Verfahren zur Herstellung eines Antibiotieums. Die Erfindung bezieht sich auf ein Ver fahren zur Herstellung eines neuen, als Fu- magillin bezeichneten Antibioticums.
In neuerer Zeit wurde gefunden, dass neue, bisher nicht verwendete chemische Stoffe mit vorher nicht erzielten Wirkungen aus Nähr medien isoliert werden können, in welchen ge wisse spezifische Mikroorganismen unter sorg fältig kontrollierten Bedingungen gezüchtet wurden.
Diese therapeutisch nutzbaren chemischen Verbindungen wurden ohne Rücksicht auf ihre chemische Struktur als Antibiotica bezeichnet und auf Grund ihrer Herstellungsmethode und ihrer wachstumshemmenden Wirkung ge genüber Mikroorganismen klassifiziert. Unter diesen antibiotischen Stoffen - soweit sie iso liert und charakterisiert wurden - konnten bisher nur wenige gefunden werden, die gegen Viren wirksam sind oder die Aktivität von Bakteriophagen hemmen.
Gegenstand der vorliegenden Erfindung ist ein Verfahren zur Herstellung eines Anti- bioticums, welches dadurch gekennzeichnet ist, dass man einen Pilz aus der Gruppe Asper- gilliis fumigatus in einem Nährmedium züch tet und dabei im Nährmedium das Antibioti- cum Fumagillin als Erzeugnis erhalten wird. Das Fumagillin ist eine antibiotische Substanz mit antibakteriophagen Eigenschaften, die gegen Viren wirksam ist.
Dieser bisher un bekannte Stoff kann als solcher oder in Form seiner Derivate für die Behandlung von Infek tionen bei Menschen bzw. Tieren Verwendung finden.
Fumagillin ist eine weisse, kristalline, feste organische Carboxylsäure mit einem pg-Wert von etwa 6,5 (-log. der Säuredissoziations- konstanteg,festgestellt durch elektrometrische Titration) und einem Schmelzpunkt von 189 bis 194 C (Kofler-Block), oder 190 bis 191 C (Kapillarrohr). Es ist optisch aktiv, [a] D = minus 26,6 (c: 0,25 % in Methanol).
Es enthält lediglich die Elemente Kohlenstoff, Wasserstoff und Sauerstoff, hat angenähert die empirische Formel C27H3r,07 und ent hält neben einer Carboxylgruppe eine Alkoxyl- gruppe. Sein Molekulargewicht beträgt 475 (berechnet aus einem Neutraläquivalent) bzw. 488 (berechnet aus der Alkoxylbestimmung). Das kristalline Produkt gibt keine Ferrichlo- rid- oder Millonsche Reaktion.
Die Salkowski- Sterol-Probe ist fraglich positiv, der Lieber- mann-Burchad-Test ist negativ. Der Legal sehe Test (Aglucone usw.) ist negativ.
Sein Ultraviolett-Absorptionsspektrum zeigt Maxi- mas bei 239, 304 (Inflexion), 332 (Inflexion), 336 und 351 m,u mit k -Werten von 7;52 bei 239 m,u, 147,8 bei 336 m,ss und 136,4 bei 351 m,u, was die Anwesenheit eines sich aus wenigstens drei, möglicherweise aus vier Dop pelbindungen zusammensetzenden Systems konjugierter Doppelbindungen anzeigt.
Das Infrarot-Spektrum zeigt Banden bei 3120, 1714, 1632, 1597, 1576, 1491, 1377, 1230, 1163; 1124, 1013 und 838 Millimikron. Fumagillin bildet einen IX ethylester, Schmelzpunkt 145 bis 147 C (Kofler-Block), ein Octabromid, Schmelzpunkt 118 bis 122 C (Koller-Block), ein Amid, welches bei 160 C (Koller) ver kohlt, und ein 2,4-Dinitrophenylhydrazon, Schmelzpunkt 123 bis 126 C (Koller-Block).
