Verfahren zur Herstellung von Verbindungen des Penicillins mit Sulfonamiden
Das vorliegende Patent bezieht sich auf ein Verfahren zur Herstellung von Verbindungen des Penicillins mit Sulfonamiden der Formel

worin R Wasserstoff oder eine Alkyl-, Pyridin-, Thiazol- oder Pyrimidingruppe bedeutet, welches dadurch gekennzeichnet ist, dass eine Lösung oder Dispersion eines Penicillinsalzes und eine Lösung oder Dispersion eines Salzes eines der genannten Sulfonamide zusammengebracht werden.
Zweckmässig werden wässerige Lösungen der beiden Komponenten verwendet, das heisst, es wird vorteilhaft von wasserlöslichen Salzen ausgegangen, z. B. vom Acetat des Sulfonamids. Im Falle der Verbindungen der Formel (a) werden die Monopenicillinderivate und im Falle der Verbindungen (b) die Bipenicillinderivate gebildet.
Die erfindungsgemäss erhältlichen Verbindungen sind sehr stabil, wenig wasserlöslich und nicht giftig, auch wenn sie in viel höheren Dosen als therapeutisch üblich verabreicht werden. Sie sind deshalb wertvoll, weil sie in ihrem Molekül gleichzeitig ein Antibiotikum und ein Sulfonamid enthalten. Ihre wässrigen Suspensionen, mit oder ohne Beifügung von oberflächenaktiven Stoffen, sind leicht einspritzbar. Die antibiotische Aktivität der Suspensionen und Dispersionen dieser Verbindungen variiert nicht mit der Zeit. Daher sind sie besonders geeignet zur Herstellung von einspritzbaren Suspensionen, von sirupösen Suspensionen, von Pillen und von Dispersionen in fettigen, seifigen oder gallertartigen Trägern.
Die Sulfonamide der Formel (a) können durch Sulfonierung des Benzylaminacetylats mit Chlorsulfonsäure, durch Behandlung des Sulfonsäurechlorids mit Ammoniak oder mit einem primären oder sekundären Amin und schliesslich durch Entacetylierung der erhaltenen Verbindung durch Kochen mit normaler Salzsäure erhalten werden.
Die Sulfonamide der Formel (b) können hergestellt werden, indem man das acetylierte N,N'-Diben- zyl-äthylendiamin der Formel

Beispiel 1 Herstellung der Penicillinverbindung des p-Amino benzylsulfonamids
15 g p-Aminobenzylsulfonamidchlorhydrat werden in 30 ml Wasser aufgelöst. Getrennt davon werden in 30 ml Wasser 22 g Natriumpenicillin-G aufgelöst. Dann mischt man beide Lösungen. Nach einiger Zeit scheidet sich eine Verbindung ab, die, nachdem sie mit kaltem Wasser gewaschen und über Phosphorsäureanhydrid getrocknet wurde, zwischen 122 und 127 C unter Gasentwicklung schmilzt. Wenn man einer der Lösungen, bevor man sie mischt, oder der Mischung selbst Kristalle der bei einem früheren Verfahren hergestellten Substanz als Katalysator beifügt, erfolgt eine raschere Kristallisation. 1 mg der Verbindung, die etwa 4,5 /o Feuchtigkeit aufweist, enthält 1037 bis 1070 internationale Einheiten Penicillin (theoretisch 1090).
Die Wasserlöslichkeit der Verbindung bei Raumtemperatur beträgt 10 0/0.
Beispiel 2 Herstellung der Bipenicillinverbìndung des a, ss-Bis- (p-aminobenzylsulf onamid)-äthans
Man löst 200 g p-Amino-benzylsulfonamid und 100 g Dibromäthan in 1 Liter absolutem Athylalkohol. Das Ganze wird während vier Stunden gekocht.
Das feste Produkt, das sich beim Abkühlen ausscheidet, wird durch Filtrieren abgetrennt und zusammen mit dem durch Eindampfen des Filtrats erhaltenen Erzeugnis aus Wasser, das mit Ammoniak leicht alkalisch gemacht wurde, wiederholt umkristallisiert.
Man erhält eine Verbindung, die zwischen 210 und 2120 C schmilzt und nach dem Trocknen bis zum konstanten Gewicht folgende Werte bei der Analyse aufweist: C=48, 28 /Q ; H-5 950/0 N-l3 640/o; S=15,700/u berechnet für C6H22O4N4S2: C=48,22o/o; H=5, 56 /o ; N=l4,060/o S=16,090/o
15 g des so erhaltenen a,B-Bis-(p-aminobenzylsul- fonamid)-äthans werden in 100 ml Wasser, dem 4,5 g Eisessig zugesetzt wurden, aufgelöst (Lösung A). In 80 ml Wasser werden 24,4 g Natriumpenicillin-G gelöst (Lösung B). Die Lösung A wird nach und nach in die Lösung B, die auf Oo C in einem Eisbad gehalten wurde, eingegossen. Es bildet sich ein voluminöser Niederschlag, welcher nach einiger Zeit kristallin wird.
Die Kristallisation wird durch Beifügung von kleinen Mengen eines durch ein früheres Verfahren hergestellten Produktes, das als Katalysator dient, besonders gefördert. Man filtriert, wäscht in kaltem Wasser und trocknet das Produkt im Vakuum.
Ein Muster des Produktes, das 4,3 O/o Feuchtigkeit enthält, erweicht bei 1050 C und zersetzt sich dann unter Gasentwicklung.
Eine Probe, die 6 O/o Wasser enthielt, wies eine Aktivität von 962 bis 1013 UI/mg auf. Das Produkt sollte theoretisch 1047 UI/mg aufweisen. Wasserlöslichkeit der Verbindung bei normaler Temperatur: 0,3 O/o.
Beispiel 3 Herstellung von a,B-Bis-(p-aminobenzylsulfonamid)- äthan
Man verteilt 72 g vom Diacetat des N,N'-Diben- zyl-äthylendiamins oder die der Base entsprechende Menge in 200 mol Wasser, das 60 ml Essigsäureanhydrid enthält, indem man stark schüttelt. Man erhält ein öliges Produkt, welches, nachdem es in Eis gekühlt wurde, zu einer weissen Kristallmasse gerinnt.
Diese Masse schmilzt, wenn sie aus Methylalkohol umkristallisiert wurde, bei 137 C. Die Ausbeute ist fast quantitativ, wenn man die essigsaure Mutterlauge, nachdem sie mit Natriumcarbonat neutralisiert wurde, konzentriert und das darin aufgelöste Produkt zurückgewinnt. Das erhaltene N,N'-Diacetyl-N,N'-dibenzyl- äthylendiamin wird nach und nach in Chlorsulfonsäure eingeschüttet. Die Mischung wird 4-5 Stunden auf 70800 C erhitzt. Dann lässt man die Flüssigkeit abkühlen und giesst sie in feinem Strahl über etwa 20 Teile zerriebenes Eis. Man erhält ein gummiarti- ges Produkt, das noch feucht, nachdem es in einer kleinen Menge eiskaltem Wasser gewaschen wurde, in 2,5 Teile einer wässrigen Lösung mit 25 O/o Ammoniak eingebracht wird.
Man verrührt es darin zu einem Brei. Nach einigen Stunden mischt man wieder, verdampft das überflüssige Ammoniak, kühlt und filtriert die Masse. Das feste Produkt wird entacetyliert, indem man es mit 6-7 Teilen normaler Salzsäure während einiger Stunden kocht. Man erhält so das Dichlorhydrat des a,ss-Bis-(p-aminoben- zylsulfonamid)-äthans, dessen freie Base bei 210 bis 2120 C schmilzt und mit dem im Beispiel 2 hergestellten Ausgangsstoff identisch ist.
Herstellung der Bipenicillinverbindung des a,ss-Bis- (p-aminosulfobenzylmethylamid)-äthans
Die durch Behandlung des N,N'-Diacetyl-N,N'dibenzyl-äthylendiamins mit Chlorsulfonsäure erhaltene Verbindung der Formel

