Verfahren zur Herstellung von Chinolizinabkömmlingen Die vorliegende Erfindung betrifft ein Verfahren zur Herstellung von Substituenten aufweisenden Chinolizinen und deren Sälzen, welches dadurch ge kennzeichnet ist, dass man ein im isocyclischen Ring durch Alkoxygruppen, welche unter sich verbunden sein können, substituiertes 2-(Cyan-carbalkoxy-methyl- iden)-3-alkyl-1,2,3,4,6,7-hexahydro-benzo [a]chinolizin hydriert, verseift und decarboxyliert,
die gebildete Carbonsäure mit einem durch Alkoxygruppen, welche unter sich verbunden sein können, kernsubstituierten ss-Phenyl-äthylamin zum entsprechenden Säureamid kondensiert, letzteres durch Einwirkung von wasser abspaltenden Mitteln cyclisiert und anschliessend hydriert, wobei ein in den isocyclischen Ringen Alkoxygruppen, die unter sich verbunden sein können, aufweisendes 2-[(1',2',3',4'-Tetrahydro-isochinolyl-1')- methyl]-3-alkyl-1,2,3,4,6,
7-hexahydro-benzo [a]chinoli- zin gebildet wird.

R1, R2, R3 und R4 bedeuten Alkylgruppen, wie z. B. Methyl, Äthyl, Propyl, Butyl, Isobutyl usw., und " eine ganze Zahl von 1-4.
Zwei benachbarte Gruppen R10 bzw. R40 können miteinander verbunden sein, z. B. zu einer Methylen- dioxybrücke.
Die Ausgangsverbindungen, die zum Teil bekannt sind, lassen sich durch Kondensation der entsprechend substituierten 2-Oxo- 3-alkyl-1, 2, 3, 4, 6, 7-hexahydro- benzo [a]chinolizine mit Cyanessigsäureester gewin nen. Ihrerseits kann man die substituierten 2-Oxo-3- alkyl-1,2,3,4,6,7-hexahydro-benzo[a]chinolizine z. B.
durch Mannich-Kondensation von im aromatischen Ring entsprechend substituierten 1-Carbalkoxymethyl- 1,2,3,4-tetrahydroisochinolinen mit monosubstituier ten Malonsäuredimethylestern und Formaldehyd, nachfolgender Dieckmann-Kondensation, Verseifung und Decarboxylierung der gebildeten tricyclischen Ketodicarbonsäureester gewinnen.
Die Hydrierung der 2-(Cyan-carbalkoxy-methyl- iden)-1,2,3,4,6,7-hexahydro-benzochinolizine I erfolgt zweckmässig katalytisch in Gegenwart von Edelmetall- katalysatoren, wie Platin, Palladium usw. Eine ge eignete Methode besteht z. B. darin, dass man das Methyliden-benzochinolizin in Form eines mineral sauren Salzes, z. B. des Hydrochlorides, mit Wasser stoff und Platinschwarz in alkoholischer Lösung schüttelt. Nach Aufnahme der berechneten Menge Wasserstoff kann man die gebildeten gesättigten Cyan- essigester z.
B. durch Einengen, Zugeben von verdünn ter Mineralsäure, zweckmässig Salzsäure, bis zur kongo sauren Reaktion und Kristallisierenlassen in Form ihrer entsprechenden mineralsauren Salze gewinnen. Sie sind sterisch einheitlich. Im Ultraviolett-Absorp- tionsspektrum (Feinsprit) weisen diese Verbindungen eine kurzwellige Bande bei 230-233 mu von bedeutend niedrigerer Extinktion als die entsprechende Bande des Ausgangsmaterials auf.
Die Verseifung und Decarboxylierung der gesättig ten Cyanessigester 1I erfolgt vorteilhaft durch Erwär men auf über etwa 80 mit starken, wässerigen Mineral- säuren, z. B. 15-30% Salzsäure oder nicht zu stark ver dünnter Schwefelsäure. Aus der mineralsauren Lösung kann man anschliessend nach Einengen und Lösen in Wasser die mineralsauren Salze der gebildeten Carbon- säuren III direkt kristallin erhalten.