Der Organismus, welcher die neue antibio tische Substanz gemäss der Erfindung erzeugt, wurde aus einer Bodenprobe, welche aus Kala- mazoo Caunty, Michigan, stammte, isoliert.
Dieser im Boden gefundene Organismus ist funktionell und strukturell ein Glied der Aspergillus fumigatus-Reihen der Aspergillus fumigatus-Gruppe, wie sie von Thom und Raper, Manual of Aspergilli, Williams und Wilkins, Baltimore, Md., 1945, Seiten 87 bis 91, beschrieben wurde;
jedoch ist er von der Type des Stammes Aspergillus fumigatus NRRL 163 hinsichtlich mehrerer morphologi scher Merkmale verschieden, wie in folgender Tabelle gezeigt wird.

Ergänzend zu den sich aus dem vorstehen den Vergleich ergebenden Unterschieden wird darauf hingewiesen, dass Aspergillus fumi- gatus NRRL 163 keine antiphage Substanz produziert, wenn er unter den gleichen Bedin gungen kultiviert wird,
unter welchen die anti biotische Substanz gemäss der Erfindung durch Aspergillus fumigatus H-3 erzeugt wird.
Der Nachweis von Fumagillin in Fermen tationsliquor sowie die angenäherte Bestim mung der in dieser Flüssigkeit oder in andern Lösungen vorhandenen Menge beruht auf der dem Fumagillin eigenen Fähigkeit, die Wir kung von Bacteriophagen zu inhibieren. Für Versuchszwecke wurde die inhibierte Wirkung des Fumagillins, auf Staphylococcus aureus 209 P als Standard gewählt.
Die Zubereitung der erforderlichen Lösun gen und ihrer Anwendung für den Nachweis von Fumagillin kann folgendermassen er folgen Herstellung <I>der</I> baeteriophagen Suspension. In mehrere Flaschen wird eine geeignete Menge einer sterilen Nährbouillon gefüllt, die 0,5 % Pepton, 0,3 % Rindfleischextrakt und 0,5 % Natriumchlorid enthält.
Diese Bouillon wird mit 0,5 Volumenprozent einer 24-Stunden- Nährbouillon von Staphilococcus aureus 209 P geimpft und bei 37 C etwa 7 Stunden lang oder bis die Bouillon trüb geworden ist, incubiert. Dann werden zum Inhalt jeder Flasche etwa 5 Volumenprozente einer aktiven bacterio- phagen Suspension zugegeben, die in der fol genden Weise bereitet wurde:
Man züchtet eine Wirteluiltur (staphilococcus aureus 209P) in einer Trypticase-Soja-Bouillon, bis sich eine Trüblmg entwickelt. Diese Bouillon Kultur wird sodann mit der Stammsuspension einer Bacteriophagenkultur, beispielsweise der American Type Culture Collection versetzt und so lange incubiert, bis die Auflösung der Wirtezellen erfolgt ist (in 12 bis 24 Stunden).
Die Bouillon wird sodann durch ein Seitz- Filter filtriert, um etwa nicht aufgelöstes Zell- material abzutrennen; das erhaltene klare Fil trat besteht aus einer Suspension von Bacterio- phagen-Teilchen in hoher Konzentration. Diese Operation lässt sich viele Male hintereinander wiederholen, bis man die gewünschte hohe Konzentration von Bacteriophagen-Teilchen je em3 erhält (gewöhnlich ist eine Suspension von 1 bis 2 Milliarden Teilchen je cms für Versuchszwecke befriedigend).
Der Flaschen inhalt wird dann abermals bei 37 C etwa 16 Stunden incubiert, in welcher Zeit die Trü bung der Bouillon entweder ganz verschwunden ist oder merklich abgenommen hat. Der Inhalt der Flaschen wird zusammengegossen, und die Zellen werden durch Filtration durch ein steriles Filter aus gesintertem Glas (Poren grösse UF) abgetrennt. Die sterile, gefilterte; bacteriophage Suspension wird auf ihre Ak tivität geprüft und.in einem Kühlschrank bis zum Gebrauch gelagert.
