Das Verfahren gemäss vorliegender Erfindung ist dadurch gekennzeichnet, dass man ein in 11 -Stellung eine Sauerstoffunktion aufweisendes 5a,6a-Oxydo- 17a,21-dioxyallopregnan 3,20-dion-bis-(alkylen-ketal) der Formel 1 mit einer Alkyl- oder Aryl-metallver- bindung, vorzugsweise einem Methylgrignardreagens umsetzt, den so erhaltenen Metall-Steroidkomplex in praktisch neutralem Medium zum entsprechenden,
eine 11-Sauerstoffunktion aufweisenden 5a,17a,21- Trioxy-6ss-alkyl (oder aryl)-allopregnan-3,20-dion- 3,20-bis-(alkylen-ketal) (1I) hydrolysiert, letzteres durch Hydrolyse mit einer Säure in das eine 11-Sauer- stoffunktion aufweisende 5a,17a,21-Trioxy-6ss-alkyl- (oder aryl)-allopregnan-3,20-dion (I11)
umwandelt und durch Wasserabspaltung das in 11-Stellung eine Sauerstoffunktion aufweisende 6-Alkyl-(oder Aryl)- 17a,21-dioxy-4-pregnen-3,20-dion (IV) herstellt. Die Wasserabspaltung kann mittels einer Säure oder Base erfolgen.
Grosse Mengen an Base oder Säure bilden das 6a-Epimere, 11-Oxy-(oder Keto)-6a-alkyl- (oder aryl)-17a,21-dioxy-4-pregnen-3,20-dion, klei nere Mengen an Säure oder Base, bei denen das Reaktionsgemisch annähernd neutral ist, das 6ss Epimer, 11-Oxy-(oder Keto)-6ss-alkyl-(oder aryl)- 17a,21-dioxy-4-pregnen-3,20-dion, das mit weiterer Säure oder Base in einer zusätzlichen Stufe in das a-Epimer umgewandelt werden kann. Man kann in der ersten Verfahrensstufe z.
B. Dialkyl-Kadmium- verbindungen, Alkyl- und Phenyl-Kadmiumhalogenid'e oder Aryl- und Alkyl-Kalziumhalogenide, wie Phenyl- Kalzium-jodid, verwenden, arbeitet jedoch vorzugs weise mit Alkyl- und Aryl-Grignardreagenzien wie Äthyl-, Propyl-, Butyl-, Phenyl-magnesium-bromid oder -jodid, wobei man 11-Oxy-(oder Keto)-5a,17a,
21- Trioxy-6ss-alkyl-(oder 6ss-aryl)-allopregnan-3,20- dione erhält, welche durch die folgenden Umsetzun gen in die entsprechenden, eine 11-Sauerstoffunktion aufweisenden 6-Alkyl- oder 6-Ary1-17a,21-dioxy-4- pregnen-3,20-dione übergehen.
Die Bezeichnung 11-Sauerstoffunktion bedeutet hierin eine in 11-Stellung befindliche Oxy- oder Ketogruppe.
Unter den erfindungsgemäss herstellbaren Ver bindungen sind besonders wertvoll aktive 6a-Methyl- hydrocortison und das 6a-Methyl-cortison.
Die neuen Verbindungen, wie z. B. 6-Methyl- cortison und 6-Methyl-hydrocortison und deren Ester, sind aktive Nebennierenrindenhormone mit gluco- corticoider Wirkung. Sie haben ausserdem den Vor teil eines schwachen Salzretentionsvermögens und einer antiphlogistischen Wirkung, die sie für die orale, parenterale wie auch äusserliche Verwendung wertvoll macht.
Insbesondere sind die 6a-Methyl- Epimeren, 6a-Methyl-hydrocortison und 6a-Methyl- cortison und deren 21-Ester wirksamer als Hydro- cortison. Die 6/3-Methylepimeren lassen sich leicht mit Säuren oder Basen, wie im Beispiel 17 gezeigt, in die 6a-Epimeren umwandeln. In der Tabelle 1 ist ein Vergleich der Aktivität dieser Verbindungen mit derjenigen des Hydrocortisons zusammengestellt.