Die Bildung der Säureamide IV aus den gewonne nen mineralsauren Salzen von<B>111</B> und dem kern substituierten ss-Phenyl-äthylamin kann nach an sich bekannten Methoden erfolgen. Zum Beispiel kann man die Carbonsäure-hydrochloride in einem inerten Lö sungsmittel, wie Chloroform, mit Thionylchlorid in die entsprechenden Säurechlorid-hydrochloride überfüh ren und letztere auf das Amin einwirken lassen.
Nach einer andern Ausführungsform kann man die mineral sauren Salze der Carbonsäuren und das Amin in einem mit Wasser nicht mischbaren Lösungsmittel, z. B. Benzol, Toluol oder Xylol, in Gegenwart eines Konden sationsmittels, wie Eisessig-Ammonacetat, erwärmen und das sich bildende Wasser kontinuierlich entfernen.
Zwecks Cyclisierung der gewonnenen Säureamide IV lässt man nun auf dieselben wasserabspaltende Mit tel einwirken. Eine geeignete Ausführungsform besteht z. B. darin, dass man die Säureamide mit einem Phos- phoroxyhalogenid, z. B. Phosphoroxychlorid, in einem inerten Lösungsmittel, z. B. Toluol oder Benzol, er wärmt.
Die gebildeten Cyclisierungsprodukte V kri stallisieren bei Anwendung. von Phosphoroxychlorid als wasserabspaltendes Mittel in der Regel aus der Reaktionslösung in Form des wasserhaltigen Dihydro- chlorids aus. Im Gegensatz zu den Säureamiden IV weisen sie im Ultraviolett-Absorptionsspektlum cha rakteristische Absorptionsmaxima bei 240-245 mu, 290 mu, 302-304 mu, 354-362 mss (in n/ 100 Salzsäure) auf.
In der letzten Reaktionsstufe werden die Cycli- sierungsprodukte V einer Hydrierung unterworfen. Dies kann z. B. katalytisch oder mittels chemischen Reduktionsmitteln, wie z. B. Lithiumborhydrid, Li- thiumaluminiumhydrid, Natriumborhydtid, erfolgen. Man gewinnt so Gemische von zwei stereoisomeren Verbindungen. Sie können durch fraktionierte Kri- stallisation in die einzelnen Komponenten aufgetrennt werden.
Die nach dem erfindungsgemässen Verfahren er haltenen, Substituenten aufweisenden Chinolizine VI stellen Basen dar, welche mit Mineralsäuren gut kri stallisierende, wasserlösliche Salze geben. Sie weisen im Ultraviolett-Absorptionsspektrum charakteristische Maxima: bei 230 und 282 mu (in n/100 salzsaurer Lösung) auf.
Sie haben ausgeprägte expektorations- fördernde Eigenschaften und zeichnen sich ferner durch ihre amoebicide Wirkung, insbesondere gegen Entamoeba histolytica, aus. Sie sollen deshalb als Heilmittel Verwendung finden.
<I>Beispiel 1</I> Metlayliden-bettzoclzinolizin <I>(Herstellung der Ausgangs-</I> <I>verbindung)</I> 1 Gewichtsteil 2-Oxo-3-äthyl-9,10-dimethoxy-1,2,3, 4,6,7-hexahydro-benzo [a]chinolizin, 0,5 Gewichtsteile Cyanessigsäureäthylester, 0,1 Gewichtsteil Ammon- acetat und 0,1 Raumteil Eisessig werden in 3 Raum teilen Toluol 4 Stunden unter Rückfluss gekocht und das gebildete Wasser azeotrop entfernt.
Nach dem Er kalten wird mit 3n-Salzsäure extrahiert, die salzsauren Auszüge werden mit Äther gewaschen und die nach dem Versetzen mit Natronlauge freigesetzten basischen Anteile in Äther aufgenommen. Nach dem Einengen, Lösen in alkoholischer Salzsäure und Versetzen mit Äther kristallisiert das 2-(Cyan-carbäthoxy-methyl- iden)- 3-äthyl-9,10-dimethoxy-1, 2, 3, 4, 6, 7-hexahydro- benzo [a]chinolizin-hydrochlorid vom Schmelzpunkt 160-162 . Hydrochlorid: Schmelzpunkt 160-162 .
Hydrobromid: Schmelzpunkt 182-184 . Freie Base aus Hydrochlorid bereitet: Schmelzpunkt 130-132 . U.V.- Absorptionsspektrum des Hydrochlorids in Sprit 2in2,X 233 mau s = 24300, Amax 283 mA s = 5450.