Vorbereitung <I>der</I> Testplatten. Geschmolzenes, halbfestes Nähragar, das 0,5 % Pepton, 0,3 % Rindfleischextrakt, 0,5 Natriumchlorid, 0,7 %Agar und Wasser ent hält, wird auf 50 C abgekühlt und mit etwa 1 Volumenprozent einer 24-Stunden-Bouillon- Kultur von S. aureus 209 P gemischt.
Zu der flüssigen Agar-Suspension wird eine durch Vorversuche bestimmte, zur Verhinderung des Wachstums von S-aureus 209 P hinreichende Menge der in bereits angegebener Weise berei teten phagen Suspension zugesetzt. Je 5 cm3 dieser bacteriophagen S-aureus-Suspension werden in mit flachem Boden ausgestattete Petrischalen gegossen und erstarren gelassen. Die so zubereiteten Testplatten werden bis zum Gebrauch unter Kühlung gelagert.
Herstellung <I>des</I> Standardpräparates <I>und der</I> illuster.
Es wird eine Acetonlösung mit 100 Mikro gramm kristallinem Fumagillin je cms her gestellt. Teile dieser Lösung werden mit ste rilem, destilliertem Wasser auf Konzentra tionen von 10, 5, 2,5 und 1,5 Mikrogramm je cms verdünnt.
Die zu untersuchenden Muster mit nicht näher bekanntem Fumagillin- gehalt werden mit Aceton und Wasser (.die Unlöslichkeit von gereinigtem Fumagillin kann es notwendig machen, Lösungen, die mehr als 100 Mikrogramm je em3 enthalten, mit Aceton zu verdünnen) bis zu einer schät zungsweisen Konzentration von 1,25 bis 10 Mikrogramm je cm3 verdünnt.
Untersuchungsmethode. In jede der obigen Lösungen wird ein 6,35 mm (1/,1 Zoll) Durchmesser besitzendes Filterpapierscheibchen (Schleicher und Schüll 740 E) getaucht, und die überschüssige Flüs sigkeit von den Scheiben wird abtropfen ge lassen. Dann werden die einzelnen Papier scheiben auf je eine in der beschriebenen Weise hergestellte Petri-Platte gelegt. Gewöhnlich werden von den vier Standardlösungen sowie der zu untersuchenden Lösung je vier Schei ben angelegt, jede auf einer besonderen Test platte.
Die Platten werden bei 37 C 16 bis 18 Stunden lang incubiert, wonach der Durch messer der Wachstiunszonen in Millimetern ge messen und die durchschnittliche Grösse der Zonen- für jede Verdünnung ermittelt wird. Die Messergebnisse für die Zonengrösse bei den Standardverdünnungen werden gegenüber ihren Konzentrationen aufgetragen; die so er haltenen Punkte werden durch Linien ver bunden und ergeben eine Standardkurve.
Nach Bestimmung der Wachstumszone inMillimetern der Testplatte der Fumagillinlösung von un bekannter Konzentration kann mit Hilfe der Standardkurve der fragliche:Fumagillingehalt in Mikrogramm je cm3 angegeben werden.
Definitionsgemäss besitzt ferner eine Lösung, die Fumagillin bzw. Antibiotikum H-3 ent hält, dann 1 Phage-Einheit je eins, wenn diese Lösung bei der Bestimmung nach der oben be schriebenen Methode nach 24stündiger In- cubationsdauer der Platte eine Wachstumszone von Staphylococcus aureus von 8 mm Durch messer ergibt.