Die erfindungsgemäss erhältlichen Verbindungen können für human- oder veterinärmedizinische Ver wendung zubereitet werden, indem man sie in für die gewünschte Verabreichung geeignete feste oder flüssige Trägerstoffe einverleibt.
Sie dienen ferner als Zwischenprodukte bei der Herstellung von 6-Alkyl-(oder Aryl)-cortisonestem, in die sie z. B. durch Oxydation der 21-Acylate mit Chromsäure überführbar sind.
Die Ausgangsprodukte für das erfindungsgemässe Verfahren, die 5a,6a-Oxydo-11-oxy-(oder keto)-17a,21-dioxy- allopregnan-3,20-dion-3,20-bis-(ketale) erhält man durch Ketalisierung von Cortison, Hydro- cortison und der Epi-F-Verbindung und anschliessende Behandlung mit einer organischen Persäure wie in Präparation 1 und 2 beschrieben.
Bei der Durchführung des erfindungsgemässen Verfahrens wird ein 5a,6a-Oxydo-11-oxy-(oder keto)- 17a,21-dioxy-allopregnan-3,20-dion-3,20-bis-(ketal), gewöhnlich ein Äthylenketal, vorzugsweise in einem organischen Lösungsmittel wie Tetrahydrofuran, Benzol, Toluol, Äthyläther, Propyläther usw.
gelöst, wobei man die höher siedenden Lösungsmittel, wie Tetrahydrofuran und Benzol, bevorzugt, mit einer Alkyl- oder Aryl-metallverbindung, insbesondere einem Methylmetallhalogenid wie Methylmagnesium- chlorid, -bromid oder -jodid, Methyllithium, Di- methylcadmium oder dergleichen umgesetzt.
Andere geeignete Alkyl- oder Arylmetalle und Alkyl- oder Aryl-metallhalogenide sind Äthyl-, Propyl-, Butyl-, Pentyl-, Hexyl-, Phenyl-, Benzyl-magnesiumchlorid, -bromid oder -jodid, Methyllithium, Phenyllithium, -natrium- oder -kalium, Phenylkalziumjodid,
die Alkylcadmiumhalogenide und Dialkylcadmiumverbin- dungen, deren Alkylgruppe 1 bis 6 Kohlenstoffatome aufweist, wobei Methyl-magnesiumbromid und Me- thyl-magnesiumjodid bevorzugt werden. Gemäss einer bevorzugten Ausführungsform zur Herstellung von 6-Methylsteroiden beginnt man die Reaktion bei Zimmertemperatur oder darunter, wobei Tempera turen zwischen 0 und 30 C bevorzugt werden.
Nach Zusatz des Methyl-magnesiumhalogenids oder eines anderen Methylmetallhalogenids erhöht man die Tem peratur und erwärmt die Mischung während 1 bis 48 Stunden auf etwa Rückflusstemperatur. Gewöhn lich verwendet man einen grossen überschuss an Grignardreagens (10 bis 500 Moläquivalente). Die Reaktionstemperatur liegt in der Regel zwischen 25 und 100 C, und der bevorzugte Bereich ist 60 C bis zur Rückflusstemperatur der Mischung, das heisst etwa 80 bis 85 C.
Nach Beendigung der Reaktion wird die Mischung in praktisch neutralem Medium zersetzt. Gemäss der bevorzugten Ausführungsform wird die Reaktions mischung mit einer gesättigten wässrigen Ammonium chloridlösung versetzt, mit Eis gekühlt und mehrere Minuten bis eine Stunde gerührt. Die wässrige und organische Schicht werden voneinander getrennt. Die organische Phase wird gewaschen, getrocknet und verdampft, wobei rohes 11-Oxy-(oder Keto)-5a,17a,21-trioxy-6ss-methyl- allopregnan-3,20-dion-3,20-(alkylenketal) zurückbleibt, das nach üblichen Verfahren wie Um kristallisieren und/oder Chromatographie mit orga nischen Lösungsmitteln gereinigt werden kann.
