Abstract
"SISTEMA DE EXPRESSãO VIRAL TRANSGêNICO BINáRIO, MéTODO DE ALTERAçãO DA EXPRESSãO DE GENE VEGETAL ENDóGENO OU TRANSGENE EM UMA PLANTA, MéTODO DE ALTERAçãO DOS NìVEIS DE UMA PROTEìNA CODIFICADA POR UM GENE ALVO EM UMA PLANTA E SISTEMA DE REPLICAçãO VIRAL" A presente invenção refere-se a um sistema de expressão viral em vegetais binária regulada. Ele compreende dois componentes cromossomicamente integrados. Um componente é um proreplicon, o qual contém seq³ências virais de ação cis necessárias para a replicação e contém um gene alvo. O outro componente é um gene de replicação de ação trans que compreende um promotor regulado operacionalmente ligado à região codificadora para uma proteína de replicação viral. O proreplicon não contém o gene de replicação essencial para a replicação de replicon, e dessa maneira não pode se submeter à replicação episomal autónoma. Entretanto, a expressão regulada da proteína de replicação de ação trans nas células de vegetais que também contém o proreplicon vai acionar a liberação do replicon livre do proreplicon integrado, resulta em sua replicação episomal in trans, e resulta na expressão do gene alvo, caso esteja presente, através da amplificação de genes. O sistema de expressão é útil para produção de proteínas estranhas, bem como o silenciamento de genes e transgenes endógenos no tecido vegetal. It comprises two chromosomally integrated components. One component is a proreplicon, which contains viral cis-action sequences necessary for replication and contains a target gene. The other component is a trans-acting replication gene that comprises a regulated promoter operably linked to the coding region for a viral replicating protein. Proreplicon does not contain the replication gene essential for replicon replication, and therefore cannot undergo autonomous episomal replication. However, regulated expression of the trans-acting replication protein in plant cells that also contains the proreplicon will trigger the release of the free replicon from the integrated proreplicon, results in its episomal replication in trans, and results in the expression of the target gene, if present. present through gene amplification. The expression system is useful for the production of foreign proteins, as well as the silencing of genes and endogenous transgenes in plant tissue. Tissue-specific expression is controlled by the choice of the promoter that controls the transcription of the trans-acting replication gene.
