Mit arbeiter, J. Amer. Chem. Soc. 78, 2023 1956)], dass ein Lacton der Formel 1, in welcher R1, R2, R4 und R6 für Wasserstoff, R3 für Methoxy und R5 für Methyl stehen, erhalten wird, wenn man das Lactam der Formel II in Form seines Methylester-acetates mit Phosphoroxychlorid behandelt,
das entstandene Dehydro-reserpsäure-methylester-acetat direkt mit Natriumborhydrid reduziert, das gebildete Iso-reserp- säure-methylester-acetat mit methanolischer Kali lauge verseift, und die entstandene Iso-reserpsäure durch Behandeln mit N,N'-Dizyklohexyl-carbodiimid in das gewünschte Lacton überführt.
Das erwähnte Verfahren hat nun den Nachteil, d'ass durch die Bildung des Lactonringes auf einer späten Stufe der Synthese, nämlich nach dem Aufbau des ein basisches Stickstoffatom enthaltenden penta- zyklischen Systems, man wegen der Empfindlichkeit dieser Verbindung gegen die üblichen Lactonisie- rungsreagenzien, wie Acetanhydrid, usw., gezwungen ist, N,N'-Dizyklo-carbodiimid für die Lactonisierung zu verwenden,
was unwirtschaftlich und unergiebig ist. Die Ausbeuten betragen etwa 40%.
Dieser Nachteil kann vermieden werden, wenn man erfindungsgemäss die Lactonisierung auf der Lactamstufe vornimmt, welche gegen energische Lactonisierungsbedingungen, wie Erhitzen mit Acet- anhydrid/Natriumacetat, völlig stabil ist, und das ge wünschte Lacton in Ausbeuten von 80-909/o lie fert.
Ein weiterer Vorteil des erfindungsgemässen Ver fahrens besteht darin, dass das bisher unbekannte 3 - Dehydro - reserpsäure-lacton überraschenderweise vorzügliche Eigenschaften besitzt und leicht in kri stalliner Form als freie Base isoliert und gereinigt werden kann. Dies war nicht vorauszusehen, denn in der Literatur werden allgemein 3-Dehydro-Verbin- dungen von Reserpsäure- oder Yohimbin-Derivaten als instabile und nur in Form ihrer Salze isolierbare Verbindungen beschrieben.
Die nach dem vorliegenden Verfahren hergestell ten Verbindungen sind bei Zimmertemperatur kri stallin und stellen wertvolle Zwischenprodukte dar. Sie sollen zur Herstellung von therapeutisch wich tigen Substanzen, z. B. für die Synthese des Re- serpins und seiner Analoga, Verwendung finden.
In den nachfolgenden Beispielen erfolgen alle Temperaturangaben in Celsiusgraden. Die Schmelz punkte sind urkorrigiert. <I>Beispiel 1</I> Iso-deserpidsäure-lacton a) Lacton-lactam: 0,644 g Oxysäure - lactam (Formel II: R,-R4 und R6 = H, R5 = CH3) (her gestellt nach Schweiz.
Patent Nr. 360058) und 0,1 g wasserfreies Natriumacetat werden in einer Lösung von 1,3 cm3 Acetanhydrid in 30 cm?, Benzol 15 Stunden am Rückfluss zum Sieden erhitzt, wobei die Substanz innert einer Stunde in Lösung geht. Nach dem Abkühlen wird im Vakuum zur Trockne ver dampft, und der Rückstand zwischen Natriumhydro- gencarbonatlösung und Chloroform verteilt.
Aus dem amorphen Rückstand der Chlöroformlösung (0,65 g) kristallisiert das Lacton-lactam in Prismen vom Smp. 177-178 (aus Essigester). [a120 = -36 (c = 0,2 in Pyridin). Ausbeute 0,497 g oder 810/0 der Theorie.
b) 3-Dehydro-deserpidsäure-lacton: 0,25 g des unter a) beschriebenen Lacton-lactams erhitzt man in einer Lösung von 8 cm3 frisch destilliertem Phosphoroxychlorid während 2 Stunden in einer Stickstoffatmosphäre am Rückfluss. Die gelbe Lö sung verdampft man im Vakuum zur Trockne, löst den Rückstand in Methylenchlorid, schüttelt die Lö sung mit verdünnter Ammoniaklösung aus, trocknet über Natriumsulfat,
worauf beim Einengen und Ver drängen des Methylenehlorids durch Methanol 3-De- hydro-deserpidsäure-lacton in Nadeln vom Smp. 309 bis 310 kristallisiert. [a]20 = + 13l (c = 0,3 in Pyridin); [a]20 = + 16,5 (c = 0,2 in Chloroform). Ausbeute 1,72g oder 9019/o der Theorie.
c) Iso-deserpidsäure-lacton: 3,3 g 3-Dehydro- deserpidsäure-lacton löst man in 90 ein-- 50 % iger wässeriger Essigsäure und schüttelt mit 0,35 g vor hydriertem Platinkatalysator in einer Wasserstoff atmosphäre, wobei innert 41/2 Stunden nahezu die für 1 Mol Wasserstoff berechnete Menge aufgenom men worden ist.
Nach beendeter Wasserstoff aufnahme wird vom Katalysator filtriert, das Filtrat im Vakuum stark eingeengt, der Rückstand in Me- thylenchlorid gelöst, die Lösung mit verdünntem Ammoniak ausgeschüttelt, über Natriumsulfat ge trocknet und im Vakuum zur Trockne verdampft. Der Rückstand liefert aus Methanol Iso-deserpid- säure-lacton der Formel I, in welcher R,-R4 und R6 = H und R5 =<B>CH,</B> bedeuten.
Nadeln, Smp. 225 bis 227 . [a]20 = -93 (Pyridin). Ausbeute 3,06 g oder 9219/o der Theorie.
<I>Beispiel 2</I> 10-Benzyloxy-iso-deserpidsäure@lacton a) Lacton-lactam: 1,04 g Oxysäure-lactam (For mel II: R1, R3, R4, R6 = H, R#, = OCH.2 - C.H5 und R5 = CH.) werden, wie in Beispiel 1 beschrieben, in Benzol mit Acetanhydrid/Natriumacetat behandelt.
Man erhält das Lacton-lactam in hexagonalen Säulen vom Smp. 198-200 (aus Essigester). [19]D =-31 (c - 0,3 in Pyridin). Ausbeute 0,89 g oder 8911/o der Theorie.
b) 3-Dehydro-10-benzyloxy-deserpidsäure-lacton: 0,8 g des unter a) beschriebenen Lacton-lactams werden, wie in Beispiel 1 beschrieben, mit Phos- phoroxychlorid behandelt und aufgearbeitet. Man erhält die Verbindung aus Methanol. Feine Nadeln vom Smp. 260-263 . [19]D = -- 104 (c = 0,3 in Pyridin). Ausbeute 0,56 g oder 7319/o der Theorie.
c) 10-Benzyloxy-iso-deserpidsäure-lacton: 1,0 g 10-Benzyloxy-dehydro-deserpidsäure-lacton löst man in einem Gemisch von 30 cmb Methanol und 70 cm-' Methylenchlorid, versetzt die Lösung bei Zimmer temperatur in 2 Portionen mit 1,5 g Natriumbor- hydrid und lässt 10 Minuten stehen.
Darauf wird die Hauptmenge des Methylenchlorids im Vakuum ver dampft, und die verbleibende methanolische Lösung mit 25 cm3 Wasser verdünnt, worauf 10-Benzyloxy- iso-deserpidsäure-lacton der Formel I, in welcher R1, R3, 1141 R6 = H, R2 = OCH2 * C6Hs und R5 =<B>CH,</B> bedeuten, in feinen Nadeln vom Smp. 147 bis 149 kristallisiert.
[a]20 = -68 (c = 0,3 in Pyridin). Ausbeute 0,88 g oder 910/0. der Theorie. <I>Beispiel 3</I> Iso-reserpsäure-lacton a) Lacton-lactam: 1,5 g Oxysäure-lactam (For mel 1I: R1, R2, R4, R6 = H, R3 = OCH3 und RS =<B>CH.)</B> werden, wie in Beispiel 1 beschrieben, in Benzol mit Acetanhydrid/Natriumacetat während 5 Stunden am Rückfluss erhitzt.