<I>Gesättigter</I> Cyanessigsäureester 1 Gewichtsteil 2-(Cyan-carbäthoxy-methyliden)-3- äthyl-9,10-dimethoxy-1,2,3,4, 6,7-hexahydro-benzo [a] chinolizin-hydrocWorid wird in 20 Raumteilen Fein sprit gelöst und in Gegenwart von 0,1 Gewichtsteil vorhydriertem Platinoxydkatalysator hydriert, bis die theoretische Menge Wasserstoff aufgenommen ist.
Nach dem Einengen wird mit Wasser versetzt, Salz säure bis zur kongosauren Reaktion zugegeben, und nach einigem Stehen filtriert. Das 2-(Cyan-carbäthoxy methyl)-3-äthyl-9,10-dimethoxy-1,2,3,4,6,7-hexahydro- benzo [a]chinolizin-hydrochlorid kristallisiert in farb losen Blättchen, die nach dem Umlösen aus 70% Sprit- Äther bei 213-214 schmelzen. U.V.-Absorptions- spektrum in Sprit AmaX 230 mu E = 7900,
AmaX 284m/,t s = 3700.
Carbonsäure 1 Gewichtstei12-(Cyan-carbäthoxy-methyl)-3-äthyl- 9,10-dimethoxy-1,2,3,4,6,7-hexahydro-benzo [a]chino- lizin-hydrochlorid wird in 50 Raumteilen 20% Salz säure suspendiert und 2 Stunden am Rückfluss gekocht. Nach dem Einengen zur Trockne wird der Rückstand in wenig Wasser gelöst und die abgeschiedenen Kri stalle filtriert.
Das bei 50 luftgetrocknete 2-Carboxy methyl-3-äthyl-9,10-dimethoxy-1,2,3,4,6,7-hexahydro- benzo [a]chinolizin-hydrochlorid schmilzt unscharf bei 100 . Methylester-Hydrochlorid : Schmelzpunkt 215 bis 216 .
Säureamid 1 Gewichtsteil 2-Carboxymethyl-3-äthyl-9,10-di- methoxy-1, 2, 3, 4, 6, 7-hexahydro-benzo [a] chinolizin- hydrochlorid, 3 Gewichtsteile Homoveratrylamin, 0,3 Gewichtsteile Ammonacetat und 0,3 Raumteile Eis essig werden in 40 Raumteilen Xylol suspendiert und 24 Stunden unter Rückfluss gekocht, wobei das gebil dete Wasser azeotrop entfernt wird.
Nach dem Er kalten wird mit der gleichen Menge Xylol verdünnt, mit Wasser gewaschen, die Xylollösung eingeengt und der Rückstand mit Äther versetzt. Das kristalline Homoveratrylamid des 2-Carboxymethyl-3-äthyl-9,10- dimethoxy-1,2, 3, 4, 6, 7-hexahydro-benzo [a] chinolizins schmilzt nach dem Umlösen aus Essigester bei 154 bis 155 . Hydrojodid: Schmelzpunkt 220-221 .
Cyclisierwtgsprodukt 1 Gewichtsteil 3-Äthyl-9,10-dimethoxy-1,2,3,4,6,7- hexahydro-benzo [a]chinolizin-(2)-essigsäure-homover- atrylamid wird in 20 Raumteilen Benzol gelöst, mit 0,5 Raumteilen Phosphoroxychlorid versetzt und 1 Stunde auf 80 erwärmt. Nach dem Abkühlen wird vom ausgeschiedenen<B>Öl</B> dekantiert und dieses mit Aceton unter Zusatz von Wasser aufgekocht.
Das kri stalline 2-[(6',7'-Dimethoxy-3',4'-dihydro-isochinolyl- 1')-methyl ]-3-äthyl-9,10-dim ethoxy-1, 2, 3, 4, 6, 7-hexa- hydro - benzo [a] chinolizin - dihydrochlorid - hydrat schmilzt, nach Umlösen aus Methanol-Äther und Trocknen bei 50 an der Luft, bei 122-124 unter Rot färbung und vorherigem Sintern.
