Verfahren zur Herstellung von neuen heterocyclischen Verbindungen
Die vorliegende Erfindung betrifft ein Verfahren zur Herstellung von neuen 1-Aza-thioxanthen=Deri- vaten der Formel I,

worin R1 ein Wasserstoff- oder ein Halogenatom, R2 eine niedere Alkylgruppe bedeutet und x für den Wert 1 oder 2 steht.
Erfindungsgemäss gelangt man zu den neuen Verbindungen der allgemeinen Formel I, indem man 1 -Aza-thioxanthen-Derivate der Formel II

oxydiert.
In den Ausgangssubstanzen der Formel II kann R1 z. B. ein Wasserstoff-, ein Chlor- oder ein Bromatom bedeuten; R2 kann für die Methyl-, Äthyl-, Propyl-, Isopropyl- oder die Butylgruppe stehen.
Die Oxydation des Schwefelatoms gelingt mit geeigneten Oxydationsmitteln, wie z. B. mit Wasserstoffperoxyd, organischen Persäuren, anorganischen Persäuren oder ihren Salzen, z. B. mit Alkalimetaperjodaten.
Die Ausführung des Verfahrens gestaltet sich beispielsweise wie folgt:
Endprodukte der Formel I, worin x für 1 steht, werden erhalten, indem man die essigsaure Lösung der Ausgangssubstanz (1 Mol), wie z. B. 9-[1-Me- thyl-piperidyliden-(4)] 1 -aza-thioxanthen, mit einem Mol einer verdünnten wässerigen Natriummetaperjodatlösung während 3 bis 5 Std. bei Raumtemperatur versetzt. Zur Vervollständigung der Reaktion lässt man das Gemisch noch 3 Tage bei Raumtemperatur stehen. Nach Entfernen von ausgefallenem Natriumjodat und nach Alkalischmachen mit Ammoniak oder einem Alkalihydroxyd wird das Endprodukt mit einem indifferenten organischen Lösungsmittel, vorzugsweise mit Methylenchlorid, extrahiert, nach bekannten Methoden daraus isoliert und gereinigt.
Anstatt mit einer wässerigen Natriummetaperjodatlösung kann die Oxydation auch mit organischen Persäuren, z. B. mit Perbenzoesäure, oder mit Wasserstoffperoxyd bei Raumtemperatur oder leicht erhöhter Temperatur ausgeführt werden, wobei jeweils ein Mol des Oxydationsmittels verwendet wird.
Endprodukte der Formel I, worin x für 2 steht, werden beispielsweise erhalten, indem man das Ausgangsprodukt der Formel II in Eisessig nach Zugabe einer starken Mineralsäure, z. B. konz. Schwefel säure, bei Raum- oder erhöhter Temperatur mit zwei Mol Wasserstoffperoxyd behandelt. Nach mehrstündiger Reaktionszeit wird das Gemisch eingeengt, der Rückstand in Wasser aufgenommen, mit Ammoniak oder mit einer Alkalilauge bis zur alkalischen Reaktion versetzt, die ausgeschiedene Base in einem indifferenten organischen Lösungsmittel, vorzugsweise mit Methylenchlorid, aufgenommen und nach Auswaschen mit Wasser das Lösungsmittel wieder eingedampft. Die Basen können direkt kristallisiert und/oder mit organischen oder anorganischen Säuren in ein geeignetes Salz übergeführt werden.
Die erfindungsgemäss hergestellten neuen l-Azathioxanthen-Derivate der Formel I sind basische, bei Raumtemperatur kristallisierte Verbindungen, die mit organischen oder anorganischen Säuren beständige kristallisierte Salze bilden. Solche Salze sind beispielsweise die Hydrochloride, Hydrobromide, Sulfate, Fumarate, Maleinate, Malate, Acetate, Tartrate.
Die erfindungsgemäss hergestellten neuen l-Azathioxanthen-Derivate besitzen mannigfaltige therapeutisch verwertbare pharmakodynamische Eigenschaften. So zeichnen sie sich einerseits durch broncholytische und antitussive, anderseits aber auch durch histaminhemmende, narkosenpotenzierende, anticholinergische und sedative Wirkung aus. Die neuen Verbindungen sollen deshalb sowohl als Antiasthmatica, Broncholytica und Antitussiva als auch als Antihistaminica in der Therapie Verwendung finden, besonders bei Rhinitis allergica, Asthma bronchiale, Bronchitis und bei den verschiedensten allergischen Erkrankungen.