Man erhält das Lac- ton-lactam als amorphen Rückstand, der nicht zur Kristallisation gebracht werden konnte. Ausbeute <B>1,35</B> g.
b) 3-Dehydro-reserpsäure-lacton: 1,05 g des unter a) beschriebenen Lacton-lactams werden, wie in Beispiel 1 beschrieben, mit Phosphoroxychlorid behandelt und aufgearbeitet. Man erhält aus Me thanol 0,7 g der Substanz. Nadeln vom Smp. 290 bis 292 . [a120 = + 114 (c = 0,3 in Pyridin). Aus beute 0,7 g oder 7011/a der Theorie.
c) Iso-reserpsäure-lacton: 0,2 g 3-Dehydro-re- serpsäure werden, wie in Beispiel 1 beschrieben, mit Platinkatalysator in 50o/oiger wässeriger Essigsäure hydriert und aufgearbeitet. Man erhält das Iso- reserpsäure-lacton der Formel I, in welcher R1, R2, R4, R6 = H, R3 = OCH3 und R5 = CH3 bedeu ten, aus Methanol in Nadeln vom Smp. 224-225 .
[11]D = -142 (c = 0,3 in Pyridin). Ausbeute 157 mg oder 85"/o, der Theorie.
<I>Beispiel 4</I> 11-Athoxy-iso-deserpidsäure-lacton a) Lacton-lactam: 2,0 g Oxy-säure-lactam (For mel 1I: R1, R2 und R4 = H, R3 = OCA), (herge stellt nach Schweiz. Patent Nr. 360058) und 0,8 g wasserfreies Natriumacetat werden in einer Lösung von 8 cm?, Acetanhydrid in 80 cm3 Benzol 15 Stun den am Rückfluss zum Sieden erhitzt, wobei die Sub stanz innert einer Stunde in Lösung geht.
Nach dem Abkühlen wird im Vakuum zur Trockne verdampft und der Rückstand zwischen Natriumhydrogen- carbonatlösung und Chloroform verteilt. Aus dem amorphen Rückstand der Chloroformlösung (2 g) kristallisiert das Lacton-lactam in Nadeln vom Smp. 169-171 (aus Essigester). [a] D = -130 (c = 0,3 in Pyridin). Ausbeute 701/o der Theorie.
b) 3 -Dehyd'ro -11 - äthoxy-deserpidsäure-lacton: 1,25 g des unter a) beschriebenen Lacton-lactams erhitzt man in einer Lösung von 25 cm3 frisch destilliertem Phosphoroxychlorid während 2 Stunden in einer Stickstoffatmosphäre am Rückfluss. Die gelbe Lösung verdampft man im Vakuum zur Trockne, löst den Rückstand in Methylenchlorid, schüttelt die Lösung mit verdünnter Ammoniak- lösung aus, trocknet über Natriumsulfat,
worauf beim Einengen und Verdrängen des Methylenchlorids durch Methanol 3-Dehydro-11-äthoxy-deserpidsäure- lacton in feinen Nadeln vom Smp. 290=293 kri stallisiert. [11]D = + 180 (c = 0,3 in Pyridin). Aus- beute 0,915 g oder 76 % der Theorie.
c) 11-Äthoxy-iso-deserpidsäure-lacton: 0,82 g 11-Äthoxy-dehydro-deserpidsäure-lacton löst man in einem Gemisch von 10 cm?, Methanol und 20 cm Methylenchlorid, und versetzt die Lösung bei Zim mertemperatur in 2 Portionen mit 1 g Natriumbor- hyd'rid. Nach beendeter Reaktion versetzt man mit wenig Essigsäure und. verdampft hierauf die Lösung zur Trockne.
Den verbleibenden Rückstand verteilt man zwischen verdünnter Ammoniaklösung und Me- thylenchlorid. Nach Verdampfen des Lösungsmittels erhält man das 11-Äthoxy-iso-deserpidsäure-lacton der Formel I, in welcher R,, R2 und R4 = H, R3 = OC.H5 bedeuten, das aus Essigester in Prismen kristallisiert. Smp. 195-198 , [a120 = -130' (c = 0,2 in Pyridin). Ausbeute 0,720 g oder 871/o, der Theorie.
Beispiel 12-Methoxy-iso-deserpidsäure < lacton a) Lacton-lactam: 2,7 g Oxy-säure-lactam (For mel II: R1, R2 und R3 = H, R4 = OCH3) werden, wie in Beispiel 1 beschrieben, in Benzol mit Acet- anhydrid/Natriumacetat über Nacht zum Sieden am Rückfluss erhitzt. Man erhält das Lacton-lactam als amorphen Rückstand, der bisher nicht zur Kristalli sation gebracht werden konnte.
b) 3-Dehydro-12-methoxy-deserpidsäure-lacton: 2,6 g des unter a) beschriebenen amorphen Lacton- lactams werden, wie in Beispiel 1 beschrieben, mit Phosphoroxychlorid behandelt und aufgearbeitet. Man erhält die Verbindung aus Methanol. Nadeln vom Smp. 240-242 , [a120 = + 186 (c = 0,2 in Pyridin). Ausbeute über zwei Stufen 6311/o der Theorie.
c) 12-Methoxy-iso-deserpidsäure-lacton: 1,5 g des unter b) beschriebenen 3-Dehydro-12-methoxy- deserpidsäure-lactons werden, wie in Beispiel 4 angegeben, mit Natriumborhydrid reduziert und auf gearbeitet. 12-Methoxy-iso-deserpidsäure-lacton der Formel I, in welcher R1, R, und R3 = H, R4 = OCH3 bedeuten, kristallisiert aus Methanol in Na deln vom Smp. 252-254 , [11]D = -148 (c = 0,2 in Pyridin). Ausbeute 601/o der Theorie.
<I>Beispiel 6</I> 10-Methoxy-iso-deserpidsäure-lacton a) Lacton-lactam: 2,5g Oxy-säure-lactam (For mel II: R1, R3 und R4 = H, R2 = OCH3) werden, wie in Beispiel 1 beschrieben, in Benzol mit Acet- anhydrid/Natriumacetat über Nacht zum Sieden am Rückfluss erhitzt.
Man erhält das Lacton-lactam aus Essigester in Nadeln vom Smp. 178-180 , [a]20 = -25 (c = 0,4 in Pyridin). Ausbeute 77o/11 der Theorie. b) 3 -Dehydro-10-methoxy-de:serpidsäure-lacton: 0,2 g des unter a) beschriebenen Lacton-lactams werden, wie in Beispiel 1 beschrieben, mit Phosphor- oxychlorid behandelt und aufgearbeitet. Man er hält die Verbindung aus Methanol.
Nadeln vom Smp. 268-270 , [a]20= + l21 (c = 0,3 in Pyridin). Ausbeute 92% der Theorie.
c) l0-Methoxy-iso-deserpidsäure-lacton: 0,17 g des unter b) beschriebenen 3-Dehydro-10-methoxy- deserpidsäure-lactons löst man in 4 cm3 50%iger wässeriger Essigsäure und schüttelt mit 0,03 g vor hydriertem Platinkatalysator in einer Wasserstoff atmosphäre, wobei innert 4 Stunden nahezu die für 1 Mol Wasserstoff berechnete Menge aufgenommen worden ist.
Nach beendeter Wasserstoffaufnahme wird vom Katalysator filtriert, das Filtrat im Vakuum stark eingeengt, der Rückstand in Methylenchlorid gelöst, die Lösung mit verdünntem Ammoniak aus geschüttelt, über Natriumsulfat getrocknet und im Vakuum zur Trockne verdampft. Der Rückstand liefert aus Benzol 10-Methoxy-iso-deserpidsäure- lacton der Formel I, in welcher R1, R3 und R4 = H, R2 = O,-"H" bedeuten.
Nadeln vom Smp. 140-142 und 212 (Doppelsmp.). [a120 = -91 (c = 0,3 in Pyridin). Ausbeute 94% der Theorie.
<I>Beispiel 7</I> 11-Benzyloxy-iso-deserpidsäure-lacton a) Lacton-lactam: 1,1g Oxy-säure-lactam (For mel 1I: R1, R: und R4 = H, R3 = OCHzCOH") wer den, wie in Beispiel 1 beschrieben, in Benzol mit AcetanhydridlNatriumacetat über Nacht erhitzt.
Man erhält das Lacton-lactam aus Essigester in Nadeln vom Smp. 203 , [a]20 = -13 (c = 0,3 in Pyridin). Ausbeute 84% der Theorie.
b) 3-Dehydro-11-benzyloxy-deserpidsäure-lacton: 0,81 g des unter a) beschriebenen Lacton-lactams werden, wie in Beispiel 1 beschrieben, mit Phosphor- oxychlorid behandelt und aufgearbeitet. Man erhält die Verbindung aus Methanol. Nadeln vom Smp. 275 , [a1-20 + 92 (c = 0,3 in Pyridin). Ausbeute 671% der Theorie.
c) 11-Benzyloxy-iso-deserpidsäure-lacton: 0,1 g des unter b) beschriebenen 3-Dehydro-11-benzyloxy- deserpidsäure-lactons werden, wie in Beispiel 6 be schrieben, katalytisch hydriert und aufgearbeitet.
