Verfahren zur Herstellung von neuen Hydrazin-Derivaten
Die vorliegende Erfindung betrifft ein Verfahren zur Herstellung von neuen Hydrazin-Derivaten der Formel I,

worin A eine Alkylengruppe und R1 und R2 Wasserstoff oder niedere Alkylgruppen bedeuten, wobei die Gruppen A, R1 und R2 zusammen mindestens zwei Kohlenstoffatome enthalten, oder R1 und R2 zusammen mit dem sie verbindenden Stickstoffatom einen 6gliedrigen heterocyclischen Rest bilden.
Erfindungsgemäss werden die neuen Hydrazin Derivate hergestellt, indem man aus acylierten Hydrazin-Derivaten der Formel II,

worin R3 eine Acylgruppe bedeutet, die Acylgruppe hydrolytisch abspaltet.
Diese Abspaltung kann in der Weise erfolgen, dass man ein acyliertes Hydrazin der Formel II, dessen Herstellung in den nachfolgenden Beispielen ausführlich beschrieben ist, mit einer wässerigen Mineralsäure, vorzugsweise mit wässeriger Salzsäure, während 4 bis 6 Stunden bei einer Temperatur von 80-120 erhitzt. Die Desacylierung wird vorteilhaft in einer Stickstoffatmosphäre ausgeführt. Die Endprodukte werden nach üblichen Methoden isoliert und gereinigt und gegebenenfalls in ein geeignetes Salz übergeführt.
Die erfindungsgemäss hergestellten neuen Hydrazin-Derivate sind bei Raumtemperatur flüssig und lassen sich im Vakuum unzersetzt destillieren. Es sind basische Verbindungen, die mit anorganischen oder organischen Säuren beständige, bei Raumtempe- ratur kristallisierte Salze bilden.
Die neuen Verfahrensprodukte dienen als Zwi schenprodukte zur Herstellung von Medikamenten.
Sie besitzen indessen auch selbst therapeutisch verwertbare pharmakodynamische Eigenschaften, die sie zu wertvollen Medikamenten machen. So zeichnen sie sich beispielsweise durch eine starke Monoaminoxydase-Hemmwirkung aus.
In den nachfolgenden Beispielen sind die Temperaturen in Celsiusgraden angegeben.
Beispiel 1
60 g 1-(Dimethylamino-isopropyl)-2-acetylhydrazin werden in einem Gemisch von 500 cm3 konz. Salzsäure und 400 cm3 Wasser gelöst und dann in einer Stickstoffatmosphäre während 5 Stunden auf 115 bis 1200 erhitzt. Darauf wird die Lösung im Vakuum bei 500 eingedampft, der zähe Rückstand in Äthanol gelöst, wobei beim Anreiben das Dimethylamino-isopropyl-hydrazin-dihydrochlorid auskristallisiert. Smp.
115-130" Nach Umkristallisieren aus Methanol und Äthanol schmilzt die Verbindung bei 130-135 (hygroskopisch).
Diese Verbindung entsteht auch beim Verseifen von 1- (Dimethylamino - isopropyl)-2-benzoylhydrazin [Smp. des Dihydrochlorids 210-220 (Zers unter den oben angegebenen Reaktionsbedingungen.
Zur Herstellung der freien Base werden 154 g rohes, gut getrocknetes Dimethylamino-isopropylhydrazin-dihydrochlorid in 200 cm3 Methanol suspendiert und unter Rühren und Kühlen mit 7-90 cm3 2,1n. Natriummethylat-Lösung versetzt. Man rührt noch 11/2 Stunden, filtriert dann das ausgefallene Natriumchlorid über hochgereinigter Diatomeenerde ab und dampft das Filtrat im Vakuum bei 350 ein, wobei sich nochmals Natriumchlorid ausscheidet. Nach Abfiltrieren wird das Filtrat im Vakuum unter Stickstoff eingedampft und der Rückstand fraktioniert, wobei das Dimethylamino-isopropylhydrazin zum grössten Teil zwischen 54-560 bei 11 mm Hg als farblose Flüssigkeit übergeht.
Zur weiteren Charakterisierung wird das Dipikrat hergestellt, das nach Umkristallisieren aus N Methanol/ Wasser (10: 1) bei 157-160 (Zers.) schmilzt.
Das als Ausgangsmaterial verwendete 1-(Dimethylamino-isopropyl)-2-acetylhydrazin wird wie folgt hergestellt:
136 g Dimethylamino-aceton, 100 g Acetylhydrazin und 400 cm3 Äthanol werden 5 Stunden am Rückfluss zum Sieden erhitzt. Darauf wird abgekühlt, wobei ein Teil des l-(Dimethylamino-isopropyliden)- 2-acetylhydrazins kristallisiert. Nach Abfiltrieren werden aus der Mutterlauge nach Abdampfen des Ätha nols noch weitere Fraktionen an reiner Substanz erhalten. 1- (Dimethylamino - isopropyliden) -2 acetyl - hydrazin schmilzt nach Umkristallisieren aus Äthanol, Essigester oder Dioxan bei 121-122".
Eine Lösung von 20 g 1-(Dimethylamino-isopropyliden)-2-acetylhydrazin in 200 cm3 Methanol wird mit Raney-Nickel und Wasserstoff bei Zimmertempe rasur und einem Überdruck von 20 at geschüttelt, wobei die berechnete Menge Wasserstoff aufgenommen wird. Man filtriert vom Katalysator ab und dampft die Lösung ein. Der feste Rückstand wird destilliert, wobei das 1- (Dimethylamino - isopropyl) -2- acetyl - hydrazin zwischen 137-138 bei 11 mm Hg übergeht.