Das erfindungsgemässe Verfahren wird vorteilhaft mit einem Fumagillin erzeugen den Stamm von Aspergillus fumigatus H-3, unter submersen, aeroben Bedingungen, in einer Nährlösung, welche ein Kohlehydrat, Kochsalz, einen Zusatz eines festen Corn-steep- Präparates, Caleiumearbonat und so viel Na- triumhydroxyd enthält,
dass der schliessliche pH-Wert des Mediums etwa 6,7 beträgt. Das erzeugte Fumagillin kann vom Kulturmedium isoliert werden, vorzugsweise so, dass man das Fumagillin vom Mycelium durch Extraktion mit Chloroform abtrennt, das Rohfumagillin aus dem Chloroform abscheidet und durch Lösen in Aceton reinigt. Hiezu wird die Acetonlösung z. B. konzentriert und gekühlt, der entstehende Niederschlag abgetrennt, mit tert. Butanol gewaschen und aus Methanol und Wasser auskristallisiert.
Für die Herstellung von Fumagiillin wird ein Nährmedium etwa folgender Zusammen setzung bevorzugt Dextrin 5-15 Teile Natriumchlorid <B>-1-</B> 7 " Festes Corn-steep-Präparat 25-40 " Calcituncarbonat 1- 2 " Wasser Ergänzung auf 1000 " Das Nährmedium wird zweckmässig durch Zusatz von Natriumhydroxydlösung auf einen pH-Wert von etwa 6,7 eingestellt, durch Er hitzen sterilisiert und dann gekühlt.
Mit die sem Medium ist es möglich, in 36 bis 72 Stun den etwa 70 bis 170 Mikrogramm Fumagiliin je cm3 zu erzeugen.
Für die Herstellilng grosser Mengen Fuma- gillin wird dem Nährmedium die Aspergillus fumigatus H-3-Kultur, vorzugsweise in einer Menge von etwa 5 Volumenprozenten, bezogen auf das Nährmedium, zugesetzt. Die Impf kultur kann dadurch erhalten werden, dass der Fümagillin erzeugende, auf Schrägagar ge züchtete Stamm von Aspergillus fumigatus H-3 in Schüttelflaschen eingebracht wird, welche ein die angegebene Zusammensetzung besitzendes Nährmedium enthalten.
Nach 2 Tagen Wachstum kann ihr Inhalt in Rühr- flaschen (Sweep-stirbottles) übertragen wer den, welche das oben angegebene Medium ent halten und dort weitere zwei Tage wachsen gelassen werden.
Die Fermentation kann bei 20 bis 30 C 36 bis 72 Stunden lang durchgeführt werden, wobei die maximale Ausbeute gewöhnlich nach etwa 42stündiger Incubationsdauer erreicht wird. Während der Fermentation kann Luft durch das bewegte, geimpfte Medium geleitet werden, vorzugsweise in einer Menge von etwa 0,4 Volumteilen in. der Minute auf einen Volumteil des Mediums.
Nachdem die Fermen tation die gewünschte, durch Prüfung des Ge misches auf den Fuunagillin-Gehalt bestimmte Zeitspanne vor sich gegangen ist, wird die Flüssigkeit zweckmässig durch Filtration ge klärt. Die geklärte Flüssigkeit kann durch Extraktion mit einer etwa i[io ihres Volumens betragenden Menge von handelsüblichem Hexan entfettet werden. Aus der entfetteten geklärten Flüssigkeit kann man das Fuma- gillin mit Chloroform extrahieren, das Chloro form aus dem Extrakt abtreiben und den Rückstand in Aceton lösen.
Die Acetonlösung scheidet nach Kühlung eine geringe Menge eines braunen Niederschlages aus, der aus Ver unreinigungen besteht und entfernt werden kann. Die Acetonlösimg wird z. B. bis auf etwa 60 % ihres ursprünglichen Volumens konzen triert, gekühlt lind der entstehende, das Anti- biotieum enthaltende Niederschlag abgetrennt. Das Präzipitat kann nach Waschen mit ter tiärem Butanol aus einer Mischung von Me thanol und Wasser umkristallisiert werden. Das so erhaltene Fumagillin besitzt die oben angegebenen Eigenschaften.
