Verfahren zur Herstellung von Yohimbe-Alkaloiden
Die vorliegende Erfindung betrifft ein Verfahren zur Herstellung neuer organischer Verbindungen, insbesondere neuer am Ring E substituierter Derivate von Yohimbe-Alkaloiden der folgenden allgemeinen Formel

worin R die Cyan-, Carboxyl-, Methyl-oder eine niedere Carbalkoxygruppe und Ri ein Wasserstoffatom oder eine niedere Alkanoylgnuppe bedeuten, sowie der Säureadditionssalze dieser Verbindungen. Geeig- nete niedere Alkylgruppen, die beim vorliegenden erfinidungsgemässen Verfahren in Betracht gezogen werden, sind diejenigen mit 1 bis 6 Kohlenstoffatomen, wobei Methyl und Athyl bevorzugt sind.
Geeig- nete niedere Carbalkoxygruppen sind Carbomethoxy, Carbäthoxy und dergl. Die wertvollen nicht-toxischen pharmazeutisch verträglichen Säureadditionssalze dieser neuen Derivate können, ebenfalls hergestellt werden. Typische Säureadditionssalze sind die Hydrochloride, Phosphate, Sulfate, Citrate und der. gl.
Das erfindungsgemässe Verfahren ist gekenn- zeichnet durch Reduktion des entsprechenden 17-Keto-yohimbealkaloids. Man kann gewünschtenfalls das so gebildete Produkt mit einem niederen Alkanoylierungsmittel behan, deln und ferner gewünschtenfalls die Säureadditionssalze herstellen.
Die Reduktion kann nach jedem beliebigen, an sich bekannten Verfahren zur Reduktion von Ketogruppen, das die anderen, am umgesetzten Yohimbekern sitzenden Gruppen nicht beeinflusst, bewirkt werden.
Die erfindungsgemäss herstellbaren Verbindungen sind im allgemeinen weisse, kristalline Festsubstanzen, deren freie Basen in organischen Lösungsmittel, wie niederen Alkanolen, Chloroform, Aceton, Athylacetat, Dimethylformamid und Ider., löslich sind, und deren Salze in polaren Lösungsmitteln, wie beispielsweise Wasser und niederen Alkanolen, lös- lich sind.
Die erfindungsgamäss herstellbaren Verbindun- gen sind wertvolle Depressoren für das Zentralnervensystem von niederer Toxizität vom Typ des Muskelrelaxans und Tranquillizers und können oral oder parenteral verabreicht werden. Bei dieser Verabrei chung zeigen sie diese Wirkung in Mengen von 25 bis etwa 350 mg/kg Körpergewicht. Die erfindungsge mäss herstellbaren Verbindungen können als solche verwendet werden, werden jedoch vorzugsweise in Form der nicht toxischen Säureadditionssalze angewandt, die leicht durch Behandeln mit einem Äquivalent einer Säure, wie beispielsweise Salzsäure, Phosphorsäure, Schwefelsäure, Zitronensäure und dergl., hergestellt werden.
Das 18a-Cyanyohimban-17-on kann durch Behandeln von Yohimban- [18, 17-d]-isoxazol mit einem basischen Reagenz, wie beispielsweise einem Alkalialkylat, Natriumhydrid oder schwachem wässrigem Alkali, hergestellt werden. Diese Reaktion wird vorzugsweise in einem niederen Alkanollösungsmittel bei Raumtemperatur über einen Zeitraum von mehreren Stunden durchgeführt. Das 18a-Cynyohimban17-on kann auch durch Behandeln von 18-Hydroxymethylenyohimban-17-on mit O, N-Bis- (trifluor acetyl)-hydroxylamin hergestellt werden,.
Diese Reaktion wirdvorzugsweise in einem inerten Lösungsmit- tel, wie beispielsweise Benzol, bei einer Temperatur von 50 bis 100 C über einen Zeitraum von 15 Minuten bis zu mehreren Stunden durchgeführt.
Yohimbano- [18, 17-d]-isoxazol kann leicht durch einige Minuten langes Behandeln von 18-Hydroxy- methylenyohimban-17-on mit Hydroxylamin-hy- drochlorid in einem Lösungsmittel, wie Eisessig, bei 100 C hergestellt werden. 18-Hydroxymethylenyo himban-17-on kann in guter Ausbeute durch Behandeln von Yohimban-17-on mit einem niederen-Alkylformiat, wie beispielsweise Methyl-oder Athylfor- miat, in Gegenwart einer geeigneten Base, wie beispielsweise einem Alkalialkylat, Natriumhydrid, Natriumamid und dergl., hergestellt werden. Yohimban-17-on wurde von Witkop, Ann. 554, 83 (1943) beschrieben.
Das 18a-Methylyohimban-17-on kann durch Entschwefeln des 18- (niedrig Alkyl)-thiomethylenyo- himban-17-ons mit Raney-Nickel hergestellt werden.
