erhalten wird. Diese Verbindung ist ein antigonadotro- phisches Mittel.
Im folgenden wird das erfindungsge- mässe Verfahren durch seine Anwendung auf das ge nannte 16-Halogen-17-oxo-Steroid als Beispiel beschrie- ben.
Falls das erfindungsgemässe Verfahren auf das 16-Halogen-17-oxoandrostan angewendet wird, kann die überführung durch die folgenden Formeln gezeigt werden:

Wie oben angeführt, kann eine Doppelbindung zwi schen den 4- und 5-Stellungen oder 5- und 6-Stellungen anwesend sein; das Wasserstoffatom in der 5-Stellung, falls die genannte Doppelbindung nicht vorhanden ist, besitzt eine a- oder ss-Konfiguration; und worin die Wellenlinie eine -a- oder ss-Konfiguration anzeigt.
In die sem Zusammenhang soll bemerkt werden, dass die oben genannten Formeln den wesentlichen, aufeinanderfol- genden Verlauf der einzelnen Stufen des Verfahrens an geben. Die Oxy- und/oder Oxoradikale, ausgenommen die Oxy- und Oxoradikale in der 17-Stellung, können in den genannten Verbindungen auch geschützt sein.
Mit andern Worten, ein Oxyradikal veranschaulicht ein freies Oxyradikal oder ein geschütztes Oxyradikal, wie z. B. ein acyliertes Oxyradikal, und das Oxoradikal steht für ein freies Oxo- oder für ein geschütztes Oxo- radikal, z.
B. ein ketalisiertes Oxoradikal. Auch soll ver standen werden, dass die Doppelbindung zwischen den 4- und 5-Stellungen, falls sie anwesend ist, in die 5- und 6-Stellungen verschoben werden kann, falls das Oxo- radikal in der 3-Stellung geschützt ist.

Das freie Oxoradikal in der 3-Stellung kann durch die im vorliegenden Verfahren verwendeten Reaktions- mittel auf unerwünschte Weise angegriffen werden. Deshalb soll zweckmässigerweise das genannte freie Oxo- radikal vorher geschützt werden. Im Gegensatz dazu ist der Schutz des freien Oxyradikals in der 3-Stellung un nötig.
Gleichwohl kann die acylierte Verbindung leich ter als die entsprechende freie Oxyverbindung erzeugt werden. In einem solchen Falle kann die acylierte Ver bindung als solche dem erfindungsgemässen Verfahren unterworfen werden. Somit können praktisch die folgen den 16-Halogen-17-oxo-androstane verwendet werden:

Die genannten 16-Halogen-17-oxo-androstane II sind bekannte Verbindungen und können gemäss einer üblichen Methode hergestellt werden [vgl. J. Fried und Mitarb.: US Patent Nrn. 2 857 403 und 2<B>831872;</B> B. Ellis und Mitarb.: J. Chem. Soc., 800 (1958); J. Fajkos und Mitarb.: Chemical Abstracts, 53, 4349 (1959)].
Das erfindungsgemässe Verfahren umfasst drei Stufen.
In der ersten Stufe wird das 16-Halogen-17-oxo- androstan II vorzugsweise mit einer Schwefel enthalten den organischen Säure, wie Thioalkansäure, z. B. Thio- essigsäure, Thiopropionsäure, Thiobuttersäure, und Alkyldithiokohlensäure, z. B. Methyldithiokohlensäure, Äthyldithiokohlensäure, Propyldithiokohlensäure, als freie Säure oder als Alkalimetallsalz derselben in einem geeigneten Medium, z. B.
Aceton, Äther, Dioxan, Tetra- hydrofuran, bei Zimmertemperatur während mehreren Stunden behandelt.
In der zweiten Stufe wird das so erhaltene 16-substi- tuierte Thioandrostan III mit einem Alkalimetallhydrid, z. B. Lithiumaluminiumhydrid, Lithiumborhydrid, Na- triumborhydrid, in einem geeigneten Medium, z.B. Äther, Dioxan, Tetrahydrofuran, Benzol, und falls notwendig, unter Erhitzen reduziert.
In der dritten Stufe wird das erhaltene 16-Mercapto- androstan IV mit einer Carbonylverbindung, wie Alde hyde und Ketone, z.
B. Acetaldehyd, Propionaldehyd, Butyraldehyd, Benzaldehyd, Aceton, Methyläthylketon, Diäthylketon, Methylbutylketon, Dipropylketon, Dibut- ylketon, Cyclohexanon, Benzophenon, in Gegenwart eines sauren Katalysators, z. B. p-Toluolsulfonsäure, Schwefelsäure, kondensiert.
Obschon das erfindungsgemässe Verfahren soeben von Stufe zu Stufe veranschaulicht wurde, können diese Stufen nacheinander, ohne Isolierung des Produktes in jeder Stufe, durchgeführt werden, was insbesondere in jenen Fällen von Vorteil ist, wo die erzeugten Zwischen produkte nicht stabil sind. Zum Beispiel ist das 16-Mer- captoandrostan IV verhältnismässig unstabil und soll zweckmässigerweise der Reaktion in der nachfolgenden Stufe, ohne Isolierung aus dem Reaktionsgemisch, unter worfen werden.
Erfindungsgemäss erzeugte Endprodukte sind vor zugsweise die cylischen Acetale I, von welchen die fol genden als Beispiele genannt werden können: 3ss,17ss-Dioxy-16ss-mercapto-5 (6)-androsten-16,17- acetonid, 3,ss-Acetoxy-16ss-mercapto- 17ss-oxy-5 (6)-androsten- 16,17-acetonid, 3,ss-Propoxy-16ss-mercapto-17ss-oxy-5(6)-androsten- 16,17-acetonid, 3ss,17.ss-Dioxy-16ss-mercapto-5(6)
-androsten- diäthylketon-16,17-cyclisches-acetal, 3a,17ss-Dioxy-16ss-mercapto-5(6)-androsten- 16,17-acetonid, 3 a-Acetoxy-16ss-mercapto- 17ss-oxy-5 (6)-androsten- benzaldehyd-16,17-cyclisches-acetal, 3,6,17ss -Dioxy-16ss-mercapto-5a-androstan-16,17- acetonid, 3a-Butoxy-16ss-mercapto-17@ oxy-5ss-androstan- cyclohexanon-16,17-cyclisches-acetal,

Von diesen Verbindungen sind die p4-3-Oxo-cycli- schen Acetale Ia als antigonadotrophischeMittel nützlich. Bei oraler Verabreichung an eine Maus vom Körperge wicht von 15-17 g, 40 Tage alt, bei einer Dosis von 1 mg, zeigt z.B. 3-Oxo-16ss-mercapto-17ss-oxy-4-androsten-16,
17-acetonid eine antigonadotrophische Wirksamkeit mit dem Inhibierungsverhältnis von 81,82 %, welches unge- fähr gleichwertig ist jenem bis jetzt bekannten anti- gonadotrophischen Steroid,
dem 17a-Äthinyl-19-nor- testosteron. Die anderen A4-3-Oxo-cyclischen Acetale Ia weisen ebenfalls ähnliche Wirksamkeiten auf.
Die cycli- schen Acetale I, welche nicht in den 04-3-Oxo-cych- schen Acetalen Ia miteingeschlossen sind, können leicht in die cyclischen Acetale la in üblicher Weise, konver tiert werden.
<I>Beispiel 1</I> a) 9,03 g 3,17-Dioxo-16ss-brom-4-androsten [vgl. J. Fajkos und Mitarb.: Chemical Abstracts, 53, 4349 (1959)] werden mit 200 ml wasserfreiem Benzol, 15 ml Äthylenglykol und 450 mg p-Toluolsulfonsäure vereinigt und die so erhaltene Lösung während 16 Stunden bei Rückfluss erhitzt. Nach Kühlen wird das Reaktionsge misch mit wässriger Natriumcarbonatlösung neutralisiert und mit Benzol geschüttelt.
Die Benzolschicht wird mit Wasser gewaschen und das Lösungsmittel unter einem reduzierten Druck entfernt. Der so erhaltene Rückstand wird aus Äther kristallisiert und aus Dichlormethan- Aceton unikristallisiert und liefert .6,83 g 3,3-Äthylendi- oxy 1@6ss-brom-17-oxo-5(6)-androsten in. Form von Kri stallen mit F = 204-206' C.