Nach Umkristallisieren einer Probe aus Hexan schmilzt die Substanz bei 63-65o.
Das Dipikrat schmilzt bei 118-121 (Zers.).
Beispiel 2
30 g 1-(Diäthylamino-isopropyl)-2-acetylhydrazin werden in 450 cm3 6-n. Salzsäure gelöst und unter Stickstoff während 5 Stunden auf 95 erwärmt; Dann wird die Lösung im Vakuum eingedampft, der Rückstand in wenig Wasser gelöst und unter Kühlung mit 50prozentiger wässeriger Kalilauge alkalisch gemacht. Das ausgefallene öl wird in seither aufgenommen, die ätherische Lösung über Magnesiumsulfat getrocknet, eingedampft, und der Rückstand im Vakuum fraktioniert, wobei das Diäthylamino-isopropylhydrazin zwischen 75-77O bei 12 mm Hg als farblose Flüssigkeit übergeht.
Zur weiteren Charakterisierung wird das Dipikrat hergestellt und dieses zweimal aus Methanol umkri stallisiert. Smp. 155-1 57o (Zers.).
Das als Ausgangsmaterial verwendete 1-(Diäthyl amino-isopropyl)-2-acetylhydrazin wird wie folgt hergestellt:
312 g Diäthylamino-aceton, 180 g Acetylhydrazin und 800 cm3 Äthanol werden 5 Stunden am Rückfluss zum Sieden erhitzt. Ein Teil des Äthanols wird abdestilliert, worauf beim Erkalten das 1-(Diäthylamino isopropyliden)-2-aeetylhydrazin kristallisiert. Aus den Mutterlaugen werden noch weitere Fraktionen an reiner Substanz erhalten. Nach Umkristallisieren aus Essigsäureäthylester schmilzt das Hydrazin-Derivat bei 107-108 .
200 g 1- (Diäthylamino - isopropyliden) -2 - acetyl - hydrazin werden in 21 Methanol gelöst und mit Raney-Nickel und Wasserstoff bei Zimmertemperatur und einem Anfangsdruck von 30 at während 24 Stunden geschüttelt, wobei die berechnete Menge Wasserstoff aufgenommen wird. Darauf wird vom Katalysator abfiltriert, das Filtrat eingedampft und der vollständig kristallisierende Rückstand destilliert, wobei das l-(Diäthylamino-isopropyl)-2-acetylhydrazin zwischen 100-105 bei 0,2 mm Hg als leicht gelb gefärbtes Öl übergeht, das in der Vorlage kristallin erstarrt. Eine Probe, umkristallisiert aus Petroläther, schmilzt bei 59-61".
Das Dipikrat schmilzt bei 103-106 (Zers.).
Beispiel 3
2,0 g 1-(Piperidyl-l'-isopropyl)-2-acetylhydrazin werden unter-Stickstoff während 4 y4 Stunden mit wässeriger 6-n. HC1 auf 95o erwärmt. Darauf wird die Lösung im Vakuum bei 500 eingedampft. Der zähe Rückstand wird in kaltem Athanol gelöst, etwas ungelöstes Hydrazin-dihydrochlorid abfiltriert, und das Filtrat langsam mit Äther versetzt. Beim Anreiben kristallisiert nach einiger Zeit rohes (Piperidyl-l-iso propyl) - hydrazin - dihydrochlorid. Die Verbindung schmilzt nach Umkristallisieren aus Athanol/Äther bei 172175" (Zers.).
Zur Herstellung der freien Base werden 11,74 g (Piperidyl-lisopropyl)-hydrazin-dihydrochlorid in wenig Methanol aufgeschlämmt und unter Kühlung mit 48 cm3 2,15-n. Natriummethylat-Lösung versetzt.
Man gibt die gleiche Menge Äther zu, filtriert das ausgefallene Natriumchlorid über hochgereinigter Diatomeenerde ab, dampft die Lösung im Vakuum ein und destilliert den Rückstand im Vakuum, wobei das (Piperidyl-1-isopropyl)-hydrazin zwischen 99-103 bei
10 mm Hg als farblose Flüssigkeit übergeht.
Zur Charakterisierung wird das Dipikrat hergestellt, das nach Umkristallisieren aus Methanol/Wasser (5:1) bei 158-1609 (Zers.) schmilzt.
Das als Ausgangsmaterial verwendete l-(Piperidyl- l'-isopropyl)-2-acetylhydrazin wird wie folgt hergestellt:
Eine Lösung von 62,1 g Piperidyl-l-aceton und 33 g Acetylhydrazin in 200 cm3 Äthanol wird während 6 Stunden am Rückfluss zum Sieden erhitzt. Beim Erkalten kristallisiert schon ein Teil des l-(Piperidyl-l'-isopro- pyliden)-2-acetylhydrazins aus. Nach Abfiltrieren wird die Mutterlauge eingedampft, und der Rückstand nach Behandeln mit Tierkohle aus Essigsäureäthylester ebenfalls kristallisiert. Das l-(Piperidyl-l'-isopropyli- den)-2-acetylhydrazin schmilzt nach Umkristallisieren aus Essigsäureäthylester bei 132-1330.
Eine Lösung von 20 g l-(Piperidyl-l'-isopropyli- den)-2-acetylhydrazin in 200 cm3 Methanol wird mit Raney-Nickel bei Zimmertemperatur und 20 at mit Wasserstoff geschüttelt, bis kein Wasserstoff mehr aufgenommen wird. Darauf wird vom Katalysator abfiltriert und die Lösung eingedampft. Der Rückstand kristallisiert vollständig durch. Das 1-(Piperidyl-l'- isopropyl)-2-acetylhydrazin kristallisiert aus Essigester und schmilzt bei 101-1040. Sdp. 126-130 bei 0,05 mm Hg.
