Verfahren zur Herstellung von neuen Aryloxyessigsäureamiden
Die vorliegende Erfindung bezieht sich auf ein Verfahren zur Herstellung neuer Derivate von Aryloxyessigsäuren mit wertvollen pharmakologischen 5 Eigenschaften.
Es wurde gefunden, dass substituierte Aryloxyessigsäureamide der Formel

worin Rt einen Alkyl-, Alkenyl- oder Cycloalkylrest oder eineh gegebenenfalls durch Halogenatome oder niedermolekulare Alkyl- oder Alkoxygruppen substi tuierten Phenyl- oder Benzylrest, R2 Wasserstoff oder einen niedermolekularen Alkyl- oder Alkenylrest, R3 und R4 niedermolekulare Alkylreste und X die direkte
Bindung oder eine Athylen- oder Vinylengruppe bedeuten, wobei Alkylreste Rj und R2 auch unter sich direkt oder über ein Sauerstoffatom verbunden sein können, pharmakologisch wertvolle Eigenschaften, insbesondere hypnotische, narkotische und an ästhetische Wirksamkeit besitzen.
Man stellt die oben definierten Verbindungen erfindungsgemäss her, indem man ein Halogenacetamid der Formel II

2-Methoxy-4-(a-hydroxy-isobutyl)-phenol,
2-Methoxy-4-(a-hydroxy-n-amyl)-phenol,
2-Methoxy-4-(a-hydroxy-isoamyl)-phenol,
2-Methoxy-4-(3'-hydroxy-butyl)-phenol,
2-Methoxy-4-(3'-hydroxy-Al .2'-butenyl)-phenol,
2-Äthoxy-4-(a-hydroxy-äthyl)-phenol,
2-Äthoxy-4-(a4iydroxy-propyl)-phenol, 2-Athoxy-4-(a-hydroxy-n-butyl)-phenol und 2-Äthoxy-4-(a-hydroxy-isoamyl)-phenol genannt.
Im nachfolgenden Beispiel bedeuten Teile Ge wichtsteile; diese verhalten sich zu Volumteilen wie g zu cm3. Die Temperaturen sind in Celsiusgraden angegeben.
Beispiel
2-Äthoxy-4( 1'-hydroxy-n-butyl)-phenoxyessig- säure-N,N-diäthylamid
In einer Lösung von 0,23 g Natrium in 40 ml abs. Athanol werden 2,1 g 2-Athoxy-4-(1'-hydroxy- n-butyl)-phenol 5 Minuten am Rückfluss gekocht.
Nach Zugabe von etwa 0,1 g Natriumjodid werden 2 g Chloressigsäure-N,N-diäthylamid in 6 ml abs.
Äthanol gelöst, zugetropft und 21/2 Stunden bis zur neutralen Reaktion am Rückfluss gekocht. Nach Abkühlen wird vom ausgefallenen Kochsalz abgenutscht und das Filtrat im Vakuum eingedampft. Der Rückstand wird in 50 ml Ather aufgenommen, mit 2n NaOH und Wasser gewaschen, über Na2SO4 getrocknet, filtriert und eingedampft. Das Rohprodukt wird im Kugelrohr bei einer Luftbadtemperatur von 180 bis 215 und 0,001 Torr destilliert. Das erhaltene Produkt, 2,3 g zähflüssiges, schwach trübes, farbloses Öl, ist nach C,H,N-Analyse, IR-Spektrum und Dünnschichtchromatogramm identisch mit der nach durch Reduktion mit NaBH4 von 2-Äthoxy-4-n- butyroyl-phenoxy-essigsäure-N,N-diäthylamid hergestellten Verbindung.

In a solution of 0.23 g of sodium in 40 ml of abs. Ethanol, 2.1 g of 2-ethoxy-4- (1'-hydroxy-n-butyl) -phenol are refluxed for 5 minutes.
After adding about 0.1 g of sodium iodide, 2 g of chloroacetic acid-N, N-diethylamide in 6 ml of abs.
Dissolved ethanol, added dropwise and refluxed for 21/2 hours until a neutral reaction. After cooling, the precipitated common salt is filtered off with suction and the filtrate is evaporated in vacuo. The residue is taken up in 50 ml of ether, washed with 2N NaOH and water, dried over Na2SO4, filtered and evaporated. The crude product is distilled in a bulb tube at an air bath temperature of 180 to 215 and 0.001 Torr. The product obtained, 2.3 g of viscous, slightly cloudy, colorless oil, according to C, H, N analysis, IR spectrum and thin-layer chromatogram is identical to that obtained after reduction with NaBH4 of 2-ethoxy-4-n-butyroyl- phenoxy-acetic acid-N, N-diethylamide produced compound.