Das erhaltene Eluat von 904 mg wird aus Methanol kristallisiert und aus Chloroform- Athanol umkristallisiert und man erhält 644 mg 3,3- Äthylendioxy-16,ss-äthoxythiocarbonylthio-17-oxo-5 (6)- androsten,
in Form von Nadeln, mit F = 208-210 C (Zers.).

Nach Entfernen des Lösungs- mittels wird der Rückstand von 406 mg mit Äther ge waschen und liefert 3:00 mg 3,3-Äthylendioxy-16ss-mer- capto-17.ss-oxy-5(6)-androsten als rohe Kristalle mit F = <B>207-211</B> C.
d) 10O0 mg 3,3 Äthylendioxy-16ss-mercapto-17ss-oxy- 5(6)-androsten werden mit 10 mg p-Toluolsulfonsäure in 5 ml Aceton während 5 Stunden bei Rückfluss erhitzt.
Das Reaktionsgemisch wird dann mit Wasser versetzt und die ausgefallenen Kristalle werden filtriert. Nach Lösen dieser Kristalle in Benzol-Petroläther (1:
1) wer den .dieselben auf 3 g Tonerde chromatographiert. Die eluierte Substanz wird aus Aceton-Methanol kristallisiert und liefert 80 mg 3-Oxo-16ss-mercapto-17ss-oxy 4-andro- sten-16,17-acetonid in Form von Blättchen mit F = 228-230 C.

<I>Beispiel 3</I> 3,686 g 3,3 Äthylendioxy-,16fl=mercapto-17fl-oxy- 5(6)-androsten, hergestellt wie im Beispiel 1 a), b) und c), werden mit 120 ml Diäthylketon und 300 mg p Toluolsulfonsäure während 13 Stunden unter Rückfluss behandelt. Danach wird das Reaktionsgemisch mit Benz ol geschüttelt, die Benzolschicht mit wässerigem Na- triumcarbonat und dann mit Wasser gewaschen und schliesslich getrocknet.
Nach Entfernen des Lösungs- mittels wird der Rückstand auf Tonerde chromatogra- phiert. Die mit Petroläther-Benzol (1:1, 1:
2) eluierte Substanz wird aus Petroläther kristallisiert und aus Meth anol umkristallisiert und liefert 1,85 g 3-Oxo-16,ss-mer- capto-17ss-oxy-4 -androsten-diäthylketon-16,17-cycli- sches-acetal, in Form von Nadeln, mit F = 144-146 C. fa]D28 + 40,0 T 2 (C = 1,043 Chloroform).

