Die Cephalosporinverbindungen, worin RI Pyridiho oder substituiertes Pyridino ist, können auch hergestellt werden, indem man die carbocyclische Carboxamido- cephalosporansäureverbindung (das beisst die Verbin dung mit R1 = Acetoxy) in Lösung mit einem Über schuss von Pyridin oder substituiertem Pyridin zum
Rückfluss erhi°zt, um das entsprechende carbocyclische Acylderivat des Kerns von Cephalosporin-CA-verbin- dungen zu bilden.
Die carbocyclischen Carboxamidocephalosporan säuren können auch zur Herstellung der entsprechenden carbocyclischen Acylderivate des Kerns von Desacetyl cephalosporin-C-verbindungen verwendet werden, in dem man die Säureverbindung m einem gepufferten wässrigen Medium mit Citrusfrucht-Acetylesterase be handalt.
RI kann die Hydroxyl-oder Acetoxy-oder Pyridino- oder eine substitwierte Pyridinogruppe sein, wie sie durch Umsetzung von Cephalosporin C mit
Nicotin, Nicotinsäure, Isonicotinsäure, Nicotinamid, 2-Aminopyridiin, 2-Amino-6-methylpyridin,
2, 4, 6-Trimethylpyridin, 2-Hydroxymethylpyridin,
Sulfapyridin, 3-Hydroxypyridin,
Pyridin-2,3-dicarbonsäure, Chinolin,
Picolinsäure und dergleichen entsteht.
R kann ein Cyclobutyl-, Cyclopentyl-, α-Naphthyl-, ss-Naphthyl-, oder Adamantylrest oder ein Substitu- tionsprodukt dieser Reste mit einem oder mehreren Chlor-, Brom-, Fluor-, Jod-, Nitro-, Trifluoromethyl-, Cl-bis C4-Alkyl-oder Cl-bis Cs-Atkoxy-Substituenten am Ring sein.
Die Struktur und Darstellung des Adamantytrestes ist von Stetter in Angewandte Chemie 66, 217 (1954) beschrieben worden.
Die erfindungsgemäss hergestellten neuen Verbin- dungen sind insofern mit dem Cephalospori'n C verwandt, als sie den 5, 6-Dihydro-2H-1, 3-thiazinring mit dem ankondensierten ss-Lactamring in der 2, 3-Stellung enthalten, der chara'kteristisch für das Cephalosporin C ist. Im Gegensatz zum Cephallosporin C jedoch, das in der 7-Stellung die 5'-Amino-N'-adipamylgruppe aufweist, sind die erfindungsgemäss hergestellten Verbin dungen in der 7-Stellung durch eine Carbocyclocarbon säureamidgruppe gekennzeichnet.
Darüber hinaus sind die erfindungsgemäss hergestellten Verbindungen im Gegensatz zum Cephalosporin C, das nur eine verhältnismässig geringe antibakterielle Wirksamkeit besitzt, hochwirksame antibakterielle Mittel, die das Wachstum zahlreicher Typen von Mikroorganismen in den verschiedensten Umgebungen zu hemmen vermögen.
Wie aus den obigen Ausführungen hervorgeht, falot eine Vielzahl von verwandten Verbindungen, die die bicyclische Ringstruktur des Cephalosporins C aufwei- sen, jedoch in bezug auf die Substituenten variieren, unter die obige Formel.
Wie bei den Penicillinen, mit denen die erfindungsgemäss hergestellten Verbindungen bis zu einem gewissen Grade verwandt sind, können durch Kombination mit nichttoxischen pharmazeutischen annehmbaren Kationen oder Anionen zahlreiche Salze hergestellt werden.
Als Beispiele seien einige Typen von kationischen Salzen genannt, die aus den Verbindungen hergestellt werden können, die den Cephalosporin-C-Kern enthal'ten : wasserlösliche Salze, wie die Natrium-, Kalium-, Lithium-, Ammonium-und substituierten Ammoniumsalze, sowie weniger wasserlösliche Salze, wie die Calcium-, Barium-, Procain-, Chinin- und Dibenzyläthylen diaminsalze. Diejenigen Verbindungen, die den Cephal'o- sporin-CA-Kern enthalten, bilden keine kationischen, sondern anionische Salze, das heisst Additionssalze mit starken Säuren, wie Salzsäure, Bromwasserstoffsäure, Phosphorsäure, Schwefelsäure und ähnliche Säuren.