11-Benzyloxy-iso-deserpidsäure-lacton der Formel I, in welcher R1, R2 und R4 = H, R3 = OCH2 - C6H,- bedeuten, kristallisiert aus Essigester in Nadeln vom Smp. 23 8-240 ; [a]20= -106 (c = 0,3 in Pyridin). Ausbeute 84% der Theorie.
<I>Beispiel 8</I> 9-Chlor-iso-deserpidsäure-lacton a) Lacton-lactam: 4,95 g Oxy-säure-lactam (For mel II: R2, R3 und R4 = H, R1 = Cl) werden, wie in Beispiel 1 beschrieben, in Benzol mit Acet- anhydridjNatriumacetat über Nacht zum Sieden am Rückfluss erhitzt.
Man erhält das Lacton-lactam aus Essigester in Polyedern vom Smp. 209-210 , [a]2() = -27 (c = 0,2 in Pyridin). Ausbeute 7711/9 der Theorie.
b) 3-Dehydro-9-chlor-deserpidsäure-lacton: 0,5 g des unter a) beschriebenen Lacton-lactams werden, wie in Beispiel 1 beschrieben, mit Phosphoroxy- chlorid erhitzt und aufgearbeitet. Man erhält die neue Verbindung aus Methanol in Nadeln vom Smp. 309-310 , [a]20 = + 156 (c = 0,2 in Pyri- din). Ausbeute 79% der Theorie.
c) 9-Chlor-iso-deserpidsäure-1'acton: 0,1 g des unter b) beschriebenen Dehyd'ro-lactons werden, wie in Beispiel 6 beschrieben, katalytisch hydriert und aufgearbeitet. 9-Chlor-iso-deserpidsäure-lacton der Formel I, in welcher R2, R3 und R4 = H, R1 = Cl bedeuten, kristallisiert aus Methanol in Rhombo- edern vom Smp. 237-239', [a]20 = -75 (c = 0,2 in Pyridin). Ausbeute 89 ; 0 der Theorie.
<I>Beispiel 9</I> 10,11-Dioxymethylen-iso-deserpidsäure-lacton a) Lacton-lactam: 1,09g Oxy-säure4actam (For mel II: R1 und R4 = H, R2 und R3 = -O-CH2- O-) werden, wie in Beispiel 1 beschrieben, in Benzol mit Acetanhydrid und Natriumacetat behandelt.
Man erhält das Lactam-lacton aus Essigester in Nadeln vom Smp. 219-220 , [cc] D = -34 (c = 0,2 in Pyridin). Ausbeute 87 % der Theorie.
b) 3 -Dehydro - 10,11 - dioxymethylen - deserpid- säure-lacton: 0,4 g des unter a) beschriebenen Lacton-lactams werden, wie in Beispiel 1 angege ben, mit Phosphoroxychlorid behandelt und aufgear beitet. Man erhält die Dehydro-Verbindung aus Methanol in Nadeln vom Smp. 276 , [a]20 = + 98 (c = 0,2 in Pyridin). Ausbeute 67 /o der Theorie.
c) 10,11-Dioxymethylen-iso-deserpidsäure-lacton: 0,113 g des unter b) beschriebenen Dehydro-lactons werden, wie in Beispiel 6 beschrieben, katalytisch hydriert und aufgearbeitet. Man erhält 10,11-Dioxy- methylen iso-deserpidsäure-lacton der Formel I, in welcher R1 und R4 = H, R2 und R3 = -O-CH#-O- bedeuten, aus Methanol in Platten vom Smp. 146 bis 147 , [a]20 = -80 (c = 0,2 in Pyridin)
. Ausbeute 85% der Theorie.
<I>Beispiel 10</I> 10,11-Dioxymethylen-iso-deserpidsäure-lacton a) Lacton-lactam: 3,6g Oxy-säure-lactam (For mel 1I: R1 und R4 = H, R2 und R3 = -O-CH Z O-) (hergestellt nach Schweiz.
Patent Nr. 360058, aus 5,6-Dioxymethylen-tryptamin und dem rechtsdrehen den Antipode der 1,2,3,4,7,8,9,10-Oktahydro-2- methoxy-3-acetoxy-7-oxo-l-naphtholsäure) werden, wie in Beispiel 1 beschrieben, in Benzol mit Acet- anhydrid#/Natriumacetat behandelt. Man erhält das Lacton-lactam aus Essigester in Nadeln vom Smp. 218-220 , [a120 =<B>+320</B> (c = 0,2 in Pyridin). Ausbeute 790/9 der Theorie.
b) 3 -Dehyd@o-- 10,11-dioxymethylen-dieserpid- säure-lacton: 2,7 g des unter a) beschriebenen. Lac- ton-lactams werden, wie in Beispiel 1 angegeben, mit Phosphoroxychlorid behandelt. Man erhält die Dehydro-Verbindung aus Methanol in Platten vom Smp. 273-275 , [a]20 = -96 (c = 0,2 in Pyridin). Ausbeute 63 9/o der Theorie.
c) 10,11-Dioxymethylen-iso-deserpidsäure lacton: 0,2 g des unter b) beschriebenen Dehydro-lactons werden, wie in Beispiel 6 angegeben, katalytisch hydriert. 10,11- Dioxymethylen - iso - deserpidsäure- lacton der Formel 1, in welcher R1 und R4 = H, R, und R3 = -O-CH"-O- bedeuten, kristallisiert aus Methanol in Platten vom Smp. 145-147 , [a]D = +78 (c = 0,2 in Pyridin)
. Ausbeute 879/o der Theorie.
<I>Beispiel 11</I> 10,11-Dimethoxy-iso-deserpidsäure-lacton a) Lacton-lactam: 1,75g Oxy-säure-lactam (For mel 1I: R1 und R4 = H, R2 und R3 = OCHS) werden, wie in Beispiel 1 beschrieben, in Benzol mit Acetanhydrid und Natriumacetat behandelt. Man erhält das Lacton-lactam als amorphen Schaum, der bisher nicht kristallisiert werden konnte. Rohausbeute 9211/o der Theorie.
b) 3-Dehydro-10,11-dimethoxy-deserpidsäure- lacton: 1,55 g des unter a) beschriebenen Lacton- lactams werden, wie in Beispiel 1 angegeben, mit Phosphoroxychlorid erhitzt. Man erhält das Dehydro- lacton aus Methanol in Nadeln vom Smp. 314-316 , [a]D = + 108 (c = 0,2 in Pyridin). Ausbeute 559/o der Theorie.
c) 10,11 - Dimethoxy - iso - deserpidsäure-lacton: 0,2 g des unter b) beschriebenen Dehydro-lactons werden, wie in Beispiel 4 beschrieben, mit Natrium- borhydrid reduziert und aufgearbeitet.
10,11-Di- methoxy-iso-deserpidsäure-lacton der Formel I, in welcher R1 und R4 = H, R2 und R3 = OCH3 be deuten, kristallisiert aus Methanol in kurzen, derben Prismen vom Smp. 217-219 , [a]20 = -110 (c = 0,2 in Pyridin)
. Ausbeute 92'% der Theorie. <I>Beispiel 12</I> 1 I-Propoxy-iso-deserpidsäure-lacton a) Lacton-lactam: 2,99g Oxy-säure-lactam (For mel 11: R1, R2 und R4 = H, R3 = OC3H7, RS = CH3, R0 = H) (hergestellt nach Schweiz.
Patent Nr. 360058) und 1,5 g wasserfreies Natriumacetat werden in einer Lösung von 10 cm-' Acetanhydrid und 100 cm-' Benzol 15 Stunden am Rückfluss erhitzt, wobei die Substanz innert 1 Stunde in Lösung geht. Nach dem Abkühlen wird im Vakuum zur Trockne ver dampft und der Rückstand zwischen Natriumhydro- gencarbonatlösung und Chloroform verteilt. Da das Lacton-lactam nicht kristallisiert, wird direkt der amorphe Rückstand weiterverarbeitet.
b) 3 -Dehydro-11-propoxy-deserpidsäure lacton: 2,8 g des unter a) beschriebenen Lacton-lactams er hitzt man in einer Lösung von 50 cm3 frisch destil liertem Phosphoroxychlorid während 2 Stunden in einer Stickstoffatmosphäre am Rückfluss. Die gelbe Lösung verdampft man im Vakuum zur Trockne, löst den Rückstand in Methylenchlorid, schüttelt die Lösung mit verdünnter Ammoniaklös.ung aus, trock net über Natriumsulfat,
worauf beim Einengen und Verdrängen des Methylenchlorids durch Methanol 3-Dehydro-11-propoxy-deserpidsäure-lacton in Na deln vom Smp. 283-285 auskristallisiert. [a]2 = + 103 (c = 0,2 in Pyridin).
c) 11-Propoxy-iso-deserpidsäure-lacton: 1,6 g 11-Propoxy-3-dehydro-deserpidsäure-lacton löst man in einem Gemisch von 60 cm3 Methylenchlorid und 30 cm3 Methanol und versetzt die Lösung bei Zim mertemperatur in zwei Portionen mit 1,7g Natrium- borhydrid. Nach beendeter Reaktion versetzt man mit wenig Essigsäure und verdampft hierauf die Lö sung zur Trockne.