Das so erhaltene 11-Oxy-(oder Keto)-Sa,17a,21-trioxy-6ss-methyl- allopregnan-3,20-dion-3,20-bis-(alkylenketal) wird mit einer Säure, vorzugsweise in einem mit Wasser mischbaren Lösungsmittel, z. B. in wässrig alkoholischem saurem Medium, hydrolysiert. Als Alkohole bevorzugt man Methanol und Äthanol, doch kann man auch tert.-Butylalkohol, Propyl- alkohol, Isopropylalkohol oder Dioxan, Aceton usw. als Lösungsmittel verwenden.
Als Säure kann man eine anorganische oder organische Säure, vorzugs weise eine Mineralsäure, wie Schwefelsäure oder Salz säure, verwenden, doch lassen sich auch organische Säuren, wie z. B. Ameisen-, Essig-, Propionsäure oder Toluolsulfonsäure, verwenden.
Die Mischung kann am Rückfluss gekocht, dann mit Natriumbikar- bonat, Natriumkarbonat, Kaliumkarbonat, Kalium hydroxyd oder andern Alkalilösungen neutralisiert und konzentriert werden, wobei man ein rohes, aus 11-Oxy-(oder Keto)-5a,17a,21-trioxy-6ss-methyl-allo- pregnan-3,20-dion bestehendes Produkt erhält. Dieses kann durch Umkristallisieren aus organischen Lö sungsmitteln wie Aceton, Äthylacetat,
Skellysolve B-Hexanen, Methanol, tert. Butylalkohol, Äther oder dergleichen und Mischungen derselben, gereinigt wer den.
Aus dem so erhaltenen 11-Oxy-(oder Keto)- 5 a,17a,21-trioxy-6ss-methyl- allopregnan- 3,20- dion wird nun Wasser abgespalten, was in alkalischer oder saurer Lösung in An- oder Abwesenheit einer Stick stoffatmosphäre erfolgen kann. Gemäss einer bevor- zagten Ausführungsform arbeitet man in alkalischem Medium.
Zu diesem Zweck wird das Steroid in Methanol, Äthanol, Dioxan oder sonst einem geeigne ten Lösungsmittel, das mit der verwendeten Base nicht reagiert, gelöst und die Lösung durch Durch leiten von Stickstoff von Sauerstoff befreit und dann mit einer ebenfalls sauerstofffreien Lösung einer Alkalimetallbase umgesetzt. Natrium- oder Kalium- hydroxyd werden bevorzugt; doch kann man auch Alkalimetallalkoholate, Bariumhydroxyd, Kalzium hydroxyd oder dergleichen verwenden.
Die Menge der verwendeten Base bestimmt das Verhältnis von 6a-Methyl-epimer zu 6P-Methyl-epimer im entstehen den Produkt. Wenn die Menge der Base genügend ist, um die Lösung während der ganzen Reaktions zeit alkalisch zu halten, wiegt das 6a-Methyl-epimer vor, wenn die Lösung nahezu neutral wird, so ent steht das 6,8-Methyl-epimer. Eine nachträgliche Be handlung mit einem enolbildenden Mittel, Säure oder Base, führt das 6ss-Methyl-hydrocortison in das wirk samere 6a-Methyl-hydrocortison über.
Man lässt dann die alkalische Mischung unter Stickstoffatmosphäre 4-48 Stunden bei Temperaturen zwischen 15 und 40 C stehen, wobei 11-Oxy-(oder I#,eto)-6-methyl- 17a,21-dioxy-4-pregnen-3,20-dion ensteht. Zu dessen Isolierung wird beispielsweise mit Essigsäure ange säuert, konzentriert und schliesslich zur Trockne eingedampft.
Der Rückstand wird aus geeigneten Lö sungsmitteln wie Aceton, Skellysolve B-Hexanen, Heptanen, Äthanol, Methanol, tert. Butylalkohol, Dioxan, Äther, Aceton und dergleichen umkristalli siert, um das reine 11-Oxy-(oder Keto)-6-methyl- 17a,21-dioxy-4-pregnen-3,20-dion zu erhalten.