Die folgende Aufzählung erläutert in Verbindung g mit den weiter unten folgenden Beispielen die Vcrbin- dungstypen, die erfindungsgemäss erhältlich sind :
7-Cyclobutancarboxamidocephalosporansäure,
7-Cyclopentancarboxamidocephalosporansäure,
7-(2'-Chlorcyclobutancarboxamido) cephalosporansäure,
7-(2'-Bromcyclopentancarboxamido) cephalosporansäure,
7-(3'-Fluorcyclopentacarboxamido) cephalosporansäure,
7-(2'-Methylcyclopentancarboxamido) cephalosporansäure,
7-(3'-Methyloxycyclopentancarboxamido) cephalosporansäure, 7-α-Nitro-ss-naphthoylaminocephalosporansäure,
7-α-Trifluormethyl-ssp-naphthoylamino- cephalosporansäure, 7-Methylaclamantancarboxamido- cephalosporansäure und dergleichen, sowie die entsprechenden Cephalosporin-CA-Analoga.
Cephallospori'n C kann durch Kultivierung eines Ce phalosporin C bildenden Organismus in einem geeigneten Nährmedium hergestellt werden, wie in der britischen Patentschrift Nr. 810 196 beschrieben i ! st.
Cephaldsporin C lässt sich auch leicht in Verbindun- gen vom Cephalosporin-CA-Typ verwandeln, z. B. durch Kochen m wässriger Losung mit einem Überschuss an Pyridin, wie es in der belgischen Patentschrift Nummer 593 777 beschrieben wird.
Die Umsetzung ist ganz all- gemein auf Pyridin und substituierte Pyridine anwend- bar, von denen oben zahlreiche Beispiele genannt wurden, wobei die entsprechenden Derivate des Cephalo- sporin-CA-Typs entstehen, bei denen die Pyridino-oder substituierte Pyridinogruppe mit der Methylgruppe in der 5-Stellung des Thiazinringes verbunden ist und mit der Carboxylgruppe in der 4-S'tellung ein inneres Salz bildet.
Desacetylcephalosporin C lässt sich bequem durch mehrstündige Behandlung von Cephalosporin C mit Citrusfrucht-Acetylesterase in wässrigem Phosphatpuffer vom pH = 6, 5-7 nach dem Verfahren von Jansen, Jang und Mac Donnell. Archiv. Biochem. 1947-48, 1516, herstellen.
Aus den verschiedenen, auf diese Weise erhältlichen Cephalosporin-C-Verbindungen lässt sich der entspre- chende Kern leicht herstellen, wenn man die 5' Amino-N'-adipamyl-Seitenkette zwischen dem Amidstickstoff und der Amidcarbonylgruppe spaltet. So kann die 7-Aminocephalosporansäure nach dem in der belgischen Patentschrift Nr. 593 777 beschriebenen Verfahren durch längeres Digerieren von Cephalosporin C mit einer Mineralsäure in Abwesenheit von Licht her- gestellt werden.
Die oben definierten Verbindungen werden durch Acylieruhg der entsprechenden Cephalosporin-C-Kerne hergestellt, sei es nun der Kern des Cephalosporins C selbst, des Cephalosporins CA oder einer anderen Variante. Wahlweise können die Verbindungen vom Ce p'halosporiin-CA-und Desacetylcephalosporin-Typ auch erhalten werden, indem man die Umwandlungsverfahren der belgischen Patentschrift Nr. 593 777 und von Jan 'sen und Mitarbeitern auf di'e entsprechenden 7-Acyl aminocephalosporansäuren anwendet, um auf diese Weise die Verbindungen mit den entsprechenden Ker nens-zu erhalten.
Für die Acylierung der 7-Aminogruppe der Cephalosporan- Kerne - wie oben angegeben-können die üblichen Acylierungsverfahren sowie irgendwelche der ver schiedenen Typen von bekannten Acylierungsmitteln n verwendet werden, die eine Zusammensetzung aufweisen, die der gewünschten Seitenkette entspricht.
Ein bequemes Acylierungsmittel ist das entspre chende Carbocyclocarbonsäurechlarid oder-bromid.
