Procédé de préparation de N-amino-alcoyl-phénothiazines
On connaît l'importance acquise en thérapeutique par la chlorpromazine ou chloro-3-N-y-diméthyl- amino-propyl-phénothiazine qui est employée pour potentialiser la narcose, l'analgésie et l'anesthésie locale ainsi qu'en raison de ses pouvoirs hypothermi- sant, antiémétique, adrénolytique et antichoc.
Ce composé fait partie d'une série de N-diméthyl- aminoalcoyl-phénothiazines, y compris celles qui portent, dans la position 3, un substituant insensible à l'action des agents alcalins, tels que l'amidure de sodium, le sodium l'hydrure de sodium et la soude caustique anhydre, utilisés selon les procédés classiques pour la fixation d'un groupe diméthylamino- alcoylique sur l'atome d'azote de la phénothiazine.
Ce mode de greffage de chaîne en milieu alcalin est, en revanche, incompatible avec la présence de substitutants tels que OH, CHO, CO-CH3, CO-C2H5, etc.
La présente invention a pour objet un procédé qui permet de préparer de tels dérivés portant un substituant sensible aux agents alcalins et qui, de plus se prête également à la préparation des dérivés de phénothiazine mentionnés en premier lieu ; ce procédé est donc d'une application très générale et, par conséquent, d'un intérêt évident, notamment par le fait qu'il ouvre un nouveau champ d'investigation et, en même temps, fournit une nouvelle voie d'accès aux composés du type de la chlorpromazine ou du type de la prométhazine (N-p-diméthylamino-propyl- phénothiazine).
Le procédé selon l'invention est caractérisé en ce qu'on estérifie un acide phénothiazine-N-carboxylique en 1'ester amino-alcoylique correspondant, puis on décarboxyle cet ester pour provoquer l'union directe de l'atome d'azote de la phénothiazine au radical amino-alcoylique.
Selon le mode d'exécution adopté de préférence, on effectue la décarboxylation par un simple traitement thermique, le cas échéant en présence d'un catalyseur ; on peut en particulier distiller 1'ester sous vide ; on peut aussi opérer par chauffage au sein d'un solvant approprié tel que le décahydro-naphta- lène.
Parmi les nouvelles N-amino-alcoyl-phénothiazines qui permettent d'obtenir le présent procédé, l'acétyl 3-N-y-diméthylamino-propyl-phénothiazine tient une place exceptionnelle en ce sens que, par ses propriétés de potentialisation de la narcose et de l'anesthésie locale, par son pouvoir antiémétique, par son pouvoir hypothermisant et par son pouvoir dépressif sur les centres nerveux, elle dépasse la chlorpromazine et qu'en outre, elle manifeste, employée seule, une action hypnotique que ne possède pas la chlorpromazine.
On constate, en outre, que les animaux d'expérience ayant reçu cette phénothiazine reviennent à leur état normal plus vite que dans le cas de l'admi- nistration de chlorpromazine, de sorte que tout laisse à penser que son élimination est plus rapide que celle de la chlorpromazine.
La titulaire a trouvé, en outre, que le sel de ce dérivé de phénothiazine qui offre le plus d'intérêt pour l'administration thérapeutique était le maléate acide de l'acétyl-3-N-y-diméthylamino-propyl-phénothiazine.
Les essais pharmacodynamiques effectués avec l'oxalate et avec le maléate acide de l'acétyl-3-N-y- diméthylamino-propyl-phénothiazine ont conduit aux résultats suivants :
Pour la souris, la toxicité aiguë de l'oxalate, administré par voie intraveineuse, est analogue à celle de la chlorpromazine et celle du maléate est moindre ; la toxicité chronique (administration sous-cutanée) de l'oxalate et celle du maléate sont toutes deux moindres que celle de la chlorpromazine.
L'action dépressive, I'action de potentialisation de la narcose, I'action hypothermisante, l'action anesthésique locale et l'action antiémétique sont toutes au moins égales et, pour la plupart, supérieures à celles de la chlorpromazine.