Diese Reaktion wird vorzugsweise bei Rückflusstem- peratur eines interten Lösungsmittels, wie beispielsweise Aceton, über einen Zeitraum von mehreren Stunden durchgeführt. Das 18- (niedrig Alkyl)-thio methylenyohimban-17-on kann durch Behandeln von
18-Hydroxymethylenyohimban-17-on mit einem geeigneten niederen Alkylmercaptan in Gegenwart eines Säurekatalysators, wie beispielsweise p-Toluolsulfonsäure, Schwefelsäure, Essigsäure und dergl., hergestellt werden. Das bevorzugte Verfahren besteht in der Behandlung von 18-Hydroxymethylenyohimban-17-on mit einem niederen Alkylmercaptan in Gegenwart von Eisessig und wasserfreiem Magnesi umsulfat.
Das 18a-Cyanyohimban-17-on und das 18a-Me thylyohimban-17-on können durch Verwendung eines Reduktionsmittels, wie beispielsweise eines Alkaliborhydrids, von Wasserstoff und einem Katalysator, einem Metall und einer Base und dergl., zum entspre chenden 18a-Cyan-17-hydroxyyohimban und 18a-Methyl-17-hydroxyyohimban reduziert werden.
Diese Reaktion wird vorzugsweise in einem niederen Alkanollösungsmittel bei Raumtemperatur über einen Zeitraum von mehreren Stunden durchgeführt.
Die 18a-Cyan-17-hydroxyyohimbane können , durch herkömmliche Hydrolyse mit wässrigem Natriumhydroxyd zu den entsprechenden 17-Hydroxyyo- himban-18a-carbonsäuren hydrolysiert werden. Die
17-Hydroxyyohimban-18a-carbonsäuren können dann nach herkömlichen Verfahren, wie beispielsweise durch Behandeln mit einem Diazoalkan, Behandeln mit einem niederen Alkanol in Gegenwart einer Mineralsäure oder Behandeln mit einem niede ren Alkanol in Gegenwart von N, N-Dicyclohexylcar- bodiimid, verestert werden.
Die 17-Hydroxyyohimban-18a-carbonsäureester können auch durch Reduktion des entsprechenden 17-Oxoyohimban-18a-carbonsäureesters mit einem
Reduktionsmittel, wie beispielsweise einem Alkaliborhydrid, Wasserstoff und einem Katalysator, einem Metall und einer Base und dergl., hergestellt werden.
Diese Reaktion wird vorzugsweise in einem niederen Alkanollösungsmittel bei Raumtemperatur über einen Zeitraum von mehreren Stunden durchgeführt. Die 17-Oxoyohimban-18a-car. bonsäure kann leicht durch Behandeln von Yohimban-17-on mit einem niederen Alkylmetallcarbonat, wie beispielsweise Methylmagnesiumcarbonat, in einem inerten Lösungsmittel, wie beispielsweise Dimethylformamid, bei 120 bis 130 C über einen Zeitraum von mehreren Stunden hergestellt werden.
Die so hergestellte 17-Oxoyohimban-18a-carbonsäure kana dann nach herkömmlichen Verfahren, wie beispielsweise durch Behandeln mit einem Diazoalkan, Behandeln mit einem niederen Alkanol in Gegenwart einer Mineralsäure oder Behandeln mit einem niederen Alkanol in Gegenwart von N, N'-Dicyclohexylcarbodiimid, ver estert werden.
Die 18a-Cyan-17-hydroxyyohimbane, 18a-Me- thyl-17-hydroxyohimbane, 17-Hdroxyyohimban- 18a-carbonsäuren und die niedrig Alkyl-17-hydr- oxyyohimban-18a-carboxylate können alle an der Hydroxylgruppe in 17-Stellung nach herkömmlichen Verfahren acyliert werden. So ergibt beispielsweise Behandeln dieser 17-Hydroxyderivate mit einem niedrigen Alkanoylsäurehalogensid oder mit einem niedrigen Alkanoylsäureanhydrid die entsprechenden 17- (niedrig Alkanoyloxy)-derivate.
Die folgenden Beispiele dienen zur näheren Erläuterung der Erfindung, ohne sie zu beschränken.
Beispiel 1
Herstellung von 1 8-Hydroxymethylenyohimban-1 7-on
Zu einer gekühlten Mischung von 10,0 g Yohimban-17-on, 10,0 g Natriummethylat und 300 cm3 mit Natrium getrocknetem Benzol wurden 14 cm3 Äthyl- formiat zu, gefügt. Die Mischung wurde unter Stick stoff bei Raumtemperatur 20 Stunden lang gerührt und auf ein Gemisch von 300g Eis unld 200cm3 Wasser gegossen. Die organische Schicht wurde abgetrennt und mit dreimal je 100cm3 0, 1 n-Natrium- hydroxyd gewaschen. Die basischen Waschwässer und die wässrige Schicht wurden vereinigt und in der Kälte mit Essigsäure neutralisiert.
Filtrieren ergab
9, 4 g 18-Hydroxymethylenyohimban-17-on-hemy- hydrat als lohfarbene Kristalle vom Smp. = 140-147 C. Beim Stehen, in der Kälte über Nacht ergaben die Mutterlaugen weitere 1, 8 g Kristalle.
