Abstract
Dans la recherche de séquences régulatrices de plantes capables d'entraîner l'expression du gène effecteur déclenchée par les nématodes dans les structures d'alimentation, le marquage par promoteurs semble être un outil précieux. Un grand nombre de plantes transgéniques Arabidopsis thaliana (L.) a été généré. Elles ont été transformées par un gène de .beta.-glucuronidase fonctionnant comme marqueur de promoteur. Deux produits de reconstruction d'ADN-T, pGV1047 et p.DELTA.gusBin19 ont été utilisés. Les réponses précoces à l'invasion par les nématodes ont été trés interessantes. Quatre lignées présentant une activité GUS dans les syncytia induite par les nématodes à kystes de la betterave ont été étudiées. L'activation du gène reporter a également été identifiée dans les galles induites par les nématodes ectoparasites et des neudosités des racines. Des études ont montré que les quatre marqueurs étaient activés de manière différentielle pendant le développement de la structure d'alimentation. Des zones adjacentes ADN-T responsables des réponses observées après la nématodose ont été isolées et caractérisées pour l'activité du promoteur.
