Verfahren zur Herstellung eines Penicillinsalzes. 1)ie vorliegende Erfindung betrifft ein Verfahren zur rlerstellung eines neuen Peni- cillinsalzes, und zwar des Cy clohexylaminsal- zes cles Penicillins, welches bei 25 C in Was ser zu 5 % und in Alkohol zu 15 /o, löslich ist und ein Spektrum aufweist, das Banden bei<B>1780</B> ein-', was die Anwesenheit eines f-Laktaniringes anzeigt, und bei 692 und 726 ein-',
was auf einen Phenylessigsäurerest schliessen lässt, enthält. Das neue Penicillin salz soll für die Reinigung von Penicillin Ver wendung finden.
Das Verfahren gemäss der vorliegenden Erfindung ist dadurch gekennzeichnet., dass man eine wässrige Lösung eines Penicillin metallsalzes, vorzugsweise bei einem pH von 6-7, mit einem mineralsauren Salz des Cyelo- hexylainins zur Umsetzung bringt.
Es wurde festgestellt, dass bei der Behand lung einer wässrigen Lösung eines rohen Peni- cillinmetallsalzes (z. B. eines Natrium- oder Calciunisalzes), welche vorteilhafterweise, aber nicht notwendigerweise, eine gussalzende Sub stanz enthalten kann, mit einem mineralsauren Salz des Cyclohexy lamins, z. B. dem Hydro chlorid, eine doppelte Umsetzung eintritt, und so das Penieilliiieyelohexylaminsalz gebildet wird.
Das Cyclohexylaniinsalz kann der wä.ssri- gen Lösung des Penieillinmetallsalzes entweder in Forni der im wesentlichen reinen Substanz oder in Form einer konzentrierten, wässrigen Lösung zugesetzt werden. Das Einstellen der Wasserstoffionenkonzentration auf einen Wert von 6 bis 7 kann z. B. dadurch erfolgen, dass eine Säure, wie z. B. Phosphorsäure, oder eine anorganische Base, wie z. B. Natriumhydro- xyd, zugesetzt wird.
Es wurde festgestellt, dass es vorteilhaft ist, die Fällung des Cyclohexyl- aminsalzes des Penicillins in Gegenwart einer im Folgenden zu beschreibenden, gussalzenden Substanz auszuführen.
Als Ausgangsmaterial wird zweckmässig eine gekühlte wässrige Lösung des rohen Peni- cillinmetallsalzes (z. B. des Natrium- oder Cal- eiumsalzes) verwendet. Die Aktivität des ver wendeten Penicillins sollte nicht kleiner als 400 Oxfordeinheiten pro mg sein und die Kon zentration des Penicillins in der wässrigen Lö sung vorteilhaft. 2 bis 10 1/a Gewicht/Volumen betragen.
Diese Lösung enthält, wie bereits oben er wähnt wurde, vorteilhaft eine gussalzende Sub stanz, das heisst ein Salz mit einer hohen Lös lichkeit in Wasser, dessen Lösung zweckmässig neutral ist. Bevorzugt wird die Verwendung von Salzen, wie z. B. Natriumchlorid, Natrium sulfat, Kaliumchlorid und dergleichen. Am moniumsalze sind nicht günstig. Salze, die auf das Penicillin oder dessen Salze eine zerstö rende Wirkung ausüben, sollten natürlich nicht verwendet werden. Die Menge der in der Lösung vorhandenen gussalzenden Sub stanz wird bevorzugterweise so bemessen sein, dass die Lösung annähernd gesättigt ist.
Wenn es erwünscht ist, als P:enicillin- inetallsalz das Calciumsalz zu verwenden, so wird man das Cy elohexylaininsalz vor dem aassalzenden Mittel zusetzen, da. sonst. durch Aussalzung ein das Penicillin enthaltender Niederschlag entstehen könnte, welcher nicht (las Cyclohexylaniinsalz des Penicillins dar stellt.
Zur Ausführung des Verfahrens gemäss vorliegender Erfindung kann beispielsweise wie folgt. vorgegangen werden Die wässrige Lösung des Penicillinmetall- salzes wird unter Rühren und Kühlen mit einer dem vorhandenen Penicillin mindestens äquivalenten Menge eines Cycloliexylamin- salzes, entweder in Form der im wesentlichen reinen Substanz oder in Form einer konzen trierten Lösung, versetzt.
Das Gemisch wird auf ein Pu von 6 bis 7 eingestellt, indem unter Rühren vorsichtig eine Säure, wie z. B. Phos phorsäure, oller eine anorganische Base, wie z. B. Natriumhy droxyd, zugesetzt wird. Dieses Gemisch wird gerührt und gekühlt, bis keine weitere Fällung mehr eintritt. Das Reaktions produkt, nämlich das rohe Cyclohexylaminsalz des Penicillins, wird durch Filtrieren oder Zentrifugieren isoliert.
Es kann mit einer kleinen Menge einer annähernd gesättigten Lösung einer aassalzenden Substanz geWa- schen werden; diese ist zweckmässig eine neu trale, wässrige Lösung. Das Reaktionsprodukt wird durch Filtrieren und Zentrifugieren so vollständig als möglich vom Wasser befreit.
Das feuchte Produkt kann auf verschie dene Arten behandelt werden. Zweckmässig wird das zurückgehaltene Wasser soweit als möglich aus :(lern so erhaltenen Material ent fernt, indem dieses während mehreren Stun den über einem Troeknungsmittel, wie z. B. Phosphorpentoxyd, in einem evakuierten Ex sikkator gehalten wird. Wenn das Produkt trocken ist, so wird es eventuell vorteilhaft mit Aceton oder Methyläthylleton gewaschen. Das auf diese Weise gewaschene oder das nicht. gewaschene Produkt. kann wie folgt umkristal lisiert werden Das getrocknete Produkt wird mit etwas mehr Chloroform erwärmt, als nötig ist, um bei 30 C das organische Material zu lösen.