Umkristallisieren aus Methanol ergab farblose
Nadeln vom Smp. = 207-210 C (Zersetzung), die bei 145-148 C zu einem Glas sinterten.
Beispiel 2
Herstellung von 18-Hydroxymethylenyohimban-17-on
Eine Mischung von 5, 0 g Yohimban-17-on, 5, 0 g
Natriummethylat, 150 cms trockenem Peroxyd- freiem Dioxan und 7 cmss Äthylformiat wurde bei Raumtemperatur unter Stickstoff 21 Stunden lang gerührt. Die Mischung wurde mit Essigsäure neutra- lisiert und fast t zur Trockne konzentriert. Der Rück- stand wurde aus wässrigem Methanol umkristallisiert und ergab 5, 3 g 18-Hydroxymethylenyohimban-
17-on-hemyhydrat als lohfarbene Kristalle vom Smp. = 207-210 (Zers.), die bei 145 bis 154 C zu einem Glas sinterten.
Beispiel 3
Herstellung von 1 7-Oxoyohimban-1 8a-carbonitril
Eine Suspension von 0,35 g 18-Hydroxymethylenyohimban-17-on in 18 cm3 Benzol wurde teilweise destilliert, um, die Feuchtigkeit zu entfernen. Zur erhaltenen Suspension wurden 0, 161. oms trockenes Pyridin, 0, 34g O, N-Bis-(trifluoracetyl)-hydroxylam, in und 1 cm3 trockenes Aceton hinzugefügt, und die erhaltene Mischung mittels eines Ölbades 2 Stunden lang auf 75 bis 80 C erhitzt. Das Lösungsmittel wurde unter vermindertem Druck entfernt, was einen dunkelbraunen Gummi ergab, der zwischen 6 cm3 gesättigtem Natriumbicarbonat und 5 cm3 Chloroform verteilt wurde.
Die wässrige Schicht wurde weiter mit Chloroform gewaschen, und die-vereinigten organischen Schichten wurden über Magnesiumsulfat getrocknet und verdampft. Der erhaltene braune Feststoff (0, 227g) wurde nacheinander aus Methanol-Chloroform, Aceton-Petroläther (Siedebereich 20 bis 40 C) und Methanol-Wasser umkristallisiert, was Kristalle von 17-Oxoyohimban-18a-carbonitril vom Smp. = 265 bis 272 C (Zers.) ergab.
Beispiel 4
Herstellung von Yohimbano- [17, 18-c]-isoxazol-hydrochlorid und Yohimbano- [18, 17-d]-isoxazol-hydrochlorid
Eine Mischung von 1, 0 g 18-Hydroxymethylenyohimban-17-on, 0, 225 g Hydroxylaminhydrochlo- rid und 15 cm. s Eisessig wurde in einem ) lbad 6 Minuten lang auf 100 C erhitzt. Die Mischung wurde abgekühlt und filtriert, was 0, 43 g farblose Nadeln ergab. Umkristallisieren aus wässrigem Methanol ergab 0, 148 g einer Mischung der Hydrochloride von Yohimbano- [17, 18-c]-isoxazol und Yohim bano- [18, 17-d]-isoxazol als farblose Nadeln vom Smp. = 310 bis 315 C (Zers.), die 1/4 Mol Kristall- wasser enthielten.
Beispiel S
Herstellung von 17-Oxoyohimban-17a-carbonitril
Eine Mischung von 0, 36g Yohimbano [17, 18-c]-isoxazol und Yohimbano- [18, 17-d]-isoxazol wurde zu einer Lösung von 0, 115g Natrium in 10 cm3 Athanol zugesetzt. Nach Stehen über Nacht wurde dia Mischung unter Stickstoff 3 Stunden lang zum Rückfluss gebracht. Die Mischung wurde mit Essigsäure neutralisiert und mit Wasser verdünnt, was 0, 281 g lohfarbener Kristalle vom Smp. = 263 bis 268 C (Zers.) ergab.
Reinigen einer Probe durch Chromatographie an Silicagel ergab 17-Oxoyohim ban-18a-aarbonitril mit 1/4 Mol Kristallwasser als loh- farbene Nadeln vom Smp. = 278 bis 280 C (Zers.).
Beispiel 6
Herstellung von 18-n-Butylthiomethylenyohimban-17-on
Zu einer Mischung von 0,663 g 18-Hydroxymethylenyohimban-17-on, 2,0 g magnesiumsulfat und 5, 0 cm3 1-Butanthiol wurden 10 cm3 Essigsäure zugefügt. Die Mischung wurde bei Raumtemperatur 20 Stunden lang geriihrt und filtriert. Das Filtrat wurde zwischen 50cm3 ChloroformXund 100cm3 4n-Natriumhydroxyd verteilt. Nach weiterer Chloroformextraktion wurden die vereinigten organischen Schichten über Magnesiumsulfat getrocknet und konzentriert, was 0, 66 g weisslicher Kristalle von 18-n-Bu- tylthiomethylenyohimban-17-on ergab. Umkristalli- sieren aus Aceton ergab farblose Kristalle vom Smp.
