Verfahren zur Herstellung von N-(ss-Phenyl-äthyl)-4-phenyl-4-carbalkoxy-piperidinen Die Erfindung betrifft ein Verfahren zur Her stellung von N-(ss-Phenyl-äthyl)-4-phenyl-4-carb- alkoxy-piperidinen und deren Salzen aus 4-Phenyl- 4-carbalkoxy-piperidinen. Diese neuen Verbindungen besitzen eine überlegene analgetische Aktivität.
Die N-(/3-Phenyl-äthyl)-4-phenyl-4-carbalkoxy- piperidine lassen sich durch folgende Strukturformel darstellen:

Die chemische Verwandtschaft von beispielsweise N-(ss-Phenyl-äthyl)-4-phenyl-4-carbomethoxy-piperidin und N-(ss-Phenyl-äthyl)-4-phenyl-4-carboäthoxy-pi- peridin mit dem bekanten Analgeticum Meperidin zeigt sich deutlich bei einem Vergleich obiger Formel mit der Strukturformel von Meperidin

Obgleich Meperidin als Analgeticum in weitem Umfang angewandt wird, ist seine Toxicität in vielen Fällen nachteilig.
Zahlreiche N-substituierte Meperi- dinanalogen wurden bisher in der Erwartung her gestellt, eine Verbindung aufzufinden, die hinsicht lich der analgetischen Aktivität mit Meperidin ver gleichbar ist und eine verminderte Toxicität aufweist; die bisher aufgefundenen Verbindungen erfüllten aber nicht die in sie gesetzten Erwartungen.
Die bisherigen Erfahrungen liessen nicht erwarten, dass die Forschungsarbeit zur Auffindung eines verbes serten Analgeticums des Meperidintypus führen würde; denn man wusste, dass mit steigendem Mole kulargewicht die Toxicität des N-Substituenten zu nimmt und die analgetische Aktivität abnimmt (so besitzt N-Benzyl-4-phenyl-4-carboäthoxy-piperidin nur 50 /o der analgetischen Aktivität von Meperidin).
Angesichts dieser Forschungsergebnisse ist es ausserordentlich überraschend, dass N-(ss-Phenyl- äthyl)-4-phenyl-4-carbomethoxy-piperidin und N-(ss- Phenyl-äthyl)-4-phenyl-4-carboäthoxy-piperidin und ihre Salze nur den dritten Teil der Toxicität, aber die doppelte analgetische Potenz des Meperidins be sitzen.
Bei vergleichbarer analgetischer Aktivität be trägt die Toxicität von N-(B-Phenyl-äthyl)-4-phenyl- 4-carbomethoxy-piperidin und N-(l-Phenyl-äthyl)-4- phenyl-4-carboäthoxy-piperidin nur 1/s von der des Meperidins.
Die N-(ss-Phenyl-äthyl)-4-phenyl-4-carbalkoxy- piperidine der Formel I werden erfindungsgemäss hergestellt, indem man ein 4-Phenyl-4-carbalkoxy- piperidin (III) mit einem fl-Phenyl-äthylhalogenid (I1) umsetzt:

worin X Halogen und R eine Alkylgruppe, insbeson dere einen Methyl- oder Äthylrest, bedeutet.
Allenfalls können durch Behandeln mit einer Säure die entsprechenden Salze der Piperidinverbin- dungen (I) erhalten werden. Es können auch schon Salze als Ausgangsstoffe, wie z. B. 4-Phenyl-4-carbo- (methoxy oder äthoxy)-piperidin-carbonat benutzt werden.
Die Reaktion zwischen dem 4-Phenyl-4-carbo- methoxy oder äthoxy)-piperidin-carbonat und dem ss-Phenyl-äthylhalogenid kann in üblicher Weise durchgeführt werden, indem z. B. die Reaktions teilnehmer in einem flüssigen Medium erhitzt wer den, welches unter den Reaktionsbedingungen prak tisch inert und ein Lösungsmittel für die Reaktions teilnehmer ist. Gewöhnlich verwendet man hierfür ein niedermolekulares Alkanol, wie z.
B. Äthanol, und führt die Reaktion vorzugsweise derart durch, dass die Reaktionsteilnehmer in dem Alkanol unter Rückfluss in Gegenwart einer Base, z. B. Natrium- bicarbonat, erhitzt werden. Als ss-Phenyläthyl- halogenid dient z.
B. das Chlorid, Bromid oder Jodid; vorzugsweise verwendet man ss-Phenyl-äthyl- chlorid oder ss-Phenyl-äthylbromid. Mit diesen be vorzugten Reaktionsteilnehmern und unter den be vorzugten Reaktionsbedingungen ist die Umsetzung gewöhnlich nach etwa 1 bis 2 Tagen abgeschlossen. N-(ss-Phenyl-äthyl)-4-phenyl-4-carbo(methoxy oder äthoxy)-piperidin wird aus dem Reaktionsgemisch z. B. in der Weise gewonnen, dass die anorganischen Salze abfiltriert werden und die alkoholische Lösung zur Trockne im Vakuum eingedampft wird.
Der Rückstand wird mit Wasser verrieben. Nach Ab giessen des Wassers erhält man so durch Trocknen des gewaschenen Produktes im Vakuum die N-(ss- Phenyl - äthyl) - 4 - phenyl - 4 - carbo(methoxy oder äthoxy)-piperidinbase. Die Umwandlung dieser Base in die entsprechenden Salze geht meistens in der Weise vor sich, dass man die Base unter praktisch wasserfreien Bedingungen mit einer Säure, z. B. Chlorwasserstoff, Bromwasserstoff, Schwefelsäure usw., umsetzt.
Diese salzbildende Reaktion wird am besten in einem niedermolekularen Alkanol, wie Äthanol, Methanol, Propanol usw., bewerkstelligt. Beim Verdünnen des alkanolischen Reaktionsme diums mit Äther fällt das Salz des N-(ss-Phenyl- äthyl)-4-phenyl-4-carbo(methoxy oder äthoxy)-piperi- dins aus, z.
B.: N-(ss-Phenyl-äthyl)-4-phenyl-4-carbomethoxy- piperidinhydrochlorid, N-(fl-Phenyl-äthyl)-4-phenyl-4-carbomethoxy- piperidinhydrobromid, N-(ss-Phenyl-äthyl)-4-phenyl-4-carbomethoxy- piperidinsulfat, N-(ss-Phenyl-äthyl)-4-phenyl-4-carboäthoxy- piperidinhydrochlorid, N-(ss-Phenyl-äthyl)-4-phenyl-4-carboäthoxy- piperidinhydrobromid, N-(ss-Phenyl-äthyl)
-4-phenyl-4-carboäthoxy- piperidinsulfat. Das so gebildete Salz wird aus der alkoholischen. Aufschlämmung zweckmässig durch Filtration oder Zentrifugieren gewonnen.
<I>Beispiel 1</I> Ein Gemisch aus 7 g 4-Phenyl-4-carboäthoxy- piperidincarbonat, 4,44 g ss-Phenyl-äthylbromid, 4,2g Natriumbicarbonat und 60 cm3 abs. Äthanol wird unter Rückfluss annähernd 48 Stunden erhitzt. Das Reaktionsgemisch wird zur Entfernung der an organischen Salze filtriert, und der Äthanol aus dem Filtrat im Vakuum verdampft. Der Rückstand wird mit Äther aufgeschlämmt und zur Entfernung wei terer anorganischer Substanz filtriert. Das unlösliche Produkt wird mit weiterem Äther gewaschen.
Die filtrierte ätherische Lösung nebst Waschäther liefert beim Eindampfen zur Trockne die N-(ss-Phenyl- äthyl)-4-phenyl-4-carboäthoxy-piperidinbase.
Beim Einleiten von Chlorwasserstoffgas in das Gemisch auf filtrierter ätherischer Lösung und Waschäther fällt sofort ein kristalliner Niederschlag aus; wenn dieser abfiltriert und durch Umkristalli- sieren aus Äthanol-Äther gereinigt wird, so erhält man annähernd 4 g praktisch reines ss-(Phenyl-äthyl)- 4-phenyl-4-carboäthoxy-piperidinhydrochlorid.
Schmelzpunkt: 190-192 C.