Beispiel 4
Die Lösung von 75 g l-(l'-Dimethylamino-butyl- 3')-2-acetylhydrazin in einem Gemisch aus 400 cm3 konz. Salzsäure und 320 cm3 Wasser wird in einer Stickstoffatmosphäre 6 Stunden auf 110 erwärmt.
Dann wird im Vakuum eingedampft, der Rückstand in wenig Wasser gelöst und die Lösung unter gutem Kühlen mit 50prozentiger Kalilauge stark alkalisch gemacht. Von ausgefallenem Kaliumchlorid wird abfiltriert, und das Filtrat viermal mit einem Gemisch aus gleichen Teilen Ather und Methylenchlorid ausgeschüttelt. Nach Trocknen über Pottasche wird das Lösungsmittel verdampft und der Rückstand im Vakuum fraktioniert destilliert. l-Dimethylamino-butyl- 3-hydrazin destilliert bei 12 mm Hg bei 800. Es ist eine farblose Flüssigkeit; ; nD =1, 4581.
Das Dipikrat kristallisiert aus Methanol/Wasser (5:1) in Prismen, die bei 151-153 unter Zersetzung schmelzen.
Das als Ausgangsmaterial verwendete l-(l'-Di methylamino-butyl-3')-2-acetylhydrazin wird wie folgt hergestellt:
Die Lösung von 14,8 g Acetylhydrazin und 23 Dimethylaminobutanon in 50 cm3 Eisessig wird mit 0,5 g Platinoxyd versetzt und dann zwei Tage bei Normaldruck mit Wasserstoff geschüttelt. Dann wird vom Katalysator abfiltriert, die Lösung im Vakuum eingedampft, der Rückstand mit Eis versetzt und unter guter Kühlung mit 50prozentiger Kalilauge alkalisch gemacht. Dann schüttelt man dreimal mit Chloroform aus, trocknet über Natriumsulfat und dampft ein.
Der Rückstand wird dann fraktioniert destilliert, wobei 1- (1'-Dimethylamino-butyl-3')-2-acetylhydrazin bei 0,2 mm Hg von 102-103 als leicht gelbliches Öl destilliert. n2D = 1,4702.
Beispiel 5
Die Lösung von 52,0 g l-(l'-Diäthylamino-butyl- 3')-2-acetylhydrazin in einem Gemisch von 270 cm3 konz. Salzsäure und 210 cm3 Wasser wird in einer Stickstoffatmosphäre 6 Stunden auf 110 erwärmt.
Dann wird im Vakuum zur Trockne eingedampft, der Rückstand in wenig Wasser gelöst und die Lösung unter gutem Kühlen mit 50prozentiger wässeriger Kalilauge stark alkalisch gemacht. Man gibt anschliessend noch so viel festes KOH zu, bis sich eine ölige Substanz abgeschieden hat. Alsdann wird von ausgefallenem Kaliumchlorid über hochgereinigter Diatomeenerde filtriert und der Filterrückstand mit einem Gemisch aus gleichen Teilen Methylenchlorid und Äther gewaschen. Das Filtrat wird anschliessend noch zweimal mit dem gleichen Lösungsmittelgemisch ausgeschüttelt.
Die vereinigten organischen Fraktionen werden nach Trocknen über Pottasche eingedampft und der Rückstand im Vakuum fraktioniert. 1-Di äthylamino-butyl-(3)-hydrazin destilliert bei 11 mm.Hg bei 95-960. nI= =1,4590.
Das als Ausgangsmaterial verwendete l-(l'-Di äthylanuno-butyl 3')-2-acetylhydrazin wird wie folgt hergestellt:
Die Lösung von 42,8 g 1-Diäthylamino-butan-3on in 150 cm3 Eisessig wird unter öfterem Schütteln mit 22,2 g Acetylhydrazin versetzt. Nach etwa 15ständigem Stehen gibt man 0,5 g Platinoxyd zu und schüttelt etwa 12 Stunden bei Zimmertemperatur und bei einem Druck von 5 at mit Wasserstoff. Darm wird vom Katalysator abfiltriert, die Lösung im Vakuum eingedampft, der ölige Rückstand in wenig Wasser aufgenommen und die Lösung bei 10-20 mit 50prozentiger wässeriger Kalilauge stark alkalisch gemacht. Nach Zusatz von festemKOH wird die Lösung mit Chloroform ausgeschüttelt.
Nach Trocknen über Pottasche wird das Lösungsmittel verdampft, und der Rückstand im Vakuum fraktioniert destilliert. 1-(1' Diäthylamino-butyl-3')-2-acetylhydrazin destilliert bei 0,06 mm Hg bei 115". Es ist eine farblose Flüssigkeit, die an der Luft Kohlendioxyd und Wasser anzieht. und35 = 1,4702.
Beispiel 6
Die Lösung von 35,0 g 1-[1'-(N-Methyl-N-isopro- pyl)-aminobutyl-37-2-acetylhydrazin in einem Gemisch von 180 cm3 konz. Salzsäure und 140 cms Wasser wird in einer Stickstoffatmosphäre 6 Stunden erwärmt. Dann wird im Vakuum zur Trockne eingedampft, der Rückstand in wenig Wasser gelöst und die Lösung unter I Kühlung mit 50prozentiger wässe- riger Kalilauge stark alkalisch gemacht. Man gibt anschliessend noch so viel festes KOH zu, bis sich eine ölige Substanz abgeschieden hat, die in Chloroform aufgenommen wird. Alsdann wird vom ausgefallenen Kaliumchlorid über hochgereinigter Diatomeenerde filtriert, und der Filterrückstand mit Chloroform gründlich ausgewaschen.