Das warme Gemisch wird rasch abfiltriert, um das ungelöste, anorganische Material zu entfernen. Das Filtrat wird mit geiiügen(l Aceton oder Metliyläthylketon versetzt, so dass beim Küh- len das gereinigte Cyeloliexylaniinsalz des Penicillins ausfällt.
Es kann auch direkt. (las feuchte Material umkristallisiert werden. Man kann hiezu z. B. das feuchte Material in Chloroform lösen und die Lösung von den wenigen Tropfen der wäss- rigen Phase abtrennen. Beire Verdünnen mit \?. oder 3 Volumen Aceton oder Methylätliyl- keton kristallisiert dann das gereinigte Cyclo- hexylaminsalz aus.
Es sei betont, dass die hier beschriebene, spezielle Lösung;smittelkombina- tion für Kristallisierzweeke natürlich keines wegs die einzig mögliche ist. Es kann iin Laboratorium leicht festgestellt werden, olr irgendein anderes Lösungsmittel oder Lösungs- mittelgemiseh befriedigende Resultate ergibt. Wenn das auf diese Weise erhaltene Produkt. nicht den gewünschten Reinheitsgrad auf weist, so kann der oben beschriebene Reini- gungsprozess wiederholt werden.
Man kann auch das feuchte Material in einer kleinen Menge Äthylalkohol lösen, z. B. so, dass 3 bis 5 eins auf 1 Megaeinheit des ursprünglich verwendeten Penicillins kommen.
Nachdem eventuell vorhandene Spuren des Aussalzungsmittels, wenn ein solches verwen det wurde, durch Filtrieren entfernt sind, kann das gereinigte Cycloliexylaniinsalz des Penicillins gefällt werden, indem die alkoho- lisehe Lösung mit der etwa 3- bis )fachen Voluinmenge Diäthyl-, Diisopropyl- oder Di- butyläther versetzt wird.
Die in den folgenden Beispielen verwen deten Abkürzungen haben folgende Bedeu tung: ME = Megaeinheit, das heisst 1000000 Oxfordeinheiten, und OE = Oxfordeinheit.
<I>Beispiel 1:</I> 2,5 AIE Penicillinnat.rium (910 OE/mg ) werden in 32 cm3 bei 0 C mit Natriumnitrat gesättigtem Wasser gelöst.
Es wird eine 50 /mige wässrige Lösung von Cz-elohexylamin- hydrochlorid in einer der doppelten Menge an vorhandenem Penicillin äquivalenten Menge zugesetzt. Die Lösung wird auf ein PH von 6 eingestellt. und vor dem Filtrieren etwa eine halbe Stunde stehengelassen. Die Fällung wird mit einigen em3 gesättigter \atrium- nitratlösung gewaschen.
Das feuchte Produkt wird zusammen mit 25 cm3 Chloroform leicht erwärmt und die Lösung durch ein trockenes Filter filtriert. um einige Tropfen noch vorhandener, wässri- ger Lösung zu entfernen. Das Filtrat wird mit. 75 cm3 Aceton versetzt. Das Gemisch wird etwa 1. Stunde im Kühlschrank stehengelassen und dann filtriert. Eine weitere Menge wird durch Einengen des Filtrates unter vermin derten Druck und durch Kristallisierenlassen erhalten.
Die Ausbeute beträgt 1,53 g (erste Fraktion) und 0,1 g (zweite Fraktion) im wesentlichen reines Peiiicillin-eyelohexylamin- salz.
<I>Beispiel 2:</I> Die Lösung von 20 ME Penicillinnatr ium (1100 OE/nig) in 21.5 ein-' Wasser wird bei etwa 0 C mit. Natriumchlorid gesättigt. Das Gemisch wird mit 20 em3 50 o/oiger wässriger Lösung von Cyelohexylamin-hy droehlorid ver setzt, so dass es ein pH von 6,2 aufweist, und wird dann bei 0 C während 1 Stunde stehen gelassen.
Das Gemisch wird hierauf im Va kuum filtriert. und das feste Produkt. durch Absaugen möglichst gut getrocknet. Das Pro dukt wird in 100 em3 Äthylalkohol gelöst und die Lösung durch Filtrieren von Natrium chloridspuren befreit. 300 cm3 Isopropyläther werden zugesetzt, worauf das Penicillincyclo- hexylaminsalz rasch in feinen Nadeln aus kristallisiert.
Nach halbstündigem Stehen bei etwa 10 C werden die Kristalle abfiltriert. Das Filtrat, wird im Vakuum zur Trockne ein gedampft., der Rückstand in 30 em3 Chloro form gelöst und die Lösung mit 80 cm3 Me- thylätliylketon versetzt. Das Gemisch wird in einer Kältemischung abgekühlt und hierauf filtriert.
Das Salz wird mit llethyläthylketon gewaschen. Die Ausbeute an reinem Penleillin- eyeloliexyiaminsalz beträgt. 7.1.,2 g bei der ersten Fraktion und 2.85 ; bei der zweiten Fraktion. 24 II/o des ursprünglichen Penicillins bleiben in der wässrigen Mutterlauge zurück.
An Stelle des Penicillinnatriums können, wie Versuche ergaben, mit gutem Erfolg auch andere Metallsalze des Penicillins verwendet werden.