<I>Beispiel 2</I> Ein Gemisch aus 3,6 g 4-Phenyl-4-carbomethoxy- piperidincarbonat, 2,8 g ss-Phenyl-äthylbromid, 2,3 g Natriumbicarbonat und 35 cm?, abs. Äthanol wird unter Rückfluss und Rühren rund 40 Stunden erhitzt. Das Reaktionsgemisch wird zur Entfernung anorga nischer Salze filtriert, die dreimal mit je 15 cm3 abs. Äthanol ausgewaschen werden.
Das Gemisch aus äthanolischem Filtrat und Waschalkohol wird im Vakuum zu einem Kristallbrei eingedampft, der in einem Gemisch aus 15 cm3 Äther und 15 cm3 Wasser aufgelöst wird. Die Schichten werden ge trennt; die wässrige Schicht wird mit 15 cm3 Äther extrahiert. Die vereinigten Ätherextrakte werden über wasserfreiem Magnesiumsulfat getrocknet. Die getrocknete ätherische Lösung kann zur Trockne eingedampft werden und liefert N-(ss-Phenyl-äthyl)- 4-phenyl-4-carbomethoxy-piperidinbase.
Wenn man aber Chlorwasserstoffgas in die ver einigten Ätherextrakte einleitet, welche die N-(ss- Phenyl-äthyl)-4-phenyl-4-carbomethoxy-piperidinbase enthalten, so fällt sogleich N-(ss-Phenyl-äthyl)-4- phenyl-4-carbomethoxy-piperidinhydrochlorid aus; durch Filtration erhält man dabei etwa 3,8 g rohes Hydrochlorid und nach Umkristallisieren aus Äthanol-Äther etwa 3,3 g praktisch reines N-(ss Phenyl-äthyl)-4-phenyl-4-carbomethoxy-piperidin- hydrochlorid.
Schmelzpunkt 224-225 C. Analyse: berechnet für CUH2P2N # HCI: C = 70,07; H = 7,28; N = 3,87 gefunden: C = 70,05; H = 7,32; N = 3,90. Im Sinne obiger Arbeitsweise kann man anstelle des wasserfreien Chlorwasserstoffes andere wasser freie Säuren, wie z. B. Schwefelsäure, Phosphor säure, Zitronensäure, Oxalsäure oder Bromwasser stoff verwenden.

Although meperidine is widely used as an analgesic, its toxicity is disadvantageous in many cases.
Numerous N-substituted meperidine analogs have so far been made in the expectation of finding a compound which is comparable with meperidine in terms of analgesic activity and has a reduced toxicity; the connections found so far did not meet the expectations placed in them.
Experience so far did not suggest that the research would lead to the discovery of an improved analgesic of the meperidine type; because it was known that with increasing molecular weight, the toxicity of the N-substituent increases and the analgesic activity decreases (N-benzyl-4-phenyl-4-carboethoxypiperidine has only 50% of the analgesic activity of meperidine).

The dried ethereal solution can be evaporated to dryness and gives N- (ss-phenyl-ethyl) -4-phenyl-4-carbomethoxy-piperidine base.
If, however, hydrogen chloride gas is introduced into the combined ether extracts which contain the N- (ss-phenyl-ethyl) -4-phenyl-4-carbomethoxy-piperidine base, N- (s-phenyl-ethyl) -4-phenyl falls immediately -4-carbomethoxy-piperidine hydrochloride from; Filtration gives about 3.8 g of crude hydrochloride and, after recrystallization from ethanol-ether, about 3.3 g of practically pure N- (ss phenyl-ethyl) -4-phenyl-4-carbomethoxypiperidine hydrochloride.