= 219 bis 222 C (Zers.).
Beispiel 7
Herstellung von 1 8a-Methylyohimban-1 7-on
Eine Mischung von 10, 0 Raney-Nickel, 0, 80 g 18-n-Butylthiomethylen-yohilmbanzl7-on und 80cm3 Aceton wurde 12 Stunden lang zum Rückfluss gebracht und dann durch Diatomeenerde filtriert. Der Filterkuchen wurde gut mit Aceton gewaschen, und die vereinigten Filtrate wurden unter vermindertem Druck konzentriert, was 0, 50 g nahezu farbloser Kristalle ergab. Eine Chloroformlösung des Produkts wurde an Aluminiumoxyd chromatographiert.
Verdampfen der dritten 25 cma-Portion des Eluats, gefolgt von Umkristallisieren des Rückstands aus Methanol-Chloroform ergab 18a-Methylyohimban-17- on, das 1/4 Mol Kristallwasser enthielt, als farblose Nadeln vom Smp. = 297 bis 302 C (Zers.).

Herstellung von
17a-Hydroxyyohimban-18a-carbonitril
Herstellung von
17α-Hydroxyyohimban-18α-carbonitril und 17ss-Hydroxyyohimban-18α-carbonitril
Zu einer gekühlten Lösung von 0,35 g Natriumborhydrid in 50 cm3 Äthanol wurden 2, 0 g 17-Oxoyohimban 18α-carbonitril zugefügt. Die Mischung wurde bei Raumtemperatur 4 Stunden lang gerührt. Das überschüssige Natriumborhydrid wurde mit Essigsäure zersetzt und das Lösungsmittel unter vermindertem Druck entfernt. Der hinterbleibende blassgelbe Feststoff wurde zwischen Chloroform und Wasser verteilt, und das chloroformlösliche Produkt wurde an Aluminiumoxyd chromatographiert.
Eluie ren mit Chloroform/Aceton (3 : 2) ergab 0, 46 g eines Feststoffes, der beim Kristallisieren aus wässrigem Methanol 0, 135 g 17a-Hydroxyyohimban-18a-carbo- nitril als lohfarbene Kristalle vom Smp. = 260 bis s 265 C (Zers.) ergab, die 1/, Mol Kristallwasser enthielten.
Weiteres Eluieren der Säulen mit Chloroform/ Methanol (99 : 1) ergab 0, 534g eines Feststoffes, der nach Kristallisieren aus Methanol 0, 36 g 17ss-Hydroxyyohimban-18a-carbonitril als weisse flockige Nadeln vom Smp. = 247 bis 250 C (Zers.) ergab, die 1/4 Mol Kristallwasser enthielten.
Beispiel 9
Herstellung von 17a-Hydroxyyohimban-18a-carbonsäure und 17a-Hydroxyyohimban-18a-carbonsäuremethylester
Eine Mischung von 0, 10g 17a-Hydroxyyohim- ban-18a-carbonitril, 4, 0cm3 Äthanol, l, 0cm3Wasser und 0, 25 g Natriumhydroxyd wurde 21 Stunden lang zum Rückfluss gebracht. Das Lösungsmittel wurde entfernt, und der Rückstand wurde in 5, 0 om3 Wasser gelöst und mit Essigsäure neutralisiert. Der Feststoff wurde durch Filtrieren entfernt und mit 3, 0 cm3 Wasser gewaschen und getrocknet. Man erhielt 0, 08 g rohe 17a-Hydroxyyohimban-18a-carbonsäure.
Eine zweite Ausbeute von Säure (0, 011 g) wurde aus den Mutterlaugen beim Stehen erhalten. Die verei nigten Ausbeuten an roher 17c !-Hydroxy-L8a-car- bonsäure wurden in Methanol suspendiert und mit überschüssigem Diazomethan in Äther behandelt.
Zersetzen des überschüssigen Diazomethans mit Essigsäure und Verdampfen des Lösungsmittels unter vermindertem Druck ergab 0, 092g eines Glases.
Chromatographieren des Glases an Aluminiumoxyd mit Chloroform als Elutionsmittel ergab 0, 012 g
17α-Hydroxyyohimban-18α-carbonsäurementhylester in lohfanbenen Nadeln vom Smp. = 206 bis 211 C (Zers.) mit einem Mol Kristallmethanol.
Beispiel 10
Herstellung von
17ss-Hydroxyyohimban-18α-carbonsäure und 1 7ss-Hydroxyyohimban-1 8a-carbonsäuremethylester
Eine Mischung von 0, 092 g 17ss-Hydroxyyohim ban-l8a-carbonitril, 4, 0 cm3 Äthanol, 1,0 cm3 Wasser und 0, 22g Natriumhydroxyd wurde 18 Stunden lang zum Rückfluss gebracht. Die Mischung wurde auf ca. 1, 5 cm3 eingeengt und der sich abscheidende Feststoff durch Zugabe von 4 ; 0 cms Wasser gelöst.