Die Wasserabspaltung kann auch erfolgen, indem man das 11-Oxy-(oder Keto)-5a,17a,21-trioxy-6ss- methylallopregnan-3,20-dion mit einer Säure oder mit Thionylchlorid in Pyridinlösung umsetzt, wobei das entsprechende 11-Oxy-(oder Keto)-6-methyl-17a,21- dioxy-4-pregnen-3,20-dion erhalten wird.
Die 21-Acylate des so erhaltenen 6-Methyl- cortisons-, -hydrocortisons und -Epi-F-Verbindungen erhält man nach den gleichen üblichen Methoden, wie sie für Cortison, Hydrocortison und Epi-F-Ver- bindung bekannt sind.
In den folgenden Präparationen wird gezeigt, wie die Ausgangsstoffe für das erfindungsgemässe Ver fahren hergestellt werden können. Präparation <I>1</I> 5a,6a-Oxydo-11ss,17a,21-trioxy-allopregnan- 3,20-dion-3,20-bis-(äthylenketal) A.
Zu einer Lösung von 0,901 g 11ss,17a,21- Trioxy-5-pregnen-3,20-dion-3,20-bis-(äthylenketal) in 18 ml Chloroform gibt man eine Lösung von 331 mg Perbenzoesäure in 5,19 ml Chloroform. Man lässt die Lösung im Kühlschrank (etwa 4 C) 24 Stunden und dann weitere 72 Stunden bei Zimmertemperatur stehen.
Die Reaktionslösung wird dann mit 5<B>%</B> iger Natriumbikarbonatlösung und Wasser gewaschen, über wasserfreiem Natriumsulfat getrocknet und zur Trockne eingedampft, wobei man 1,031 g rohes festes Produkt erhält. Nach dem Umkristallisieren aus Aceton erhält man 431 mg eines bei 230-247 C schmelzenden Produktes.
Die Mutterlauge wird zur Trockne verdampft, der Rückstand in Methylen- chlorid gelöst und über 25g mit Säure gewaschenem Aluminiumoxyd chromatographiert. Die Säule wird mit je 3 Fraktionen Methylenehlorid plus 5, 10, 15, 20, 25 und 50 0!o Aceton,
Aceton und Aceton plus 5 % Methanol entwickelt. Das Eluat mit Aceton plus 5 % Methanol gibt weitere 279 mg hochschmelzendes Produkt.
Das hochschmelzende Material, 5a,6a-Oxydo-11 ss,17a,21-trioxy-allopregnan- 3,20-dion-3,20-bis-(äthylenketal) wird dreimal aus Aceton und Methanol umkristalli siert, wonach sein Schmelzpunkt 263-268 C ist. Andere Eluatfraktionen mit niedrigerem Schmelz punkt enthalten das 5ss,6ss-Isomere.
B. In genau gleicher Weise wie unter A be schrieben, wird lla,17a,21-Trioxy-5-pregnen-3,20- dion-3,20-bis-(äthylenketal) mit Perbenzoesäure in Chloroform eiloxydiert, wobei eine Mischung von 5a,6a-Oxydo- und 5f,6ss-Oxydo-lla,l7a,21-trioxy- pregnan-3,20-dion-3,20-bis-(äthylenketal), erhalten wird, die man auf chromatographischem Wege in die beiden Isomeren zerlegt.
Präparation <I>2</I> 5 a,6a-Oxydo-17a,21-dioxy-allopregnan-3,11,20- trion-3,20-bis-(1,2-propylenketal) Zu einer Lösung von<B>19</B> 17a,21-Dioxy-5-pregnen- 3,11,20-trion-3,20-bis-(1,2-propylenketal), [Cortison- 3,20-bis-(1,2-propylenglykolketal)] in Chloroform gibt man eine Chloroformlösung von Perbenzoesäure und lässt wie in Präparation 1A zunächst im Kühl schrank und dann bei Zimmertemperatur stehen, wäscht, trocknet und verdampft wie dort.
