Verfahren zur Herstellung von Acetyl-salicylsäurediäthylaminoäthylesterShydrochlorid
Acetylsalicylsäure ist eine relativ instabile Verbindung und die Herstellung ihrer Ester, insbesondere in hoher Reinheit, ist mit grossen Schwierigkeiten verbunden.
Ein bekanntes Verfahren zur Herstellung von Acetylsalicylsäure-diäthylaminoäthylester-hydrochlo- rid (s. Z. Jerzmanowska und Z. Orchowicz, Acta Polon. Pharm. 13, 11-25 [1956]) besteht in der Umwandlung von Acetylsalicylsäure in ihr Chlorid durch Einwirkung von Phosphorpentachlorid in einer Ben zinkohlenwasserstoff-Fraktion als Reaktionsmedium und anschliessende Veresterung des Chlorids mit Diäthylaminoäthanol in Benzol, gefolgt von einer Umkristallisierung aus absolutem Äthanol. Dieses Verfahren erwies sich jedoch in technischem Massstab als ungeeignet.
Gemäss vorliegender Erfindung wurde nun festgestellt, dass Acetylsalicylsäure-diäthylaminoäthylester- hydrochlorid leicht hergestellt werden kann durch Umesterung leicht zugänglicher Salicylsäureester mit Diäthylaminoäthanol, Acetylierung mit Acetanhydrid und folgende Umwandlung des resultierenden Acetylsalicylsäure-diäthylaminoäthylesters in das Hydrochlorid in mit Chlorwasserstoff gesättigtem absolutem Äthanol.
So verläuft z. B. die Umesterung von Salicylsäurephenylester mit Diäthylaminoäthanol leicht bei 40 bis 50 und Normaldruck unter 90-95 Obiger Pro duktausbeute; aliphatische Salicylsäureester, die unter üblichen Bedingungen nur schwierig reagieren und niedrige Ausbeuten von etwa 12-14 O/o ergeben, werden erfindungsgemäss unter genau bestimmten Bedingungen, nämlich vermindertem Druck und Abdestillieren des resultierenden Alkohols, umgeestert unter 85-90 Obiger Produktausbeute.
Der bei der Umesterung erhaltene Salicylsäurediäthylaminoäthylester wird entweder direkt oder nach vorgängiger Destillation im Vakuum mit Acetanhydrid bei weniger als 55 , zweckmässigerweise bei 40-50 , acetyliert. Der resultierende Acetylsali cylsäure-diäthylaminoäthylester wird bei vermindertem Druck destilliert, wonach man das Hydrochlorid durch Zusatz von trockener äthanolischer Chlorwasserstofflösung in 5-25 0/oigem Überschuss ausfällt.
Das Hydrochlorid ist als Lokalanästhetikum verwendbar.
Beispiel 1
130 Gewichtsteile Diäthylaminoäthanol werden mit 214 Gewichtsteilen Salicylsäurephenylester 3 Stunden lang bei 40 und weitere 2 Stunden bei 50-600 verrührt. Überschüssiges Diäthylaminoäthanol und das resultierende Phenol werden bei verrnr dertem Druck abdestilliert wonach man den Salicylsäure-diäthylaminoäthylester bei 148-1520 und 15 mm Hg. abdestilliert. Die Ausbeute beträgt 210 bis 220 Gewichtsteile. Dann werden 105 Gewichtsteile Acetanhydrid und gegebenenfalls noch einige Tropfen konzentrierte Schwefelsäure zu 200 Ge wichtsteilen des Salicylsäureesters gegeben, wonach man 2 Stunden auf 500 erwärmt.
Das Gemisch wird bei 15 mm Hg. destilliert und die Fraktion von reinem Acetyls alicylsäure-diäthylaminoäthylester wird bei 170-1780 gesammelt. Die Ausbeute beträgt 205 bis 210 Gewichtsteile. 300 Gewichtsteile einer 10 0/obigen Chlorwasserstofflösung in absolutem Sitha- nol werden 205 Gewichtsteilen des Esters unter kräftigem Rühren und starkem Kühlen zugesetzt, wobei eine Temperatur unterhalb 200 aufrecht zu erhalten ist; dann wird das Gemisch auf -10" abgekühlt, das sich abscheidende Acetylsalicylsäure-diäthylamino äthylester-hydrochlorid abfiltriert und mit kaltem absolutem Äthanol gewaschen. Die Ausbeute an trockenem Produkt beträgt 200 Gewichtsteile.
Das Produkt kann aus 250 Gewichtsteilen absolutem Äthanol umkristallisiert werden, wobei man eine erste Fraktion von 180 Gewichtsteilen trockener Kristalle vom Schmelzpunkt 134-136 erhält.
