Es wurde gefunden, dass man tertiäre Amine der Formel

erhalten kann, wenn man tertiäre Aminoacetonitrile der Formel

mit Organomagnesiumhalogeniden der Formel Hal-Mg-Z-CsH4-R in Gegenwart von Lösungsmitteln umsetzt und die Reaktionsprodukte anschliessend hydrolytisch zersetzt.
In diesen Formeln bedeuten die Reste Ri und R2 gleiche oder verschiedene Alkyl-, Oxyalkyl-, Cycloalkyl-, Aryl-oder Aralkylgruppen, oder auch zusammen mit dem N einen gegebenenfalls ein weiteres Heteroatom enthaltenden heterocyclischen Ring, zum Beispiel den Pyrrolidin-, Piperidin-oder Morpholinring, R3 Wasserstoff oder einen Substituenten erster Ord'nung, beispielsweise eine Hydroxyl-, Alkoxy-, Alkyl-, Alkylamino-oder Dialkylaminogruppe, R4 Wasserstoff oder zusammen mit R3 eine Methylendioxygruppe, Z einen Alkylenrest mit 2 bis 4 Kohlenstoffatomen, der gerade oder verzweigt sein kann, und R5 Wasserstoff oder eine Alkyl-oder Alkoxy- gruppe mit 1 bis 3 Kohlenstoffatomen.
Die Umsetzung kann in der gleichen Weise und unter den gleichen Bedingungen wie in der Hauptpatentschrift angegeben durchgeführt werden.
Die Arylaminoacetonitrile der Formel II lassen sich, soweit sie nicht bekannt sind, nach bekannten Methoden (vgl. Houben-Weyl, Methoden der organischen Chemie, 4. Aufl., 8. Band, Seite 279 ff.) herstellen.
Die nach dem erfindungsgemässen Verfahren erhältlichen Verbindungen sind neue Substanzen, die stark wirksame Spasmolytika mit vorwiegend muskulotropem Angriffspunkt darstellen, deren Wirkung diejenige des Papaverins erreicht oder übertrifft.
Beispiel 1 1-Phenyl-l-di-n-propylamino-3-phenyl-pro pan

Zu der aus 4,6 g Magnesiumspänen und 37 g p-Phenyl-äthylbromid in abs. Ather bereiteten Grignardverbindung lässt man 20,2 g Phenyl-di-n-propyl- amino-acetonitril (Kpo4 = 105 bis 107"), in 50 cm3 abs. Ather gelöst, unter Rühren zutropfen und erwärmt nach beendigter Zugabe etwa 2 Stunden unter Rückfluss. Nach dieser Zeit wird das Reaktionsgemisch durch Zufügen von Eis und verdünnter Salzsäure zersetzt, die Atherschicht abgetrennt und verworfen. Die salzsaure wässrige Lösung versetzt man mit etwas Ammoniumchlorid, dann mit Ammoniak bis zur alkalischen Reaktion und nimmt das ab geschiedene Öl in Ather auf.
Nach dem Trocknen über Natriumsulfat wird der Äther verjagt und der Rückstand im Vakuum destilliert. Man erhält die Verbindung als farblose Flüssigkeit vom Kpo, l = 131 in einer Ausbeute von 71 I/o d. Th. Bei Verwendung von Benzol als Lösungsmittel erhält man dasselbe Ergebnis.
Beispiel 2 1-Phenyl-l-diäthylamino-3-phenyl-butan

Zu einer in üblicher Weise bereiteten Grignardverbindung aus 4,6 g Magnesiumspänen und 39,8 g P-Phenyl-propylbromid in 50 cm3 abs. Ather werden 14,1 g Phenyl-diäthylaminoacetonitril, in etwa 30 cm3 abs. Äther gelöst, zugetropft. Die Weiterverarbeitung erfolgt wie in Beispiel 1 angegeben. Man erhält eine farblose Flüssigkeit vom Kpo, 6 = 155 in einer Ausbeute von 81 % d. Th.
Beispiel 3 1-Phenyl-1-(methyl-[ss-oxy-äthyl]-amino)-4-phenyl-butan

Eine Grignardlösung aus 4, 6 g Magnesiumspänen und39, 8gy-Phenyl-propylbromid inAther setzt man, wie in Beispiel 1 beschrieben, mit 17,6 g Phenyl (methyl-[ss-oxy-äthyl]-amino)-acetonitril (Kp0.8 = 75 bis 77 ) um und erhält ein gelbliches, viskoses 01 vom KP0.25 = 170 bis 172 in einer Ausbeute von 78 le d. Th.
Beispiel 4 1- (4-Methoxy-phenyl)-l-dimethylamino-5-phenyl-pentan

