Description

Ausbeute 35-40 b Calcium-2-keto- 1-idonat =70 - 800/a der Theorie.

Additional patent to main patent no. 307624. Process for the production of 2-keto-l-idonic acid. The main patent is. a process for the production of 2-keto-l-idonic acid described, which consists in subjecting 1-idonic acid or a salt thereof to oxidative fermentation with Pseudomonas aeruginosa as the fermentation agent.

The subject of the present additional patent is also a process for the production of 2-keto-l-idonic acid by way of the oxidative fermentation of 1-idonic acid or salts thereof. This process is characterized by the fact that Aeetobacter welanogenum is used as a fermentation agent.

This pathogen, too, can easily be assimilated to 1-idonic acid through suitable passage breeding to such an extent that the fermentation effect is equivalent to that with known pathogens.

For example, the calcium, potassium or ammonium salt of 1-idonic acid can be used in concentrations of up to 10% for fermentation; the fermentation is expediently carried out with aeration at temperatures of 28-30 C.

Example: 50 g calcium l-idonate are dissolved in 1 liter 10 / aigeni yeast extract or yeast autolysate, 5 gd = glucose are added, sterilized and with 50 cm3 of a culture activated by passage cultivation on the same nutrient medium inoculated by Acetobacter melanogenum. After a fermentation period of 3 - 5 days with simultaneous ventilation with sterile air or oxygen and a fermentation temperature of 28 to 30 C, the maximum fermentation is enough.

After filtration of the bacteria, the mixture is concentrated in vacuo at 400 ° C. to a syrup and this is stirred into 1 liter of methanol. The calcium 2-keto-1-idonate which precipitates out in flakes is filtered and dried in vacuo. Yield 35-40 b calcium 2-keto-1-idonate = 70 - 800 / a of theory. The potassium or ammonium salt can also be used instead of the calcium salt of 1-idonic acid.
