Description
<B>Zusatzpatent</B> zum Hauptpatent Nr. 307624. Verfahren <B>zur Herstellung von</B> 2-Keto-l-idonsäure. Im Hauptpatent ist. ein Verfahren zur llerstellung von 2-Keto-l-idonsäure beschrie ben, welches darin besteht, dass man 1-Idon- säure oder ein Salz derselben der oxydativen Gärung mit Pseudomonas aeruginosa als Gärungerreger unterwirft.
Gegenstand des vorliegenden Zusatzpaten tes ist ebenfalls ein Verfahren zur Herstellung von 2-Keto-l-idonsäure auf dem Wege der oxy dativ en Vergärung von 1-Idonsäure oder von Salzen derselben. Dieses Verfahren ist da.- dureh gekennzeichnet., da.ss man Aeetobacter welanogenum als Gärungserreger verwendet.
auch dieser Erreger lässt sich leicht durch ge eignete Passagenzüchtung an die 1-Idonsäure So weit assimilieren, dass der Gäreffekt dem jenigen mit bekannten Erregern gleichkommt.
Für die Vergärung kann zum Beispiel das Calcium-, Kalium- oder Ammoniumsalz der 1-Idonsäure in Konzentrationen bis zu 10% angewendet werden; die Vergärung wird zweckmässig unter Belüftung bei 'Temperatu ren von 28 - 30 C durchgeführt.
<I>Beispiel:</I> 50 g Calcium-l-idonat werden in 1 Liter 10/aigeni Hefeextrakt oder Hefeautolysat ge löst, 5 g d=Glukose zugesetzt, sterilisiert und mit 50 cm3 einer durch Passagenzüchtung auf dem gleichen Nährboden aktivierten Kul tur von Acetobacter melanogenum beimpft. Nach einer Gärdauer von 3 - 5 Tagen bei gleichzeitiger Belüftung mit steriler Luft oder Sauerstoff und einer Gärtemperatur von 28 bis 30 C ist das Maximum der Gärung er reicht.
Nach Filtration der Bakterien wird im Vakuum bei 400 C zu einem Sirup eingeengt und dieser in 1 Liter Methanol eingerührt. Das in Flocken ausfallende Calcium-2-keto- 1-idonat wird filtriert und im Vakuum ge trocknet. Ausbeute 35-40 b Calcium-2-keto- 1-idonat =<B>70</B> - 800/a der Theorie. An Stelle des Calciumsalzes der 1-Idonsäure kann auch das Kalium- oder Ammoniumsalz verwendet werden.

For example, the calcium, potassium or ammonium salt of 1-idonic acid can be used in concentrations of up to 10% for fermentation; the fermentation is expediently carried out with aeration at temperatures of 28-30 C.
