Verfahren zur Herstellung von Oxazolin-4-onen
Die vorliegende Erfindung betrifft ein Verfahren zur Herstellung von 5-Aryl-2-amino-2-oxazolin-4onen der allgemeinen Formel:

Auf Grund der Beweglichkeit der Wasserstoffatome ist es ganz selbstverständlich, dass die neuen Verbindungen in verschiedenen weiteren tautomeren Formen vorliegen können, von denen eine nachfolgend gezeigt ist:

Selbstverständlich umfasst die vorliegende Erfindung die Herstellung sämtlicher dieser tautomeren Formen. Der einfacheren Beschreibung wegen werden jedoch die neuen Verbindungen hier als 5-(o Halogen- phenyl)-2-amino-2-oxazolin-4-one bezeichnet.
Die neuen erfindungsgemäss hergestellten Verbindungen sind sehr wertvolle Stimulantien des Zentral nervensystems. Sie zeigen eine milde, stimulierende Wirkung und ausgezeichnete appetitzügelnde Wirkung innerhalb eines weiten Dosierungsbereiches und besitzen bestimmte Vorteile gegenüber andern Stimulierungsmedikamenten, wie z. B. den Amphetaminen und Pipradrol.
Amphetamin und eng verwandte Verbindungen, wie z. B. Methamphetamin, werden seit vielen Jahren als Stimulatien des Zentralnervensystems verwendet, jedoch begleiten zahlreiche unerwünschte Nebenreaktionen ihre Anwendung. Zum Beispiel verursachen sie einen mehr oder weniger ausgeprägten Anstieg des Blutdrucks, und es besteht eine Tendenz im Hinblick auf die Verträglichkeit bei fortgesetztem Gebrauch. Die erfindungsgemäss hergestellten Verbindungen besitzen diese ernsten Nebeneffekte nicht und sind infolgedessen gegenüber den Amphetaminen bemerkenswert überlegen. Die erfindungsgemäss hergestellten Verbindungen sind ebenso, selbst bei hohen Dosierungen, frei von unerwünschten adrenergischen und cardiovasculären Wirkungen, die den Amphetaminen eigentümlich sind. Wenn die Amphetamine in gesteigerten Dosierungen gegeben werden, werden üblicherweise Krämpfe beobachtet.
Die erfindungsgemäss hergestellten Verbindungen verursachen keine Krämpfe, wenn die Dosierung gesteigert wird. Die neuen Verbindungen besitzen einen niedrigen Grad der Toxizität und eine wünschenswert grosse Breite zwischen wirksamer und letaler Dosis, das heiss einen hohen therapeutischen Index. Die erfindungsgemäss hergestellten Verbindungen weisen einen weit grö sseren Sicherheitsgrad auf als Pipradol, welches einen sehr engen Bereich zwischen wirksamer und toxischer Dosis aufweist.
Weiterhin sind die neuen erfindungsgemäss hergestellten Verbindungen beträchtlich stärker aktiv als das in der USA-Patentschrift Nr. 2 892 753 beschriebene 5-Phenyl-2-imino-4-oxo-oxazolidin. Zum Beispiel sind die erfindungsgemäss hergestellten Verbindungen 2-Dimethylamino-5-phenyl-2-oxazolin-4-on und 2-Athylmethylamino -5- phenyl-2-oxazolin-4-on etwa neunmal so wirksam bei der Erzeugung eines 50 % igen Anstiegs der motorischen Aktivität bei nichttoxischer Dosis. Zum Beispiel ist das erfindungsgemäss hergestellte 5-(o-Fluorphenyl)-2-amino 2-oxazolin-4-on etwa siebenmal so wirksam bei der Hervorrufung einer 50% eigen Steigerung der motorischen Aktivität bei nichttoxischer Dosis.
Diese Art von Versuch ist als brauchbare Methode zur Bestimmung der stimullerenden Aktivität bekannt und ist durch P. B. Dews, British Journal of Pharmacology, Bd. 8, S. 46 (1953) und durch G. Chen et al., Journal of Pharmacology and Experimental Therapeutics, Bd. 127, S. 241 (1959) beschrieben.
Die erfindungsgemäss hergestellten Verbindungen sind im allgemeinen weisse, kristalline Stoffe, die nur mässig löslich in Wasser sind. Sie sind basische Sul > stanzen, die in wässrigen Mineralsäuren bei Raumtemperatur löslich sind und die in bestimmten Fällen isolierbare Säureadditionssalze bilden.
Die erfindungsgemäss hergestellten Verbindungen können in Form ihrer freien Basen oder als nichttoxische Säureadditionssalze, wie z. B. als Hydrochloride, Sulfate, Phosphate, Citrate und ähnliche, verabreicht werden. Die Verbindungen können oral oder parenteral gegeben werden, und bei derartiger Verabreichung sind sie Stimulatien des Zentralnervensystems bei Einzeldosen im Bereich von etwa 1 bis 100 mg. Die Dosierungsart kann eingeregelt werden, um den optimalen therapeutischen Effekt zu ergeben. Zum Beispiel können verschiedene Dosen täglich gegeben werden, oder die Dosis kann in dem Mass erniedrigt werden, wie es durch die Gegebenheit der therapeutischen Situation geboten ist.
Ein spezieller Vorteil der erfindungsgemäss hergestellten Verbindungen ist deren niedrige Toxizität.
Die LD50 des 2-Dimethylamino-5-phenyl-2-oxazolin4-ons ist z. B. 570 mg/kg bei oraler Verabreichung.
So liegt bei einer Dosis von etwa 65 mg/kg als wirksamer Dosis der therapeutische Index dieser Verbindung bei etwa 9. Die so des 5-(o-Chlorphenyl)2-amino-2-oxazolin-4-ons ist z. B. grösser als 4000 mg/kg bei oraler Verabreichung. So ist bei einer Dosis von etwa 400 mg/kg als wirksamer Dosis der therapeutische Index dieser Verbindung etwa 10.
Zur therapeutischen Verabreichung können die neuen Verbindungen mit pharmazeutischen Trägerstoffen eingebaut werden und z. B. in Form von Tabletten, Dragees, Kapseln, Suppositoiren oder Flüssigkeiten, die in Form von Tropfen, Emulsionen, Suspensionen, Sirupen, Schokolade, Kandis, Kaugummi und dergleichen, verabreicht werden, gebraucht werden. Derartige Mischungen und Präparate sollten mindestens 0,1 % des aktiven Bestandteils enthalten. Der Prozentsatz in den Mischungen und Präparaten kann natürlich variiert werden und kann üblicherweise zwischen 2 und etwa 60 S oder mehr des Gewichtes der Einheit ausmachen. Die Menge aktiven Bestandteils in diesen therapeutisch brauchbaren Mischungen oder Präparaten wird derartig gewählt, dass eine geeignete Dosierung erzielt wird.
Bevorzugte Mischungen oder Präparate werden in der Weise hergestellt, dass eine Dosierungseinheitsform zwischen etwa 1 mg und etwa 100 mg der neuen Verbindungen enthält.
In der nachfolgenden Beschreibung besitzen X, R ; L und R2 die oben für Formel I angegebene Bedeutung.
Das erfindungsgemässe Verfahren zur Herstellung der Verbindungen der Formel I besteht darin, dass ein Phenylessigsäurederivat der folgenden allgemeinen Formel:

worin Z 0 bedeutet, wenn Y Halogen oder Sulfonyloxy ist, und worin Z NH oder S ist, wenn Y OH ist, während Rt, R2 und X die oben angegebene Bedeu- tung besitzen, cyclisiert wird. Gegebenenfalls werden die nichttoxischen Säureadditionssalze hergestellt.

Die Reaktion nach obigem Schema ist nach einer Vielzahl von Bedingungen durchführbar unter Verwendung verschiedener unterschiedlicher Phenylessigsäurederivate zur Umsetzung mit dem l,l-Dialkyl- harnstoff. Die Bedingungen und Ausgangsmaterialien werden in der Absicht ausgewählt, dass die bestmögliche Umwandlung des offenkettigen Acylharnstoff-Zwischenproduktes mit nachfolgendem (oder gleichzeitigem3 Ringschluss zur Herstellung der gewünschten neuen Produkte erzielt wird. Geeignete Ausgangsmaterialien sind
2-Phenyl-2-chloracetylchlorid,
2-Phenyl-2-bromacetylbromid, Äthyl-2-phenyi-2-brom acetat,
2-Phenyl-2-(p-toluolsufonyloxy)-acetylchlorid und ähnliche.
Die Reaktion wird im allgemeinen in einem Lösungsmittel durchgeführt. Ein inertes Lösungsmittel, wie z. B. Benzol, Toluol und dergleichen, wird allgemein angewandt, wenn man das Acylharnstoff- Zwischenprodukt zu isolieren wünscht. In einigen Fällen, insbesondere, wenn gleichzeitiger Ringschluss gewünscht wird, ist ein wässriges Lösungsmittel brauchbar. Die Acylharnstoff- Zwischenprodukte werden bequemerweise mit wässrigen oder alkoholischen Basen cyclisiert. Die Temperatur liegt gebräuchlicherweise in Bereich von 25 bis 1500 C.