Zur näheren Erläuterung der Erfindung diene folgendes Beispiel: a) Herstellung <I>einer</I> Impfkultur.
Man bringt vegetative Individuen und Sporen von auf Schrägagar gezüchtetem Aspergillus fumigatus H-3 in einige Halbliter flaschen, von welchen jede 100 eins eines Nähr mediums folgender Zusammensetzung enthält Dextrin 10 g Kochsalz 5 g Festes Corn-steep-Präparat 32 g Calcilimearbonat 1 g Leitungswasser, Ergänzung auf 1 Liter Der pH-Wert des Mediums wird durch Zu satz von Natriumhydroxydlösung auf 6,7 ein gestellt.
Dieses Medium wird sterilisiert, mit Schräg agar versetzt und 48 Stunden lang bei einer Temperatur von 24 C geschüttelt (belüftete submerse Kultur). Diese Kultur von Asper- gillus fumigatus H-3 kann entweder unmittel bar zur Impfung eines Ansatzes im Grossen oder zur Erzeugung von grösseren Mengen Impfstoff verwendet werden.
<I>b)</I> Herstellung von grösseren <I>Mengen</I> Impfmaterial. Zu 6 Litern eines Mediums, das die unter a) angeführten Bestandteile enthält, werden in einer 18,9 Liter (5 Gallonen) fassenden Flasche, die für Bewegung und Belüftung ein gerichtet ist, 5 Volumenprozente einer nach a) erhaltenen Impfkultur gegeben. Das geimpfte Medium wird bei einer Temperatur von 24 C 48 Stunden incubiert und dabei gelüftet. Nach dieser Zeit ist die Brühe zur Impfung grosser Behälter geeignet. <I>c) Ansatz im</I> Grossen.
6781 Liter (1500 Gallonen) Nährmedilun der unter a) angegebenen Zusammensetzung werden in einem mit Glas ausgefütterten, 7570,8 Liter (2000 Gallonen) fassenden Tank mit 281,8 Liter (75 Gallonen) einer vege tativen 48-Stunden-Kultur von Aspergillus fumigatus H-3, die wie unter b) angegeben hergestellt wurde, geimpft. Das geimpfte Nähr medium wird 42 Stunden bei einer Tempera tur von 240 C incubiert und dabei pro Minute etwa 2200 Liter (etwa 80 Kubikfuss) Luft durchgeblasen. Die Untersuchung einer Probe des Mediums nach 42 Stunden zeigte, dass 170 Phage-Einheiten je eins vorhanden waren.
(Definition der Phage-Einheit: siehe oben.) d) <I>Isolierung des</I> h'umagillins.
Am Ende der Fermentationsperiode wer den 68,04 kg (150 Pfund) Diatomeenerde als Filtrierhilfe dem Inhalt des Fermentations- behälters zugesetzt und dann mit Hilfe einer Platten- und Rahmenfilterpresse filtriert.
Die geklärte Flüssigkeit enthält 26,6 mg Fest Stoff je eins und ergab 142 Phage-Einheiten je eins. Die geklärte Flüssigkeit wurde .mit 670 Liter (177 Gallonen) handelsüälichexn Hexan innig gemischt.
Dabei wurde ein Pod- bielniak-Extraktor verwendet. Die Hexan- schicht, welche die unerwünschten Fettstoffe enthält, wurde abgezogen. Die entfettete Flüs sigkeit wurde dann mit 587 Liter (155 Gal lonen) Chloroform extrahiert. Die Ohloro- formschicht wurde abgetrennt; sie enthielt 1190-g Feststoffe und 35g Fumagillin. Das Chloroform wurde unter vermindertem Druck, ohne Wärmezufuhr von aussen, -abgetrennt.