Die Acylierung wiird in Wasser oder einem geeigneten organischen Lösungsmittel ausgeführt, vorzugsweise unter praktisch neutralen Bed'ingungen sowiie vorzugsweise bei verringerter Temperatur, das heisst oberhalb des Gefrierpunktes des Reaktionsgemisches und unterhalb von etwa 20 C. Bei einem typischen Verfahren wird die 7-Aminocephalosporansäure bzw. eines ihrer ge nannten Derivate zusammen mit einer ausreichenden Menge Natriumbicarbonat oder einer anderen geeigneten alkalischen Verbindung zur Beschleunigung der Lösung in wässrigem 50volumprozentigem Aceton gelöst, wobei die Konzentration der 7-Aminocephalospo- ransäure etwa 1-4 Gew. % beträgt.
Die Lösurig wird auf etwa 0-5 C abgekühlt, und es wird unter Rühren und
Kühlen eine Lösung des Acylierungsmittels in etwa
20 % igem Überschuss hinzugegeben. Falls der pH-Wert der Mischung nicht konstant bleibt, kann er durch Ein- leiten von Kohlendioxyd im Neutralbereich gehalten werden. Nach beendeter Zugabe des Acylierungsmittels wird das Reaktionsgemisch weiter gerührt und'auf Raumtemperatur erwärmen gelasse. Das Reaktions- gemisch wird sodann bis auT etwa pH = 2 angesäuert und mit einem organischen Lösungsmittel, wie Athyl- acetat, extrahiert.
Der Äthylacetat-Extrakt wird mit einer Base, die das gewünschte Kation des Endproduktes aufweist, auf etwa pH = 5, 5 eingestellt und mit Wasser extrahiert. Die wässrige Lösung wird abgetrennt und zur Trockne verdampft. Der Rückstand wird in der Mcmst- möglichen Menge Wasser aufgenommen und das gewünschte Produkt durch Zugabe eines grossen Uberschusses an Aceton und gegebenenfalls Äther gefällt.
Das dabei erhaltene kristalline Produkt wird abfiltriert, mit Aceton gewaschen und getrocknet.
Die Acylierung kann auch mit der entsprechenden Carbocyclocarbonsäure selbst in Verbindung mit einer äquimolaren Menge eines Carbodiimids, wie
N, N'-Diisopropylcarbodiimid,
N, N'-Dicyclohexylcarbodiimid,
N,N'-Bis-(p-dimethylamidophenyl)-carbodiimid,
N-Äthyl-N'-(4"-äthylmorpholinyl)-carbodiimid oder dergleichen, ausgeführt werden, wobei die Acylierung in solchen Fällen bei Normaltemperatur verläuft.
Wahlweise kann die Carbocyclocarbonsäure in das entsprechende Säureanhydirid, das Azid oder einen akti- vierten Ester verwandelt werden, wobei jedes dieser Derivate zur Erzielung der gewünschten AcyHerung verwendet werden kann. Weitere bekannte Mittel brauchen hier nicht aufgezahlt zu werden.
In vielen Fällen kann das Acylierungsmittel eines oder mehrere asymmetrische Kohlenstoffatome enthalten und daher in optisch aktiven Formen vorkommen. Bei der gewöhnlichen chemischen Synthese werden dliese Verbindungen gewöhnlich in Form des Racemats, das heisst eines äquimolaren Gemisches der optischen Antipoden, das keine optische Drehung zeigt, gewonnen.
Werden die genannten optischen Isomeren gewünscht, so kann das Acylierungsmittel in üblicher Weise gespalten werden, z. B. durch Umsetzung der freien Säure mit Cinchonin, Strychnih, Brucin und dergleichen, fraktio nierte Kristallisation der dia'stereoisomeren Salze und getrenntes Ansäuern der festen Phase und der fliissigen Phase, um die optischen Antipoden freizusetzen.
Die auf diese Weise erhaltenen freien Säuren können als solche für die Acylierung verwendet werden, und zwar vorzugsweise in Verbindung mit einem Carbodiimid, oder können nach üblichen Verfahren in das entspre- chende Säurehalogenid oder ein gemischtes Anhydrid umgewandelt werden, wobei natürliche extreme Bedin gungen vermieden werden müssen, damit keine Racemi sierung eintritt.
