worin die 3 mit Ph bezeichneten Phenylreste zusam men mindestens einen Tertiäraminoalkoxyrest und je höchstens zwei Alkyl-, Alkylsulfonyl-, Alkylthio-, Alk- oxy-, Tertiäraminoalkoxy-, Dialkylamino- oder ver- esterte Hydroxygruppen oder Halogenatome tragen, und von Salzen dieser Verbindungen.
Die mit Ph bezeichneten Phenylreste können die selben oder verschiedene Substituenten aufweisen. Der mindestens an einem Phenylrest vorhandene Tertiär- aminoalkoxyrest stellt zweckmässig ein Radikal der Formel R2-(Methylen)n-O- dar, bei welchem n eine ganze Zahl von 1 bis 7, insbe sondere 2 oder 3 und R2 eine tertiäre Aminogruppe be deuten. Letztere ist zweckmässigerweise eine Dialkyl- aminogruppe, wie die Dimethylamino- oder vorzugs weise Diäthylaminogruppe. Sie kann jedoch auch in Form einer heterocyclischen Gruppe, wie z. B. in Form des Morpholino-, Pyrrolidino- oder Piperidinoradikals, vorliegen.
Sie befindet sich zweckmässig in p-Stellung des Phenylrestes.
In jedem der 3 Phenylringe können daneben bis zu insgesamt 2 Substituenten, vorzugsweise z. B. niedere Alkylgruppen, wie Methyl, Äthyl, Isopropyl; niedere Alkylsulfonylgruppen, wie Methylsulfonyl, Äthylsulfo- nyl; niedere Alkylthiogruppen, wie Methylmercapto, Äthylmercapto; niedere Alkoxygruppen, wie Methoxy, Äthoxy; niedere Dialkylaminogruppen, wie Dimethyl- amino, Diäthylamino; Hydroxygruppen, die mit niede- ren Alkancarbonsäuren verestert sein können, wie Acet- oxy; Halogenatome, wie Chlor, Brom, Jod oder Fluor; oder weitere Tertiäraminoalkoxygruppen der vorstehend diskutierten Zusammensetzung, vorhanden sein.

Das erfindungsgemässe Verfahren ist dadurch ge kennzeichnet dass man eine Verbindung der allge meinen Formel Ph-CO-R (II) mittels einer metallorganischen Reaktion mit einem Halogenid der allgemeinen Formel Ph-Hal (III)
wobei in den Formeln II und III Hal ein Halogenatom und R einen Alkoxy-, Aralkoxy- oder Aryloxyrest be deuten und Ph die vorstehend angegebene Bedeutung haben, kondensiert und das Kondensationsprodukt hy- drolysiert. Im erhaltenen Reaktionsprodukt können allenfalls vorhandene schützende Gruppen abgespaltet und vorhandene Hydroxygruppen gegebenenfalls zu Acyloxygruppen verestert werden. Man kann das so erhaltene Produkt in ein Salz überführen..
Durch die metallorganische Reaktionen führt man zwei Ph-Reste ein.
Die vorstehend angeführte metallorganische Reak tion kann z. B. in Form einer Grignardreaktion durch geführt werden. Die Verbindung der Formel II wird da bei in einem inerten Lösungsmittel, wie z. B. absolutem Äther, Benzol, Tetrahydrofuran, mit dem entsprechen den Phenylmagnesiumhalogenid, das man aus einem Phenylhalogenid und Magnesium herstellen kann, kon densiert. Die Reaktion kann, wie an sich bekannt, z. B. bei einer Temperatur zwischen 0 und dem Siedepunkt des Lösungsmittels durchgeführt werden. Geeignete Halogenide sind z. B. die entsprechenden Chloride, Bromide oder Jodide.
In einer anderen Ausführungs form des erfindungsgemässen Verfahrens wird die Ver bindung der Formel Hin Gegenwart eines inerten Lö sungsmittels mit einer alkalimetallorganischen Verbin dung, welche man z. B. durch Umsetzung eines Phe nylhalogenids mit Lithium oder Alkyllithium herstellen kann, kondensiert.
