Process for the preparation of new N-heterocyclic compounds The present invention relates to a process for the preparation of new N-heterocyclic compounds with valuable pharmacological properties.
10,11-dihydro-5H-dibenzo (b, f) azepine and 5H-dibenzo (b, f) azepine monoalkylated in an aromatic ring and derivatives thereof have not yet become known.
It has now been found that N-substituted derivatives of such compounds of the formula

wherein R is an alkyl radical with 2-6 carbon atoms, X is the ethylene or vinylene group, Y is an alkylene radical with 2-6 carbon atoms, of which 2-4. in the direct chain between N and Am, and Am is a low molecular weight dialkylamino group, where one of the two alkyl radicals of Am with the alkylene radical Y,
or both alkyl radicals can be linked directly or via an oxygen atom, an alkyl imino, hydroxyalkylimino or alkanoyloxyalkyl imino group, have valuable pharmacological properties, in particular antiallergic and sedative activity, and u. a. for the treatment of certain forms of mental illnesses, in particular mood depressions, as well as for potentiating the effect of other medicinal substances, in particular narcotics, come into consideration.
Quaternary ammonium salts, which are derived from the tertiary bases defined above, act as ganglioplegics. The process according to the invention for preparing the new compounds is characterized in that a compound of the formula

and reaction of the resulting 3-alkyl-5-chlorocarbonyl-iminodibenzyl or -iminostilbene with an amino alcohol of the formula HO-Y-Am IV.
By addition of reactive esters, especially halides or sulfates of aliphatic or araliphatic alcohols, e.g.
B. of methyl iodide, dimethyl sulfate, ethyl bromide, ethyl iodide or benzyl chloride, monoquaternary ammonium compounds can be obtained from the tertiary amines of the formula I in the usual way, the group Am reacting.
With inorganic or organic acids, such as hydrochloric acid, hydrobromic acid, sulfuric acid, phosphoric acid, methanesulfonic acid, ethane disulfonic acid, acetic acid, citric acid, malic acid, succinic acid, fumaric acid, maleic acid, tartaric acid, benzoic acid and phthalic acid, the tertiary bases form salts, some of which are water-soluble.
In the following example, parts mean parts by weight; these relate to parts by volume as g to cm3. The temperatures are given in degrees Celsius.
Example 30 parts of 5-chlorocarbonyl-3-n-propyl-imino-dibenzyl (prepared from 3-n-propyliminodibenzyl and phosgene) in 50 parts by volume of abs. Benzene are added dropwise to 25 parts of 3-dimethylaminopropanol within one hour. After the dropwise addition, the reaction mixture is refluxed for 16 hours. Then you cool it down and dissolve it with water.
The benzene layer is separated off, washed thoroughly with water and the basic components are extracted with 2N hydrochloric acid. The acidic solution is made alkaline with potassium carbonate solution and extracted with ether. After drying, the solvent is distilled off. The residue is the crude 5- (γ-dimethylamino-carbo-propoxy) -3-n-propyl-iminodibenzyl.
30 parts of the above crude product are heated in an oil bath to 230-240 for about 6 hours until the evolution of carbon dioxide has ceased. The residue is then distilled in vacuo.
The 5 - (y - dimethylamino - propyl) - 3 n - propyl iminodibenzyl passes under 0.001 mm pressure at 142 to 144.