Umkristal- lisieren aus Aceton gefolgt von einer Fraktionierung der Mutterlauge auf einer Säule aus mit Säure ge waschenem Aluminiumoxyd nach der unter Präpa- ration 1A beschriebenen Methodik ergibt 5a,6a-Oxydo-17a,21-dioxy-allopregnan-3,11,20- trion-3,20-bis-(1,2-propylenketal) und sein 5ss,6ss-Oxydo-Isomeres.
Bei Verwendung des leichter zugänglichen Corti- son-3,20-bis-(äthylenketals) erhält man 5a,6a-Oxydo-17a,21-dioxy-allopregnan-3,11,20- trion-3,20-bis-(äthylenketal). In gleicher Weise wie in den Präparationen 1 und 2 kann man durch Umsetzung der entsprechen den Diketale des Cortisons, Hydrocortisons und Epi-F, deren Ketalgruppe durch Umsetzung des Steroid-3,20-dions mit Äthylen-, Propylen-, 1,2-,
1,3- oder 2,3-Butylenglykol oder Pentan-, Hexan-, Heptan- oder Octandiolen, deren Alkoholgruppen vicinal, z.
B. in 1,2-, 2,3 -, 3,4-Stellung oder durch ein Kohlen stoffatom getrennt, wie in 1,3-, 2,4-, 3,5-Stellung sind, gebildet wurden, mit einer organischen Per säure, wie Perameisen-, Peressig-, Perbenzoe-, Mono- perphthalsäure oder dergleichen Sa,6a-Oxydo-1 I a,17a,21-trioxy-allopregnan- 3,20-dion-3,20-bis-(alkylenketale), 5a,6a-Oxydo-11 ss3,17a,
21-trioxy-allopregnan- 3,2-dion-3,20-bis-(alkylenketale) und 5a,6a-Oxydo-17a,21-dioxy-allopregnan- 3,11,20-trion-3,20-bis-(alkylenketale) erhalten. Für die Zwecke der vorliegenden Erfindung werden Ausgangssteroide mit der Äthylenketalgruppe bevorzugt, da diese Ketale im allgemeinen leichter herstellbar sind als die durch Umsetzung der 3,20- Diketoverbindungen mit höheren Alkandiolen erhält lichen.
In den nachfolgenden Beispielen 1 bis 5 wird die erste Stufe, in den Beispielen 6 bis 10 die zweite Stufe und in den Beispielen 11 bis 15 die dritte Stufe des erfindungsgemässen Verfahrens beschrieben.
<I>Beispiel 1</I> 3,20-bis-(Äthylenketal) des 5a,11 j3,17cc,21-Tetraoxy- 6/3-methyl-allopregnan-3,20-dions Eine Lösung von 1,115 g 5a,6a-Oxydo-11f-17a,21-trioxy-allopregnan- 3,20-dion-3,20-bis-(äthylenketal)
in 165 ml durch Destillation über Lithiumaluminium- hydrid gereinigtem Tetrahydrofuran wird tropfen weise zu einer Lösung von 95 ml Methylmagnesium- bromid (vierfach molare Konzentration an Methyl- magnesiumbromid) gegeben.
Zu dieser Mischung gibt man 575 ml Benzol und erhitzt 26 Stunden unter Rühren am Rückfluss. Nach dem Abkühlen giesst man in 700 ml eisgekühlte, gesättigte Ammonium chloridlösung, rührt 30 Minuten und trennt die Benzolphase von der wässrigen Phase. Die wässrige Phase wird dreimal mit je 200 ml Äthylacetat extra hiert und die Extrakte zur Benzolphase gegeben. Die organische Phase wird mit Wasser gewaschen, über wasserfreiem Natriumsulfat getrocknet und zur Trockne eingedampft. Man erhält 1,314 g festes Rohprodukt.
Nach Verreiben mit Äthyläther verblei ben 1,064 g kristallines Produkt vom Smp. 221 bis 230 C. Durch Umkristallisieren erhält man eine Analysenprobe vom Smp. 228-233 C; [a]v - 11 in Chloroform, des 5a,11(3,17a,21-Tetraoxy-6ss-methyl-allopregnan- 3,20-dion-3,20-bis-(äthylenketals).