Das Kondensationsprodukt wird am besten ohne Reinigung und in üblicher Weise hydro- lysiert, beispielsweise durch Eingiessen in ein Gemisch von Eis und verdünnter Salzsäure und nachfolgende Alkalisch-Stellung oder bevorzugt durch Eingiessen in kalte, wässrige Ammoniumchloridlösung, wobei der ent sprechende Alkohol erhalten wird. Dieser kann durch Behandlung mit geeigneten, mit Wasser urmischbaren organischen Lösungsmitteln, wie z. B. Chloroform, Äther, Methylenchlorid, und Waschen mit Wasser von den Nebenprodukten der Reaktion abgetrennt und iso liert werden.
Allenfalls vorhandene Hydroxylgruppen müssen vor dem Umsatz mit metallorganischen Verbindungen ge schützt werden, um eine Reaktion dieser Gruppen mit dem metallorganischen Reagens zu verhüten. Die Ab spaltung dieser Schutzgruppen, z. B. Dihydropyranyl- äther, kann gleichzeitig mit der hydrolytischen Zer legung der organischen Metallverbindung erfolgen. Wei- ters können vorhandene Hydroxygruppen gegebenenfalls zu Acyloxygruppen verestent werden. Dies kann in übli cher Weise durch Behandeln mit Säurechloriden oder -anhydriden erreicht werden.
Es können auch Salze der gemäss dem erfindungs gemässen Verfahren erhältlichen basisch substituierten Triphenylcarbinole hergestellt werden. Solche Salze sind z. B. diejenigen mit anorganischen Säuren, wie Chlor wasserstoffsäure, Bromwasserstoffsäure, Schwefelsäure und ähnliche; mit organischen Säuren, wie Oxalsäure, Essigsäure, Milchsäure, Weinsäure und insbesondere Zitronensäure und ähnliche; und mit quaternisierenden Mitteln, z. B. mit Alkylhalogeniden, wie Methylbromid, Äthyljodid, mit Dialkylsulfaten, wie Dimethylsulfat, und mit Aralkylhalogeniden, wie Benzylbromid.
Die Endprodukte des erfindungsgemässen Verfahrens können durch Kristallisation oder durch Chromato graphie an Aluminiumoxyd gereinigt werden, wobei man in der Regel farblose Kristalle erhält. Es wurde nun festgestellt, dass diese Verbindungen den Cholesteringe halt des Blutes und der Leber senken. Im Tierversuch weisen die Verbindungen bei therapeutisch geeigneter Dosierung keinen störenden Einfluss auf die normale Entwicklung (Gewichtszunahme) den Tiere auf. Sie sol len daher als Heilmittel zur Behandlung von Arterio sklerose verwendet werden.
Die Verfahrensprodukte können als Heilmittel, z. B. in Form pharmazeutischer Präparate, Verwendung fin den, welche sie oder ihre Salze in Mischung mit einem für die enterale oder parenterale Applikation geeigneten pharmazeutischen, organischen oder anorganischen iner- ten Trägermaterial, wie z. B. Wasser, Gelatine, Milch zucker, Stärke, Magnesiumstearat, Talk, pflanzliche Öle, Gummi, Polyalkylenglykole, Vaseline usw., enthalten. Die pharmazeutischen Präparate können in fester Form, z.B. als Tabletten, Dragees, Suppositorien, Kapseln, oder in flüssiger Form, z.
B. als Lösungen, Suspensionen oder Emulsionen, vorliegen. Gegebenenfalls sind sie sterilisiert und bzw. oder enthalten Hilfsstoffe, wie Kon- servierungs-, Stabilisierungs-, Netz- oder Emulgiermittel, Salze zur Veränderung des osmotischen Druckes oder Puffer. Sie können auch noch andere therapeutisch wert volle Stoffe enthalten.