Diese Reaktion wird im allgemeinen in einem Lösungsmittel, wie z. B. einem niederen Alkanol, durchgeführt. Bequemerweise wird Äthanol verwendet, und es wird eine Rückflusstemperatur aufrechterhalten, um die Reaktion, insbesondere den Ringschluss, in einem vernünftigen Zeitraum, üblicherweise einige Stunden, hervorzurufen. Verschiedene Guanidinsalze, wie z.
B. das Hydrochlorid und Carbonat, die im Handel erhältlich sind, können verwendet werden, und diese werden im allgemeinen mit einem Alkali neutralisiert, um einen wirksamen Umsatz des Guanidins mit dem Ester zu ermöglichen.
Die offene Kettenstruktur (III) kann als Zwischenprodukt betrachtet werden, welches Ammoniak verliert und die gewünschten Produkte ergibt. Jedoch ist eine Isolierung dieser Zwischenprodukte, die in situ gebildet werden, nicht notwendig, und sie ist im allgemeinen schädlich und unerwünscht.
Anstelle von Guanidin kann hierbei ein substituierter Thioharnstoff in einem niederen Alkanol als Lösungsmittel, z. B. Äthanol, mit einem Gehalt von einem Äquivalent Alkalialkoholat eingesetzt werden.
Anderseits kann Cyanamid in Form eines Alkalisalzes, z. B. Natrium- oder Kaliumcyanamid, verwendet werden anstelle von Guanidin.
Das allgemeine Verfahren zur Herstellung der neuen Verbindungen wird ebenso in dem nachfolgenden Schema erläutert:

B. p-Toluolsulfonyloxy, und Z eine Gruppe, die durch Harnstoff ersetztbar ist, wie z. B. ein reaktives Halogen, wie Chlor und Brom, einen niederen Alkoxyrest und dergleichen, bedeuten.
Die Reaktion nach obigem Schema ist unter einer Vielzahl von Bedingungen unter Verwendung einiger unterschiedlicher a-(o-Halogen-phenyl)-essig säurederivaten zur Umsetzung mit Harnstoff durchführbar. Die Bedingungen und Ausgangsmaterialien werden in der Hinsicht gewählt, dass die bestmögliche Umwandlung des offenkettigen Zwischenproduktes mit nachfoIgendem (oder gleichzeitigem) Ringschluss (-HY) zur Herstellung der gewünschten neuen Produkte erzielt wird. Geeignete Ausgangsmaterialien sind 2- (o-Chlorphenyl) -2-chloracetylchlorid,
2-(o-Fluorphenyl)-bromacetylbromid, Äthal-2-(o-bromphenyl)-2-bromacetat, 2- (o-Fiuorphenyl)-2- (p-toluolsulfonyloxy) - acetylchlorid und ähnliche.
Die Reaktion wird im allgemeinen in einem Lösungsmittel durchgeführt, allgemein wird ein inertes Lösungsmittel, wie Benzol, Toluol und ähnliche, verwendet, wenn es gewünscht wird, das Acylharnstoff Zwischenprodukt zu isolieren. In einigen Fällen, insbesondere, wenn gleichzeitiger Ringschluss gewünscht wird, ist ein wässriges Lösungsmittel brauchbar. Die Acylharnstoff-Zwischenprodukte werden bequemerweise mit wässrigen oder alkoholischen Basen cyclisiert. Die Temperatur liegt üblicherweise im Bereich von 25 bis 150 .
Die Erfindung wird ausführlicher auf Grund der folgenden spezifischen Beispiele beschrieben.
Beispiel 1
Herstellung von 5- (o-Chlorphenyl)-2-amino-
2-oxazolin-4-on
Ein Gemisch von 21,5 g o-Chlormandelsäure äthylester, 9,6 g Guanidin-hydrochlorid und 5,6 g Kaliumhydroxydplätzchen in 100 ml abs. Äthanol wird etwa 1 Stunde am Rückfluss gehalten und dann gekühlt. Die Mischung wird mit etwa 350 ml Wasser verdünnt und auf 0 C abgekühlt, worauf sich das Rohprodukt als Niederschlag ausscheidet. Das Rohprodukt wird durch Filtration abgetrennt und getrocknet, F. = 258-2600 C (Zersetzung). Das Rohprodukt wird durch Umkristallisation aus wässrigem Alkohol gereinigt und ergibt das 5-(o-Chlorphenyl)2-amino-2-oxazolin-4-on, F. = 259-261 C (Zersetzung).
Beispiel 2
Herstellung von 5-(o-Fluorphenyl)-2-amino 2-oxazolin-$on
Eine Lösung von 4,9 g o-Fluormandelsäureäthylester in 12 ml abs. Athanol wird bei Rückflusstemperatur mit 12,2 ml 2,05molarem äthanolischem Guanidin (hergestellt durch Behandlung von Guanidinhydrochlorid mit einer äquivalenten Menge Natriumäthylat in Äthanol und Abfiltrieren des ausgefällten Natriumchlorids) während etwa 2 Stunden erhitzt. Das Äthanol wird durch Destillation entfernt und der Rückstand während 15 Minuten mit 25 ml Molarem Natriumhydroxyd gerührt. Die Suspension wird filtriert und das Filtrat mit 30 ml 2molarer Essigsäure behandelt.
Der Niederschlag wird abfiltriert und aus Äthanol umkristallisiert und ergibt 5-(o-Fluorphenyl) -2- amino-2-oxazolin-4n, F. = 235-237 C (Zersetzung).
Beispiel 3 o-Fluormandelsäure-äthylester
Bekannte substituierte Mandelsäuren werden in die entsprechenden Estern nach einem durch 5. L.
Shapiro et al. (J. Am. Chem. Soc. 81, 5646 [1959]) beschriebenen Verfahren umgewandelt, nach welchem eine molare Lösung der Säure in abs. Äthanol während einiger Stunden mit 2,5 milli-Äquivalenten p-Toluolsulfonsäuremonohydrat am Rückfluss erhitzt wird. Das wässrige Äthanol wird durch Destillation entfernt und das hinterbliebene Öl in ausreichend Ather gelöst, um eine etwa molare Lösung des Esters zu ergeben. DieAtherlösung wird mit 10% der Natriumcarbonatlösung und einer geringen Menge Wasser gewaschen und dann über Magnesiumsulfat getrocknet. Nach Entfernung des Lösungsmittels und Destillation des Rückstandes unter vermindertem Druck werden die Ester in einem im wesentlichen reinen Zustand erhalten.
Der nach diesem Verfahren hergestellte o - Fluormandelsäureäthylester destilliert bei 71-72 C (0, 2 mm).
Beispiel 4 2-Amino-5- (o-chIorphenyl)-2-oxazolin-Son
Zu einer Mischung von 2,3 g Natriummetall, gelöst in abs. Äthanol, werden 7,6 g Thioharnstoff und anschliessend 21,4 g o-Chlormandelsäureäthyl- ester in 50 ml Athanol zugegeben. Die Mischung wird bei Rückflusstemperatur während etwa 1 Stunde erhitzt und dann zur Trockne konzentriert. Der Rückstand wird in 50 ml Wasser aufgelöst und mit 50 ml 2n Essigsäure angesäuert. Die Mischung wird abgekühlt, über Nacht stehengelassen und das Rohprodukt gesammelt. Nach Reinigung durch Umkristallisation aus wässrigem Dimethylformamid erhält man 2-Amino-5-(o-chlorphenyl)-2-oxazolin-4-on, F. = 260w2630 C (Zersetzung).

Due to the mobility of the hydrogen atoms, it goes without saying that the new compounds can exist in various other tautomeric forms, one of which is shown below:

Of course, the present invention encompasses the preparation of all of these tautomeric forms. For the sake of simpler description, however, the new compounds are referred to here as 5- (o halophenyl) -2-amino-2-oxazolin-4-ones.
The new compounds prepared according to the invention are very valuable stimulants of the central nervous system. They show a mild, stimulating effect and excellent appetite suppressing effect within a wide dosage range and have certain advantages over other stimulating drugs, such as. B. the amphetamines and pipradrol.
Amphetamine and closely related compounds, such as. Methamphetamine, for example, have been used as central nervous system stimulants for many years, but numerous undesirable side reactions accompany their use. For example, they cause a more or less pronounced rise in blood pressure, and there is a tendency for tolerability with continued use. The compounds prepared according to the invention do not have these serious side effects and are consequently remarkably superior to the amphetamines. The compounds prepared according to the invention are also free from undesirable adrenergic and cardiovascular effects which are peculiar to amphetamines, even at high dosages. When the amphetamines are given in increased doses, convulsions are usually observed.
The compounds prepared according to the invention do not cause cramps when the dosage is increased. The new compounds have a low degree of toxicity and a desirably large range between effective and lethal dose, that is to say a high therapeutic index. The compounds prepared according to the invention have a far greater degree of safety than pipradol, which has a very narrow range between the effective and toxic dose.
Furthermore, the new compounds prepared according to the invention are considerably more active than the 5-phenyl-2-imino-4-oxo-oxazolidine described in U.S. Patent No. 2,892,753. For example, the compounds 2-dimethylamino-5-phenyl-2-oxazolin-4-one and 2-ethylmethylamino -5-phenyl-2-oxazolin-4-one made in accordance with the present invention are about nine times as effective in producing a 50% increase motor activity at a non-toxic dose. For example, the 5- (o-fluorophenyl) -2-amino 2-oxazolin-4-one prepared according to the invention is about seven times as effective in inducing a 50% increase in motor activity at a non-toxic dose.
This type of experiment is known as a useful method for determining stimulatory activity and is described by PB Dews, British Journal of Pharmacology, Vol. 8, p. 46 (1953) and by G. Chen et al., Journal of Pharmacology and Experimental Therapeutics , Vol. 127, p. 241 (1959).
The compounds prepared according to the invention are generally white, crystalline substances which are only sparingly soluble in water. They are basic sulphates which are soluble in aqueous mineral acids at room temperature and which, in certain cases, form isolable acid addition salts.
The compounds prepared according to the invention can be in the form of their free bases or as non-toxic acid addition salts, such as. B. as hydrochlorides, sulfates, phosphates, citrates and the like, can be administered. The compounds can be administered orally or parenterally, and when administered in such a manner are central nervous system stimuli at single doses ranging from about 1 to 100 mg. The dosage type can be adjusted to give the optimal therapeutic effect. For example, different doses can be given daily, or the dose can be reduced to the extent that the circumstances of the therapeutic situation require.
A particular advantage of the compounds prepared according to the invention is their low toxicity.
The LD50 of 2-dimethylamino-5-phenyl-2-oxazolin4-one is z. B. 570 mg / kg when administered orally.
Thus, at a dose of about 65 mg / kg as the effective dose, the therapeutic index of this compound is about 9. The 5- (o-chlorophenyl) 2-amino-2-oxazolin-4-one is e.g. B. greater than 4000 mg / kg for oral administration. Thus, at a dose of about 400 mg / kg effective dose, the therapeutic index of this compound is about 10.
For therapeutic administration, the new compounds can be incorporated with pharmaceutical carriers and z. B. in the form of tablets, coated tablets, capsules, suppositoirs or liquids, which are administered in the form of drops, emulsions, suspensions, syrups, chocolate, candy, chewing gum and the like, are used. Such mixtures and preparations should contain at least 0.1% of the active ingredient. The percentage in the mixtures and preparations can, of course, be varied and can usually be between 2 and about 60% or more of the weight of the unit. The amount of active ingredient in these therapeutically useful mixtures or preparations will be chosen such that a suitable dosage will be achieved.
Preferred mixtures or preparations are made such that a dosage unit form contains between about 1 mg and about 100 mg of the new compounds.
In the description below, X, R; L and R2 have the meaning given above for formula I.
The process according to the invention for the preparation of the compounds of the formula I consists in that a phenylacetic acid derivative of the following general formula:

where Z is 0 when Y is halogen or sulfonyloxy, and where Z is NH or S when Y is OH, while Rt, R2 and X have the meaning given above, cyclization is carried out. Optionally, the non-toxic acid addition salts are prepared.