Der nach Entfernung des Chloroforms zurück bleibende Sirup wurde mit Aceton bis zu einem Volturen von 3700 cm- aufgefüllt. Die Ace- tonlösung wurde auf 5 C gekühlt, wobei eine geringe Menge eines braunen Niederschlages ausfiel, der durch Filtration abgesondert wurde. Der Niederschlag wurde mit Aceton ge waschen, die Waschflüssigkeit wurde dem ersten Filtrat zugeschlagen.
Das Volumen vom ersten Filtrat und Waschflüssigkeit be trug zusammen 3800 em3. Diese Lösung ent hielt 1062 g Feststoffe mit einem Gehalt an antibakteriophagem Wirkstoff von 300 Micro- gramm je mg.
1500 ems der erwähnten Acetonlösung wer den bei vermindertem Druck und bei Raum temperatur unter Stickstoffatmosphäre auf ein Volturen von 900 cm3 konzentriert. Die sich ergebende dicke Suspension wurde unter Stickstoff in eine 1 Liter fassende Zentri- fugiertasse gebracht und 19 Stunden auf minus 30 C gekühlt. Die Suspension wurde 1 Stunde mit einer Geschwindigkeit von 1500 bis 1700 UmdrehungenjMin. zentrifugiert.
Die über stehende Flüssigkeit würde vom festen Rück stand dekantiert und letzterer fünfmal bei Raumtemperatur mit je 1525 cms tert. Butanol gewaschen.
Der nach dem Waschen mit ter tiärem Butanol zurückbleibende feste Riick- stand betrug nach Trocknung bei Raumtem peratur 22,2 g: Nach Rekristallisation dieses Materials aus einer Mischung von gleichen Teilen Methanol und Wasser wog es 19,8 g und hatte einen Schmelzpunkt von 190 bis 191 C (Kapillarrohr) sowie die bereits erwähnten andern physikalischen und chemischen Eigen schaften.

Die Ultraviolett- und Infrarotabsorptions- spektren zeigen die Anwesenheit von Absorp tionsbändern im Bereich von 3285 em-1, 3100 cin-1, 1710 cm-1, 1618 cm-', 1596 cm-', 1520 em-1 und 1505 cm-1.
Method of making an antibiotic. The invention relates to a method for the production of a new antibiotic called Fumagillin.
More recently it has been found that new, previously unused chemical substances with previously unachievable effects can be isolated from nutrient media in which certain specific microorganisms were grown under carefully controlled conditions.
These therapeutically useful chemical compounds were referred to as antibiotics regardless of their chemical structure and classified on the basis of their production method and their growth-inhibiting effect against microorganisms. Among these antibiotic substances - insofar as they have been isolated and characterized - so far only a few have been found that are effective against viruses or inhibit the activity of bacteriophages.
The present invention relates to a method for producing an antibiotic which is characterized in that a fungus from the group Aspergilliis fumigatus is grown in a nutrient medium and the antibiotic fumagillin is obtained as a product in the nutrient medium. Fumagillin is an antibiotic substance with antibacteriophage properties that is effective against viruses.
This previously unknown substance can be used as such or in the form of its derivatives for the treatment of infections in humans or animals.
Fumagillin is a white, crystalline, solid organic carboxylic acid with a pg value of about 6.5 (-log. The acid dissociation constant, determined by electrometric titration) and a melting point of 189 to 194 C (Kofler block), or 190 up to 191 C (capillary tube). It is optically active, [a] D = minus 26.6 (c: 0.25% in methanol).

The organism which produces the new antibiotic substance according to the invention was isolated from a soil sample which came from Kalamazoo Caunty, Michigan.
This organism found in the soil is functionally and structurally a member of the Aspergillus fumigatus series of the Aspergillus fumigatus group, as described by Thom and Raper, Manual of Aspergilli, Williams and Wilkins, Baltimore, Md., 1945, pages 87 to 91 has been;

Investigation method.
