Description
Verfahren zur Darstellung eines neuen Esters. Es wurde gefunden, dass man zu einem neuen Ester des Desoxycorticosterons gelan gen kann, wenn man ein 21-Halogenpregnen- 3,20-dion mit einem palmitylierenden Mittel behandelt.
Geeignete palmitylierende Mittel sind z. B. die Salze der Palmitinsäure, wie Na- triumpalmitat.
Das so gewonnene Desoxycorticosteron- palmitat schmilzt bei 60-61 . Die neue Ver bindung zeichnet sich durch besonders gün stige physiologische Wirkung aus und soll deshalb therapeutische Verwendung finden. <I>Beispiel:</I> Zu einer Lösung von 0,5 Teilen 21-Chlor- pregnen-3,20-dion in 20 Teilen absolutem Alkohol wird 1 Teil Natriumpalmitat zu gefügt. Man erwärmt hierauf das Gemisch bis zur beendeten Kochsalzabscheidung und filtriert. Das durch Wasserzusatz zum Filtrat ausfallende Reaktionsprodukt wird auf der Nutsche mit Wasser gewaschen.
Man gewinnt so das Desoxycorticosteron-palmitat, das nach Umkristallisation aus Petroläther bei 60-61 schmilzt und eine spezifische Drehung von [a] D + 128 (in Chloroform) aufweist.
Process for the preparation of a new ester. It has been found that a new ester of deoxycorticosterone can be obtained by treating a 21-halopregnene-3,20-dione with a palmitylating agent.
Suitable palmitylating agents are e.g. B. the salts of palmitic acid, such as sodium palmitate.
The deoxycorticosterone palmitate obtained in this way melts at 60-61. The new connection is characterized by a particularly favorable physiological effect and should therefore be used therapeutically. <I> Example: </I> 1 part of sodium palmitate is added to a solution of 0.5 part of 21-chloro-pregnen-3,20-dione in 20 parts of absolute alcohol. The mixture is then heated until the separation of sodium chloride has ended and filtered. The reaction product precipitated by adding water to the filtrate is washed with water on the suction filter.
The deoxycorticosterone palmitate is obtained in this way, which after recrystallization from petroleum ether melts at 60-61 and has a specific rotation of [a] D + 128 (in chloroform).