In der Literatur sind die erfindungsgemäss verwendeten AcyTierungsmittel zusammen mit Verfahren zu ihrer Herstellung beschrieben worden, und eine Anzahl von ihnen ist im Handel erhältlich. Sämtliche Verbindungen lassen sich nach bekannten Verfahren leicht herstellen.
Die Erfindung wird weiter durch die folgenden Ar beitsbeispiele erläutert, die jedoch nicht als Begrenzung des Erfindungsbereiches aufzufassen sind. Die in den Beispielen angegebenen chemischen Analysen wurden nach dem Verfahren von Ford, Analytical Chemistry 19, 1004 (1947), durchgeführt, das auf der quantitativen Bestimmung des ss-Lactamringes des Cephalosporin- Moleküls durch Umsetzung mit Hydroxylamin beruht.
Die antibiotíschen Wirksamkeiten wurden an Staphylo- coccus aureus 209 P un'ter Anwendung einer geeigneten Abwandlung der Papierscheibenplatten-Verfa'hren von Higgins und Mitarbeitern, Antibiotie & Chemitherapy 3, 50-54 (Januar 1953), und Loo und Mitarbeitern, Journal of Bacteriology 50, 701-709 (1945), bestimmt. Die pKa-Werte der Säuren wurden durch Titration in wässrigem 66 % igem Diemethylformamid bestimmt.
Beispiel 1 7-G-NaphthoylaminocephalosporaQsäure
1,0 g 7-Aminocephalosporansäure und 680 mg Natriumbicarbonat wurden in einem Gemisch von 50 cm3 Wasser und 40 cm3 Aceton gelöst. Die erhaltene Lösung wurde in einem Eisbad geküt undinnerhalb von 45 Min. mit einer Lösung von 670 mg α-Naphthoylchlorid in 10 cm3 Aceton versetzt, wonach noch weitere 1, 5 Std'. lang gerührt wurde. Aus dem Reaktionsgemisch wurde das Aceton abgezogen, und die zurückbleibende Lösung wurde mit 50 cm3 Äthylacetat überschiechtet und mit Salzsäure auf pH = 2, 0 angesäuert.
Die Afhylacetat- schicht wurde abgetrennt und bei pH = 5, 5 mit Wasser ausgeschüttelt, wobei zur pH-Einstellung verdünnte wässrige Kalilauge verwendet wurde. Der wässrige Extrakt wurde bis zu einem Sirup eingeengt, der durch Verdünnen mit Aceton fest wurde. Der Niederschlag wurde aus einem Gemisch von Methylalkohol und ho- propylalkohol umkristallisiert. Es wurden 250 mg 7-ce- Naphthoyllaminocephalosporansäure in Form des Ka liumisalzes gewonnen. Der pKa-Wert der Säure beträgt 4,85. Bei der chemischen Analyse entspricht 1 mg Produkt 1330 Einheiten Penicillin G. Die Verbindung zeigt UV-Maxima bei 219 und 269 m (# = 47 600 bzw.

Beispiel 2
7-ss-Naphthoylaminocephalosporansäure
1, 0 g 7-Aminocephalosporansäure und 1 g Natrium bicarbona't wurden in einem Gemisch von 50 cm3 Wasser und 40 cm3 Aceton gelös't. Die Lösung wurde in einem Eisbad gerührt und innerhalb von etwa 30 Min. mit einer Lösung von 670 mg ss-Naphthoylchlorid in 10 cm3 Aceton versetzt, wonach noch weiter 2-3 Std. lang in der Kälte gerührt wurde. Das Reaktionsgemisch wurde im Vakuum von dem Aceton befreit, und es wur- den 100 cm Äthylacetat zugegeben, gefolgt von 1normaler Salisäure, um das Gemisch auf pH = 2 anzusäuern. Die wässrige Phase wurde abgetrennt, mit 50 cm3 Athylacetat gewaschen und verworfen.