Das Filtrat wird anschliessend noch mit dem gleichen Lösungsmittel ausgeschüttelt.
Die vereinigten organischen Fraktionen werden nach Trocknen über Magnesiumsulfat im Vakuum ein geengt, und der Rückstand destilliert. 1-[1'-(N-Methyl- N-isopropyl)-aminobutyl- 3'] -hydrazin destilliert bei einem Druck von 11 mm Hg bei 96-98".
Das als Ausgangsmaterial verwendete 1-[1'-(N Methyl - N-isopropyl)-aminobutyl-3']-2-acetylhydrazin wird wie folgt hergestellt:
Die Lösung von 36,3 g N-Methyl-N-isopropyl- aminobutan-3-on in 220 cm3 Eisessig wird unter gu- tem Schütteln und Kühlen mit 18,8 g Acetylhydrazin versetzt. Nach Stehen über Nacht wird mit 0,43 g Platinoxyd und Wasserstoff bei Zimmertemperatur und einem Anfangsdruck von 5 at geschüttelt, wobei die berechnete Menge Wasserstoff aufgenommen wird.
Dann wird vom Katalysator abfiltriert und nach Einengen der Lösung im Vakuum mit 50prozentiger wässeriger Kalilauge stark alkalisch gemacht. Nach Zusatz von festem KOH wird das ausgeschiedene Ö1 mit Chloroform extrahiert. Man trocknet den Chloroformextrakt über Magnesiumsulfat, verdampft das Lösungsmittel und destilliert den Rückstand, wobei das 1 - [1'- (N-Methyl-N-isopropyl)-aminobutyl-3']-2acetylhydrazin unter einem Druck von 0,3 mm Hg bei 139-147 destilliert.
Das als Ausgangsprodukt verwendete N-Methyl N-isopropyl-aminobutan-3 -on wird hergestellt, indem man 64,0 g N-Methyl-N-isopropylamin-hydrochlorid mit 175 cm3 Aceton und 24,4 g Paraformaldehyd in 116 cm3 Athanol während 16 Stunden zum Sieden am Rückfluss erhitzt. Nach Eindampfen der Lösung im Vakuum wird der Rückstand mit etwa 150 cm3 Wasser versetzt, mit 50prozentiger wässeriger Kalilauge alkalisch gemacht, und die Lösung mehrmals mit Äther extrahiert. Die vereinigten Ätherauszüge werden über Magnesiumsulfat getrocknet, eingedampft, und der Rückstand im Vakuum fraktioniert destilliert.
N-Methyl-N-isopropyl-aminobutan-3 on siedet bei einem Druck von 11 mg Hg bei 63-640. und =1,4342.
Beispiel 7
4,4 g g 1-(2'-Diäthylamino-isohexyl-1')-2-acetyl- hydrazin werden in 20 cm3 wässeriger 20prozentiger Salzsäure während 5 Stunden am Rückfluss in einer Stickstoffatmosphäre auf 1200 erhitzt. Nach Abkühlen wird die Reaktionslösung mit Pottasche alkalisch gemacht und gesättigt, durch hochgereinigte Diatomeenerde filtriert und das Filtrat mit total 200 cm3 Chloroform extrahiert. Nach Trocknen des Extraktes über Magnesiumsulfat wird das Chloroform im Vakuum bei 30O entfernt, und der Rückstand im inerten Gasstrom destilliert, wobei das 1-(2'-Diäthylamino-iso- hexyl-l')
-hydrazin zwischen 105-108 bei 9 mm Hg als farblose Flüssigkeit überdestilliert. n =1,4540.
Das als Ausgangsmaterial verwendete 1-(2'-Di äthylamino-isohexyl-1')-2-acetylhydrazin wird wie folgt hergestellt:
Zu einer Lösung von 9,6 g Acetylhydrazin in 120 cm3 Wasser lässt man unter Rühren bei einer Temperatur von 18-20 innert 20 Minuten 29,5 g 2-Diäthylamino-isohexylchlorid zutropfen. Anschlie ssend wird das Reaktionsgemisch während 100 Stunden bei Zimmertemperatur weitergerührt. Gegen Ende der Reaktionszeit hat sich die Emulsion in eine klare Lösung umgewandelt. Die Reaktionslösung wird mit Pottasche gesättigt und mit total 350 cm3 Chloroform extrahiert.
Nach Trocknen der vereinigten Chloroformauszüge über Maguesiumsulfat wird das Chloroform im Hochvakuum abgedampft und der Rückstand im Hochvakuum fraktioniert, wobei das 1-(2'-Diäthyl amino-isohexyl-1')-2-acetylhydrazin zwischen 125 bis 1300 bei 0,08 mm Hg übergeht. nD2 =1,4668.
Beispiel 8
3,1 g 1-(ss-Diäthylamino-äthyl)-2-acetylhydrazin werden in 15 cm3 wässeriger 20prozentiger Salzsäure während 5 Stunden am Rückfluss in einer Stickstoffatmosphäre auf 100" erhitzt. Nach Abkühlen wird die Reaktionslösung mit Pottasche alkalisch gemacht und gesättigt, durch hochgereinigte Diatomeenerde filtriert und das Filtrat mit total 150 cm3 Chloroform extrahiert. Nach dem Trocknen des Extraktes über Magnesiumsulfat wird das Chloroform im Vakuum entfernt, und der Rückstand im inerten Gasstrom destilliert, wobei das ss-Diäthylamino-äthylhydrazin zwischen 76 bis 77O bei 9 mm Hg übergeht. n2r, 2 = 1, 4590.
