Verfahren zur Gewinnung eines wasserlöslichen Anthrachinonglykosidgemisches aus Drogen. Die abführende Wirkung der Anthra- chinonglykoside von Drogen, wie rheum, senna, cascara., sagra.da u. a., ist in der Li teratur bekannt und beschrieben. Es be stehen daher zahlreiche Verfahren, um diese Glykoside in möglichst reiner Form zu ge winnen.
Die gebräuchlichen Methoden ver wenden hierzu Zinkoxyd, Magnesiumoxyd, P1)-,0, und anorganische Salze, um die Bal laststoffe aus den Rohextrakten zu entfernen. Keines dieser Verfahren erzielt jedoch ein reines Glykosid.
Gegenstand vorliegender Erfindung ist ein Verfahren zur Gewinnung eines wasser löslichen Anthrachinonglykosidgemisches, das dadurch gekennzeichnet ist, dass man den wasserlöslichen Teil des aus Cascara sagrada durch Extraktion mit einem Lösungsmittel, zum Beispiel Wasser oder Alkohol, erhält lichen Auszuges mit einem wasserunlöslichen Meta.llhydroxyd,insbesondere mit<B>Al</B> uminium- hydroxyd, behandelt, wobei man zu einem reinen Anthra.chinonglykosid gelangt.
Das Verfahren kann so ausgeführt werden, da.ss man in die durch Eindampfen des wässerigen oder alkoholischen Extraktes zur Trockne und Aufnehmen in Wässer gewonnene wäs serige Lösung das betreffende Mela.11- hydroxyd unter Rühren einträgt: Zweck mässig wird hierbei so viel Metallhydroxyd zugegeben, bis eine Probe der Lösung weder durch Bleiacetat, noch durch Kochsalz ge trübt wird; die Ballaststoffe werden dadurch aus der Droge entfernt.
Ist dieser Punkt er reicht - das Metallhydroxyd wirkt hierbei chemisch durch Salzbildung mit den im Reaktionsgemisch als Verunreinigungen vor handenen :Säuren - so wird das itIetall- hydroxyd filtriert, also aus dem Reaktions gemisch entfernt. Die nunmehr durch Ent fernung der unlöslich gewordenen Verun reinigungen gereinigte Glykosidlösung wird in geeigneter Weise zur Trockne gebracht.
Die in feinen Blättchen erhaltene Anthra: chinonglykosidtrockensubstanz, qualitativ stets gleich zusammengesetzt, ist gelb ge färbt, leicht löslich in Wasser und Eisessig, schwer löslich in Sprit und unlöslich in Ben zol. Sie gibt in wässeriger Lösung die für die Anthrachinonglykoside charakteristische Bromverbindung (nach Aweng, Deutsche Apotheker-Zeitung, 1901, Seite 538) und enthält keine Spur von Ballaststoffen.
Das Aluminiumhydroxyd kann durch andere Me- tallhydroxyde, wie Mangan- und Eisen- hy droxyd etc., ersetzt werden.
<I>Beispiel 1:</I> 1000 Gewichtsteile von Cascara sagrada werden mit 96 %igem Sprit oder einem an dern Lösungsmittel erschöpft, die Kolatur zur Troekne eingedampft, in Wasser auf genommen, filtriert und mit frisch gefälltem Aluminiumhydroxyd eine bis zwei Stunden lang gerührt. Man filtriert, bringt die Lösung in der Leere bei 40' zur Trockne und er hält 100 Teile eines gelblichen Pulvers von folgenden Eigenschaften: Es ist sehr hygro skopisch, unlöslich in Benzol, Äther und Chloroform, etwas löslich in Sprit, leicht lös lich in Wasser und Eisessig.
Die wässerige Lösung ist hellbraun, schwach sauer und wird weder durch Kochsalz, noch durch ver dünnte Mineralsäure gefällt (Abwesenheit; von Ballaststoffen). , Beispiel <I>2</I> 1000 Gewichtsteile von Casca.ra sagrada werden mit 96 loigem Sprit erschöpft und der Extrakt zur Trockne verdampft, dann wird in Wasser aufgenommen, filtriert und mit friseh gefälltem Eisenhydroxyd eine bis drei Stunden lang gerührt.
Man setzt so viel Eisenhydroxyd zu, bis eine Probe des Fil trates mit Kochsalzlösung keine Trübung mehr zeigt. Dann filtriert man die Lösung, verdampft sie in der Leere zur Trockne lind erhält ungefähr 100 Teile eines gelblich braunen Pulvers von den in Beispiel 1 be schriebenen Eigenschaften.