Den verbleibenden Rückstand verteilt man zwischen verdünnter Ammoniaklösung und Methylenchlorid. Nach Verdampfen des Lö- sungsmittels erhält man das 11-Propoxy-iso-deserpid- säure-lacton der Formel 1, in welcher R1, R2 und R4 = H, R3 = OC3H7, R5 = CH3, R6 = H, das aus Methanol in sechseckigen Blättchen kristallisiert.
Smp. 185-187 (sint. bei 130 ). MM = -117 c = 0,2 in Pyridin).
<I>Beispiel 13</I> 11-Chlor-iso-deserpidsäure-lacton a) Lacton lactam: 2,34g Oxy-säure-lactam (For mel II: R1, R2 und R4 = H, R3 = Cl, RS = CH3, R6 = H) werden, wie in Beispiel! 1 angegeben, in Benzol mit Acetanhydrid-Natriumacetat über Nacht zum Sieden am Rückfluss erhitzt.
Aus dem amorphen Rückstand kristallisiert das Lacton-lactam aus Essig ester in Rhomboedarn vom Smp. 145-147 . [a]D = -31 (c = 0,2 in Pyridin).
b) 3 - Dehydro -11 -chlor - deserpidsäure-lacton: 1,77 g des unter a) beschriebenen Lacton-lactams werden, wie in Beispiel 1 beschrieben, mit Phosphor oxychlorid behandelt und aufgearbeitet. Man erhält die Verbindung aus Methanol in Nadeln vom Smp. 300-304 . [a]20 = + 124 (c = 0,2 in Pyridin).
c) 11-Chlor-iso-deserpidsäure"lacton: 1,34 g des unter b) beschriebenen 3-Dehydro-11-chlor-deserpid- säure-lactons werden:, wie in Beispiel 12 angegeben, mit Natriumborhydrid reduziert und aufgearbeitet.
11-Chlor-iso-deserpidsäure-lacton der Formel 1, in welcher R1, R2 und R4 = H, R3 = Cl, R5 = CH3, RE = H bedeuten, kristallisiert aus Methanol in Prismen vom Smp. 222-224 . [a]20 = -140 (c = 0,2 in Pyridin).
<I>Beispiel 14</I> 11-Brom-iso-deserpidsäure-lacton a) Lacton-lactam: 2,86 g Oxy-säure-lactam (For mel II: R1, R2 und R4 = H, R3 = B.r, RS = CH3, RG = H) werden, wie in Beispiel 1 angegeben, in Benzol mit Acetanhydrid-Natriumacetat über Nacht am Rückfluss zum Sieden erhitzt.
Man erhält das Lacton-lactam aus Essigester in Polyedern vom Smp. 168-171 . [a]D = -29 (c = 0,2 in Pyridin).
b) 3 -Dehydro - 11 -Brom - deserpidsäure-lacton: 2,1 g des unter a) beschriebenen Lacton-lactams werden, wie in Beispiel 1 angegeben, mit Phosphor oxychlorid behandelt und aufgearbeitet. Man erhält die Verbindung aus Methanol in Nadeln vom Smp. 295-297 . [a]D = + 97" (c = 0,2 in Pyridin).
c) 11-Brom-iso-deserpidsäure-lacton: 1,67 g des unter b) beschriebenen 3-Dehydro-11-brom-deserpid- säure-lactons werden, wie in Beispiel 12 angegeben, mit Natriumborhydrid reduziert und aufgearbeitet. 11 Brom-iso-deserpidsäure-lacton der Formel I, in welcher R1 R2, R4 und R, = H, R3 = Br und R5 =<B>CH"</B> bedeuten, kristallisiert aus Methanol in Prismen vom Smp. 231 (sirrt.
bei 140-150 ). [a]D -- -132 (c = 0,2 in Pyridin).
<I>Beispiel 15</I> 9-Brom-iso-deserpidsäure-1acton a) Lacton-lactam: 2.,65 g Oxy-säure-lactam (For mel II: R1 = Br, R2, R3 und R4 = H, R5 = CH3, R6 = H) werden, wie in Beispiel 1 angegeben, in Benzol mit Acetanhydrid und Natriumacetat über Nacht am Rückfluss zum Sieden erhitzt.
Man er hält das Lacton-lactam aus Essigester in kugeligen Kristallaggregaten vom Smp. 225-226 . [a] =-27 D (c = 0,2 in Pyridin).
b) 9-Brom-3-dehydro-deserpidsäure-lacton: 1,9 g des unter a) beschriebenen Lacton-lactams werden, wie in Beispiel 1 beschrieben, mit Phosphoroxy- chlorid behandelt und aufgearbeitet. Man erhält die Verbindung aus Methanol in Nadeln vom Smp. 294 bis 295 . [a]D = + 150 (c = 0,2 in Pyridin).
c) 9-Brom-iso-deserpidsäure-lacton: 1,4 g des unter b) beschriebenen' 3-Dehydro-9-brom-deserpidsäure- lactons werden, wie in Beispiel 12 angegeben, mit Natriumborhydrid reduziert und aufgearbeitet. 9-Brom-iso-deserpidsäuro-lacton der Formel I, in welcher R1 = Br, R2 R3, R4 und R5 = H und R5 =<B>CH,</B> bedeuten, kristallisiert aus Methanol in rhombischen Blättchen vom Smp. 213-215 (sirrt.
bei 140 ). [a]D = -34" (c = 0,2 in Pyridin).
<I>Beispiel 16</I> 9-Methoxy-iso-deserpidsäure-lacton a) Lacton-lactam: 2,37g Oxy-säure-lactam (For mel 1I: R1 = OCHS, R2, R3 und R4 = H, RS = CH3, R6 = H) werden, wie in Beispiel 1 angegeben, in Benzol mit Acetanhydrid-Natriumacetat über Nacht am Rückfluss zum Sieden erhitzt.
Man erhält das Lacton-lactam aus Essigester in rhombischen Blätt chen vom Smp. 175-l77 . [a]D = -135" (c = 0,2 in Pyridin).
b) 3-Dehydro - 9 - methoxy-deserpids.äure lacton: 1,8 g des unter a) beschriebenen Lacton-lactams wer den, wie in Beispiel 1 angegeben, mit Phosphoroxy- chlorid behandelt und aufgearbeitet. Man erhält die Verbindung aus Methanol in Nadeln vom Smp. 278 bis 280 . [a]D = + 168 (c = 0,2 in Pyridin).
c) 9-Methoxy-iso-deserpidsäure-lacton.: 1,15 g des unter b) beschriebenen 3 Dehydro-9-methoxy-de- serpidsäure-lactons werden, wie in Beispiel 12 an gegeben, mit Natriumborhydrid reduziert und auf gearbeitet.
9-Methoxy-iso-deserpidsäure-lacton der Formel I, in welcher R1 = OCH3, R2, R3 und R4 = H, R5 = CH3, R6 = H, kristallisiert aus Methanol in Polyedern vom Smp. 193-195 (sirrt. bei 130 ). [a]D = -61" (c = 0,2 in Pyridin).
<I>Beispiel 17</I> 11-Isopropoxy-i:so-deserpidsäure-lacton a) Lacton-lactam: 2,29 g Oxy-säure-lactam (For mel Il: R1, RZ und R4 = H, R3 - OCH(CH3)2, R5 = CH3, R6 = H) werden, wie in Beispiel 1 an gegeben, in Benzol mit Acetanhydrid-Natriumacetat über Nacht am Rückfluss zum Sieden erhitzt.
Man erhält das Lacton-lactam als amorphen Rückstand, der bisher nicht zur Kristallisation gebracht werden konnte.
b) 3 - Dehydro -11 - isopropoxy - deserpidsäure - lacton: 2,25 g des unter a) beschriebenen amorphen Lactonlactams werden, wie in Beispiel 1 angegeben, mit Phosphoroxychlorid behandelt und aufgearbei tet. Man erhält die Verbindung aus Methanol in Na deln vom S.mp. 287-289 . [a]D = + 101 (c = 0,2 in Pyridin).
c) 11-Isopropoxy-iso-deserpidsäure-lacton: 1,08 g des unter b) beschriebenen 3-Dehydro-11-isopropoxy- deserpidsäure-lactons werden, wie in Beispiel 12 an gegeben, mit Natriumborhydrid reduziert und aufge arbeitet.