Das als Ausgangsmaterial verwendete l-(ss-Di- äthylamino-äthyl)-2-acetylhydrazin wird wie folgt hergestellt:
Zu einer Lösung von 6,6 g Acetylhydrazin in 40 cm3 Wasser lässt man unter Rühren bei einer Temperatur von 18-200 innert 15 Minuten 12,0 g ss-Di- äthylamino-äthylchlorid zutropfen, wobei sich das Reaktionsgemisch trübt. Nach Entfernen der Kühlung tritt leichte Erwärmung ein, und es entsteht eine klare Lösung. Zur Beendigung der Reaktion wird noch 24 Stunden bei Zimmertemperatur weitergerührt.
Darauf wird die Reaktionslösung unter starkem Kühlen mit Pottasche gesättigt und mit total 175 cm3 Chloroform extrahiert. Nach Trocknen der vereinigten Chloroformextrakte über Magnesiumsulfat wird das Chloroform im Vakuum entfernt und der Rückstand im Hochvakuum destilliert, wobei das l-(ss-Diäthyl- amino-äthyl)-2-acetylhydrazin zwischen 120-1220 bei 0,02 mm Hg als farbloses Öl übergeht. nD = 1,4699.
Zur weiteren Charakterisierung wird das saure Naphthalin-1,5-disulfonat hergestellt. Smp. 248-2490 (Zers.) nach Umkristallisieren aus Wasser/Methanol.
Beispiel 9
3,0 g 1-(3'-Amino-n-propyl)-2-benzoylhydrazin werden in 15 cm3 20prozentiger wässeriger Salzsäure während 6 Stunden am Rückfluss auf 110" erhitzt.
Darauf werden 30 cm3 Wasser zugegeben, die Reak tionsmischung gekühlt, die ausgeschiedene Benzoesäure abfiltriert und das Filtrat im Vakuum zur Trockne eingedampft. Zur vollständigen Entfernung des Wassers versetzt man den Rückstand mit 20 cm3 abs. Äthanol und verdampft das Lösungsmittel nochmals im Vakuum zur Trockne. Der kristalline Rückstand, das 1-(3 '-Amino-n-propyl)-hydrazin-dihydro- chlorid, wird aus Methanol/Äther umkristallisiert und schmilzt bei 119-121 (Zers.).
Das als Ausgangsmaterial verwendete l-(3'-Amino- n-propyl)-2-benzoylhydrazin wird wie folgt hergestellt:
Zu einer Lösung von 11,0 g Benzoylhydrazin in 20 cm3 Wasser lässt man unter Rühren bei einer Temperatur von 100" innert 45 Minuten 4,4 g Acrylnitril zutropfen. Anschliessend wird unter weiterem Rühren die Temperatur noch während 2 Stunden auf 90O gehalten. Beim Abkühlen scheidet sich das Reaktionsprodukt kristallin aus. Man versetzt mit Wasser und kristallisiert das 1-(2'-Cyanoäthyl)-2-benzoylhydrazin direkt ohne weitere Reinigung aus Wasser um. Smp.

Zu einer auf 0O abgekühlten Suspension von 2,2 g LiA1H4 in 120 cm3 abs. Tetrahydrofuran lässt man langsam eine Lösung von 6,0 g 1-(2'-Cyanoäthyl)-2 benzoylhydrazin in 40 cm3 Tetrahydrofuran so zutropfen, dass die Temperatur nie über +10 steigt.
Darauf wird das Reaktionsgemisch noch 11/2 Stunden zum Sieden am Rückfluss erhitzt. Nach Abkühlen gibt man unter weiterem Rühren 4 cm5 Wasser und 16 cm3 Sprozentige wässerige Natronlauge zu und rührt anschliessend noch 15 Minuten bei Zimmertemperatur.
Sodann wird filtriert, der Filterrückstand mehrmals gut mit abs. Tetrahydrofuran ausgewaschen und das Filtrat im Vakuum zur Trockne verdampft. Der Rückstand, das 1 (3'-Amino-n-propyl)-2-benzoylhydrazin, wird in wenig abs. Methanol aufgenommen und daraus mit methanolischer Salzsäure das Dihydrochlorid hergestellt. Smp. 224-2260 (Zers.).
Beispiel 10
Eine Lösung von 2,7 g 1-(3'-Amino-n-propyl)-2acetylhydrazin in 13,5 cm3 20prozentiger wässeriger Salzsäure wird während 6 Stunden am Rückfluss auf 110 erhitzt. Darauf wird die Reaktionslösung im Vakuum zur Trockne eingedampft, der kristalline Rückstand zur vollständigen Entfernung des Wassers noch zweimal mit je 30 cm3 abs. Äthanol versetzt, und das Lösungsmittel im Vakuum verdampft. Der Rückstand, das 1-(3'-Amino-n-propyl)-hydrazin-dihydro- chlorid, wird aus Methanol/Ather umkristallisiert.
Smp. 117-119 (Zers.). Der Mischschmelzpunkt mit einem Präparat, hergestellt nach Beispiel 9 durch Verseifung von 1-(3 '-Amino-n-propyl)-2-benzoylhydrazin, zeigt keine Depression.