The reaction according to the above scheme can be carried out under a variety of conditions using various different phenylacetic acid derivatives for reaction with the l, l-dialkyl urea. The conditions and starting materials are chosen with the intention of achieving the best possible conversion of the open chain acylurea intermediate with subsequent (or simultaneous3 ring closure to produce the desired new products. Suitable starting materials are
2-phenyl-2-chloroacetyl chloride,
2-phenyl-2-bromoacetyl bromide, ethyl-2-phenyi-2-bromo acetate,
2-phenyl-2- (p-toluenesufonyloxy) acetyl chloride and the like.
The reaction is generally carried out in a solvent. An inert solvent such as. Benzene, toluene, and the like, are generally used when it is desired to isolate the acylurea intermediate. In some cases, particularly when simultaneous ring closure is desired, an aqueous solvent is useful. The acylurea intermediates are conveniently cyclized with aqueous or alcoholic bases. The temperature is usually in the range of 25 to 1500 C.

This reaction is generally carried out in a solvent such as e.g. B. a lower alkanol performed. Ethanol is conveniently used and a reflux temperature is maintained to cause the reaction, particularly ring closure, in a reasonable period of time, usually a few hours. Various guanidine salts, such as

The open chain structure (III) can be viewed as an intermediate product which loses ammonia and gives the desired products. However, isolation of these intermediates which are formed in situ is not necessary and is generally deleterious and undesirable.
Instead of guanidine, a substituted thiourea in a lower alkanol can be used as a solvent, e.g. B. ethanol, with a content of one equivalent of alkali metal alcoholate can be used.
On the other hand, cyanamide in the form of an alkali salt, e.g. B. sodium or potassium cyanamide, can be used instead of guanidine.

example 1
Preparation of 5- (o-chlorophenyl) -2-amino-
2-oxazolin-4-one
A mixture of 21.5 g of o-chloromandelic acid ethyl ester, 9.6 g of guanidine hydrochloride and 5.6 g of potassium hydroxide biscuits in 100 ml of abs. Ethanol is refluxed for about 1 hour and then cooled. The mixture is diluted with about 350 ml of water and cooled to 0 C, whereupon the crude product separates out as a precipitate. The crude product is separated off by filtration and dried, F. = 258-2600 C (decomposition). The crude product is purified by recrystallization from aqueous alcohol and gives 5- (o-chlorophenyl) 2-amino-2-oxazolin-4-one, m.p. = 259-261 ° C. (decomposition).
Example 2
Preparation of 5- (o-fluorophenyl) -2-amino 2-oxazolin- $ one
A solution of 4.9 g of ethyl o-fluoromandelate in 12 ml of abs. Ethanol is heated at reflux temperature with 12.2 ml of 2.05 molar ethanolic guanidine (prepared by treating guanidine hydrochloride with an equivalent amount of sodium ethylate in ethanol and filtering off the precipitated sodium chloride) for about 2 hours. The ethanol is removed by distillation and the residue is stirred for 15 minutes with 25 ml of molar sodium hydroxide. The suspension is filtered and the filtrate is treated with 30 ml of 2 molar acetic acid.
The precipitate is filtered off and recrystallized from ethanol and yields 5- (o-fluorophenyl) -2-amino-2-oxazoline-4n, mp = 235-237 ° C. (decomposition).
Example 3 Ethyl o-fluoromandelate
Known substituted mandelic acids are converted into the corresponding esters after a through 5. L.
Shapiro et al. (J. Am. Chem. Soc. 81, 5646 [1959]), according to which a molar solution of the acid in abs. Ethanol is refluxed for a few hours with 2.5 milli-equivalents of p-toluenesulfonic acid monohydrate. The aqueous ethanol is removed by distillation and the remaining oil dissolved in sufficient ether to give an approximately molar solution of the ester. The ether solution is washed with 10% of the sodium carbonate solution and a small amount of water and then dried over magnesium sulfate. After removal of the solvent and distillation of the residue under reduced pressure, the esters are obtained in a substantially pure state.
The o-fluoromandelic acid ethyl ester produced by this process distills at 71-72 ° C. (0.2 mm).
Example 4 2-Amino-5- (o-chlorophenyl) -2-oxazoline-Son
To a mixture of 2.3 g of sodium metal dissolved in abs. Ethanol, 7.6 g of thiourea and then 21.4 g of o-chloromandelic acid ethyl ester in 50 ml of ethanol are added. The mixture is heated at reflux temperature for about 1 hour and then concentrated to dryness. The residue is dissolved in 50 ml of water and acidified with 50 ml of 2N acetic acid. The mixture is cooled, left to stand overnight and the crude product collected. After purification by recrystallization from aqueous dimethylformamide, 2-amino-5- (o-chlorophenyl) -2-oxazolin-4-one, m.p. = 260w2630 C (decomposition) is obtained.