11-Isopropoxy-iso-deserpidsäure-1'acton der Formel I, in welcher R1, R, und R4 = H, R3 = OCH(CH)3)2, R5 = CH3, Rr - H, konnte bisher nicht kristallisiert erhalten werden.
<I>Beispiel 18</I> 11-Butoxy-iso-deserpidsäure-lacton a) Lacton-läctam: 3,26g Oxy-säure-laetam (For mel 1I: R1, R2 und R4 = H, R3 = OCHZCH.CH"CH3, R5 = CH3, R6 = H) werden, wie in Beispiel 1 an gegeben, in Benzol mit Acetanhydrid-Natriumacetat über Nacht am Rückfluss zum Sieden erhitzt.
Man erhält das Lacton-lactam als amorphen Rückstand, der bisher nicht kristallisiert werden konnte.
b) 3 -Dehydro-11-butoxy-deserpidsäure-lacton: 2,99 g des unter a) beschriebenen amorphen Lacton- lactams werden, wie in Beispiel 1 angegeben, mit Phosphoroxychlorid behandelt und aufgearbeitet. Man erhält die Verbindung aus Methanol in Nadeln vom Smp. 263-265 . [a]D = + 116 (c = 0,2 in Pyridin).
c) 11-Butoxy-iso-deserpidsäure-lacton: 1,39 g des unter b) beschriebenen 3-Dehydro-11-butoxy-deserpid- säure-lactons werden, wie in Beispiel 12 angegeben, mit Natriumborhydrid reduziert und aufgearbeitet.
11-Butoxy-iso-deserpidsäure-lacton der Formel I, in welcher R1, R2 und R4 = H, R3 = OCH2CHZCHZCH3, R5 = CH3, R6 = H, kristallisiert aus verdünntem Methanol in Prismen vom Smp. 158-164 . [a120 -117" (c = 0,2 in Pyridin). <I>Beispiel 19</I> 11-Methoxy-17-desmethoxy-17-äthoxy-iso- deserpidsäure-lacton a) Lacton lactam:
2,0 g Oxy-säure-lactam(For- mel 1I: R1, R2 und R4 = H, R3 = OCH3, R5 = CH2CH3, RG = H) werden, wie in Beispiel 1 be schrieben, in Benzol mit Acetanhydrid-Natriumacetat über Nacht am Rückfluss zum Sieden erhitzt. Man erhält das Lacton-lactam als amorphen Rückstand, der bisher nicht kristallisiert werden konnte.
b) 3 -Dehydro-11-methoxy-17-desmethoxy-17- äthoxy-deserpid'sä#ure-lacton: 0,86 g des unter a) be schriebenen amorphen Lacton-lactam:s werden, wie in Beispiel 1 angegeben, mit Phosphoroxychlorid be handelt und aufgearbeitet. Man erhält die Verbin dung aus Methanol in feinen Nadeln vom Smp. 235 bis 237 . [a120 = + 134 (c = 0,2 in Pyridin).
c) 3 -Dehydro-11-methoxy-17-desmethoxy-17- äthoxy-iso-deserpidsäure lacton: 0,8 g des unter b) beschriebenen 3-Dehydro lactons werden, wie in Beispiel 12 angegeben, mit Natriumborhydrid redu ziert und aufgearbeitet.
Die neue Verbindung (For mel I: R1, R2 und R4 = H, R3 = OCHS, R5 = CH,CH3, R6 = H) kristallisiert aus Methanol in Rhomboedem vom Smp. 1150 (unscharf). [a]20 = r 106 (c = 0,2 in Pyridin).
<I>Beispiel 20</I> 11-Methoxy-17-desmethoxy-17-propoxy-iso- deserpidsäure-lacton a) Lacton-lactam: 4,02 g Oxy-säurerlactam (For mel 1I: R1, R2 und R4 = H, R3 = OCHS, R5 = CH,CH2CH3, R" = H) werden, wie in Beispiel 1 angegeben, in Benzol mit Acetanhydrid-Natrium- acetat über Nacht am Rückfluss zum Sieden erhitzt.
Da das Lacton-lactam nicht kristallisiert, wird direkt der amorphe Rückstand weiterverarbeitet.
b) 3 -Dehydro-11-methoxy-17-desmethoxy-17- propoxy-deserpidsäure-lacton: 1,42 g des unter a) beschriebenen Lacton-lactams werden, wie in Bei spiel 1 angegeben, mit Phosphoroxychlorid erhitzt und aufgearbeitet. Man erhält die Verbindung aus Methanol in feinen Prismen vom Smp. 238-2400. [a] D = + 1220 (c = 0,2 in Pyridin).
c) 11-Methoxy-17-desmethoxy-17-propoxy-iso- deserpidsäure-lacton: 1,26 g des unter b) beschrie benen Dehydro-lactons werden, wie in Beispiel 12 angegeben, mit Natriumborhydrid reduziert und auf gearbeitet.
Die neue Verbindung (Formel I: R1, R2 und R4 = H, R3= OCHS, R5 = CH2CH2CH3, RG = H) kristallisiert aus Methanol in Prismen vom Smp. 870. [,]D = -1090 (c = 0,2 in Pyridin).
<I>Beispiel 21</I> 11-Methoxy-17-desmethoxy-17-isopropoxy-iso-. deserpidsäure-lacton a) Lacton-lactam: 2,92 g Oxy-säure-lactam (For mel 1I: R1, R2 und R4 = H, R3 = OCHS, R5 = CH(CH3)2, R6 = H) werden, wie in Beispiel 1 be schrieben, in Benzol mit Acetanhydrid-Natriumacetat über Nacht am Rückfluss zum Sieden erhitzt.
Man erhält das Lacton-lactam als gelben Schaum, der bis jetzt nicht kristallisiert werden konnte.
b) 3-Dehydro-11-methoxy-17-desmethoxy-17-iso- propoxy-deserpidsäure-lacton: 2,74 g des unter a) be schriebenen Lacton-lactams werden, wie in Beispiel 1 angegeben, mit Phosphoroxychlorid erhitzt und aufgearbeitet. Man erhält die neue Verbindung aus Methanol in Plättchen vom Smp. 215-220'. [a]D = + l05 (c = 0,2 in Pyridin).
c) 11-Methoxy-17 -desmethoxy-17-isopropoxy- iso-deserpidsäure-lacton: 2,32 g des unter b) beschrie benen 3-Dehydro-lactons werden, wie in Beispiel 12 angegeben, mit Natriumborhydrid reduziert und auf gearbeitet.
Die neue Verbindung (Formel I: R1, R2 und R4 = H, R3 = OCH3, R5 = CH(CH3)2, R, H) kristallisiert aus Aceton-Ather in Prismen vom Smp. 210-212 . [,]20 = -113 (c = 0,2 - in Pyridin).
<I>Beispiel 22</I> (d)-5-Methyl-iso-deserpidsäure-lacton 1- a) Lacton-lactam: 2,87 g (d)-Oxy-säure-lactam (Formel 1I: R1, R2, R3, R4 = H, R5 und R6 = CH3) werden, wie in Beispiel 1 angegeben, in Benzol mit Acetanhydrid und Natriumacetat über Nacht am Rückfluss zum Sieden erhitzt.
Man erhält das Lacton- lactam aus Essigester in Prismen vom Smp. 193 bis 1940. [a]20 = -180 (c = 0,2 in Pyridin).
b) (d)-3Dehydro-5-methyl-deserpidsäure-lacton: 1,56 g des unter a) beschriebenen Lacton-lactams werden, wie in Beispiel 1 angegeben, mit Phosphor- oxychlorid behandelt und aufgearbeitet. Man erhält die neue Verbindung als amorphen Rückstand, der nicht näher charakterisiert wurde.
c) (d)-5-Methyl-iso-deserpidsäure-lacton: 1,5 g des unter b) beschriebenen (d)-3-Dehydro-5-methyl- deserpidsäure-lactons werden, wie in Beispiel 12 an gegeben, mit Natriumborhydrid reduziert und auf gearbeitet.
(d)-5 Methyl-iso-deserpidsäure=lacton der Formel I, in welcher R1, R2, R3, R4 = H, R5 und RE =<B>CH.,</B> kristallisiert aus Methanol in feinen Na deln vom Smp. 2610. [a]20 = -960 (c = 0,2 in Pyridin).
<I>Beispiel 23</I> (1)-5-Methyl-iso-deserpidsäure-lacton 2 a) Lacton-lactam: 1,72 g (1)-Oxy-säure-lactam (Formel II: R1, R2, R3, R4 = H, R5 und R, = CH3) werden, wie in Beispiel 1 angegeben, in Benzol mit Acetanhydrid und Natriumacetat über Nacht am Rückfluss gekocht.
