Process for the preparation of new 4-substituted 1,2-diaryl-3,5-dioxo-pyrazolidines The present invention relates to a process for the preparation of new 1,2-diphenyl-3,5-dioxopyrazolidine derivatives with valuable pharmacological properties.
It has been found that 4-substituted 1,2-diaryl-3,5-dioxo-pyrazolidines of the formula I,
EMI0001.0008
wherein R1 is a lower alkyl radical, R, hydrogen, a lower alkyl, alkoxy or alkylsulfonyl radical, a halogen atom or a hydroxyl group, and R #, hydrogen or a hydrocarbon radical, wel cher in a chain and / or a possible ring by O or SO ., be interrupted and can be substituted on carbon atoms with multiple bonds by halogen atoms,
mean, and their salts with inorganic and organic bases have valuable uricosuric, anti-inflammatory and analgesic activity with low toxicity.
The compounds defined above are prepared by adding a compound of the formula 11,
EMI0001.0020
wherein R2 'signifies a lower alkyl mercapto radical or the same as R, converted into a compound of the formula 1 by oxidation.
Hydrogen peroxide in a solvent such as, for example, glacial acetic acid at temperatures between about 40 and 100 is particularly suitable as an agent for the oxidation of thio compounds of the formula II to corresponding sulfonyl compounds of the formula I.
The compounds obtained according to the invention and their salts with inorganic and organic bases are suitable, for example, for the treatment of rheumatic diseases and gout, whereby they can be administered per os or rectally or in the form of aqueous solutions of their salts also parenterally, for example intramuscularly or intravenously, can be administered.
In the compounds of the formula I, R1 can be, for example, methyl or ethyl, R2, for example, hydrogen, the methylsulfonyl, ethylsulfonyl, methyl, ethyl, isopropyl, isobutyl, tertiary butyl, methoxy, ethoxy or n-propoxy radical or Chlorine, bromine or fluorine and R3, for example, methyl, ethyl, n-propyl,
Isopropyl, n-butyl, isobutyl, sec. Butyl, tertiary butyl, n-amyl, isoamyl, n-hexyl, n-heptyl, n-octyl, n-decyl, n-dodecyl, n-hexadecyl, crotyl, 3-chloro crotyl, methallyl, cyclopentyl, cyclohexyl,
2,5-endomethylene-1-cyclohexenyl-, 2,5-endomethylene-cyclohexylmethyl-, cycloheptenyl-, phenyl-, p-chlorophenyl, p-methoxyphenyl-, 3,4-dimethyl-phenyl-, Benzyl-, p-chlorobenzyl-, 3,4-dimethoxy-benzyl-, 3,4-methylene-dioxybenzyl-, ss-phenyl-ethyl-,
Y-phenyl-propyl, s-methoxy-ethyl, s-ethoxy-ethyl, s-n-butoxy-ethyl, s-phenoxy-ethyl, s-benzyloxy-ethyl, or s-phenylsulfonyl-ethyl radical or also mean hydrogen.
The new compounds of formula 1 form fairly easily to easily soluble monobasic salts with alkali metal hydroxides. Solutions of the same can be prepared directly by dissolving the new compounds in the calculated amount of alkali, i.e. in solutions of lithium, sodium or potassium hydroxide. Most of the new compounds also dissolve in alkali hydrogen carbonate solutions.
The novel compounds obtained according to the invention or their alkali metal salts or salts with organic bases, such as dimethylaminoethanol, diethylaminoethanol or piperazine, can be used as medicaments in combinations with suitable pharmaceutical carriers. The above-mentioned solutions of the alkali salts of the new compounds have a neutral to slightly alkaline reaction and are therefore well suited for injection.
The pharmaceuticals can be used, for example, in the form of capsules, powders, tablets or other forms suitable for oral administration and can be used, for example, by mixing the active ingredients, i.e. the new substituted 1,2-diphenyl-3,5-dioxo Pyrazolidine, with pharmaceutical excipients such as starch, lactose, stearic acid, talc, magnesium stearate, etc. are made. However, they can also be processed into suppositories with semi-solid substances with a suitable melting range, for example cocoa butter.
In the example below, parts mean parts by weight; these relate to parts by volume like g to cm? The temperatures are given in degrees Celsius.
Example 4 parts of 1,2-bis- (o-methylthio-phenyl) -3,5-dioxo-4-n-butyl-pyrazolidine are dissolved in 80 parts by volume of glacial acetic acid and mixed with 4.1 parts 30 / a iger aqueous hydrogen peroxide solution added. The temperature is increased to 70 within four hours and held at this temperature until the oxidizing agent is consumed (about 2 hours). The mixture is concentrated as much as possible at 11 torr and the residue is partitioned between ethyl acetate and dilute sodium hydrogen carbonate solution.
The organic phase is drawn out with dilute sodium carbonate solution. Acidification of these extracts on Congo blue gives crystals which are recrystallized from ethyl acetate. 1,2-bis (o-methylsulfonyl-phenyl) -3,5-dioxo-4-n-butyl-pyrazolidine, melting point 189, is obtained.
In an analogous manner one obtains: 1- (p-Methylsulfonylphenyl) -2-phenyl-3,5-dioxo-4-n-butyl-pyrazolidine melts at 164, the 1,2-bis- (m-methylsulfonyl-phenyl) -3,5-dioxo-4-n-butyl-pyrazolidine of melting point 145 (from ethanol), the 1,2-bis- (p-methylsulfonyl-phenyl) -3,5-dioxo-4-n-butyl- Pyrazolidine melts at 222, 1- (m-methylsulfonyl-phenyl) -2-phenyl-3,5-dioxo-4-n-butyl-pyrazolidine has a melting point of 9 8 (from alcohol),
1- (p-methylsulfonyl-phenyl) -2- (p-tolyl) -3,5-dioxo-4-n-butyl-pyrazolidine with a melting point of 128 (from alcohol), 1- (p-methylsulfonyl-phenyl ) -2- (m-tolyl) -3,5-dioxo-4-n-butyl-pyrazolidine with a melting point of 160 (from alcohol), 1- (p-methylsulfonyl-phenyl) -2-phenyl-3,5 -dioxo- 4-phenyl-thioethyl-pyrazolidine with a melting point of 123 (from ethanol / ethyl acetate) and 1- (p-methylsulfonyl-phenyl) -2-phenyl- 3,
5-dioxo-4-benzylpyrazolidine, m.p. 154.