Die Lösung wurde mit Essigsäure neutralisiert und der abgeschiedene Feststoff durch Filtrieren entfernt und getrocknet, was 0, 13 g rohe 17A-Hydroxyyohim- ban-18a-carbonsäure ergab. Diese wurde in Methanol suspendiert und mit überschüssigem Diazomethan in Äther behandelt. Zersetzen des überschüssi- gen Diazomethans mit Essigsäure und Konzentrieren der Lösung unter vermindertem Druck ergab 0, 13 g eines braunen Glases. Das Glas wurde aus wässrigem Methanol kristallisiert, was 0, 03 g 17ss-Hydroxyyo- himban-18a-carbonsäuremethylester als weisse Kristalle vom Smp. = I45 bis 148 C mit 1/4 Mol Kristallwasser ergab.
Beispiel 11
Herstellung von 17-Oxoyohimban-18α-carbonsäurehydrochlorid
Eine Mischung von 2, 0g Yohimban-17-on und 25 cm3 einer etwa 2m-Lösung von Methylmagnesi umcarbonat in Dimethylformamid wurde 3 Stunden lang unter Stickstoff auf 120 bis 130 C erhitzt und gerührt. Die Mischung wurde in einem Eisbad gekühlt und langsam unter Rühren zu einer Mischung zugefügt, die in einem Eissalzbad gekühlt war. Der abgeschiedene Feststoff wurde filtriert und mit 2 cm3 kalter 6n-Salzsäure gewaschen. Trocknen über Phosphorpentoxyd im Vakuum bei Raumtemperatur ergab 2, 71 g lohfarbener Kristalle vom Smp. = 292 bis 294 C (Zers.) (beim Einsetzen in ein auf 288 C vorerhitztes Ölbad).
Ein Anteil von 1, 91 g dieses Feststoffes wurde mit einer Mischung von 650 cm3 von 50 g Eis und 30 cm3 konzentrierter Salzsäure Methanol und 600 cm3 Äther verrieben, und die Suspension wurde filtriert, was 0, 86g 17-Oxoyohim- iban, 18-carbonsäurehydrochlorid als weisse Kristalle vom Smp. = 314 bis 317 C (Zers.) (nach Einsetzen in ein auf 310 C vorerhitztes Ölbad) ergab.
Beispiel 12
Herstellung von 17-Oxoyohimban-18α-carbonsäuresulfat
0, 294 g Yohimban-17-on wurden mit 4, 0 cm3 einer Lösung von Methylmagnesiumcarbonat in Dimethylformamid, wie in Beispiel 11 beschrieben, carboxyliert. Die gekühlte Reaktionsmischung wurde zu einer Mischung von 12 g Eis und 2 cm3 98 %iger Schwefelsäure gefügt und der abgeschiedene Feststoff abfiltriert und mit Wasser und 1 cm3 Äthanol gewaschen. Mehrstündiges Trocknen über Phosphorpentoxyd unter Vakuum bei Raumtemperatur ergab 0, 364 g 17-Oxoyohimban-18a-carbonsäuresulfat als blassgelbe Kristalle vom Smp. = 230 bis 260 C (Gasentwicklung bei 260 C).
Beispiel 13
Herstellung von 17-Oxoyohimban-18a-carbonsauremethylester
Zu einer Suspension von 0, 50 g 17-Oxoyohimban 18a-carbonsäurehydrochlorid in 50 cm3 eiskaltem Methanol wurden 50 cm3 diazomethanhaltiger eiskalter Äther (Diazomethan hergestellt aus 4, 0 g Nitrosomethylharnstoff und 8,0 cm3 40 %iger Kalilauge und über KOH+Plätzchen getrocknet) zugefügt.
Die Mischung wurde bei Raumtemperatur 10 Minuten lang stehengelassen und der Uberschuss an Di- azomethan durch tropfenweise Zugabe von Eisessig zersetzt. Das Lösungsmittel wurde unter Vakuum entfernt, was 0, 598 g eines hygroskopischen Glases ergab. Das Glas wurde in 15 cm3 siedendem Methanol gelöst und tropfenweise Wasser zugegeben, bis sich weisse Kristalle abschieden. Kühlen und Filtrieren ergaben 0, 201 g 17-Oxoyohimban-18a-carbonsäure-methylester als weisse Kristalle vom Smp. = 186 bis 188 C (Zers.) (nach Einsetzen in ein auf 180 C vorerhitztes ölbad).
Eine zweite Ausbeute an Kristallen (0, 0234 g) wurde durch Verdünnen des Filtrates mit Wasser erhalten. Extraktion des Filtrats mit 5 x 10 cm3 Chloroform und Verdampfen des Ex traktes im Vakuum ergab ein Glas. Dieses Glas wurde in 2, 0cm3 Methanol gelöst und die Lösung mit 1, 0 cm3 Wasser verdünnt. Abkühlen und Filtrie- ren ergaben eine dritte Ausbeute (0, 102 g) an Kristallen, Smp. = 181 bis 183 C (Zers.) (nach Einsetzen in ein auf 180 C vorerhitztes Ölbad0
Die drei Ausbeuten wurden vereinigt, in 45 cm3 Methanol gelöst und die Lösung mit 3, 0 cm3 Wasser verdünnt.