Di'e Äthylacetatextrakte wurden vereinigt und mit 50 cm3 Wasser gewaschen. Die gewaschene Athylacetatphase wurde mit 100 cm3 Wasser gerührt und mit wässriger 0, 5h Kalilauge auf pH = 5, 5 eingestellt. Der erhaltene wässrige Extrakt wurde abgotrennt und im Vakuum zur Trockene eingedampft. Der Rückstand wurde mit wässrigem Aceton verrieben, die erhaltene feste Masse abfiltriert und im Vakuum getrocknet. Es wurden 440 mg 7-ss-Naph t'hoyiaminocephalosporansäurein Form des Kaliumsal- zes erhalten. Der pKa-Wert der Säure beträgt 4, 90. Die Verbindung zeigt ein UV-Maximum bei 236 m, u (e= 46 300) mit Schultern bei 269 und 330 m .
Auf Grund der chemischen Analyse entsprach 1 mg Produkt 1510 Einheiten Penicillin G ; die biologische Wirksamkeit von 1 mg Produkt entsprach 43 Einheiten Penicillin G. Die Verbindung war gegenüber resistenten Staphylokokken in der Kulturbrühe wirksam und zeigte ferner gute Wirksamkeit gegeniiber Staphylococcus albus, BaciRus substilis und Sarcina lutea.
Beispiel 3
7-Adamantancarboxamidocephalosporansäure
Die 7-Adamantancarboxamidocephalosporansäure wurde in Form des Kaliumsalzes durch Umsetzung von 1, 0 g 7-Aminocephalosporansäure mit 1, 0 g Natriumbicarbonat und 700 mg Adamantancarbonsäurechlorid hergestellt, wobei im allgemeinen nach dem Verfahren von Beispiel 1 gearbeitet, das Reaktionsgemisch jedoch 12 Std. stehengelassen wurde. Die Ausbeute betrug nach Umkristallisieren aus Methanol-Äther 300 mg. Der pKa Wert der Säure'beträgt 4, 90. Die Verbindung zeigt ein UV-Maximum bei 260 m, (e = 7750). Auf Grund der chemischen Analyse entsprach 1 mg Produkt 1260 Ein- heiten Penicillin G.

The structure and representation of the adamanty residue has been described by Stetter in Angewandte Chemie 66, 217 (1954).
The novel compounds prepared according to the invention are related to the Cephalosporin C insofar as they contain the 5, 6-dihydro-2H-1, 3-thiazine ring with the condensed β-lactam ring in the 2,3-position, the chara 'Cteristic for the Cephalosporin C is. In contrast to cephallosporin C, however, which has the 5'-amino-N'-adipamyl group in the 7-position, the compounds prepared according to the invention are characterized in the 7-position by a carbocyclocarboxylic acid amide group.
In addition, in contrast to cephalosporin C, which has only a relatively low antibacterial activity, the compounds prepared according to the invention are highly effective antibacterial agents which are able to inhibit the growth of numerous types of microorganisms in the most varied of environments.
As can be seen from the above, a variety of related compounds, which have the bicyclic ring structure of cephalosporin C, but vary with respect to the substituents, fall under the above formula.
As with the penicillins, to which the compounds prepared according to the present invention are related to some extent, numerous salts can be prepared by combination with non-toxic pharmaceutically acceptable cations or anions.
Examples are some types of cationic salts which can be prepared from the compounds which contain the cephalosporin C nucleus: water-soluble salts, such as the sodium, potassium, lithium, ammonium and substituted ammonium salts, and less water-soluble salts, such as the calcium, barium, procaine, quinine and dibenzylethylene diamine salts. Those compounds which contain the Cephal'osporin-CA core do not form cationic, but anionic salts, that is to say addition salts with strong acids such as hydrochloric acid, hydrobromic acid, phosphoric acid, sulfuric acid and similar acids.
The following list, in conjunction with the examples below, explains the types of compounds that can be obtained according to the invention:
7-cyclobutane carboxamidocephalosporanic acid,
7-cyclopentanecarboxamidocephalosporanic acid,
7- (2'-chlorocyclobutane carboxamido) cephalosporanic acid,

The reaction can be applied quite generally to pyridine and substituted pyridines, numerous examples of which have been given above, the corresponding derivatives of the cephalosporin-CA type being formed in which the pyridino or substituted pyridino group is in contact with the methyl group the 5-position of the thiazine ring and forms an inner salt with the carboxyl group in the 4-S 'position.