L'expérience clinique effectuée jusqu'ici en chirurgie (administration préopératoire ou post-opéra- toire), en neuropsychiatrie et en dermatologie (action antiprurigineuse) a confirmé, dans l'ensemble, la su périorité du maléate sur la chlorpromazine ; on a noté en particulier une bien meilleure tolérance et l'absence complète des troubles (palpitations, sécheresse de la bouche) fréquents avec les autres neuroplégiques connus.
Exemple 1 :
a) Préparation préalable du chlorure
de l'acide acétyl-3-phénothiazine-N-carbo-
xylique.
On utilise comme matière première I'acétyl-3- phénothiazine ou éthylone-3-phénothiazine (point de fusion 188-1 890).
Dans un autoclave, on introduit 70 g d'acétyl-3- phénothiazine et 245 cm3 d'une solution à 20 tao de phosgène dans du toluène et on le chauffe au four électrique à 110-115 , avec agitation, pendant une heure et demie. On sort l'autoclave du four et on le refroidit rapidement. Après détente de la pression in térieure, on filtre la solution toluénique et on lave l'autoclave au toluène. On concentre jusqu'à siccité les solutions toluéniques réunies et l'on extrait trois fois le résidu gommeux et noir avec 250 cm3 d'éther à reflux. Après concentration des solutions éthérées jusqu'à très petit volume, une cristallisation se produit. On essore les cristaux et on les sèche.
On recueille 70 g (rendement 80 /0) d'une poudre beige que l'on peut utiliser directement pour la transformation en ester.
Recristallisé dans de l'acétate d'éthyle, le chlorure d'acide fond à 116-117 .
b) Préparation du chlorhydrate de 1'ester
y-diméthylamino-propylique de l'acide acétyl- 3-phénothiazine-N-carboxylique.
Dans un ballon muni d'une garde à chlorure de calcium, on place 30 g de chlorure d'acide acétyl-3- phénothiazine-N-carboxylique préparé comme indique ci-dessus et 11 g (5 /o d'excès) de y-diméthyl- amino-propanol dans 100 cm3 d'acétone anhydre (au lieu d'acétone, on peut utiliser un autre solvant organique anhydre tel que le benzène). On maintient le tout au reflux pendant une nuit. Il se forme un pré cipité beige clair de chlorhydrate d'ester y-diméthyl- amino-propylique de l'acide acétyl-3-phénothiazine-
N-carboxylique. On l'essore, on le lave à l'éther et on le sèche. On en recueille 30 g.
Recristallisé dans l'éthanol absolu, ce chlorhydrate fond à 213-215"avec décomposition.
c) Préparation de l'acétyl-3-N-y-di- méthylamino-propyl-phénothiazine.
On dissout dans l'eau 30 g du chlorhydrate d'es
ter basique préparé selon b) et on le décompose avec
de la soude caustique 10 N. On extrait la base (ester y-diméthylamino-propylique de l'acide acétyl-3- phénothiazine-N-carboxylique) par de l'éther, on lave la solution éthérée avec de l'eau et on la sèche sur du sulfate de sodium.
Après élimination de l'éther au bain-marie, on recueille 21 g d'une huile rouge que l'on dissout dans 42cm3 de décahydro-naphtalène (aulieudedéca hydro-naphtalène, on peut aussi utiliser, en même quantité, d'autres solvants tels que le diméthoxy-1, 3benzène, la quinoléine ou la collidine) ; on porte la solution au reflux jusqu'à cessation du dégagement de CO2. Après refroidissement, on reprend par de l'éther et on extrait à l'eau chlorhydrique. On décompose la solution de chlorhydrate par de la soude caustique 10 N et on extrait la base à l'éther. On lave la solution éthérée à l'eau et on la sèche sur du sulfate de sodium.
Après élimination du solvant, on obtient 16 g d'une huile jaune orangé. Rapporté au chlorure d'acide, le rendement est de 50 fl/o et, rapporté au chlorhydrate d'ester basique, il est de 78 /o.
On a préparé les sels suivants de I'acétyl-3-N-y- diméthylamino-propyl-phénothiazine :
Oxalate acide :
On dissout 8 g de cette phénothiazine dans 80 cm3 d'alcool absolu et on ajoute à la solution une solution de 3, 1 g d'acide oxalique dans 31 cm3 d'alcool absolu. On chauffe en agitant, pour bien mélanger et on laisse cristalliser une nuit. On essore le produit cristallisé, de couleur jaune pale et on le recristallise dans l'alcool méthylique. On obtient ainsi 9, 2 g (rendement 90 /o) de cristaux d'oxalate de couleur jaune pâle : leur point de fusion est de 187-189ç} (avec décomposition).