Das als Ausgangsmaterial verwendete 1 (3'-Amino- n-propyl)-2-acetylhydrazin wird wie folgt hergestellt:
Zu einer Lösung von 24,0 g Acetylhydrazin in 80 cm3 Wasser werden unter Rühren bei einer Temperatur von 90O innert 45 Minuten 17,6 g Acrylnitril zugegeben. Anschliessend wird unter weiterem Rühren die Temperatur noch 2 Stunden auf 1000 gehalten.
Sodann werden im Vakuum etwa 40 cm3 Wasser abgedampft, der Destillationsrückstand mit Kochsalz gesättigt und mit total 180 cm3 Chloroform extrahiert.
Nach dem Trocknen des Extraktes über Magnesiumsulfat wird das Chloroform im Vakuum entfernt und der Rückstand destilliert, wobei das 1-(2'-Cyano äthyl) 2-acetylhydrazin zwischen 135-1402 bei 0,1 mm Hg übergeht und in der Vorlage kristallin erstarrt. Die Verbindung lässt sich aus Essigester/ Petroläther umkristallisieren und schmilzt dann bei 54-560.
7,3 g 1-(2'-Cyanoäthyl)-2-acetylhydrazin, gelöst in 145 cm3 bei 0O mit Ammoniakgas gesättigtem Methanol, werden mit Raney-Nickel 2 Stunden bei 800 und einem Anfangsdruck von 100 at hydriert. Nach Abfiltrieren vom Katalysator wird das Methanol im Vakuum entfernt und der Rückstand im Hochvakuum destilliert, wobei das 1-(3 '-Amino-n-propyl)-2-acetyl- hydrazin zwischen 12w125 bei 0,003 mm Hg überdestilliert. Zur Charakterisierung wird das Naphthalin1,5-disulfonat hergestellt, das nach Umkristallisieren aus Methanol/Ather bei 185-186 (Zers.) schmilzt.

According to the invention, the new hydrazine derivatives are prepared by converting acylated hydrazine derivatives of the formula II,

where R3 is an acyl group which splits off the acyl group hydrolytically.
This cleavage can be carried out by heating an acylated hydrazine of the formula II, the preparation of which is described in detail in the following examples, with an aqueous mineral acid, preferably with aqueous hydrochloric acid, for 4 to 6 hours at a temperature of 80-120 . The deacylation is advantageously carried out in a nitrogen atmosphere. The end products are isolated and purified by customary methods and, if necessary, converted into a suitable salt.
The new hydrazine derivatives prepared according to the invention are liquid at room temperature and can be distilled without decomposition in vacuo. They are basic compounds which, with inorganic or organic acids, form stable salts that crystallize at room temperature.
The new process products serve as intermediate products in the manufacture of drugs.
However, they themselves also have pharmacodynamic properties that can be used therapeutically, which make them valuable drugs. For example, they are characterized by a strong monoamine oxidase inhibitory effect.
In the following examples, the temperatures are given in degrees Celsius.
example 1
60 g of 1- (dimethylamino-isopropyl) -2-acetylhydrazine are concentrated in a mixture of 500 cm3. Dissolved hydrochloric acid and 400 cm3 of water and then heated to 115 to 1200 in a nitrogen atmosphere for 5 hours. The solution is then evaporated in vacuo at 500, the viscous residue is dissolved in ethanol, the dimethylaminoisopropylhydrazine dihydrochloride crystallizing out on trituration. M.p.

This compound is also formed when 1- (dimethylamino-isopropyl) -2-benzoylhydrazine [mp. of the dihydrochloride 210-220 (decomposition under the reaction conditions given above.
To prepare the free base, 154 g of crude, well-dried dimethylaminoisopropylhydrazine dihydrochloride are suspended in 200 cm3 of methanol and, with stirring and cooling, 7-90 cm3 of 2.1N. Sodium methylate solution added. The mixture is stirred for a further 11/2 hours, the precipitated sodium chloride is then filtered off over highly purified diatomaceous earth and the filtrate is evaporated in vacuo at 350, whereupon sodium chloride separates out again. After filtering off, the filtrate is evaporated in vacuo under nitrogen and the residue is fractionated, the majority of the dimethylaminoisopropylhydrazine passing over as a colorless liquid between 54-560 at 11 mm Hg.
For further characterization, the dipikrate is produced which, after recrystallization from N methanol / water (10: 1), melts at 157-160 (decomp.).
The 1- (dimethylamino-isopropyl) -2-acetylhydrazine used as starting material is prepared as follows:
136 g of dimethylamino-acetone, 100 g of acetylhydrazine and 400 cm3 of ethanol are refluxed for 5 hours. It is then cooled, with part of the 1- (dimethylamino-isopropylidene) -2-acetylhydrazine crystallizing. After filtration, further fractions of pure substance are obtained from the mother liquor after evaporation of the ethanol. 1- (Dimethylamino-isopropylidene) -2 acetyl-hydrazine melts at 121-122 "after recrystallization from ethanol, ethyl acetate or dioxane.
A solution of 20 g of 1- (dimethylamino-isopropylidene) -2-acetylhydrazine in 200 cm3 of methanol is shaken with Raney nickel and hydrogen at room temperature and an overpressure of 20 atm, the calculated amount of hydrogen being absorbed. The catalyst is filtered off and the solution is evaporated. The solid residue is distilled, the 1- (dimethylamino-isopropyl) -2-acetyl-hydrazine passing over between 137-138 at 11 mm Hg.
After recrystallizing a sample from hexane, the substance melts at 63-65o.
The dipicrate melts at 118-121 (dec.).
