Procédé d'obtention d'une cellulase La présente invention -se rapporte à un procédé d'obtention d'une préparation de cellulase très pure, à partir d'un milieu de culture solide, composé de son de blé, ensemencé par un micro-organisme du groupe dit Aspergillus Noir tel que l'Aspergillus piger. Par la suite,
un tel milieu de culture sera désigné sous le terme koji , employé communément au Japon pour de tels milieux.
Dans le cas où l'on extrait le koji de son. de blé d'un Aspergillus Noir, par exemple l'Aspergillus piger, avec l'eau, la solution extraite a, en général, une cou leur brun noirâtre due à l'influence des spores noirs et à l'humidité du milieu de culture. En outre, l'extrait est difficile à clarifier, même par centrifugation et même si la centrifugation est réalisée à grande vitesse.
Il en résulte que la préparation de cellulase brute précipitée par un solvant organique hydrophyle tel que l'éthanol a une couleur noire ou blanc cendré et ceci diminue la valeur du produit.
D'autre part, il est bien connu qu'une préparation enzymatique blanche peut être obtenue en traitant ]'extrait coloré selon des procédés classiques de décoloration antérieurement à la précipitation par un agent organique hydrophyle. Cependant, ces procédés réduisent généralement l'activité enzymatique de la cel lulase.
Il serait .donc utile de pouvoir, d'une part, décolorer et purifier la solution d'enzyme brute de la cellulase extraite du koji de son de blé ensemencé par un micro- organisme du groupe Aspergillus Noir, tel que l'Asper gillus uiger, et, d'autre part, obtenir l'enzyme avec une activité relativement élevée si on la compare à celle de l'enzyme préparée à partir d'une solution extraite par l'eau.
On doit noter qu'au Japon on désigne par koji p le nom d'un milieu de culture solide ou d'un milieu solide cultivé, sur lequel on fait croître un champignon, pour la production de ferments. En général, le koji est constitué par du riz, du blé, du soja, du son de blé, ou leurs mélanges humidifiés par de l'eau, stérilisés et ense mencés par les spores de champignons et encastrés sur des plateaux en bois ou en métal et la culture est pour suivie à une température et avec une teneur en humidité convenables.
Selon l'invention, le procédé pour obtenir une pré paration de cellulase décolorée de grande pureté et à activité accrue est caractérisé en ce qu'on ,ensemence un milieu de culture solide, humide, par une couche d'Aspergillus piger produisant de la cellulase, on incube ce dit milieu de culture et on soumet ce dit milieu incubé à un séchage pendant 2 à 4 heures à 55-65 C, on le soumet ensuite à une extraction par solvant et on sépare la cellulase de la solution ainsi obtenue.
Dans l'exemple de réalisation préféré, on emploie un micro-organisme producteur de cellulase et très actif, l'Aspergillus piger TPR-3801. Le koji de son de blé incubé est extrait après séchage avec de l'acide lactique, de l'acide acétique ou de l'acide chlorhydrique 1/10- 1/50M. La solution d'enzyme est obtenue à l'état très clair. La solution d'enzyme ainsi obtenue est traitée par un solvant organique hydrophile pour précipiter la pré paration d'enzyme.
Le traitement de séchage est nécessaire pour obte nir un résultat satisfaisant car il améliore l'effet de déco loration. En outre, si le traitement de séchage est réa lisé pendant plusieurs heures entre 55 et 650 C, on ne trouve qu'une réduction faible de l'activité enzymatique. Dans le cas où l'on extrait le son de blé cultivé avec de l'acide après le traitement de séchage, on trouve un résultat plus efficace lorsqu'on utilise de l'acide dilué que, lorsque l'on utilise de l'acide concentré.
Le séchage selon des caractéristiques de la présente invention est également indispensable dans le cas où l'extraction du koji incubé est effectuée avec de l'eau.