Maléate acide :
On dissout 10 g de cette phénothiazine dans 100 cm3 d'acétate d'éthyle et on ajoute une solution de 3, 9 g (10 /o d'excès) d'acide maléique dans 40 cm3 d'acétate d'éthyle. On porte à ébullition et on laisse cristalliser lentement. On essore le produit cristallisé et on le lave avec le minimum d'acétate d'éthyle.
On obtient ainsi 12, 2 g (rendement 89"/a) d'écaillés jaunes, fondant à 136-137 .

Iodométhylate :
Dans 15 cm3 d'alcool absolu, on dissout 5 g de cette phénothiazine puis on ajoute à la solution 2 cm3 d'iodure de méthyle dissous dans 8 cm8 d'alcool absolu et on laisse cristalliser. On essore le précipité jaune, on le lave à l'alcool et à l'éther et on le recristallise dans de l'alcool absolu. On obtient ainsi 6, 5 g de petits cristaux jaune pâle d'iodométhylate d'acÚtyl-3-N-?-dimÚthylamino-propyl-phÚnothiazine fondant à 99-101 .

On a préparé également le chlorhydrate de la dinitro-2, 4-phénylhydrazone de la phénothiazine en cause et cette phénylhydrazone elle-même ; le chlorhydrate se présente en petits cristaux rouge foncé, fondant au-dessus de 265¯ et la phénylhydrazone en petits cristaux grenat foncé fondant à 164-168 .
Fxemple 2 :
En opérant selon l'exemple 1 mais en partant de méthoxy-3-phénothiazine, on prépare préalablement le chlorure de l'acide méthoxy-3-phénothiazine-N- carboxylique (prismes incolores fondant a 96-97 après recristallisation dans du cyclohexane) et, à partir de ce dernier on obtient successivement :
a) le chlorhydrate de l'ester-diméthylamino-
propylique de l'acide méthoxy-3-phénothia-
zine ;
b) cet ester lui-même (huile brun-rose) ;
c) la méthoxy-3-N-y-diméthylamino-propyl-phé-
nothiazine (par simple distillation sous vide
de 1'ester (b), huile rosée qui distille à 225

d) l'oxalate acide de la phénothiazine c) qui,
après recristallisation dans l'alcool, se pré
sente en paillettes incolores fondant à 180

Exemple 3 :
En opérant comme dans l'exemple 2 mais en remplaçant le ?-dimÚthylamino-propanol par du (3-di- méthylamino-éthanol et le chlorure d'acide méthoxy- 3-phénothiazine-N-carboxylique par celui de l'acide acétyl-3-phénothiazine-N-carboxylique, on obtient successivement le chlorhydrate de l'ester p-diméthyl- amino-éthylique de l'acide acétyl-3-phénothiazine-N- carboxylique qui, après purification, se présente en cristaux blancs fondant vers 2450 (décomposition), 1'ester basique lui-même (huile brun-rougeâtre), puis l'acétyl-3-N-p-diméthylamino-éthyl-phénothiazine sous la forme d'une huile jaune orange ;
son oxalate acide se présente en prismes jaune d'or qui fondent à 196-1980 (décomposition).
Exemple 4 :
En opérant selon les exemples précédents et en partant de chlorure d'acide acétyl-3-phénothiazine-N- carboxylique, on prépare successivement le chlorhydrate de 1'ester y-diéthylamino-propylique de l'acide acétyl-3-phénothiazine-N-carboxylique, cet ester lui-même (poudre beige fondant à 178-179o avec décomposition après recristallisation dans l'isopro- panol), l'acétyl-3-N-y-diéthylamino-propyl-phénothia- zine (huile rouge orangé bouillant à 225-2350 sous 0, 3 mm) et l'oxalate acide de cette base, qui fond à 146-148o et se présente en prismes jaunes.
Exemple 5 :
On part de chlorure de l'acide propionyl-3-phéno- thiazine-N-carboxylique, préparé suivant les données de l'exemple 1 et se présentant, après recristallisation dans l'acétate d'éthyle, en prismes incolores qui fondent à 156-157 .
En traitant ce chlorure par du y-diméthylamino propanol en excès on obtient, en cristaux jaunes, le chlorhydrate de 1'ester y-diéthylamino-propylique de l'acide propionyl-3-phénothiazine-N-carboxylique qui fond à 189-190 (décomposition). Après isolement de cet ester à partir de son chlorhydrate et décarboxylation de celui-ci à 200 sous vide, on obtient la pro pionyl-3-N-y-diéthylamino-propyl-phénothiazine dont l'oxalate (cristaux jaune clair) fond à 147-149 (décomposition).
Exemple 6:
En partant du chlorure de l'acide propionyl-3phénothiazine-N-carboxylique, et en procédant comme il est décrit dans les exemples précédents, on prépare successivement :
a) le chlorhydrate de l'ester y-diméthylamino-
propylique de l'acide propionyl-3-phénothia-
zine-N-carboxylique, en cristaux beiges fon
dant à 207-2090 avec décomposition, après
recristalisation dans l'éthanol absolu ;