Man erhält das Lacton-lactam aus Essigester in feinen Nadeln vom .Smp. 1830 (sirrt. bei 126-1280). [a]D = -52o (c = 0,2 in Pyridin). 1 Die Bezeichnung d bezieht sich auf den rechtsdrehenden Antipoden des a-Methyl-triptamins.

(1)-5-Methyl-iso-deserpid äure-lacton der Formel I, in welcher R1, R2, R3, R4 = H, R5 und R6 =<B>CH.,</B> kristallisiert aus Methanol in Prismen vom Smp. 138-140 .

<I>Beispiel 24</I> (d)-5-Methyl-11-methoxy-iso-deserpidsäure-lacton 1 a) Lacton-lactam: 3,3g (d,1)-Oxy-säure lactam (Formel 1I: R1, R2, R4 = H, R3 = OCHS, R5 und RE =<B>CH.)</B> werden, wie in Beispiel 1 angegeben, in Benzol mit Acetanhydrid und Natriumacetat über Nacht am Rückfluss gekocht.
Nach Abtrennung des kristallinen (1)-Lacton-lactams erhält man das (d)- Lacton-lactam als amorphen Rückstand.
b) (d)-3-Dehydro-5-methyl-11-methoxy-deserpid- säure-lacton: 1,3 g des unter a) beschriebenen (d)-Lacton-lactams werden, wie in Beispiel 1 an gegeben, mit Phosphoroxychlorid behandelt und auf gearbeitet. Man erhält die Verbindung als kristalli nen Rückstand, der jedoch nicht näher charakteri siert wurde.
c) (d)-5-Methyl-11-methoxy-iso-deserpidsäure- lacton: 1,2g des unter b) beschriebenen (d)-3-De- hydro - 5 -methyl-11-methoxy-deserpidsäure-lactons werden, wie in Beispiel 12 angegeben, mit Natrium- borhydrid reduziert und aufgearbeitet.
(d)-5-Methyl- 11-methoxy-iso-deserpidsäure-lacton der Formel I, in welcher R1, R2, R4 = H, R3 = OCHS, R5 und R5 = CH3, kristallisiert aus Essigester in feinen Nadeln vom Smp. 246-247 . [a] = -118 (c D = 0,2 in Pyridin).

Co-worker, J. Amer. Chem. Soc. 78, 2023 1956)] that a lactone of the formula 1 in which R1, R2, R4 and R6 are hydrogen, R3 is methoxy and R5 is methyl, is obtained if the lactam of the formula II is in the form of its methyl ester acetates treated with phosphorus oxychloride,
the resulting dehydro-reserps acid methyl ester acetate is reduced directly with sodium borohydride, the iso-reserps acid methyl ester acetate formed is saponified with methanolic potassium hydroxide, and the iso-reserps acid formed by treating with N, N'-dicyclohexyl carbodiimide in the desired lactone transferred.
The above-mentioned process now has the disadvantage that, because of the sensitivity of this compound to the usual lactonizing reagents, the formation of the lactone ring at a later stage of the synthesis, namely after the construction of the pentacyclic system containing a basic nitrogen atom, like acetic anhydride, etc., is forced to use N, N'-dicyclo-carbodiimide for lactonization,
what is uneconomical and unproductive. The yields are about 40%.
This disadvantage can be avoided if, according to the invention, the lactonization is carried out at the lactam stage, which is completely stable against vigorous lactonization conditions, such as heating with acetic anhydride / sodium acetate, and delivers the desired lactone in yields of 80-909 / o.
Another advantage of the process according to the invention is that the previously unknown 3-dehydro-reserps acid-lactone surprisingly has excellent properties and can easily be isolated and purified in crystalline form as a free base. This could not be foreseen, because in the literature 3-dehydro compounds of reserp acid or yohimbine derivatives are generally described as unstable compounds that can only be isolated in the form of their salts.
The compounds produced by the present process are kri stallin at room temperature and are valuable intermediates. They should be used for the production of therapeutically important substances, eg. B. for the synthesis of serpin and its analogs, use.
In the following examples, all temperatures are given in degrees Celsius. The melting points are corrected. <I> Example 1 </I> Iso-deserpidic acid lactone a) Lactone lactam: 0.644 g oxyacid lactam (formula II: R, -R4 and R6 = H, R5 = CH3) (manufactured in Switzerland.
Patent No. 360058) and 0.1 g of anhydrous sodium acetate are refluxed for 15 hours in a solution of 1.3 cm3 of acetic anhydride in 30 cm3 of benzene, the substance dissolving within one hour. After cooling, it is evaporated to dryness in vacuo and the residue is partitioned between sodium hydrogen carbonate solution and chloroform.
The lactone-lactam crystallizes from the amorphous residue of the chloroform solution (0.65 g) in prisms with a melting point of 177-178 (from ethyl acetate). [a120 = -36 (c = 0.2 in pyridine). Yield 0.497 g or 810/0 of theory.
b) 3-dehydro-deserpidic acid-lactone: 0.25 g of the lactone-lactam described under a) is refluxed in a solution of 8 cm3 of freshly distilled phosphorus oxychloride for 2 hours in a nitrogen atmosphere. The yellow solution is evaporated to dryness in vacuo, the residue is dissolved in methylene chloride, the solution is extracted with dilute ammonia solution, dried over sodium sulfate,
whereupon, when the methylene chloride is concentrated and displaced by methanol, 3-dehydro-deserpidic acid lactone crystallizes in needles with a melting point of 309 to 310. [a] 20 = + 131 (c = 0.3 in pyridine); [a] 20 = + 16.5 (c = 0.2 in chloroform). Yield 1.72g or 9019 / o of theory.
c) Iso-deserpidic acid lactone: 3.3 g of 3-dehydro- deserpidic acid-lactone are dissolved in 90 - 50% aqueous acetic acid and shaken with 0.35 g of hydrogenated platinum catalyst in a hydrogen atmosphere, whereby within 41 / 2 hours almost the amount calculated for 1 mole of hydrogen has been included.
When the hydrogen uptake is complete, the catalyst is filtered off, the filtrate is concentrated strongly in vacuo, the residue is dissolved in methylene chloride, the solution is extracted with dilute ammonia, dried over sodium sulfate and evaporated to dryness in vacuo. From methanol, the residue yields iso-deserpidic acid lactone of the formula I, in which R, -R4 and R6 = H and R5 = CH,.
Needles, m.p. 225-227. [a] 20 = -93 (pyridine). Yield 3.06 g or 9219 / o of theory.
<I> Example 2 </I> 10-Benzyloxy-iso-deserpidic acid @ lactone a) Lactone-lactam: 1.04 g oxy-acid-lactam (Formula II: R1, R3, R4, R6 = H, R #, = OCH.2 - C.H5 and R5 = CH.) Are treated as described in Example 1 in benzene with acetic anhydride / sodium acetate.
The lactone-lactam is obtained in hexagonal columns with a melting point of 198-200 (from ethyl acetate). [19] D = -31 (c-0.3 in pyridine). Yield 0.89 g or 8911 / o of theory.
b) 3-Dehydro-10-benzyloxy-deserpidic acid-lactone: 0.8 g of the lactone-lactam described under a) are, as described in Example 1, treated with phosphorus oxychloride and worked up. The compound is obtained from methanol. Fine needles of m.p. 260-263. [19] D = -104 (c = 0.3 in pyridine). Yield 0.56 g or 7319 / o of theory.
c) 10-Benzyloxy-iso-deserpidic acid-lactone: 1.0 g of 10-benzyloxy-dehydro-deserpidic acid-lactone is dissolved in a mixture of 30 cmb of methanol and 70 cm- 'methylene chloride, the solution is added in 2 portions at room temperature with 1.5 g sodium borohydride and let stand for 10 minutes.
The majority of the methylene chloride is then evaporated in vacuo, and the remaining methanolic solution is diluted with 25 cm3 of water, whereupon 10-benzyloxy-iso-deserpidic acid lactone of the formula I, in which R1, R3, 1141 R6 = H, R2 = OCH2 * C6Hs and R5 = <B> CH, </B> mean, crystallized in fine needles with a melting point of 147 to 149.
[a] 20 = -68 (c = 0.3 in pyridine). Yield 0.88g or 910/0. the theory. <I> Example 3 </I> Iso-reserps acid-lactone a) Lactone-lactam: 1.5 g oxy-acid-lactam (Formula 1I: R1, R2, R4, R6 = H, R3 = OCH3 and RS = <B > CH.) </B> are, as described in Example 1, refluxed in benzene with acetic anhydride / sodium acetate for 5 hours.
