Verfahren zur Darstellung von 4-Benzyl-6-methyl-3-keto-3,4-dihydro-1,4-berrzoxazin. Es ist bekannt, dass durch Einwirkung von Alkyljodid auf ein Alkalisalz des 3-geto-3,4- dihydro-1,4-benzoxazins eine Alkylgruppe an der 4-Stellung eintritt. So wurde durch Wheeler und Burnes (American Chemical Journal, 20, Seite 555 ff.) die Xethyl- und Äthylverbin- dung beschrieben.
Andere an der 4-Stellung alkylierte Abkömmlinge des 3-geto-3,4-di- hydro-1,4-benzoxazins, sowie insbesondere die Aralkylverbindungen sind bis jetzt nicht her gestellt worden.
Es wurde nun gefunden, dass man durch Einwirkung von Benzylierungsmitteln auf 6 Methyl-3-keto-3,4-dihydro-1,4-benzoxazin oder ein 6-Methyl-3-keto-3,4-dihydro-1,4-benzoxa- zinat zum 4-Benzyl-6-methyl-3-keto-3,4-di- hydro-1,4-benzoxazin gelangt, das wertvolle therapeutische Eigenschaften besitzt. Die Ben- zylierung des 6 - Methyl- 3 -keto- 3,4 - dihydr o- 1,4-benzoxazins kann in der verschiedensten Art und Weise erfolgen.
So ist es beispiels weise keineswegs erforderlich, Benzyljodid zu verwenden. Mit Hilfe des technisch leichter zugänglichen und billigeren 13enzylbromids und -chlorids kann man die gewünschte Um setzung ebenfalls sehr leicht durchführen und vorzügliche Ausbeuten erhalten.
Das 4-Benzyl-6-methyl-3-keto-3,4-dibydro- 1,4-benzoxazin kristallisiert in farblosen Na-- deln vom Schmelzpunkt<B>118'.</B> Es ist unlös lich in Wasser und verdünnten Alkalien. ziemlich leicht löslich in Alkohol, löslich in Äther. Währenddem die 4-All,ylderivate nur ganz schwache analgetische Wirkungen auf weisen, ist das 4-Benzyl-6-methyl-3-keto-3,4- dihydro - 1,4 - benzoxazin ein ausgezeichnet krampflösend wirkendes Mittel von sehr ge ringer Giftigkeit. Es soll deshalb therapeu tische Verwendung finden.
<I>Beispiel:</I> 163 Gewichtsteile 6 -3Iethyl - 3 -keto - 3,4 dihydro-1,4-benzoxazin werden in einer alko holischen Natriumäthylatlösung gelöst, die bereitet ist aus 35 Gewichtsteilen metalli schem Natrium und<B>700</B> Gewichtsteilen AI- kohol. Darauf fügt man 200 Gewichtsteile Benzylchlorid zu und kocht die Lösung unter Rückfluss. Nach etwa drei Stunden ist die Urisetzung vollendet, was man an dem Ein tritt phenolphthaleinneutraler Reaktion er kennt.
Nach Zusatz von 60 Gewichtsteilen 30 %iger Natronlauge und Destillation mit Wasserdampf lässt man erkalten. Das in Wasser und Natronlauge unlösliche Reak tionsprodukt erstarrt, worauf man es von der alkalischen Flüssigkeit trennt. Zur Reinigung kann das Reaktionsprodukt im Vakuum de stilliert und aus Methylalkohol oder Petrol- äther umkristallisiert werden.
Man erhält das 4-Benzyl- 6 -methyl - 3 - keto - 3,4 - dihydro -1,4- benzoxazin in farblosen Nadeln vom Schmelz punkt 118 0.
Das als Ausgangsverbindung dienende substituierte Benzoxazin kann ganz analog wie das nicht substituierte erhalten werden. Sein Schmelzpunkt liegt bei 205 0.
Process for the preparation of 4-benzyl-6-methyl-3-keto-3,4-dihydro-1,4-berrzoxazine. For example, it is by no means necessary to use benzyl iodide. With the help of the technically more easily accessible and cheaper 13enzyl bromide and chloride, the desired conversion can also be carried out very easily and excellent yields are obtained.
4-Benzyl-6-methyl-3-keto-3,4-dibydro-1,4-benzoxazine crystallizes in colorless needles with a melting point of <B> 118 '. </B> It is insoluble in water and diluted alkalis. fairly easily soluble in alcohol, soluble in ether. While the 4-all, yl derivatives have only very weak analgesic effects, 4-benzyl-6-methyl-3-keto-3,4-dihydro-1,4-benzoxazine is an excellent antispasmodic agent of very low toxicity . It should therefore find therapeutic use.
<I> Example: </I> 163 parts by weight of 6 -3Iethyl - 3 -keto - 3,4 dihydro-1,4-benzoxazine are dissolved in an alcoholic sodium ethylate solution which is prepared from 35 parts by weight of metallic sodium and <B> 700 parts by weight of alcohol. 200 parts by weight of benzyl chloride are then added and the solution is refluxed. After about three hours, the urination is complete, which is known from the phenolphthalein-neutral reaction.
After adding 60 parts by weight of 30% strength sodium hydroxide solution and distilling with steam, the mixture is allowed to cool. The reaction product, which is insoluble in water and sodium hydroxide solution, solidifies, whereupon it is separated from the alkaline liquid. For cleaning, the reaction product can be distilled in vacuo and recrystallized from methyl alcohol or petroleum ether.
The 4-benzyl-6-methyl-3-keto-3,4-dihydro-1,4-benzoxazine is obtained in colorless needles with a melting point of 118 °.
The substituted benzoxazine used as the starting compound can be obtained entirely analogously to the unsubstituted one. Its melting point is 205 0.