Das saure Oxalat- hydrat schmilzt bei 144-146 unter Rotfärbung und vorherigem Sintern. Das U.V.-Absorptionsspektrum in n/ 100 HCl zeigt Maxima bei 240 mlt, 290 my, 304 ma und 354 mu.
Hydrierung sprodukt 1 Gewichtsteil 2-[(6',7'-Dimethoxy-3',4'-dihydro- isochinolyl-1')-methyl]-3-äthyl-9,10-dimethoxy-1,2,3,4, 6, 7 - hexahydro - benzo [a] chinolizin - dihydrochlorid- hydrat wird in Wasser gelöst. Die nach dem Alkalisch stellen mit Ammoniak abgeschiedenen Basen werden in Äther aufgenommen, nach dem Einengen in 100 Raumteilen absolutem Äther gelöst, von Ungelöstem filtriert und mit 0,2 Gewichtsteilen Lithiumaluminium- hydrid versetzt.
Nach Abklingen der momentanen Reaktion wird noch %2 Stunde unter Rückfluss gekocht, mit Wasser versetzt, die Ätherlösung dekantiert und eingeengt. Der Rückstand wird in 90% Methanol gelöst, mit alkoholischer Salzsäure kongosauer gestellt und Äther bis zur Trübung zugegeben. Es kristallisieren grobe Prismen eines 2-[(6',7'-Dimethoxy-l',2',3',4' tetrahydro-isochinolyl-1')-methyl]-3-äthyl-9,10-dimeth- oxy-1,2,3,4,6,7-hexahydro-benzo [a]chinolizin-dihydro- chlorid-hydrates, die nach dem Umlösen aus Sprit- Äther unter vorherigem Sintern bei 253-255 schmel zen.
Das U.V.-Absorptionsspektrum zeigt Maxima bei 231 und 282 mlC. Das saure Oxalat-hydrat schmilzt bei 163-165 unter vorherigem Sintern. Die Mutterlauge aus obiger Kristallisation liefert nach dem Einengen und Umlösen aus Methanol-Äther ein Dihydrochlorid- hydrat einer zweiten stereoisomeren Verbindung vom Schmelzpunkt 213-2l5 unter vorherigem Sintern. Saures Oxalat-hydrat: Schmelzpunkt 190-192 .
Das U.V.-Absorptionsspektrum in n/100 Salzsäure zeigt Maxima bei 231 und 282 mu.
Beispiel <I>2</I> Die katalytische Hydrierung von 2-[(6',7'-Dimeth oxy-3',4'-dihydro-isochinolyl-1')-methyl]-3-äthyl-9,10- dimethoxy-1,2,3,4,6,7-hexahydro-benzo [a]chinolizin- oxalat-hydrat, das nach Beispiel 1 gewonnen wurde, über vorhydriertem Platinoxydkatalysator, liefert nach der Aufarbeitung nach den Angaben in Beispiel 1 die beiden stereoisomeren Dihydrochlorid-hydrate vom Schmelzpunkt 253-255 resp. 213-215 in analoger Weise.

The acidic oxalate hydrate melts at 144-146 with a red color and previous sintering. The U.V. absorption spectrum in n / 100 HCl shows maxima at 240 mlt, 290 my, 304 ma and 354 mu.
Hydrogenation product 1 part by weight of 2 - [(6 ', 7'-dimethoxy-3', 4'-dihydroisoquinolyl-1 ') - methyl] -3-ethyl-9,10-dimethoxy-1,2,3,4 , 6, 7 - hexahydro - benzo [a] quinolizine dihydrochloride hydrate is dissolved in water. The mother liquor from the above crystallization gives, after concentration and redissolution from methanol-ether, a dihydrochloride hydrate of a second stereoisomeric compound with a melting point of 213-215 with prior sintering. Acid oxalate hydrate: melting point 190-192.

Example <I> 2 </I> The catalytic hydrogenation of 2 - [(6 ', 7'-Dimethoxy-3', 4'-dihydro-isoquinolyl-1 ') - methyl] -3-ethyl-9.10 - Dimethoxy-1,2,3,4,6,7-hexahydro-benzo [a] quinolizine oxalate hydrate, which was obtained according to Example 1, over a pre-hydrogenated platinum oxide catalyst, gives the two after working up according to the information in Example 1 stereoisomeric dihydrochloride hydrates with a melting point of 253-255, respectively. 213-215 in an analogous manner.