The lactone lactam is obtained as an amorphous residue which could not be made to crystallize. Yield <B> 1.35 </B> g.
b) 3-Dehydro-reserps acid-lactone: 1.05 g of the lactone-lactam described under a) are, as described in Example 1, treated with phosphorus oxychloride and worked up. 0.7 g of the substance is obtained from methanol. Needles from m.p. 290 to 292. [a120 = + 114 (c = 0.3 in pyridine). From booty 0.7 g or 7011 / a of theory.
c) Lactone iso-reserps acid: 0.2 g of 3-dehydroserpsic acid are, as described in Example 1, hydrogenated with platinum catalyst in 50% aqueous acetic acid and worked up. The isoreserp acid lactone of the formula I, in which R1, R2, R4, R6 = H, R3 = OCH3 and R5 = CH3, are obtained from methanol in needles with a melting point of 224-225.
[11] D = -142 (c = 0.3 in pyridine). Yield 157 mg or 85 "/ o of theory.
<I> Example 4 </I> 11-ethoxy-iso-deserpidic acid-lactone a) Lacton-lactam: 2.0 g oxy-acid-lactam (formula 1I: R1, R2 and R4 = H, R3 = OCA) , (manufactured in Switzerland. Patent No. 360058) and 0.8 g of anhydrous sodium acetate are refluxed for 15 hours in a solution of 8 cm ?, acetic anhydride in 80 cm3 of benzene, the substance being heated to boiling within one hour Solution works.
After cooling, it is evaporated to dryness in vacuo and the residue is partitioned between sodium hydrogen carbonate solution and chloroform. The lactone-lactam crystallizes from the amorphous residue of the chloroform solution (2 g) in needles with a melting point of 169-171 (from ethyl acetate). [a] D = -130 (c = 0.3 in pyridine). Yield 701 / o of theory.
b) 3-Dehyd'ro -11 - ethoxy-deserpidic acid-lactone: 1.25 g of the lactone-lactam described under a) are heated in a solution of 25 cm3 of freshly distilled phosphorus oxychloride for 2 hours in a nitrogen atmosphere at reflux. The yellow solution is evaporated to dryness in vacuo, the residue is dissolved in methylene chloride, the solution is extracted with dilute ammonia solution, dried over sodium sulfate,
whereupon when the methylene chloride is concentrated and displaced by methanol, 3-dehydro-11-ethoxy-deserpidic acid lactone crystallizes in fine needles with a melting point of 290 = 293. [11] D = +180 (c = 0.3 in pyridine). Yield 0.915 g or 76% of theory.
c) 11-ethoxy-iso-deserpidic acid-lactone: 0.82 g of 11-ethoxy-dehydro-deserpidic acid-lactone is dissolved in a mixture of 10 cm ?, methanol and 20 cm of methylene chloride, and the solution is added at room temperature in 2 Portions with 1 g sodium borohydride. After the reaction has ended, a little acetic acid is added and. the solution then evaporates to dryness.
The remaining residue is distributed between dilute ammonia solution and methylene chloride. After evaporation of the solvent, the 11-ethoxy-iso-deserpidic acid lactone of the formula I, in which R ,, R2 and R4 = H, R3 = OC.H5, crystallizes from ethyl acetate in prisms. 195-198, [a120 = -130 '(c = 0.2 in pyridine). Example 12-Methoxy-iso-deserpid acid <lactone a) Lactone-lactam: 2.7 g of oxy-acid-lactam (For mel II: R1, R2 and R3 = H, R4 = OCH3) are as described in Example 1, in benzene with acetic anhydride / sodium acetate heated to reflux overnight. The lactone-lactam is obtained as an amorphous residue, which so far could not be brought to crystallization.
b) 3-Dehydro-12-methoxy-deserpidic acid-lactone: 2.6 g of the amorphous lactone-lactam described under a) are, as described in Example 1, treated with phosphorus oxychloride and worked up. The compound is obtained from methanol. Needles of m.p. 240-242, [a120 = + 186 (c = 0.2 in pyridine). Yield over two stages 6311 / o of theory.
c) 12-methoxy-iso-deserpidic acid-lactone: 1.5 g of the 3-dehydro-12-methoxy-deserpidic acid-lactone described under b) are, as indicated in Example 4, reduced with sodium borohydride and worked up. 12-methoxy-iso-deserpidic acid-lactone of the formula I, in which R1, R, and R3 = H, R4 = OCH3, crystallized from methanol in needles of melting point 252-254, [11] D = -148 ( c = 0.2 in pyridine). Yield 601 / o of theory.
<I> Example 6 </I> 10-methoxy-iso-deserpidic acid-lactone a) Lacton-lactam: 2.5 g oxy-acid-lactam (Formula II: R1, R3 and R4 = H, R2 = OCH3) , as described in Example 1, heated to reflux in benzene with acetic anhydride / sodium acetate overnight.
The lactone-lactam is obtained from ethyl acetate in needles with a melting point of 178-180, [a] 20 = -25 (c = 0.4 in pyridine). Yield 77o / 11 of theory. b) 3-Dehydro-10-methoxy-de: serpidic acid-lactone: 0.2 g of the lactone-lactam described under a) are treated with phosphorus oxychloride as described in Example 1 and worked up. He keeps the compound from methanol.
Needles of m.p. 268-270, [a] 20 = +121 (c = 0.3 in pyridine). Yield 92% of theory.
c) 10-methoxy-iso-deserpidic acid-lactone: 0.17 g of the 3-dehydro-10-methoxy-deserpidic acid-lactone described under b) is dissolved in 4 cm3 of 50% aqueous acetic acid and pre-shaken with 0.03 g hydrogenated platinum catalyst in a hydrogen atmosphere, with almost the amount calculated for 1 mol of hydrogen being absorbed within 4 hours.
After the hydrogen uptake has ended, the catalyst is filtered off, the filtrate is concentrated strongly in vacuo, the residue is dissolved in methylene chloride, the solution is shaken out with dilute ammonia, dried over sodium sulfate and evaporated to dryness in vacuo. From benzene, the residue gives 10-methoxy-iso-deserpidic acid lactone of the formula I, in which R1, R3 and R4 = H, R2 = O, - are "H".
Needles of m.p. 140-142 and 212 (double-sp.). [a120 = -91 (c = 0.3 in pyridine). Yield 94% of theory.
<I> Example 7 </I> 11-Benzyloxy-iso-deserpidic acid-lactone a) Lacton-lactam: 1.1g oxy-acid-lactam (Formula 1I: R1, R: and R4 = H, R3 = OCHzCOH " ) who, as described in Example 1, heated in benzene with acetic anhydride / sodium acetate overnight.
The lactone-lactam is obtained from ethyl acetate in needles with a melting point of 203, [a] 20 = -13 (c = 0.3 in pyridine). Yield 84% of theory.
b) 3-Dehydro-11-benzyloxy-deserpidic acid-lactone: 0.81 g of the lactone-lactam described under a) are treated with phosphorus oxychloride and worked up as described in Example 1. The compound is obtained from methanol. Needles of m.p. 275, [a1-20 + 92 (c = 0.3 in pyridine). Yield 671% of theory.
c) 11-Benzyloxy-iso-deserpidic acid lactone: 0.1 g of the 3-dehydro-11-benzyloxydeserpidic acid lactone described under b) are catalytically hydrogenated and worked up as described in Example 6.
11-Benzyloxy-iso-deserpidic acid lactone of the formula I, in which R1, R2 and R4 = H, R3 = OCH2 - C6H, - are crystallized from ethyl acetate in needles with a melting point of 23 8-240; [a] 20 = -106 (c = 0.3 in pyridine). Yield 84% of theory.
<I> Example 8 </I> 9-chloro-iso-deserpidic acid-lactone a) Lacton-lactam: 4.95 g oxy-acid-lactam (Formula II: R2, R3 and R4 = H, R1 = Cl) are, as described in Example 1, heated to reflux overnight in benzene with acetic anhydride sodium acetate.
The lactone-lactam is obtained from ethyl acetate in polyhedra with a melting point of 209-210, [a] 2 () = -27 (c = 0.2 in pyridine). Yield 7711/9 of theory.
b) 3-Dehydro-9-chloro-deserpidic acid-lactone: 0.5 g of the lactone-lactam described under a) are, as described in Example 1, heated with phosphorus oxychloride and worked up. The new compound is obtained from methanol in needles with a melting point of 309-310, [a] 20 = + 156 (c = 0.2 in pyridine). Yield 79% of theory.
c) 9-chloro-iso-deserpidic acid 1'actone: 0.1 g of the dehyd'ro-lactone described under b) are, as described in Example 6, catalytically hydrogenated and worked up. Lactone 9-chloro-iso-deserpidic acid of the formula I, in which R2, R3 and R4 = H, R1 = Cl, crystallizes from methanol in rhombohedra of melting point 237-239 ', [a] 20 = -75 (c = 0.2 in pyridine). Yield 89; 0 of theory.
