Abstract

L'invention concerne un procédé qui permet de tester rapidement une pluralité de composés à la recherche d'une interaction non covalente avec au moins une cible. Le procédé comprend les étapes suivantes: on introduit un flux continu constitué d'une phase mobile dans un spectromètre de masse; on règle les paramètres dudit spectromètre de façon à détecter au moins une cible contenue dans la phase mobile; on injecte progressivement dans ladite phase mobile différents échantillons d'une pluralité de mélanges, chaque mélange comprenant au moins la cible et au moins un composé d'intérêt, de façon à les faire passer dans le spectromètre; et on obtient un spectre de masse qui indique pour chaque mélange la présence ou l'absence d'un complexe constitué de la cible et du composé d'intérêt. De préférence, on injecte la totalité de chaque échantillon dans la phase mobile en moins de 5 minutes, encore mieux en moins d'une minute et idéalement en moins de 30 secondes. On peut multiplexer le procédé de l'invention en constituant un mélange qui contient une seule cible et une pluralité de composés d'intérêt, ou bien un seul composé d'intérêt et une pluralité de cibles de protéines récepteurs ou de complexes récepteur-ligand. Typiquement, on forme la pluralité de mélanges au niveau d'une pluralité de sites adressables, tels que les puits d'une plaque multipuits. On retire les échantillons de mélanges des sites adressables, puis on les injecte progressivement dans la phase mobile de façon à les amener au spectromètre de masse. On peut également déterminer la force relative de l'interaction entre la cible et le composé d'intérêt au sein de chaque complexe en dissociant au moins un complexe constitué d'une cible et d'un composé d'intérêt.