Zu dieser Lö sung gibt man eine in gleicher Weise Sauerstoff frei gemachte Lösung einer n/10 Natronlauge (150 ml) und lässt bei Zimmertemperatur (etwa 22-24 C) unter Stickstoffatmosphäre 20 Stunden stehen, worauf man mit 14 ml Essigsäure ansäuert.
Die so erhaltene saure Lösung wird bei 50-60 C im Vakuum einge dampft, der Rückstand in 200 m1 Äthylacetat gelöst und mit 200 ml Wasser behandelt. Die von der orga nischen Schicht abgetrennte wässrige Schicht wird verworfen, die organische Schicht mit 350 ml 5 % iger wässriger Natriumbikarbonatlösung, dann dreimal mit Wasser gewaschen, über wasserfreiem Natriumsulfat getrocknet und auf 180 ml eingeengt. Nach Kühlung des Konzentrats im Kühlschrank (etwa 5 C) wird filtriert und man erhält 11,9g Material.
Dieses wird wieder in 500 ml Äthylacetat gelöst, die Lösung auf 150 ml eingeengt und wie vorher gekühlt. Ausbeute <B>6,15</B> g rohes 6ss-Methyl-hydrocortison vom Smp. 220 bis 223 C.
Nach dreimaligem weiterem Umkristallisieren dieses Rohproduktes aus Äthylacetat erhält man eine analysenreine Probe des 6ss-Methyl-hydrocortisons vom Smp. 223-227 C.

Dazu gibt man bei Zimmertemperatur 15 ml 2n-wässrige Schwe felsäure.
Man lässt über Nacht (18 Stunden) bei Zimmertemperatur stehen, neutralisiert mit 225 ml wässriger 5 /aiger Natriumbikarbonatlösung und kon zentriert dann auf dem Wasserbad bei etwa 50 bis 55 C auf ein Volumen von etwa 175 ml. Dann wird filtriert und der Niederschlag mit 250 ml Wasser gewaschen und im Vakuumtrockenschrank bei 65 C getrocknet. Man erhält 7,72 g festes Produkt vom Smp. 195-205 C. Die wässrige Lösung wird dreimal mit 150 ml Äthylacetat extrahiert, die Extrakte ver einigt, dreimal mit je 150 ml Wasser gewaschen, über wasserfreiem Natriumsulfat getrocknet und unter ver mindertem Druck bei 55 C zur Trockne eingedampft.
Der Rückstand wird mit den vorher erhaltenen 7,72 g vereinigt und aus Äthylacetat umkristallisiert. Man erhält in zwei Fraktionen 6,44 g 6a-Methyl-hydro- cortison von 95 % Reinheit. Kristallisiert man 1 g dieses Produktes zweimal aus Äthylacetat, so erhält man 6a-Methyl-hydrocortison vom Smp. 200 bis 207 C; [a] D + 114 in Aceton.

Man lässt über Nacht 16 Stunden stehen, verdünnt dann mit 200 ml Was- ser,.neutralisiert mit 5 1/aiger wässriger Salzsäure und extrahiert dreimal mit je 75 ml Methylenchlorid. Die Methylenchloridextrakte werden mehrmals mit Was ser gewaschen, über wasserfreiem Natriumsulfat ge trocknet, eingedampft und der so erhaltene Rück stand aus Aceton umkristallisiert. Man erhält 6a- Methyl-hydrocortison.
Erfindungsgemäss hergestellte Verbindungen kön nen folgendermassen verestert werden: Man stellt eine Mischung aus 164 mg 6a-Methyl- hydrocortison in 1 ml Pyridin und 1 ml Essigsäure anhydrid her. Diese Mischung lässt man bei Zimmer temperatur (22-24 C) 16 Stunden stehen und giesst sie dann in 10 ml Eiswasser. Die so erhaltene wäss- rige Mischung wird dreimal mit je 25 ml Methylen- chlorid extrahiert.
