Abstract

"MéTODO DE DETERMINAçãO DAS CONCENTRAçõES DE AMILASE". Método para a determinação das concentrações de amilase é proporcionado, compreendendo as etapas de: (a) incubar uma parte da amostra possuindo uma concentração desconhecida de amilase a uma temperatura especificada em uma solução de amido tamponada miscível em água contendo amido a uma concentração entre 20-200 mg/litro; (b) deixar a mistura obtida desse modo reagir durante um período especificado de tempo; (c) acrescentar uma parcela de uma solução aquosa de iodo à essa mistura para se obter uma primeira amostra; (d) preparar uma segunda amostra mediante a adição de outra parte da amostra a uma mistura compreendendo uma segunda parcela da solução de iodo e uma segunda parcela da solução de amido; (e) medir a densidade ou a remissão ótica de ambas as amostras a um comprimento de onda entre 300-800 nm; e (f) calcular a atividade amilase na amostra usando uma curva de calibração; onde ambas a primeira e a segunda amostras contêm concentrações iguais da amostra e da solução de amido, e onde o tempo necessário para a realização do método é menor que 10 minutos, preferivelmente menos de 6 minutos. This method is suitable for monitoring amylase concentration in industrial cleaning processes.