Example 2
30 g of 1- (diethylamino-isopropyl) -2-acetylhydrazine are in 450 cm3 of 6-n. Dissolved hydrochloric acid and heated to 95 for 5 hours under nitrogen; Then the solution is evaporated in vacuo, the residue is dissolved in a little water and made alkaline with 50 percent aqueous potassium hydroxide solution while cooling. The precipitated oil has since been absorbed, the ethereal solution dried over magnesium sulfate, evaporated, and the residue fractionated in vacuo, the diethylaminoisopropylhydrazine passing over as a colorless liquid between 75-77O at 12 mm Hg.
For further characterization, the dipikrate is produced and this is recrystallized twice from methanol. M.p. 155-1 57o (dec.).
The 1- (diethylamino-isopropyl) -2-acetylhydrazine used as starting material is prepared as follows:
312 g of diethylamino-acetone, 180 g of acetylhydrazine and 800 cm3 of ethanol are heated to boiling under reflux for 5 hours. Some of the ethanol is distilled off, whereupon the 1- (diethylamino isopropylidene) -2-aetylhydrazine crystallizes on cooling. Further fractions of pure substance are obtained from the mother liquors. After recrystallization from ethyl acetate, the hydrazine derivative melts at 107-108.
200 g of 1- (diethylamino-isopropylidene) -2-acetyl-hydrazine are dissolved in 21% methanol and shaken with Raney nickel and hydrogen at room temperature and an initial pressure of 30 atm for 24 hours, the calculated amount of hydrogen being absorbed. The catalyst is then filtered off, the filtrate is evaporated and the residue, which has completely crystallized, is distilled, the l- (diethylamino-isopropyl) -2-acetylhydrazine passing over between 100-105 at 0.2 mm Hg as a pale yellow oil that is in the template solidified crystalline. A sample, recrystallized from petroleum ether, melts at 59-61 ".
The dipicrate melts at 103-106 (dec.).
Example 3
2.0 g of 1- (piperidyl-l'-isopropyl) -2-acetylhydrazine are under nitrogen for 4 and 4 hours with aqueous 6-n. HC1 heated to 95o. The solution is then evaporated in vacuo at 500. The viscous residue is dissolved in cold ethanol, some undissolved hydrazine dihydrochloride is filtered off, and the filtrate is slowly mixed with ether. When rubbed, crude (piperidyl-1-isopropyl) hydrazine dihydrochloride crystallizes after a while. After recrystallization from ethanol / ether, the compound melts at 172175 "(decomp.).
To prepare the free base, 11.74 g of (piperidyl lisopropyl) hydrazine dihydrochloride are slurried in a little methanol and, while cooling, with 48 cm 3 of 2.15 n. Sodium methylate solution added.
The same amount of ether is added, the precipitated sodium chloride is filtered off over highly purified diatomaceous earth, the solution is evaporated in vacuo and the residue is distilled in vacuo, the (piperidyl-1-isopropyl) hydrazine between 99-103 at
10 mm Hg passes over as a colorless liquid.
For characterization, the dipikrate is produced which, after recrystallization from methanol / water (5: 1), melts at 158-1609 (decomp.).
The l- (piperidyl- l'-isopropyl) -2-acetylhydrazine used as starting material is prepared as follows:
A solution of 62.1 g of piperidyl-1-acetone and 33 g of acetylhydrazine in 200 cm 3 of ethanol is refluxed for 6 hours. On cooling, some of the 1- (piperidyl-l'-isopropylidene) -2-acetylhydrazine crystallizes out. After filtering off, the mother liquor is evaporated and the residue is likewise crystallized from ethyl acetate after treatment with animal charcoal. The l- (piperidyl-l'-isopropylidene) -2-acetylhydrazine melts after recrystallization from ethyl acetate at 132-1330.
A solution of 20 g of l- (piperidyl-l'-isopropylidene) -2-acetylhydrazine in 200 cm3 of methanol is shaken with Raney nickel at room temperature and 20 atm with hydrogen until no more hydrogen is absorbed. The catalyst is then filtered off and the solution is evaporated. The residue crystallizes completely. The 1- (piperidyl-l'-isopropyl) -2-acetylhydrazine crystallizes from ethyl acetate and melts at 101-1040. Bp 126-130 at 0.05 mm Hg.
Example 4
The solution of 75 g of l- (l'-dimethylamino-butyl-3 ') - 2-acetylhydrazine in a mixture of 400 cm3 of conc. Hydrochloric acid and 320 cm3 of water are heated to 110 for 6 hours in a nitrogen atmosphere.
It is then evaporated in vacuo, the residue is dissolved in a little water and the solution is made strongly alkaline with 50 percent potassium hydroxide solution while cooling well. The precipitated potassium chloride is filtered off, and the filtrate is extracted four times with a mixture of equal parts of ether and methylene chloride. After drying over potash, the solvent is evaporated and the residue is fractionally distilled in vacuo. l-Dimethylamino-butyl-3-hydrazine distills at 12 mm Hg at 800. It is a colorless liquid; ; nD = 1.4581.
The dipicrate crystallizes from methanol / water (5: 1) in prisms which melt at 151-153 with decomposition.
The l- (l'-dimethylamino-butyl-3 ') -2-acetylhydrazine used as starting material is prepared as follows:
The solution of 14.8 g of acetylhydrazine and 23 dimethylaminobutanone in 50 cm3 of glacial acetic acid is mixed with 0.5 g of platinum oxide and then shaken with hydrogen for two days at normal pressure. The catalyst is then filtered off, the solution is evaporated in vacuo, the residue is mixed with ice and made alkaline with 50 percent potassium hydroxide solution while cooling well. Then it is extracted three times with chloroform, dried over sodium sulfate and evaporated.