Als Heilmittel können die neuen Verbindungen bzw. ihre wasserlöslichen, physiologisch verträglichen Säureadditionssalze allein oder in entsprechenden Arzneiformen für enterale oder parenterale Verabreichung verwendet werden. Zwecks Herstellung geeigneter Arzneiformen werden die Wirkstoffe mit anorganischen oder organischen, pharmakologisch indifferenten Hilfsstoffen verarbeitet; man verwendet z. B. für Tabletten und Dragees: Milchzucker, Stärke, Talk, Stearinsäure usw.; für Injektionspräparate: Wasser, Alkohole, Glycerin, pflanzliche Öle und dergleichen; für Suppositorien: Natürliche oder gehärtete Öle und Wachse und anderes mehr. Ausserdem können die Zubereitungen geeignete Konservierungs-, Stabilisierungsmittel, Netzmittel, Lösungsvermittler, Süss- und Farbstoffe, Aromantien usw. enthalten.
Ausgangsprodukte der Formel II, worin R1 Halogen und R2 eine niedere Alkylgruppe bedeuten, erhält man aus 6-Halogen-l-aza-thioxanthenen und l-Alkyl-4-halogenpiperidinen durch Grignard Reaktion, Hydrolyse und anschliessende Wasserabspaltung.
In den nachfolgenden Beispielen, welche die Ausführung des Verfahrens erläutern, die Erfindung aber in keiner Weise einschränken sollen, erfolgen alle
Temperaturangaben in Celsiusgraden, ohne Korrek turen.
Beispiel 1 9-[1 -Methyl-pipendyllden-(4)J4 -aza- thioxanthen-l O-oxyd
Zu einer Lösung von 5,88 g 9-[1-Methyl-piper idyliden-(4)]- 1 -aza-thioxanthen (Smp. 154-156 aus
Aceton) in 20 cm3 Wasser und 1,2 cm3 Eisessig lässt man unter gutem Rühren während 4 Std.
45 cm3 0,Smolare Nat:iummetaperjodatlösung zu tropfen und rührt anschliessend 3 Tage bei Raumtemperatur. Darauf wird ausgefallenes Natriumjodat abfiltriert, das Filtrat mit Kalilauge alkalisch gestellt und mehrmals mit Methylenchlorid extrahiert. Nach
Trocknen der vereinigten Extrakte über Magnesium sulfat und Abdampfen des Lösungsmittels wird der
Rückstand in Aceton gelöst, worauf das 9-[1-Me thyl-piperSdyliden- (4)]-1-aza-thioxanthen-10-oxyd kristallisiert. Es schmilzt nach zweimaligem Um kristallisieren aus Aceton bei 146-147 .
Beispiel 2 6-Chlor-9- -methyl-piperidyliden-(4)j- 1 -aza-thioxanthen-1 O-oxyd
Die Lösung von 2,7 g 6-Chlor-9-[1-methyl-pi peridyliden-(4)] - 1 -aza-thioxanthen in 40 cm3 Wasser und 0,56 cm3 Eis essig wird unter gutem Rühren bei Raumtemperatur innerhalb von 75 Min. mit der Lösung von 2, 03 g Natriummetaperjodat in 20 cm3 Wasser versetzt. Nach 128stündigem Rühren bei Raumtemperatur und 4stündigem Rühren bei 500 stellt man das Reaktionsgemisch mit konz.
Ammoniumhydroxydlösung alkalisch und extrahiert mehrmals mit Methylenchlorid. Nach Trocknen der vereinigten Extrakte über Natriumsulfat und Abdampfen des Lösungsmittels wird der Rückstand zweimal aus-2ithanol/Aceton (etwa 1: 2) umkristallisiert. Das 6- Chlor - 9-[1-methyi-piperidyliden-(4)] 1-aza-thioxanthen - 10- oxyd schmilzt bei 179-180 unter schwacher Zersetzung.