Zu dem Grignardreagens aus 2,3 g Magnesiumspänen und 16,8 g b-Phenyl-butylchlorid gibt man tropfenweise eine Lösung von 6,4 g (4-Methoxy phenyl)-dimethylamino-acetonitril in 30 cm3 abs.
Äther, erhitzt 11/a Stunden unter Rückfluss zum Sieden und arbeitet wie in Beispiel 1 beschrieben auf.
Man erhält die oben angegebene Verbindung als gelbliches Öl vom Kapo, = 180 bis 182 in einer Ausbeute von 66 ouzo d. Th.
Besipel 5 1-(4-Methoxy-phenyl)-1-dimethylamino-3-(4-methoxy-phenyl)-propan

19 g 4-methoxy-phenyl-dimethylamino-acetonitril werden, wie in Beispiel 1 beschrieben, mit dem Gri gnardreagens aus 34 g ss-(p-Methoxy-phenyl)-äthyl- chlorid und 4, 6 g Magnesiumspänen in abs. Ather umgesetzt und aufgearbeitet. Man erhält die oben angegebene Verbindung als gelbliches 41 vom KPo*06 = 165 bis 166 in einer Ausbeute von 66,5 11/o d. Th.
Beispiel 6 1- (4-Methyl-phenyl)-1-piperidino- (1)-3-phenyl-propan

Aus 18,5 g lS-Phenyl-äthylbromid und 2,3 g Magnesiumspänen in abs. Ather wird in üblicher Weise eine Grignardverbindung bereitet, die mit 7,4 g 4-Methyl-phenyl-piperidino-(1)-acetonitril (Kp0.4 = 120 ) gemäss Beispiel 1 umgesetzt und aufgearbeitet wird. Man erhält das Produkt in einer Ausbeute von 63 I/o d. Th. als gelbliches O1 vom Kpo, 4 = 175 bis 176 , aus dem man mittels ätherischer Salzsäure das farblose Hydrochlorid vom F. = 201 erhält.
Beispiel 7 1- (4-Methyl-phenyl)-1-(methyl-[ss-oxy-äthyl]-amino-4-phenyl-butan

Eine Grignardverbindung aus 19,9 g y-Phenylpropylbromid und 2,3 g Magnesium setzt man, wie in Beispiel 1 angegeben, mit 6,8 g 4-Methyl-phenyl (methyl-[ss-oxy-äthyl]-amino)-acetonitril Kp0.15 = 120 ) um und arbeitet auf. Das tertiäre Amin wird als gelbliches Öl vom Kp0.15 = 205 in einer Ausbeute von 71"/ & d. Th. erhalten.
Beispiel 8 1-(3,4-Methylendioxy-phenyl)-1-piperidino-(1)-4-phenyl-butan

Zu einem aus 2,3 g Magnesiumspänen und 19,9 g j-Phenyl-propylbromid in 50 cm3 abs. Äther hergestellten Grignardreagens lässt man unter Rühren 8 g 3,4-Methylendioxy-phenyl-piperidino-(1)-acetonitril, in 40 cm3 abs. Ather gelöst, zutropfen, erhitzt 1'/2 Stunden zum Sieden und zersetzt dann das Reaktionsgemisch durch Zugabe von Eis und verdünnter Salzsäure. Nach dem Abtrennen der Atherschicht wird die wässrige Lösung mit etwas Ammoniumchlorid versetzt und mit Ammoniak alkalisch gemacht. Das abgeschiedene 01 wird in Ather aufgenommen und mit Natriumsulfat getrocknet. Nach dem Verjagen des Athers erhält man in einer Ausbeute von 63 Ïlo d.
Th. das tertiäre Amin, aus welchem man nach dem Lösen in Äther durch Zugabe von ätherischer Salzsäure das farblose, kristalline Hydrochlorid vom F. = 165 bis 167 erhält, (N ber. : 3,75, gef. 3, 78%.)
Beispiel 9 1- (4-Oxy-phenyl)-1-piperidino- (1)-4-phenyl-butan

Aus 2,3 gMagnesiumspänen und 19,9 g y-Phenylpropylbromid bereitetes Grignardreagens wird, wie zuvor beschrieben, mit 7,5 g 4-Oxy-phenyl-piperidino (l)-acetonitril umgesetzt und aufgearbeitet. Das nach dem Verjagen des Athers verbleibende, aus Methanol umkristallisierte tertiäre Amin schmilzt bei 138 bis 139 . Es wird in einer Ausbeute von 59 % d. Th. erhalten.
Beispiel 10 1-(4-Isopropyl-phenyl)-1-diäthylamino-4-phenyl-butan

