Abstract

Selon la présente invention, on a mis au point des procédés destinés à modifier des vecteurs rétroviraux dérivés de virus qui ne sont pas connus comme étant pathogènes chez l'homme (par exemple le virus de la leucémie murine de Moloney), afin que ces vecteurs puissent transduire des séquences hétérologues dans des cellules ne se divisant pas. Ainsi, on a découvert que l'on pouvait rendre des rétrovirus capables d'infecter des cellules ne se divisant pas, en introduisant dans la particule virale l'une de ces modifications déterminées bien précisément. Par exemple, on peut associer un élément reconnu par le mécanisme d'importation nucléaire d'une cellule ne se divisant pas, au complexe de nucléoprotéines du rétrovirus. Dans un autre mode de réalisation, on modifie au moins une protéine codée par l'acide nucléique viral gag ou pol, de façon que cette protéine soit reconnue par le mécanisme d'importation nucléaire d'une cellule ne se divisant pas.