Herstellung von 17-Oxoyohimban-18a-carbonsauremethylester
Eine Mischung von 0, 35 g 17-Oxoyohimban-18α- carbonsäure-hydrochlorid, 40 cm3 Methanol und 1Q cm3 mit trockenem Chlorwasserstoff gesättigtem Methanol wurde 4 Stunden lang zum Rückfluss gebracht. Die wässrige Schicht wurde mit zusätzlichen 10 cm3 Chloroform extrahiert. Verdampfen der vereinigten Chloroformextrakte unter Vakuum ergab 0, 098 g eines Glases, das in 5 cm3 heissem Methanol gelöst wurde. Die Lösung wurde mit Wasser verdünnt, bis sich Kristalle abschieden. Abkühlen und Filtrieren ergaben 0, 073 g weisser Nadeln vom Smp. = 248 bis 255 C (Zers.) Aherstellung von 17-Oxoyohimban-18α-carbonsäurementhylester
Zu einer eis-kalten M-ischung von 10, 0, g rohem 17-Oxoyohimban-l8a-carbonsäurehydrochlorid und
100 cm3 Dimethylformamid wurden 100 cni Methanol und 11, 0 g N,N'-Dicyclohexylcarbodiimid zugesetzt. Die Mischung wurde bei Raumtemperatur 20 Stunden lang gerührt, mit 10lcm3 Wasser und 3 cm' Essigsäure behandelt und eine weitere Stunde lang gerührt. Der ausgefallene Niederschlag wurde durch Filtrieren entfernt und das Filtrat unter Vakuum auf 100 cms konzentriert.
Die restliche Lösung wurde mit 50 cm3 gesättigter Natriumbicarbonatlösung behandelt und 4 x mit je 50 cm3 Chloroform extrahiert.
Die Extrakte wurden mit 3 x je 100 cm3 Wasser gewaschen, über Natriumsulfat getrocknet und unter Vakuum konzentriert. Der Rückstand wurde mit 50''/oigem wässrigem Methanol erwärmt, und nach Abkühlen und Filtrieren erhlielt man 6, 0 g 17-Oxoyo himban-18a-carbonsäuremethylester, der mit etwas Yohimban-17-on undl N, N'çDicyclohexyl-harnstoff verunreinigt war. Die Trennung dieser Bestandteile wurde durch Chromatographie an Silicagel und Umkristallisieren aus wässrigem Methanol durchgeführt.

Herstellung von 17a-Hydroxyyohimban-18a-carbonsduremethylester 17α-Hydroxyyohimban-18α-carbonsäuremethylester 17ss-Hydroxyyohimban-18α-carbonsäuremethylester
Zu einer gekühltern Lösung von 2,38 g Natriumborhydri, d in 300 om3 Methanol wurden 11, 6 g 17-Oxoyohimban-18a-carbonsäuremethylester innerhalb von 10 Minuten zugefügt. Die Mischung wurde unter Stickstoff 50 Minuten lang bei 0 C gerührt.

Weiteres Eluieren der Säule mit Chloroform Methanol (99 : 1) ergab 2, 25g eines Feststoffes, der nach Umkristallisieren aus wässrigem Methanol 0, 896 g 17ss-Hydroxyyohimban-18α-carbonsäureme- thylester als weisse Kristalle vom Smp. = 136 bis 140 C ergab, die 1/4 Mol Kristallwasser enthielten.
Beispiel 17
Herstellung von 17a-, Hydroxyyohimban-18a-carbonsauremethylester
Zu einer gekühlten Lösung von 5, 0kg Natrium- borhydrid in 300 cm3 Methanol wurden 10, Og 17-Oxoyohimban-18a-carbonsäuremethylester in kleinen Anteilen innerhalb von 5 Minuten zugefügt.
Die Mischung wurde abgekühlt und unter Stickstoff 11/2 Stunden lang gerührt, sorgfältig durch Zugabe von Essigsäure neutralisiert und unter vermindertem Druck verdampft. Herstellung von 17a-Acetyloxyyohimban-18a-carbonsäuremethylester
Eine Mischung von 0, 225 g 17a-Hydroxyyohim- ban-18a-carbonsäuremethylester, 4, 0 cm3 trockenem Pyridin und 2, Ocm3 Essigsäureanhydrid wurde bei Raumtemperatur 66 Stunden lang stehengelassen.

The deposited solid was filtered and washed with 2 cm 3 of cold 6N hydrochloric acid. Drying over phosphorus pentoxide in vacuo at room temperature gave 2.71 g of tan crystals with a melting point of 292 to 294 ° C. (decomposition) (when placed in an oil bath preheated to 288 ° C.).