Das als Ausgangsmaterial verwendete 6-Chlor-9 [1 -methyl-piperidyliden-(4)]- 1 -aza-thioxanthen wird wie folgt hergestellt: 2, 43 g mit Jod aktivierte Magnesiumspäne werden mit 10 cm3 abs. Tetrahydrofuran überschichtet und mit 0,2 cm3 Äthylenbromid versetzt. Sobald die Reaktion in Gang kommt, lässt man die Lösung von 14,7 g 1-Methyl4-chlor-piperidin in 25 cm3 abs. Tetrahydrofuran zutropfen, wobei das Reaktionsgemisch ständig siedet.
Anschliessend erhitzt man noch etwa 2 Std. unter Rückfluss, bis sich das Magnesium zum grössten Teil gelöst hat. Nach dem Abkühlen auf 200 versetzt man das Gemisch portionenweise mit 12,4 g fein zerriebenem 6-Chlor-1-aza-thioxanthon und rührt noch 30 Min. bei Raumtemperatur. Das Reaktionsgemisch wird sodann in 300 cm3 10proz.
Ammoniunchloridlösung gegossen und dreimal mit Methylenchlorid extrahiert. Die vereinigten, über Kaliumcarbonat getrockneten Extrakte werden einge dampft und der Rückstand in Aceton gelöst, woraus das 6-Chlor-9- [1-methyl-piperidyl-(4)]-1-aza4hlo- xanthydrol kristallisiert. Smp. 166-1670 nach zweimaligem Umkristallisieren aus Äthanol.
3,75 g der erhaltenen Verbindung werden mit einem Gemisch aus 22,5 cm3 konz. Schwefelsäure und 7,5 cm3 Wasser 15 Min. auf 1400 erwärmt. Anschliessend giesst man das Reaktionsgemisch in Eiswasser und extrahiert die mit Natronlauge alkalisch gestellte Lösung mit Methylenchlorid. Nach dem Trocken des Methylenchloridextraktes über Kaliumcarbonat und Abdampfen des Lösungsmittels wird der Rückstand aus Aceton kristallisiert. 6-Chlor 9- [1 - methyl - piperidyliden - (4)]-l-aza-thioxanthen schmilzt bei 110-1120.
Beispiel 3 9-[1 -Methyl-piperidyliden-(4)1 1 -aza-thioxanthen-1 0, O-dioxyd
Das Gemisch von 9,5 g 9-[1-Methyl-piperidyli den - (4)] - 1 - aza-thioxanthen (Smp. 154-1560 nach Sintern ab 152"), 140 cm3 Eisessig und 38 cm3 konz. Schwefelsäure wird auf 600 erwärmt und unter Rühren tropfenweise mit der Lösung von 17,1 cm3 38,5proz. wässeriger Wasserstoffperoxydlösung in 17 cm3 Eisessig versetzt. Nach 16stündigem Rühren bei 600 engt man das Reaktionsgemisch unter einem Druck von 15 mm Hg ein. Anschliessend löst man den Rückstand in Wasser, stellt die wässerige Lösung mit konz. Ammoniumhydroxydlösung alkalisch und extrahiert mehrmals mit Methylenchlorid.
Die vereinigten Extrakte werden mit Wasser neutral gewaschen, über Natriumsulfat getrocknet und unter einem Druck von 15 mm Hg zur Trockne verdampft. Nach zweimaligem Umkristallisieren aus Methanol schmilzt das 9-C1- Methyl-piperidyliden -(4)1-l-aza-thio- xanthen-1 0-dioxyd bei 188-1890 (Zers.).

The procedure is carried out, for example, as follows:

The new compounds or their water-soluble, physiologically tolerable acid addition salts can be used as medicaments alone or in appropriate medicinal forms for enteral or parenteral administration. For the purpose of producing suitable dosage forms, the active ingredients are processed with inorganic or organic, pharmacologically indifferent auxiliaries; one uses z. B. for tablets and dragees: lactose, starch, talc, stearic acid, etc .; for injectables: water, alcohols, glycerine, vegetable oils and the like; for suppositories: natural or hydrogenated oils and waxes and more. In addition, the preparations can contain suitable preservatives, stabilizers, wetting agents, solubilizers, sweeteners, colorants, flavorings, etc.
Starting materials of the formula II, in which R1 is halogen and R2 is a lower alkyl group, are obtained from 6-halo-1-aza-thioxanthenes and 1-alkyl-4-halopiperidines by a Grignard reaction, hydrolysis and subsequent dehydration.