Man lässt, wie in Beispiel 1 angegeben, 23 g 4-Isopropyl-phenyl-diäthylamino-acetonitril (po, 4 = 110 ) in Ather auf die Grignardverbindung aus 4,6 g Magnesium und 39,8 g y-Phenyl-propylbromid einwirken und arbeitet wie beschrieben auf. Man erhält das tertiäre Amin vom Kp0.5 = 169 in einer Ausbeute von 74 lo d. Th.
Beispiel 11 1- (4-Dimethylamino-phenyl)-1-diäthylamino-4-phenyl-butan

Zu einer Grignardverbindung aus 2,3 g Magnesium und 19,9 g y-Phenyl-propylbromid in 50 cm3 abs. Ather lässt man unter Rühren 7,7 g 4-Dimethyl- amino-phenyl-diäthylamino-acetonitril in 30 cm3 abs.
Ather zutropfen und erhitzt 1¸ Stunden unter Rückfluss. Anschliessend wird das Reaktionsgemisch durch Zugabe von Eis und verdünnter Salzsäure zersetzt, die Atherschicht abgetrennt und verworfen und die wässrige Lösung durch Zugabe von Ammoniak alkalisch gemacht. Das hierbei abgeschiedene Amin nimmt man in Äther auf und trocknet die Lösung über Natriumsulfat. Der nach dem Verjagen des Athers verbleibende Rückstand wird aus Petroläther umkristallisiert. Man erhält das tertiäre Amin in einer Ausbeute von 69 % d. Th., F. = 71 .
Beispiel12 1-Phenyl-l-diäthylamino-4-phenyl-butan

Zu einem aus 2,3 g Magnesium und 19,9 g y-Phenyl-propylbromid in üblicher Weise bereiteten und dann vom Ather befreiten Grignardreagens gibt man 30 cm3 Dibutyläther, lässt 14,1 g Phenyl-diäthyl- amino-acetonitril, in 20 cm3 Dibutyläther gelöst, unter Rühren zutropfen und erhitzt dann 1 Stunde auf 75 bis 100 . Nach dem Abkühlen wird, wie in Beispiel 1 angegeben, aufgearbeitet. Man erhält das tertiäre Amin vom Kpo, 35 = 141 bis 142 in einer Ausbeute von 67 lo.
Beispiel13 1- (4-Methoxy-phenyl)-1-morpholino- (4)-4-phenyl-butan

Wie in Beispiel 1 angegeben, setzt man 10,8 g 4-Methoxy-phenyl-morpholino- (4)-acetonitril (F. = 80 ) mit einer Grignardlösung aus 2,3 g Magnesium und 19,9 g y-Phenyl-propylbromid um. Das entstandene tertiäre Amin ist ein gelbes Ol vom Kpo, 03 = 179 . Ausbeute 68 O/D d. Th.
Beispiel 14 1- (4-Methoxy-phenyl)-l-dimethylamino-3- (4-methyl-phenyl)-propan

Die Grignardverbindung aus 4,6 g Magnesiumspänen und 30,8g p-(4-Methyl-phenyl)-äthylchlorid wird in der üblichen Weise mit 19 g 4-Methoxy phenyl-dimethylamino-acetonitril in Ather umgesetzt.
Man arbeitet den Reaktionsansatz nach Beispiel 1 auf und erhält das tertiäre Amin vom Kpo, 6 = 158 in einer Ausbeute von 64 % d. Th.

Process for the preparation of tertiary amines
It has been found that one can use tertiary amines of the formula

can be obtained if one tertiary aminoacetonitriles of the formula

with organomagnesium halides of the formula Hal-Mg-Z-CsH4-R in the presence of solvents and then hydrolytically decomposes the reaction products.
In these formulas, the radicals R 1 and R 2 denote identical or different alkyl, oxyalkyl, cycloalkyl, aryl or aralkyl groups or, together with the N, a heterocyclic ring which may contain a further hetero atom, for example the pyrrolidine, piperidine or Morpholine ring, R3 is hydrogen or a substituent of the first order, for example a hydroxyl, alkoxy, alkyl, alkylamino or dialkylamino group, R4 is hydrogen or, together with R3, a methylenedioxy group, Z is an alkylene radical with 2 to 4 carbon atoms which is straight or branched can be, and R5 is hydrogen or an alkyl or alkoxy group having 1 to 3 carbon atoms.
The reaction can be carried out in the same way and under the same conditions as indicated in the main patent specification.
The arylaminoacetonitriles of the formula II, if they are not known, can be prepared by known methods (cf. Houben-Weyl, Methods of Organic Chemistry, 4th edition, 8th volume, page 279 ff.).
The compounds obtainable by the process according to the invention are new substances which represent highly effective spasmolytics with a predominantly musculotropic point of attack, the effect of which reaches or exceeds that of papaverine.
Example 1 1-Phenyl-1-di-n-propylamino-3-phenyl-pro pan