Beispiel 2
146 Gewichtsteile Diäthylaminoäthanol werden 3 Stunden lang mit 152 Gewichtsteilen Salicylsäuremethylester bei 100-200 mm Hg auf 70 bis 750 erwärmt, wonach die Temperatur allmählich auf 900 gesteigert und der Druck auf 30mm Hg reduziert wird. Für das gesamte Verfahren benötigt man 15 Stunden. Überschüssiges Diäthylaminoäthanol wird sodann bei 15 mm Hg abdestilliert, wonach die aus Salicylsäure.diäthylaminoäthylester bestehende Fraktion bei 148-152 gesammelt wird. Man erhält 200 Gewichtsteile des Esters, der sodann wie in Beispiel 1 beschrieben, weiterbehandelt werden kann.

Acetylsalicylic acid is a relatively unstable compound and the production of its esters, especially in high purity, is associated with great difficulties.
A known process for the preparation of acetylsalicylic acid diethylaminoethyl ester hydrochloride (see Z. Jerzmanowska and Z. Orchowicz, Acta Polon. Pharm. 13, 11-25 [1956]) consists in the conversion of acetylsalicylic acid into its chloride by the action of Phosphorus pentachloride in a benzine hydrocarbon fraction as the reaction medium and subsequent esterification of the chloride with diethylaminoethanol in benzene, followed by recrystallization from absolute ethanol. However, this method proved unsuitable on an industrial scale.
According to the present invention, it has now been found that diethylaminoethyl acetylsalicylic acid hydrochloride can easily be prepared by transesterification of easily accessible salicylic acid esters with diethylaminoethanol, acetylation with acetic anhydride and subsequent conversion of the resulting diethylaminoethyl acetylsalicylic acid into the hydrochloride in absolute hydrochloride saturated with hydrogen chloride.
So z. B. the transesterification of salicylic acid phenyl ester with diethylaminoethanol easily at 40 to 50 and normal pressure below 90-95 above product yield; Aliphatic salicylic acid esters, which react only with difficulty under customary conditions and give low yields of about 12-14%, are, according to the invention, under precisely defined conditions, namely reduced pressure and distilling off the resulting alcohol, transesterified with 85-90 above product yield.
The diethylaminoethyl salicylate obtained in the transesterification is acetylated either directly or after prior distillation in vacuo with acetic anhydride at less than 55, conveniently at 40-50. The resulting acetylsali cylic acid diethylaminoethyl ester is distilled under reduced pressure, after which the hydrochloride is precipitated in a 5-25% excess by adding dry ethanolic hydrogen chloride solution.
The hydrochloride can be used as a local anesthetic.

130 parts by weight of diethylaminoethanol are stirred with 214 parts by weight of phenyl salicylate for 3 hours at 40 and a further 2 hours at 50-600. Excess diethylaminoethanol and the resulting phenol are distilled off under reduced pressure, after which the diethylaminoethyl salicylate is distilled off at 148-1520 and 15 mm Hg. The yield is 210 to 220 parts by weight. Then 105 parts by weight of acetic anhydride and optionally a few drops of concentrated sulfuric acid are added to 200 parts by weight of the salicylic acid ester, after which the mixture is heated to 500 for 2 hours.
The mixture is distilled at 15 mm Hg. And the fraction of pure acetylsalicylic acid diethylaminoethyl ester is collected at 170-1780. The yield is 205 to 210 parts by weight. 300 parts by weight of a 10% hydrogen chloride solution in absolute sithanol are added to 205 parts by weight of the ester with vigorous stirring and vigorous cooling, a temperature below 200 being maintained; The mixture is then cooled to -10 ", the acetylsalicylic acid diethylamino ethyl ester hydrochloride which separates out is filtered off and washed with cold absolute ethanol. The yield of dry product is 200 parts by weight.
The product can be recrystallized from 250 parts by weight of absolute ethanol to give a first fraction of 180 parts by weight of dry crystals melting at 134-136.
Example 2
146 parts by weight of diethylaminoethanol are heated for 3 hours with 152 parts by weight of methyl salicylate at 100-200 mm Hg to 70 to 750, after which the temperature is gradually increased to 900 and the pressure is reduced to 30 mm Hg. The entire procedure takes 15 hours. Excess diethylaminoethanol is then distilled off at 15 mm Hg, after which the fraction consisting of salicylic acid diethylaminoethyl ester is collected at 148-152. 200 parts by weight of the ester are obtained, which can then be treated further as described in Example 1.