Die vereinigten Methylenchlorid- extrakte werden gewaschen, über wasserfreiem Na triumsulfat getrocknet und über 15g Florisil chro- matographiert. Die mit Skellysolve B plus 20 und 30 /o Aceton eluierten Fraktionen (92 mg) werden eingedampft und das erhaltene Produkt dreimal aus Aceton-Skellysolve B Hexan umkristallisiert, um das reine 6a-Methyl-hydrocortisonacetat vom Smp. 213 bis 214 zu erhalten.

6a-Methyl-hydrocortison-21-hemisuccinat Zu einer Lösung von 2,5 g Bernsteinsäure anhydrid in 25 ml Pyridin gibt man unter Rühren 2,0 g 6a-Methyl-hydrocortison und rührt weiter, bis alles 6a-Methyl-hydrocortison in Lösung gegangen ist. Nach Stehenlassen über Nacht wird die Mischung langsam in eine kräftig gerührte Mischung von 30 ml konzentrierter Salzsäure, 102 ml Wasser und 127 g Eis gegossen. Man rührt eine Stunde weiter und sammelt das rohe 6a-Methyl-hydrocortison-21-hemi- succinat auf dem Filter.
Das Produkt wird auf dem Filter so lange mit Wasser gewaschen, bis das p11 des Filtrats 4,0 ist, getrocknet und aus 45 ml Methyläthylketon und 36 ml Skellysolve B Hexanen umkristallisiert, um den reinen Ester zu erhalten.
Ausgehend vom 6ss-Methyl Epimer erhält man das entsprechende 6ss-Methyl-hydrocortison-21-hemi- succinat.
Das Natriumsalz des 6a-Methyl-hydrocortison-21- hemi-succinates kann erhalten werden durch Um setzung von 6a - Methyl - hydrocortison - 21- hemi- succinat mit Natronlauge. Gleicherweise kann das Natriumsalz des 6ss-Methyl-hydrocortison-21-hemi- succinates erhalten werden.
In der gleichen Weise kann man das Hemi- succinat des 6a-Methyl-cortisons und dessen Na triumsalz oder des 6ss-Methyl-cortisons und dessen Natriumsalz herstellen.
6a-Methyl-hydrocortison-21-(ss,ss-dimethyl-glutarat) Zu einer Lösung von 260 mg ss,ss-Dimethyl- glutarsäureanhydrid in 2 ml Pyridin gibt man 200 mg 6a-Methyl-hydrocortison. Man rührt, bis das 6a- Methyl-hydrocortison vollständig in Lösung gegangen ist und spült den Kolben mit Stickstoff. Die Mi schung wird dreimal mit je 5 ml Äthylacetat extra hiert, die Extrakte mit verdünnter Salzsäure und Wasser gewaschen, über wasserfreiem Magnesium sulfat getrocknet und unter vermindertem Druck auf 1,5 ml eingeengt. Hierzu gibt man 1 ml Skellysolve B Hexane und lässt auf 0 C abkühlen. Nach 24 Stun den wird filtriert und die Kristalle, bestehend aus 6 a-Methyl-hydrocortison-21- (ss,P <I>-</I> dimethyl - glutarat) genommen.
Das Infrarotspektrum dieser Verbindung in Nujol Mineralöl bestimmt, entspricht der ange nommenen Struktur.
Bei Verwendung von, 6ss-Methyl-hydrocortison als Ausgangsstoff erhält man 6ss-Methyl-hydrocortison- 21-(ss,ss-dimethyl-glutarat).

6a-methyl-hydrocortisone-21- (ss, ss-dimethyl-glutarate) 200 mg of 6a-methyl-hydrocortisone are added to a solution of 260 mg of ss, ss-dimethylglutaric anhydride in 2 ml of pyridine. A metal salt of 6a-methyl-hydrocortisone-21- (ss, ss-dimethyl-glutarate) is obtained as follows: The sodium salt of 6a-methyl-hydrocortisone-21- (ss, ss-dimethyl-glutarate) can be obtained by reaction of 6a-methyl-hydrocortisone-21- (ss, ss-dimethyl-glutarate) with sodium hydroxide solution.
The sodium salt of 6a-methylcortisone-21- (ss, ss-dimethyl-glutarate) can be obtained in the same way.