Das so hergestellte 3ss,17ss-Dioxy-16ss-mercapto-5 (6)- androsten-16,17-acetonid kann in 3-Oxo-16ss-mercapto- 17ss-oxy-4-androsten-16,17-acetonid, gemäss einem üb lichen Oxydationsverfahren, z. B. mit Hilfe der Oppen- auer-Oxydation, übergeführt werden.
<I>Beispiel 8</I> 3,17-Dioxo-16ss-brom-5a-androstan [vgl. J. Fajkos und Mitarb.: Chemical Abstracts, 53, 4349 (1959)] wird der Ketalisierung, wie im Beispiel 1 a), unterworfen.
Das so erhaltene 3,3-Äthylendioxy-16ss-brom-17-oxo-5a- androstan wird mit Kaliumthioacetat, wie in Beispiel 2 b), umgesetzt und liefert 3,3-Äthylendioxy-16,8-acetyl- thio-17-oxo-5a-androstan, wonach die letztere Verbin dung mit Lithiumaluminiumhydrid reduziert wird;
nach anschliessender Reaktion mit Aceton in Gegenwart von p-Toluolsulfonsäure, wie in Beispiel 1 c) und d), erhält man 3-Oxo-16ss-mercapto-17ss-oxy-5a-androstan-16,17- acetonid in Nadeln mit F = 210-212 C.
Das so erzeugte 3-Oxo-16ss-mercapto-17ss-oxy-5a- androstan-16,17-acetonid weist eine Natriumretentions- aktivität sowie eine antigonadotrophische Wirksamkeit auf.
<I>Beispiel 9</I> Zu einer Suspension von 3,079 g 3ss-Acetoxy-16ss- brom-17-oxo-5a-androstan [vgl. J. Die Ätherschicht wird über wasserfreiem Natriumsulfat getrocknet und nach Abdampfen des ,Äthers erhält man den Rückstand, der 3 -Methoxy-16ss-mercapto-17ss -oxy-1, 3,5 (10) -östratrien enthält.
c) Zu dem oben erhaltenen Rückstand werden 20 mg p Toluolsulfonsäure und 10 ml Aceton zugesetzt und das so erhaltene Gemisch während 4 Stunden unter Feuchtigkeitsabschluss bei Rückfluss behandelt. Die nie dergeschlagenen weissen Nadeln werden heiss filtriert. Zum Filtrat gibt man 30 ml Wasser zu und filtriert den Niederschlag, welcher nach Trocknen 146,6 mg rohe Kristalle ergibt.
Diese rohen Kristalle werden auf 7 g Tonerde, unter Verwendung von Benzol-Petroläther 1:1 als Lösungsmittel, chromatographiert. Die so erhaltenen 102,7 mg an rohem 3-Methoxy-16ss-mercapto-17ss-oxy- 1,3,5(10)-östratrien-16,17-acetonid werden aus Aceton umkristallisiert und ergeben 7 mg weisser Nadeln mit F = 169-170 C.

1) kristallisiert und liefert 18 g 3 -Methoxy-166B-acetylthio-17-oxo-1,3,5(10) -östratrien in weissen Kristallen mit F = 184-187 C.

In the second stage, the 16-substituted thioandrostane III obtained in this way is treated with an alkali metal hydride, e.g. Lithium aluminum hydride, lithium borohydride, sodium borohydride, in a suitable medium, e.g. Ether, dioxane, tetrahydrofuran, benzene, and if necessary, reduced with heating.

Although the process according to the invention has just been illustrated from step to step, these steps can be carried out one after the other without isolating the product in each step, which is particularly advantageous in those cases where the intermediate products produced are not stable. For example, the 16-mercaptoandrostane IV is relatively unstable and should expediently be subjected to the reaction in the subsequent stage without being isolated from the reaction mixture.

Androstene diethyl ketone 16,17-cyclic acetal, 3,3, ethylenedioxy-16ss-mercapto-17ss-oxy-5a-androstan- 16,17-acetonide, 3-oxo-16ss-mercapto-17ss-oxy-5a -androstan- 16,17-acetonide, etc.
Of these compounds, the p4-3 oxo-cyclic acetals Ia are useful as antigonadotropic agents. When administered orally to a mouse weighing 15-17 g, 40 days old, at a dose of 1 mg, e.g. 3-Oxo-16ss-mercapto-17ss-oxy-4-androsten-16,
17-acetonide has an antigonadotrophic activity with an inhibition ratio of 81.82%, which is roughly equivalent to the antigonadotrophic steroid known to date,
the 17a-ethynyl-19-nor-testosterone. The other A4-3-oxocyclic acetals Ia also have similar activities.
The cyclic acetals I, which are not included in the 04-3-oxo-cychic acetals Ia, can easily be converted into the cyclic acetals 1a in the usual way.
<I> Example 1 </I> a) 9.03 g of 3,17-dioxo-16ss-bromo-4-androstene [cf. J. Fajkos et al .: Chemical Abstracts, 53, 4349 (1959)] are combined with 200 ml of anhydrous benzene, 15 ml of ethylene glycol and 450 mg of p-toluenesulfonic acid and the resulting solution is refluxed for 16 hours. After cooling, the reaction mixture is neutralized with aqueous sodium carbonate solution and shaken with benzene.
The benzene layer is washed with water and the solvent is removed under a reduced pressure. The residue obtained in this way is crystallized from ether and unicrystallized from dichloromethane-acetone and yields 6.83 g of 3,3-ethylenedioxy 1 @ 6ss-bromo-17-oxo-5 (6) -androsten in the form of crystals with F = 204-206 'C.