A portion of 1.91 g of this solid was triturated with a mixture of 650 cm3 of 50 g of ice and 30 cm3 of concentrated hydrochloric acid, methanol and 600 cm3 of ether, and the suspension was filtered, yielding 0.86 g of 17-oxoyohimiban, 18- carboxylic acid hydrochloride as white crystals with a melting point of 314 to 317 ° C. (decomposition) (after placing in an oil bath preheated to 310 ° C.).
Example 12
Preparation of 17-oxoyohimbane-18α-carboxylic acid sulfate
0.294 g of yohimban-17-one were carboxylated as described in Example 11 with 4.0 cm 3 of a solution of methyl magnesium carbonate in dimethylformamide. The cooled reaction mixture was added to a mixture of 12 g of ice and 2 cm3 of 98% strength sulfuric acid and the deposited solid was filtered off and washed with water and 1 cm3 of ethanol. Drying over phosphorus pentoxide for several hours at room temperature gave 0.364 g of 17-oxoyohimbane-18a-carboxylic acid sulfate as pale yellow crystals with a melting point of 230 to 260 ° C. (evolution of gas at 260 ° C.).
Example 13
Preparation of 17-oxoyohimban-18a-carboxylic acid methyl ester
50 cm3 of diazomethane-containing ice-cold ether (diazomethane prepared from 4.0 g of nitrosomethylurea and 8.0 cm3 of 40% potassium hydroxide solution and dried over KOH + cookies) were added to a suspension of 0.50 g of 17-oxoyohimbane 18a-carboxylic acid hydrochloride in 50 cm3 of ice-cold methanol added.
The mixture was left to stand at room temperature for 10 minutes and the excess of diazomethane was decomposed by the dropwise addition of glacial acetic acid. The solvent was removed in vacuo to give 0.598 g of a hygroscopic glass. The glass was dissolved in 15 cm3 of boiling methanol and water was added dropwise until white crystals separated out. Cooling and filtering gave 0.1201 g of 17-oxoyohimbane-18a-carboxylic acid methyl ester as white crystals with a melting point of 186 to 188 ° C. (decomposition) (after placing in an oil bath preheated to 180 ° C.).
A second crop of crystals (0.0234 g) was obtained by diluting the filtrate with water. Extraction of the filtrate with 5 × 10 cm 3 of chloroform and evaporation of the extract in vacuo gave a glass. This glass was dissolved in 2.0 cm3 of methanol and the solution was diluted with 1.0 cm3 of water. Cooling and filtering gave a third crop (0.12 g) of crystals, mp = 181 to 183 ° C (dec.) (After placing in an oil bath preheated to 180 ° C)
The three yields were combined, dissolved in 45 cm3 of methanol and the solution was diluted with 3.0 cm3 of water.
Cooling and filtering gave 0.211 g of methyl 17-oxoyohimbane-18a-carboxylate as white crystals with a melting point of 186 to 188 ° C. (decomposition) (after placing in an oil bath preheated to 180 ° C.).
Example 14
Preparation of 17-oxoyohimban-18a-carboxylic acid methyl ester
A mixture of 0.35 g of 17-oxoyohimbane-18α-carboxylic acid hydrochloride, 40 cm 3 of methanol and 10 cm 3 of methanol saturated with dry hydrogen chloride was refluxed for 4 hours. The solution was concentrated in vacuo to approximately 5 cm3 and the remaining solution was diluted with ether. Cooling and filtering gave 0.23 g of white crystals, which were washed with ether. Some of the crystals (0.110g0 were distributed between 10 cm3 of saturated sodium bicarbonate solution and 10 cm3 of chloroform.
The aqueous layer was extracted with an additional 10 cm 3 of chloroform. Evaporation of the combined chloroform extracts in vacuo gave 0.098 g of a glass which was dissolved in 5 cm3 of hot methanol. The solution was diluted with water until crystals separated out. Cooling and filtering gave 0.073 g of white needles with a melting point of 248 to 255 ° C. (decomposition) (after insertion into an oil bath preheated to 180 ° C.). This solid was identified by infrared analysis as a mixture of yohimban-17-one and 17-oxoyohimban-18a-carboxylic acid methyl ester. This mixture was separated into the components by chromatography on silica gel.
Example 15
A preparation of 17-oxoyohimbane-18α-carboxylic acid menthyl ester
To an ice-cold mixture of 10.0 g of crude 17-oxoyohimbane-18a-carboxylic acid hydrochloride and
100 cm 3 of dimethylformamide were added to 100 ml of methanol and 11.0 g of N, N'-dicyclohexylcarbodiimide. The mixture was stirred at room temperature for 20 hours, treated with 10 ml of water and 3 cm of acetic acid, and stirred for an additional hour. The deposited precipitate was removed by filtration and the filtrate was concentrated to 100 cms in vacuo.
The remaining solution was treated with 50 cm3 of saturated sodium bicarbonate solution and extracted 4 times with 50 cm3 of chloroform each time.
