (1957) Zentalblatt 1958, 5663, on the other hand, indicate: Melting point 26-27 C, Sdp. 140-145 C / 4 mm Hg).

Reaction with potassium ethylate in the heat
A solution of 4.45 g (22.6 mM) of the 2-chloro compound in 25 ml of abs. Ethanol was with a solution of 1.30 g (33.3 mg atom) of potassium metal in 50 ml of abs. Combined ethanol and heated to 125-130 C in a bomb tube for 10 hours. The reaction mixture was then evaporated in vacuo, taken up in 100 ml of ether and washed six times with 5 ml of water each time until the pH had dropped to 8.
The combined aqueous phases were acidified and extracted with ether. After drying and evaporation, the ethereal extract yielded 2.37 g of residue with a melting point of 141-144 ° C. (yield: 58.8% of theory), from which 2-hydroxy-5-phenyl-1 was recrystallized from carbon tetrachloride , 3, 4-thiadiazole in the form of slightly brownish, feathery needles with a melting point of 145-146 ° C. (melting point according to J. chem.
Soc. 1961, 1617: 146-148 C / chloroform petroleum ether).

Reaction with ammonia
A solution of 7.00 g (35.6 mM) of the 2-chloro compound in 50 ml of abs. Ethanol were combined with 150 ml of an ethanolic ammonia solution saturated at 0 C and heated to 1000 C for 10 hours in an autoclave with a glass insert. The reaction mixture was then evaporated to dryness in vacuo on a water bath, whereupon the residue (8.2 g) with a mixture of 100 mol of ether and 10 ml of water and then twice with a mixture of 100 ml of benzene and 10 ml of water shakes and the undissolved material filtered off.
After evaporating the dried organic phase, 5.66 g of unreacted starting material remained (melting point 79-83 ° C., identified by mixed melting points; 80.90 / o of the charge). The residue (1.07 g; yield: 17% of theory), which was insoluble in the organic solvents, consisted of a colorless powder with a melting point of 200-204 ° C., from which 2-amino-5-phenyl1 was obtained by recrystallization from ethyl acetate , 3,4-thiadiazole in the form of fine needles with melting point 212213 C (melting point according to Chem. Ber. 96, 1059 (1963): 223 C).
