Abstract

Ce procédé permettant de déterminer les concentrations d'amylase comprend les étapes suivantes : (a) on incube une première partie d'un spécimen dont la concentration en amylase est inconnue à une température spécifiée, dans une solution d'amidon tamponnée contenant de l'amidon à une concentration comprise entre 20 et 200 mg/litre; (b) on laisse réagir ce mélange pendant un intervalle de temps spécifié; (c) on ajoute une portion de solution aqueuse d'iode à ce mélange de manière à obtenir un premier échantillon; (d) on prépare un second échantillon en ajoutant une seconde partie du spécimen à un mélange comprenant une seconde portion de solution d'iode et une seconde portion de solution d'amidon, (e) on mesure la densité optique des deux échantillons à une longueur d'ondes comprise entre 300 et 800 nm; et (f) on calcule l'activité de l'amylase dans le spécimen à l'aide d'une courbe d'étalonnage. Le premier et le second échantillon contiennent des concentrations identiques en spécimen et en solution d'amidon. Le temps nécessaire à la mise en oeuvre de ce procédé est inférieur à 10 minutes, et de préférence inférieur à 6 minutes. Ce procédé convient pour surveiller la concentration d'amylase dans les procédés de nettoyage industriel.