Process for preparing N-amino-alkyl-phenothiazines
We know the importance acquired in therapy by chlorpromazine or chloro-3-Ny-dimethyl-amino-propyl-phenothiazine which is used to potentiate narcosis, analgesia and local anesthesia as well as because of its hypothermic powers. - health, antiemetic, adrenolytic and shockproof.
This compound is one of a series of N-dimethyl-aminoalkyl-phenothiazines, including those which carry, in the 3-position, a substituent insensitive to the action of alkaline agents, such as sodium amide, sodium l sodium hydride and anhydrous caustic soda, used according to conventional methods for the attachment of a dimethylaminoalkyl group to the nitrogen atom of the phenothiazine.
This mode of chain grafting in an alkaline medium is, on the other hand, incompatible with the presence of substitutes such as OH, CHO, CO-CH3, CO-C2H5, etc.
The present invention relates to a process which makes it possible to prepare such derivatives bearing a substituent sensitive to alkaline agents and which, moreover, also lends itself to the preparation of the phenothiazine derivatives mentioned in the first place; this method is therefore of very general application and, consequently, of obvious interest, in particular by the fact that it opens a new field of investigation and, at the same time, provides a new route of access to the compounds of the chlorpromazine type or of the promethazine type (Np-dimethylamino-propyl-phenothiazine).
The process according to the invention is characterized in that a phenothiazine-N-carboxylic acid is esterified to the corresponding amino-alkyl ester, then this ester is decarboxylated to cause the direct union of the nitrogen atom of the phenothiazine to the amino-alkyl radical.
According to the embodiment preferably adopted, the decarboxylation is carried out by a simple heat treatment, where appropriate in the presence of a catalyst; the ester can in particular be distilled under vacuum; it is also possible to operate by heating in a suitable solvent such as decahydro-naphthalene.
Among the new N-amino-alkyl-phenothiazines which make it possible to obtain the present process, acetyl 3-Ny-dimethylamino-propyl-phenothiazine holds an exceptional place in that, by its properties of potentiation of narcosis and of local anesthesia, by its antiemetic power, by its hypothermic power and by its depressive power on the nervous centers, it exceeds chlorpromazine and that, moreover, it manifests, employed alone, a hypnotic action which chlorpromazine does not possess.
It is further observed that the experimental animals which received this phenothiazine return to their normal state more quickly than in the case of the administration of chlorpromazine, so that everything suggests that its elimination is faster than. that of chlorpromazine.
The patentee has further found that the salt of this phenothiazine derivative which is of most interest for therapeutic administration is acetyl-3-N-y-dimethylamino-propyl-phenothiazine acid maleate.
The pharmacodynamic tests carried out with the oxalate and with the acid maleate of acetyl-3-N-y-dimethylamino-propyl-phenothiazine led to the following results:
In mice, the acute toxicity of oxalate, administered intravenously, is similar to that of chlorpromazine and that of maleate is less; the chronic toxicity (subcutaneous administration) of oxalate and that of maleate are both less than that of chlorpromazine.
The depressive action, the potentiation action of narcosis, the hypothermic action, the local anesthetic action and the antiemetic action are all at least equal to and, for the most part, superior to those of chlorpromazine.
Clinical experience carried out so far in surgery (preoperative or postoperative administration), neuropsychiatry and dermatology (antipruritic action) has generally confirmed the superiority of maleate over chlorpromazine; in particular, a much better tolerance and the complete absence of the disorders (palpitations, dry mouth) frequent with other known neuroplegics were noted.
Example 1:
a) Prior preparation of the chloride
acetyl-3-phenothiazine-N-carbo- acid
xylic.
Acetyl-3-phenothiazine or ethylone-3-phenothiazine (melting point 188-1 890) is used as starting material.
70 g of acetyl-3-phenothiazine and 245 cm3 of a 20 tao solution of phosgene in toluene are introduced into an autoclave and heated in an electric oven at 110-115, with stirring, for an hour and a half. . The autoclave is removed from the oven and cooled rapidly. After relaxation of the internal pressure, the toluene solution is filtered and the autoclave is washed with toluene. The combined toluene solutions are concentrated to dryness and the gummy, black residue is extracted three times with 250 cm3 of ether at reflux. After concentration of the ethereal solutions to a very small volume, crystallization occurs. The crystals are filtered off and dried.
70 g (yield 80/0) of a beige powder are collected which can be used directly for the conversion into an ester.
Recrystallized from ethyl acetate, the acid chloride melts at 116-117.
b) Preparation of the ester hydrochloride
Acetyl-3-phenothiazine-N-carboxylic acid y-dimethylamino-propylic acid.
In a flask fitted with a calcium chloride guard, 30 g of acetyl-3-phenothiazine-N-carboxylic acid chloride prepared as indicated above and 11 g (5 / o excess) of y are placed. -dimethylamino-propanol in 100 cm3 of anhydrous acetone (instead of acetone, another anhydrous organic solvent such as benzene can be used). The whole is maintained at reflux overnight. A light beige precipitate of acetyl-3-phenothiazine acid y-dimethyl-amino-propyl ester hydrochloride formed.
N-carboxylic. It is filtered off, washed with ether and dried. 30 g are collected.
Recrystallized from absolute ethanol, this hydrochloride melts at 213-215 "with decomposition.
c) Preparation of acetyl-3-N-y-dimethylamino-propyl-phenothiazine.
30 g of es hydrochloride are dissolved in water.