The residue is then fractionally distilled, with 1- (1'-dimethylamino-butyl-3 ') -2-acetylhydrazine distilling at 0.2 mm Hg from 102-103 as a slightly yellowish oil. n2D = 1.4702.
Example 5
The solution of 52.0 g of l- (l'-diethylamino-butyl- 3 ') - 2-acetylhydrazine in a mixture of 270 cm3 of conc. Hydrochloric acid and 210 cm3 of water are heated to 110 for 6 hours in a nitrogen atmosphere.
It is then evaporated to dryness in vacuo, the residue is dissolved in a little water and the solution is made strongly alkaline with 50 percent aqueous potassium hydroxide solution while cooling well. You then add enough solid KOH until an oily substance has separated out. The precipitated potassium chloride is then filtered through highly purified diatomaceous earth and the filter residue is washed with a mixture of equal parts of methylene chloride and ether. The filtrate is then extracted twice more with the same solvent mixture.
After drying over potash, the combined organic fractions are evaporated and the residue is fractionated in vacuo. 1-Diethylamino-butyl- (3) -hydrazine distills at 11 mm.Hg at 95-960. nI = = 1.4590.
The l- (l'-diethylanuno-butyl 3 ') - 2-acetylhydrazine used as starting material is prepared as follows:
The solution of 42.8 g of 1-diethylamino-butan-3one in 150 cm3 of glacial acetic acid is mixed with 22.2 g of acetylhydrazine, with frequent shaking. After standing for about 15 hours, 0.5 g of platinum oxide is added and the mixture is shaken with hydrogen for about 12 hours at room temperature and at a pressure of 5 atm. The catalyst is then filtered off, the solution evaporated in vacuo, the oily residue taken up in a little water and the solution made strongly alkaline at 10-20 with 50 percent aqueous potassium hydroxide solution. After adding solid KOH, the solution is extracted with chloroform.
After drying over potash, the solvent is evaporated and the residue is fractionally distilled in vacuo. 1- (1 'Diethylamino-butyl-3') -2-acetylhydrazine distills at 0.06 mm Hg at 115 ". It is a colorless liquid that attracts carbon dioxide and water in the air. And 35 = 1.4702.
Example 6
The solution of 35.0 g of 1- [1 '- (N-methyl-N-isopropyl) -aminobutyl-37-2-acetylhydrazine in a mixture of 180 cm3 of conc. Hydrochloric acid and 140 cms of water are heated in a nitrogen atmosphere for 6 hours. It is then evaporated to dryness in vacuo, the residue is dissolved in a little water and the solution is made strongly alkaline while cooling with 50 percent aqueous potassium hydroxide solution. Then enough solid KOH is added until an oily substance has separated out, which is taken up in chloroform. The precipitated potassium chloride is then filtered through highly purified diatomaceous earth, and the filter residue is washed out thoroughly with chloroform.
The filtrate is then extracted with the same solvent.
The combined organic fractions are dried over magnesium sulfate and concentrated in vacuo, and the residue is distilled. 1- [1 '- (N-Methyl-N-isopropyl) -aminobutyl-3'] -hydrazine distilled at a pressure of 11 mm Hg at 96-98 ".
The 1- [1 '- (N methyl - N-isopropyl) aminobutyl-3'] - 2-acetylhydrazine used as starting material is prepared as follows:
The solution of 36.3 g of N-methyl-N-isopropyl-aminobutan-3-one in 220 cm3 of glacial acetic acid is admixed with 18.8 g of acetylhydrazine, with good shaking and cooling. After standing overnight, the mixture is shaken with 0.43 g of platinum oxide and hydrogen at room temperature and an initial pressure of 5 atm, the calculated amount of hydrogen being absorbed.
The catalyst is then filtered off and, after concentrating the solution in vacuo, it is made strongly alkaline with 50 percent aqueous potassium hydroxide solution. After adding solid KOH, the oil which has separated out is extracted with chloroform. The chloroform extract is dried over magnesium sulphate, the solvent is evaporated and the residue is distilled, the 1 - [1'- (N-methyl-N-isopropyl) -aminobutyl-3 '] -2acetylhydrazine under a pressure of 0.3 mm Hg 139-147 distilled.
The N-methyl N-isopropyl-aminobutan-3-one used as the starting product is prepared by adding 64.0 g of N-methyl-N-isopropylamine hydrochloride with 175 cm3 of acetone and 24.4 g of paraformaldehyde in 116 cm3 of ethanol for 16 Heated to reflux for hours. After evaporation of the solution in vacuo, the residue is mixed with about 150 cm3 of water, made alkaline with 50% aqueous potassium hydroxide solution, and the solution is extracted several times with ether. To a solution of 9.6 g of acetylhydrazine in 120 cm3 of water, 29.5 g of 2-diethylamino-isohexyl chloride are added dropwise with stirring at a temperature of 18-20 minutes. The reaction mixture is then stirred for a further 100 hours at room temperature. Towards the end of the reaction time, the emulsion has turned into a clear solution. The reaction solution is saturated with potash and extracted with a total of 350 cm3 of chloroform.

Example 9
3.0 g of 1- (3'-amino-n-propyl) -2-benzoylhydrazine are refluxed at 110 "in 15 cm3 of 20 percent aqueous hydrochloric acid for 6 hours.
30 cm3 of water are then added, the reaction mixture is cooled, the benzoic acid which has separated out is filtered off and the filtrate is evaporated to dryness in vacuo. 4.4 g of acrylonitrile are added dropwise to a solution of 11.0 g of benzoylhydrazine in 20 cm3 of water while stirring at a temperature of 100 "over 45 minutes.