<I> Example 9 </I> 10,11-Dioxymethylene-iso-deserpidic acid-lactone a) Lacton-lactam: 1.09g oxy-acid-lactam (Formula II: R1 and R4 = H, R2 and R3 = -O- CH2-O-) are treated as described in Example 1 in benzene with acetic anhydride and sodium acetate.
The lactam-lactone is obtained from ethyl acetate in needles with a melting point of 219-220, [cc] D = -34 (c = 0.2 in pyridine). Yield 87% of theory.

c) 10.11-Dimethoxy-iso-deserpidic acid-lactone: 0.2 g of the dehydro-lactone described under b) are, as described in Example 4, reduced with sodium borohydride and worked up.

Patent No. 360058) and 1.5 g of anhydrous sodium acetate are refluxed for 15 hours in a solution of 10 cm- 'acetic anhydride and 100 cm-' benzene, the substance dissolving within 1 hour. After cooling, it is evaporated to dryness in vacuo and the residue is partitioned between sodium hydrogen carbonate solution and chloroform. Since the lactone-lactam does not crystallize, the amorphous residue is processed further directly.
b) 3-Dehydro-11-propoxy-deserpidic acid lactone: 2.8 g of the lactone-lactam described under a) are heated in a solution of 50 cm3 of freshly distilled phosphorus oxychloride for 2 hours in a nitrogen atmosphere under reflux. The yellow solution is evaporated to dryness in vacuo, the residue is dissolved in methylene chloride, the solution is extracted with dilute ammonia solution and dried over sodium sulfate,
whereupon when the methylene chloride is concentrated and displaced by methanol, 3-dehydro-11-propoxy-deserpidic acid-lactone crystallizes out in needles of melting point 283-285. [a] 2 = + 103 (c = 0.2 in pyridine).
c) 11-propoxy-iso-deserpidic acid-lactone: 1.6 g of 11-propoxy-3-dehydro-deserpidic acid-lactone are dissolved in a mixture of 60 cm3 of methylene chloride and 30 cm3 of methanol and the solution is added in two portions at room temperature with 1.7g sodium borohydride. When the reaction has ended, a little acetic acid is added and the solution is then evaporated to dryness.
The remaining residue is distributed between dilute ammonia solution and methylene chloride. After evaporation of the solvent, the 11-propoxy-iso-deserpidic acid lactone of the formula 1, in which R1, R2 and R4 = H, R3 = OC3H7, R5 = CH3, R6 = H, is obtained from methanol in hexagonal leaflets crystallized.
M.p. 185-187 (sint. At 130). MM = -117 c = 0.2 in pyridine).
<I> Example 13 </I> 11-chloro-iso-deserpidic acid-lactone a) Lactone lactam: 2.34 g oxy-acid-lactam (Formula II: R1, R2 and R4 = H, R3 = Cl, RS = CH3, R6 = H), as in example! 1 indicated, heated to reflux in benzene with acetic anhydride sodium acetate overnight.
The lactone-lactam crystallizes from the amorphous residue from ethyl acetate in rhomboedarn with a melting point of 145-147. [a] D = -31 (c = 0.2 in pyridine).
b) 3 - dehydro -11-chloro-deserpidic acid-lactone: 1.77 g of the lactone-lactam described under a) are, as described in Example 1, treated with phosphorus oxychloride and worked up. The compound is obtained from methanol in needles with a melting point of 300-304. [a] 20 = + 124 (c = 0.2 in pyridine).
c) 11-chloro-iso-deserpidic acid "lactone: 1.34 g of the 3-dehydro-11-chloro-deserpidic acid-lactone described under b) are: as indicated in Example 12, reduced with sodium borohydride and worked up.

<I> Example 17 </I> 11-Isopropoxy-i: so-deserpidic acid-lactone a) Lacton-lactam: 2.29 g of oxy-acid-lactam (Formula II: R1, RZ and R4 = H, R3 - OCH (CH3) 2, R5 = CH3, R6 = H) are, as given in Example 1, refluxed in benzene with acetic anhydride-sodium acetate overnight.

2.0 g of oxy-acid lactam (Formula 1I: R1, R2 and R4 = H, R3 = OCH3, R5 = CH2CH3, RG = H) are, as described in Example 1, in benzene with acetic anhydride sodium acetate Heated to boiling under reflux overnight. The lactone-lactam is obtained as an amorphous residue which up to now could not be crystallized.
b) 3-Dehydro-11-methoxy-17-desmethoxy-17-ethoxy-deserpid'sä # ure-lactone: 0.86 g of the amorphous lactone-lactam described under a) are, as indicated in Example 1, treated with phosphorus oxychloride and worked up. c) 3-Dehydro-11-methoxy-17-desmethoxy-17-ethoxy-iso-deserpidic acid lactone: 0.8 g of the 3-dehydro lactone described under b) are, as indicated in Example 12, reduced with sodium borohydride and worked up .
The new compound (formula I: R1, R2 and R4 = H, R3 = OCHS, R5 = CH, CH3, R6 = H) crystallizes from methanol in rhombohedra with a melting point of 1150 (indistinct). [a] 20 = r 106 (c = 0.2 in pyridine).
<I> Example 20 </I> 11-Methoxy-17-desmethoxy-17-propoxy-iso-deserpidic acid-lactone a) Lactone-lactam: 4.02 g of oxy-acid lactam (Formula 1I: R1, R2 and R4 = H, R3 = OCHS, R5 = CH, CH2CH3, R "= H) are, as stated in Example 1, refluxed in benzene with acetic anhydride-sodium acetate overnight.
Since the lactone-lactam does not crystallize, the amorphous residue is processed further directly.
b) 3-Dehydro-11-methoxy-17-desmethoxy-17-propoxy-deserpidic acid-lactone: 1.42 g of the lactone-lactams described under a) are, as indicated in Example 1, heated with phosphorus oxychloride and worked up. The compound is obtained from methanol in fine prisms with a melting point of 238-2400. [a] D = + 1220 (c = 0.2 in pyridine).
c) 11-methoxy-17-desmethoxy-17-propoxy-iso-deserpidic acid-lactone: 1.26 g of the dehydro-lactone described under b) are, as indicated in Example 12, reduced with sodium borohydride and worked up.
The new compound (formula I: R1, R2 and R4 = H, R3 = OCHS, R5 = CH2CH2CH3, RG = H) crystallizes from methanol in prisms with a melting point of 870. [,] D = -1090 (c = 0.2 in pyridine).
<I> Example 21 </I> 11-Methoxy-17-desmethoxy-17-isopropoxy-iso-. deserpidic acid-lactone a) Lacton-lactam: 2.92 g of oxy-acid-lactam (Formula 1I: R1, R2 and R4 = H, R3 = OCHS, R5 = CH (CH3) 2, R6 = H) are as in Example 1 be written, heated to boiling under reflux in benzene with acetic anhydride-sodium acetate overnight.
The lactone-lactam is obtained as a yellow foam which up to now could not be crystallized.
b) 3-Dehydro-11-methoxy-17-desmethoxy-17-isopropoxy-deserpidic acid-lactone: 2.74 g of the lactone-lactam described under a) are, as indicated in Example 1, heated with phosphorus oxychloride and worked up . The new compound is obtained from methanol in flakes with a melting point of 215-220 '. [a] D = + 105 (c = 0.2 in pyridine).
c) 11-methoxy-17-desmethoxy-17-isopropoxy-iso-deserpidic acid-lactone: 2.32 g of the 3-dehydro-lactone described under b) are, as indicated in Example 12, reduced with sodium borohydride and worked up.
The new compound (formula I: R1, R2 and R4 = H, R3 = OCH3, R5 = CH (CH3) 2, R, H) crystallizes from acetone-ether in prisms with a melting point of 210-212. [,] 20 = -113 (c = 0.2 - in pyridine).
<I> Example 22 </I> (d) -5-methyl-iso-deserpidic acid-lactone 1- a) lactone-lactam: 2.87 g (d) -oxy-acid-lactam (formula 1I: R1, R2 , R3, R4 = H, R5 and R6 = CH3) are, as indicated in Example 1, refluxed in benzene with acetic anhydride and sodium acetate overnight.
The lactone lactam is obtained from ethyl acetate in prisms with a melting point of 193 to 1940. [a] 20 = -180 (c = 0.2 in pyridine).
b) (d) -3-Dehydro-5-methyl-deserpidic acid-lactone: 1.56 g of the lactone-lactam described under a) are, as indicated in Example 1, treated with phosphorus oxychloride and worked up. The new compound is obtained as an amorphous residue which has not been further characterized.
c) (d) -5-methyl-iso-deserpidic acid-lactone: 1.5 g of the (d) -3-dehydro-5-methyl-deserpidic acid-lactone described under b) are given as in Example 12 with Sodium borohydride reduced and worked on.