<I>Beispiel 1</I> 2,5g Magnesiumspäne werden mit 10 ml trockenem Äther und einem Körnchen Jod versetzt. Hierauf gibt man unter Rühren eine Lösung von 17,3 g p-Bromtoluol in 60 ml absolutem Äther zu und erhitzt anschliessend eine halbe Stunde am Rückfluss.
Zu der erhaltenen Grignardlösung setzt man eine Lösung von 12 g p-Di- äthylaminoäthoxy - benzoesäuremethylester (hergestellt am p-Hydroxybenzoesäuremethylester durch Umsetzen mit ss-Dimethylamino-äthylchlorid; Siedepunkt bei 0,04 mm, 120 C) in 60 ml als Äther zu und erhitzt anschliessend 11/2 Stunden am Rückfluss. Man giesst das erhaltene Reaktionsprodukt auf Eis und versetzt mit einer Ammoniumchloridlösung.
Die organische Schicht wird abgetrennt, die wässrige Lösung mit Äther ge schüttelt und die vereinigten organischen Schichten drei mal mit Wasser ausgeschüttelt, über Natriumsulfat ge trocknet und eingedampft. Der Rückstand wird aus Aceton umkristallisiert, wobei man p-(Dimethylamino- äthoxy)-phenyl-p'-tolyl-p''-tolylcarbinol vom Schmelz punkt 113-115' C erhält.
<I>Beispiel 2</I> Eine aus 4,9g Magnesium und 40 g p-Bromchlor- benzol in 120 ml abs. Äther bereitete Grignardlösung wird nach den Angaben in Beispiel 2 mit 25 g p-Di- äthylamino-benzoesäuremethylester (hergestellt aus p- Hydroxybenzoesäuremethylester durch Umsetzung mit ,B - Diäthylamino - äthylchlorid, Siedepunkt bei 120 0,04 mm) umgesetzt. Man erhält p-(Diäthylamino- äthoxy)-phenyl-p'-chlorphenyl-p''-chlor-phenyl-carbinol, welches nach dem Umkristallisieren aus Petroläther bei 90-92 C schmilzt.
In analoger Weise erhält man unter Verwendung von p-Bromanisol: p-(Diäthylaminoäthoxy)-phenyl-p'-methoxy-phenyl- p''-methoxy-phenyl-carbinol vom Schmelzpunkt 62 bis 64 C; p-Bromcumol: p-(Diäthylaminoäthoxy)-phenyl-p'-isopropyl-phenyl- p''-isopropyl-phenyl-carbinol vom Schmelzpunkt 66 bis 68 C; Brombenzol: p-(Diäthylaminoäthoxy)-phenyl-diphenyl-carbinol vom Schmelzpunkt 69 biss 70 C; o-Bromdimethylanilin: p-(Diäthylaminoäthoxy)-phenyl-o'-dimethylamino- phenyl-o''-dimethylamino-phenyl-carbinol vom Schmelzpunkt 78 bis 80 C.
<I>Beispiel 3</I> 2,7 g Lithium werden zu 46 g p-Bromdimethylanilin in 150 ml abs. Äther gegeben. Innerhalb einer Stunde hat sich das Lithium praktisch vollständig gelöst. Diese Lösung wird tropfenweise mit 20 g p-Diäthylamino äthoxy-benzoesäuremethylester in 50 ml abs. Äther ver setzt und über Nacht gekocht. Nach der Aufarbeitung gemäss Beispiel 2 erhält man p-(Diäthylaminoäthoxy) phenyl-p'-dimethylamino-phenyl-p''-dimethylamino-phe nyl-carbinol vom Schmelzpunkt 91 bis 93 C.

The end products of the process according to the invention can be purified by crystallization or by chromatography on aluminum oxide, colorless crystals being obtained as a rule. It has now been found that these compounds lower the levels of cholesterol in the blood and liver. In animal experiments, the compounds, at a therapeutically suitable dosage, have no disruptive influence on the normal development (weight gain) of the animals.
