EMI0001.0021

Process for the preparation of an intermediate product. It has been found that a previously unknown condensation product, 1-oxy-3,7-dimethyl-6-oxy-9 - [2 ', 6', 6'-trimethyl-cyclohexen- (1 ') -yl] -nonadiene, can be obtained - (2,7) -in- (4), because it can be obtained that 1-oxy-3-methyl-pentene- (2) -in- (4) is reacted with 2 equivalents of an alkylmagnesium halide,
the Grignard compound obtained is condensed with 4- [2 ', 6', 6'-trimethyl-cyclohexene- (1 ') -y1] -2 methyl-buten- (2) -a1- (1) and the condensation product is hydrolyzed.
The procedure is based on the following equation
EMI0001.0021
The required starting materials, namely 4- [2 ', 6', 6'-trimethyl-cyclohexen- (1 ') - y1] - 2-methyl-buten- (2) -al- (1) and 1-oxy- 3-methyl-pentene- (2) -in- (4) can be represented as follows:
4- [2 ', 6', 6'-trimethyl-cyclohexen- (1 ') - yl] -2-methyl-buten- (2) -al- (1) can be obtained from, B-ionone by glycidate synthesis, saponification and Decarboxylation of the glycidic ester formed can be obtained.
1-Oxy-3-methyl-penten- (2) -in- (4) is formed from 3-oxy-3-methyl-penten- (1) -in- (4) (the condensation product of methyl vinyl ketone and acetylene) by action of mineral acids with rearrangement of the double bond and the hydroxyl group. For example, 10% aqueous sulfuric acid is suitable for 4 to 5 days of shaking at room temperature.
The present process is a Grignard reaction in which 1-oxy-3-methylpentene- (2) -yne- (4) is first reacted with 2 equivalents of alkyl magnesium halide (eg ethyl magnesium bromide) . The resulting magnesium compound is now e.g. B. in a manner known per se, with 4- [2 ', 6', 6'-trimethyl cyclohexene - (1 ') -y1] -2 methyl-butene (2) -al- (1) condensed.
The implementation can be carried out in the usual solvents, such as ethyl ether. The reaction product can be hydrolyzed in a conventional manner, e.g. B. with an ammonium salt solution. Unconverted aldehyde can be in the form of a crystallizing derivative, e.g. B. phenyl semicarbazone, separate.
The condensation product (III) formed, 1-oxy-3,7-dimethyl-6-oxy-9-trimethylcyclolhexenyl-nonadiene (2,7) -in - (4), can be purified by chromatography or by vacuum distillation. It is also advantageous to separate off impurities using two immiscible solvents. For this purpose you can z.
B. pull out a solution of the crude condensation product in low-boiling petroleum ether with 75% methanol. The diol migrates into the aqueous methyl alcohol, while the impurities mainly remain in the petroleum ether.
The process provides a previously unknown intermediate for the preparation of a pentaene (vitamin A alcohol). It is a very viscous, yellowish oil that has not yet been crystallized. It does not absorb any ultraviolet rays with a wavelength greater than 260 mu; the solution in arsenic trichloride is purple in color.
The Zerewitinoff determination gives values for two active hydrogen atoms. nD = 1.527.
Example 1: While stirring vigorously, a boiling ethylmagnesium bromide solution (made from 10 parts by weight of magnesium, 46 parts by weight of ethyl bromide and 150 parts by volume of ether) is converted into a solution of 20 parts by weight of 1-oxy-3-methyl- penten- (2) -in- (4) ether added in 50 parts of space.
To complete the conversion it is refluxed for 3 hours at an oil bath temperature of 60-70 ° C. This separates the magnesium. Compound of 1-oxy-3-methyl-pentene- (2) - in (4) as gray sludge. While cooling with ice and passing in nitrogen, 34 parts by weight of 4 - [2 ', 6', 6 '- trimethyl - cyclohexen - (1') yl] -2-methylbutene- (2) -al- ( 1) Aether added in 60 parts of the room.
The reaction mixture is then stirred at 20 ° C. and refluxed for a further hour. Now add 200 parts by volume of ether and hydrolyze with ice and ammonium chloride. After washing, drying and evaporation of the ether solution, 51 parts by weight of crude, yellow-brown colored condensation product are obtained. Unreacted starting material is then distilled off in a high vacuum at 130 ° C. oil bath temperature. 40 parts by weight of crude condensation product are obtained as residue.
This is dissolved in petroleum ether with a boiling point of 60-80 C and adsorbed on a column of 800 parts by weight of deactivated aluminum oxide. The same solvent is used to wash out dark-colored components, which mainly consist of dehydration products.
The ether eluate consists of pure 1-oxy-3,7- di, methyl - 6 - oxy - 9 - [2 ', 6', 6'-trimethyl-cyclohexen- (1 ') - yl] -nonadiene- ( 2.7) -in- (4). The yield, based on 4-trimethylcyclohexenyl-2-methyl-butenal, is about 50%.
Light yellow, very viscous oil with nD = 1.527. The compound contains 2 active hydrogen atoms and shows no absorption above 260 my in the ultraviolet absorption spectrum. Example 2:
While stirring vigorously, a solution of 20 parts by weight of 1-oxy-3-methyl-pentene- (2) - is added to a boiling ethylmagnesium bromide solution (made from 10 parts by weight of magnesium, 47 parts by weight of ethyl bromide and 100 parts by volume of ether) over the course of one hour in (4) in. 100 parts of space ether added. To complete the reaction, it is then refluxed for 4 hours at an oil bath temperature of 60 ° C. The magnesium compound is deposited as gray sludge.
While cooling with ice and passing in nitrogen, 22 parts by weight of 4- [2 ', 6', 6'-trimethyl-cycloIiexen- (1 ') -yl] -2-methyl-butene (2) -al are added in the course of one hour - (1) ether added in 50 parts of space. The reaction mixture is then stirred for a further 12 hours at 0 ° C. and then refluxed for 2 hours.
200 parts by volume of ether are added and the well-cooled solution is slowly mixed with 100 parts by volume of a 10 0 strength ammonium chloride solution. After washing, drying and evaporating the ether solution, 42 parts of crude, brown-colored condensation product are obtained.
The 1-oxy-3-methyl-pentenine used in excess is now distilled off in a high vacuum. The residue is distilled in a malecular distillation apparatus at 1.0-5 mm Hg and 120-130 "C. This gives almost pure 1-oxy-3,7-dimethyl-6-oxy-9-trimethylcyclohexenyl-rionadiene- (2, 7) -in- (4)
. The yield, based on 4 - trimethylcyclohexenyl - 2 - methyl - butenal, is 60-706. Yellow, very viscous 01.