Desacetylcephalosporin C can be conveniently prepared by treating cephalosporin C for several hours with citrus fruit acetylesterase in an aqueous phosphate buffer of pH 6, 5-7 according to the method of Jansen, Jang and Mac Donnell. Archive. Biochem. 1947-48, 1516.
The corresponding core can easily be prepared from the various cephalosporin C compounds obtainable in this way if the 5 'amino-N'-adipamyl side chain between the amide nitrogen and the amide carbonyl group is cleaved. For example, 7-aminocephalosporanic acid can be produced by the process described in Belgian patent specification No. 593 777 by digesting cephalosporin C for a long time with a mineral acid in the absence of light.
The compounds defined above are produced by acylation of the corresponding cephalosporin C nuclei, be it the nucleus of cephalosporin C itself, of cephalosporin CA or another variant. Alternatively, the Ce p'halosporin-CA and desacetylcephalosporin-type compounds can also be obtained by applying the conversion procedures of Belgian Patent Specification No. 593 777 and by Jan'sen et al. To the corresponding 7-acylaminocephalosporanic acids in this way the connections with the corresponding core are to be maintained.
For the acylation of the 7-amino group of the cephalosporan nuclei - as indicated above - the usual acylation processes and any of the various types of known acylating agents can be used, which have a composition which corresponds to the desired side chain.
A convenient acylating agent is the corresponding carbocyclocarboxylic acid chloride or bromide.
The acylation is carried out in water or a suitable organic solvent, preferably under practically neutral conditions and preferably at reduced temperature, i.e. above the freezing point of the reaction mixture and below about 20 C. In a typical process, the 7-aminocephalosporanic acid or one of their named derivatives together with a sufficient amount of sodium bicarbonate or another suitable alkaline compound to accelerate the dissolution in aqueous 50 percent by volume acetone, the concentration of 7-aminocephalosporanic acid being about 1-4% by weight.

Added 20% excess. If the pH value of the mixture does not remain constant, it can be kept in the neutral range by introducing carbon dioxide. After the addition of the acylating agent has ended, the reaction mixture is stirred further and allowed to warm to room temperature. The reaction mixture is then acidified to about pH = 2 and extracted with an organic solvent such as ethyl acetate.
The ethyl acetate extract is adjusted to about pH 5.5 with a base containing the desired cation of the end product and extracted with water. The aqueous solution is separated off and evaporated to dryness. The residue is taken up in the maximum possible amount of water and the desired product is precipitated by adding a large excess of acetone and, if necessary, ether.
The crystalline product obtained is filtered off, washed with acetone and dried.
The acylation can also be carried out with the corresponding carbocyclocarboxylic acid itself in conjunction with an equimolar amount of a carbodiimide such as
N, N'-diisopropylcarbodiimide,
N, N'-dicyclohexylcarbodiimide,
N, N'-bis (p-dimethylamidophenyl) -carbodiimide,
N-ethyl-N '- (4 "-ethylmorpholinyl) -carbodiimide or the like, the acylation in such cases proceeding at normal temperature.
Optionally, the carbocyclocarboxylic acid can be converted into the corresponding acid anhydiride, the azide or an activated ester, it being possible for any of these derivatives to be used to achieve the desired acylation. Further known means do not need to be listed here.
In many cases the acylating agent can contain one or more asymmetric carbon atoms and therefore exist in optically active forms. In the usual chemical synthesis these compounds are usually obtained in the form of the racemate, that is, an equimolar mixture of the optical antipodes which shows no optical rotation.
If the optical isomers mentioned are desired, the acylating agent can be cleaved in a customary manner, e.g. B. by reaction of the free acid with cinchonine, strychnid, brucine and the like, fractional crystallization of the dia'stereoisomeric salts and separate acidification of the solid phase and the liquid phase in order to set free the optical antipodes.
The free acids obtained in this way can be used as such for the acylation, preferably in conjunction with a carbodiimide, or can be converted into the corresponding acid halide or a mixed anhydride by customary processes, with extreme natural conditions being avoided must so that no racemization occurs.
In the literature, the acytating agents used in the present invention have been described along with processes for their preparation, and a number of them are commercially available. All of the compounds can be easily made using known methods.