wird, noch feucht, in zwei Teile einer wässrigen Lösung von 35 O/o Methylamin eingebracht und während einiger Stunden zu Brei gerührt. Durch Destillation wird das überflüssige Methylamin zurückgewonnen, und das filtrierte Produkt wird entacetyliert, indem man es mit 6-7 Teilen normaler Salzsäure kocht.
Man erhält das Dichlorhydrat des a,ss-Bis-(p-amino- sulfobenzylmethylamid)-äthans, welches sich nach der Behandlung mit der stöchiometrischen Menge einer Base in das symmetrische a,B-Bis-(p-amino- sulfobenzylmethylamid)-äthan umwandelt. 1 g dieser Verbindung in einer essigsauren Lösung (0,28 g Essigsäure in 60 ml Wasser) wird mit einer wässrigen Lösung (5 ml) von 1,65 g Natriumpenicillin-G behandelt. Man erhält eine mikrokristalline Verbindung, die im Vakuum in Gegenwart von Phosphorsäureanhydrid getrocknet wird. Eine Probe, die an der Luft getrocknet wurde, wies bei der Analyse einen Wassergehalt von 9,3 o/o auf und besass eine Aktivität von 951 UI/mg (berechnet für das wasserfreie Produkt:
1085 UI/mg, berechnet für ein 9,3 O/o wasserhaltiges Produkt: 984).
Wasserlöslichkeit der Verbindung bei normaler Temperatur: 0,06 0/0.
Beispiel 4 Herstellung der Bipenicillinverbindung des a,fl-Bis- (p-aminosulfondimethylamid)-äthans
Die nach den Angaben des Beispiels 3 durch Behandlung des N,N'-Diacetyl- N,N'-dibenzyl- äthylen- diamins mit Chlorsulfonsäure erhaltene und noch feuchte Verbindung wird in zwei Teile einer wässrigen Lösung von 30 /o Dimethylamin eingebracht und zu Brei gerührt. Durch Destillation wird das überflüssige Dimethylamin zurückgewonnen, und das filtrierte Produkt wird entacetyliert, indem man es mit 6-7 Teilen normaler Salzsäure kocht.
Man erhält das Dichlorhydrat des a,B-Bis-(p-aminosulfonbenzyl- dimethylamid)-äthans, welches sich nach der Behandlung mit der stöchiometrischen Menge einer Base in wässriger Suspension in das a,-Bis-(p-aminosulfon- benzyldimethylamid)-äthan umwandelt, 1,9 g des Diacetates dieser Verbindung in wässriger Lösung werden mit 2,3 g Natriumpenicillin-G in einer wässrigen Lösung behandelt. Sofort bildet sich eine mikrokristalline Verbindung, die an der Luft getrocknet wird.
Eine Probe dieses Produktes, die noch 17 O/o Wasser enthielt, wies eine Aktivität von 876 UI/mg auf (theoretische Aktivität der wasserfreien Verbindung: 1058 UI/mg; theoretische Aktivität einer Verbindung, die 17 O/o Wasser enthielt: 978 UI/mg). Wasserlöslichkeit der Verbindung bei Raumtemperatur: 0,02 O/o.

Solution A is gradually poured into solution B, which has been kept at 0o C in an ice bath. A voluminous precipitate forms, which becomes crystalline after a while.
Crystallization is particularly promoted by the addition of small amounts of a product made by an earlier process which serves as a catalyst. It is filtered, washed in cold water and the product is dried in vacuo.