The extracts were washed with 3 × 100 cm3 of water, dried over sodium sulfate and concentrated under vacuum. The residue was heated with 50% aqueous methanol, and after cooling and filtering, 6.0 g of methyl 17-oxyohimban-18a-carboxylate were obtained, which was contaminated with a little yohimban-17-one and 1 N, N'çdicyclohexylurea was. The separation of these components was carried out by chromatography on silica gel and recrystallization from aqueous methanol.
Example 16
Preparation of 17a-hydroxyyohimbane-18a-carboxylic acid methyl ester, 17α-hydroxyyohimbane-18α-carboxylic acid methyl ester, 17ss-hydroxyyohimbane-18α-carboxylic acid methyl ester
11.6 g of methyl 17-oxoyohimbane-18a-carboxylate were added over the course of 10 minutes to a cooled solution of 2.38 g of sodium borohydride in 300 om3 of methanol. The mixture was stirred at 0 ° C. under nitrogen for 50 minutes.
The excess sodium borohydride was decomposed with acetic acid and the solvent removed under reduced pressure to give a yellow glass.
The glass was distributed between 100 cm3 of chloroform and 100 cm3 of 2.5% sodium bicarbonate solution.
The chloroform layer was separated, washed with 100 cm3 of 2.5 "/% sodium bicarbonate solution, dried over magnesium sulfate and concentrated under reduced pressure to a glass. The glass was chromatographed on 900 g of neutral alumina. Elution with chloroform gave 1.45 g of solid which after crystallization from methanol, 0.705 g of 17a-hydroxyyohimbane-18a-carboxylate resulted as whitish needles with a melting point of 212 to 216 ° C. (decomp.).
Further elution of the column with chloroform methanol (99: 1) gave 2.25 g of a solid which, after recrystallization from aqueous methanol, 0.896 g of methyl 17ss-hydroxyyohimbane-18α-carboxylate as white crystals with a melting point of 136 to 140.degree which contained 1/4 mol of water of crystallization.
Example 17
Production of 17a-, hydroxyyohimban-18a-carboxylic acid methyl ester
To a cooled solution of 5.0 kg of sodium borohydride in 300 cm3 of methanol, 10.0 g of methyl 17-oxoyohimbane-18a-carboxylate were added in small portions over the course of 5 minutes.
The mixture was cooled and stirred under nitrogen for 11/2 hours, carefully neutralized by the addition of acetic acid, and evaporated under reduced pressure. The residue was partitioned between chloroform and 2.5 l / 0 strength sodium bicarbonate. The aqueous phase was washed with additional chloroform and the combined organic layers dried over magnesium sulfate and evaporated to give 6.1 g of a partially crystalline yellow solid. This mixture was separated into its components by crystallization followed by chromatography on neutral alumina.
Elution with chloroform and crystallization from methanol gave 17α-hydroxyyohimbane-18α-carboxylic acid methyl ester as colorless needles with a melting point of 210 to 214 ° C. (decomposition) which contained 1 mol of crystal methanol.
Example 18
Production of 17a-acetyloxyyohimban-18a-carboxylic acid methyl ester
A mixture of 0.225 g of methyl 17a-hydroxyyohimban-18a-carboxylate, 4.0 cm3 of dry pyridine and 2.0 cm3 of acetic anhydride was left to stand at room temperature for 66 hours.
The dark brown mixture was concentrated to a viscous mass under reduced pressure. The residue wound in a mixture of 8.0 cm3 was cooled and dissolved with concentrated ammonia water and 2.0 cm3 methanol, and the solution was made basic with hydroxide. The solid obtained was removed by filtration and washed with 10 om3 water. After partially air drying, the solid was dissolved in methanol, treated with activated charcoal, filtered and the filtrate concentrated to a glass under reduced pressure.
The glass was recrystallized from aqueous methanol, which gave 0.08 g of 17a-acetyloxyyohimbanel-8a-carbonic acid methyl ester as tan crystals with a melting point of 232 to 234 ° C. (dec.). Recrystallization from aqueous methanol gave 0.05 g of tan crystals with a melting point of 236 to 238 ° C. (dec.).
Example 19
Production of 17ss-Acetyloxyyohlmban-18a-carboxylic acid methyl ester
A mixture of 0.250 g of 17ss-hydroxyyohimban-18α-carboxylic acid menthyl ester, 4.0 cm3 of anhydrous pyridine and 2.0 cm3 of acetic anhydride was left to stand at room temperature for 66 hours. The dark mixture was then concentrated to a viscous mass under reduced pressure. The residue was dissolved in a mixture of 8.0 cm3 of water and 2.0 cm3 of methanol and the solution was made basic with concentrated ammonium hydroxide. The deposited solid was removed by filtration and washed with water.
The solid was dissolved in methanol, treated with activated charcoal, filtered and the filtrate concentrated under reduced pressure to give 0.225 g of a glass. The glass was crystallized from aqueous methanol, which gave 0.118 g of methyl 17ss-acetyloxyyohimbane-18a-carboxylate as tan crystals with a melting point of 122 to 125.degree. Recrystallization from aqueous methanol gave 0.079 g of tan crystals with a melting point of 125 to 129 C.