The invention is further illustrated by the following work examples, which, however, are not to be construed as limiting the scope of the invention. The chemical analyzes given in the examples were carried out according to the method of Ford, Analytical Chemistry 19, 1004 (1947), which is based on the quantitative determination of the β-lactam ring of the cephalosporin molecule by reaction with hydroxylamine.
The antibiotic activities were demonstrated on Staphylococcus aureus 209 P using a suitable modification of the paper disk plate method by Higgins and coworkers, Antibiotie & Chemitherapy 3, 50-54 (January 1953), and Loo and coworkers, Journal of Bacteriology 50, 701-709 (1945), determined. The pKa values of the acids were determined by titration in aqueous 66% strength dimethylformamide.
Example 1 7G-NaphthoylaminocephalosporaQ acid
1.0 g of 7-aminocephalosporanic acid and 680 mg of sodium bicarbonate were dissolved in a mixture of 50 cm3 of water and 40 cm3 of acetone. The resulting solution was cooled in an ice bath and a solution of 670 mg of α-naphthoyl chloride in 10 cm 3 of acetone was added over a period of 45 minutes, followed by a further 1.5 hours. was stirred for a long time. The acetone was drawn off from the reaction mixture and the remaining solution was covered with 50 cm3 of ethyl acetate and acidified to pH = 2.0 with hydrochloric acid.
The ethyl acetate layer was separated off and shaken out with water at pH 5.5, using dilute aqueous potassium hydroxide solution to adjust the pH. The aqueous extract was concentrated to a syrup which solidified upon dilution with acetone. The precipitate was recrystallized from a mixture of methyl alcohol and homopropyl alcohol. 250 mg of 7-ce-naphthoyllaminocephalosporanic acid were obtained in the form of the potassium salt. The pKa value of the acid is 4.85. In the chemical analysis, 1 mg of product corresponds to 1330 units of penicillin G. The compound shows UV maxima at 219 and 269 m (# = 47 600 and

Example 2
7-ss-naphthoylaminocephalosporanic acid
1.0 g of 7-aminocephalosporanic acid and 1 g of sodium bicarbonate were dissolved in a mixture of 50 cm3 of water and 40 cm3 of acetone. The solution was stirred in an ice bath and a solution of 670 mg of ss-naphthoyl chloride in 10 cm 3 of acetone was added over the course of about 30 minutes, after which the mixture was stirred in the cold for a further 2-3 hours. The reaction mixture was freed of the acetone in vacuo, and 100 cm of ethyl acetate were added, followed by 1 normal salic acid in order to acidify the mixture to pH = 2. The aqueous phase was separated off, washed with 50 cm3 of ethyl acetate and discarded.
The ethyl acetate extracts were combined and washed with 50 cm3 of water. The washed ethyl acetate phase was stirred with 100 cm3 of water and adjusted to pH = 5.5 with aqueous 0.5 h of potassium hydroxide solution. The aqueous extract obtained was separated off and evaporated to dryness in vacuo. The residue was triturated with aqueous acetone, the solid mass obtained was filtered off and dried in vacuo. 440 mg of 7-ss-naphthoyiaminocephalosporanic acid in the form of the potassium salt were obtained. The pKa value of the acid is 4.90. The compound shows a UV maximum at 236 m, u (e = 46,300) with shoulders at 269 and 330 m.
On the basis of the chemical analysis, 1 mg of product corresponded to 1510 units of penicillin G; the biological activity of 1 mg of product corresponded to 43 units of penicillin G. The compound was active against resistant staphylococci in the culture broth and also showed good activity against Staphylococcus albus, BaciRus substilis and Sarcina lutea.
Example 3
7-adamantane carboxamidocephalosporanic acid
The 7-adamantanecarboxamidocephalosporanic acid was prepared in the form of the potassium salt by reacting 1.0 g of 7-aminocephalosporanic acid with 1.0 g of sodium bicarbonate and 700 mg of adamantanecarboxylic acid chloride, generally following the procedure of Example 1, but allowing the reaction mixture to stand for 12 hours . After recrystallization from methanol-ether, the yield was 300 mg. The pKa value of the acid is 4.90. The compound shows a UV maximum at 260 m (e = 7750). Based on the chemical analysis, 1 mg of product corresponded to 1260 units of penicillin G.