10 N caustic soda. The base (acetyl-3-phenothiazine-N-carboxylic acid y-dimethylamino-propyl ester) is extracted with ether, the ethereal solution is washed with water and dried over sodium sulfate.
After elimination of the ether in a water bath, 21 g of a red oil are collected which are dissolved in 42 cm3 of decahydro-naphthalene (aulieudedéca hydro-naphthalene, one can also use, in the same quantity, other solvents such as 1, 3-dimethoxybenzene, quinoline or collidine); the solution is brought to reflux until the evolution of CO 2 ceases. After cooling, the residue is taken up in ether and extracted with hydrochloric water. The hydrochloride solution is decomposed with 10 N caustic soda and the base is extracted with ether. The ethereal solution is washed with water and dried over sodium sulfate.
After removal of the solvent, 16 g of an orange-yellow oil are obtained. Based on the acid chloride, the yield is 50 fl / o and, based on the basic ester hydrochloride, it is 78 / o.
The following salts of acetyl-3-N-y-dimethylamino-propyl-phenothiazine were prepared:
Acid oxalate:
8 g of this phenothiazine are dissolved in 80 cm3 of absolute alcohol and a solution of 3.1 g of oxalic acid in 31 cm3 of absolute alcohol is added to the solution. The mixture is heated while stirring, to mix well and left to crystallize overnight. The crystalline product, which is pale yellow in color, is filtered off and recrystallized from methyl alcohol. 9.2 g (yield 90 / o) of pale yellow oxalate crystals are thus obtained: their melting point is 187-189% (with decomposition).

Acid maleate:
10 g of this phenothiazine are dissolved in 100 cm3 of ethyl acetate and a solution of 3.9 g (10 / o excess) of maleic acid in 40 cm3 of ethyl acetate is added. It is brought to a boil and left to crystallize slowly. The crystallized product is filtered off and washed with the minimum amount of ethyl acetate.
There is thus obtained 12.2 g (yield 89 "/ a) of yellow scales, melting at 136-137.

Iodomethylate:
In 15 cm3 of absolute alcohol, 5 g of this phenothiazine are dissolved and then 2 cm3 of methyl iodide dissolved in 8 cm8 of absolute alcohol are added to the solution and the mixture is left to crystallize. The yellow precipitate is filtered off, washed with alcohol and with ether and recrystallized from absolute alcohol. 6.5 g of small pale yellow crystals of acetyl-3-N -? - dim ?thylamino-propyl-phÚnothiazine iodomethylate are thus obtained, melting at 99-101.

Also prepared were the hydrochloride of dinitro-2, 4-phenylhydrazone of the phenothiazine in question and this phenylhydrazone itself; the hydrochloride occurs as small dark red crystals, melting above 265¯ and the phenylhydrazone as small dark garnet crystals, melting at 164-168.
Example 2:
By operating according to Example 1 but starting from methoxy-3-phenothiazine, the chloride of methoxy-3-phenothiazine-N-carboxylic acid is prepared beforehand (colorless prisms melting at 96-97 after recrystallization from cyclohexane) and , from the latter we obtain successively:
a) ester-dimethylamino- hydrochloride
methoxy-3-phenothia- propylic acid

b) this ester itself (brown-pink oil);
c) methoxy-3-N-y-dimethylamino-propyl-phe-
nothiazine (by simple vacuum distillation

Example 3:
By operating as in Example 2 but replacing? -DimÚthylamino-propanol with (3-dimethylamino-ethanol and methoxy-3-phenothiazine-N-carboxylic acid chloride by that of acetyl- acid. 3-phenothiazine-N-carboxylic acid, the hydrochloride of the p-dimethyl-amino-ethyl ester of acetyl-3-phenothiazine-N-carboxylic acid is obtained successively which, after purification, appears in white crystals melting around 2450 (decomposition), the basic ester itself (reddish-brown oil), then acetyl-3-Np-dimethylamino-ethyl-phenothiazine as an orange-yellow oil;
its acid oxalate appears in golden yellow prisms which melt at 196-1980 (decomposition).
Example 4:
By operating according to the preceding examples and starting from acetyl-3-phenothiazine-N-carboxylic acid chloride, the hydrochloride of the y-diethylamino-propyl ester of acetyl-3-phenothiazine-N- acid is successively prepared. carboxylic acid, this ester itself (beige powder melting at 178-179o with decomposition after recrystallization in isopropanol), acetyl-3-Ny-diethylamino-propyl-phenothiazine (orange-red oil boiling at 225- 2350 under 0, 3 mm) and the acid oxalate of this base, which melts at 146-148o and appears in yellow prisms.
Example 5:
The starting point is propionyl-3-phenothiazine-N-carboxylic acid chloride, prepared according to the data of Example 1 and appearing, after recrystallization from ethyl acetate, in colorless prisms which melt at 156 -157.
By treating this chloride with excess γ-dimethylamino propanol, the hydrochloride of the y-diethylamino-propyl ester of propionyl-3-phenothiazine-N-carboxylic acid is obtained in yellow crystals, which melts at 189-190 ( decomposition). After isolation of this ester from its hydrochloride and decarboxylation thereof at 200 under vacuum, pro pionyl-3-Ny-diethylamino-propyl-phenothiazine is obtained, the oxalate (light yellow crystals) of which melts at 147-149. (decomposition).
Example 6:
Starting from the chloride of propionyl-3phenothiazine-N-carboxylic acid, and proceeding as described in the preceding examples, the following are successively prepared:
a) γ-dimethylamino ester hydrochloride
propylic acid propionyl-3-phenothia-
zine-N-carboxylic, in dark beige crystals
from 207-2090 with decomposition, after
recrystallization from absolute ethanol;
b) the free base (brown-red oil);
c) by retort decarboxylation under vacuum at

d) the acid oxalate of phenothiazine c) under
yellow crystalline powder form melting at
147-149 with decomposition, after recruits
tallization in methanol.
