Abstract

"CéLULA DE LEVEDURA E MéTODOS DE DETECçãO DA INTERAçãO DE UM PRIMEIRO PEPTìDIO COM UM SEGUNDO PEPTìDIO DE UM PAR DE LIGAçãO DE PEPTìDIOS, DE DETERMINAçãO DA INTERAçãO DE UMA AMOSTRA DE TESTE COM UM PRIMEIRO OU UM SEGUNDO PEPTìDIOS DE UM PAR DE LIGAçãO DE PEPTìDIOS E DE DETECçãO SIMULTâNEA DA INTERAçãO DE DOIS PARES DE LIGAçãO DE PEPTìDIOS DIFERENTES". Esta invenção refere-se a novas células hospedeiras modificadas que expressam proteínas fundidas heterólogas e métodos de triagem de amostras de teste com atividade de ligação de peptídios em que a célula hospedeira modificada compreende: (a) uma seq³ência de genes que codifica uma proteína de fusão heteróloga, compreendendo a referida proteína de fusão um primeiro peptídio de um par de ligação de peptídios ou segmento do citado primeiro peptídio, que se junta, quer a um domínio de ligação de DNA, quer ao seu correspondente domínio de ativação transcricional de uma proteína de ativação transcricional; (b) uma seq³ência de genes que codifica uma proteína de fusão heteróloga, compreendendo a referida proteína de fusão um segundo peptídio do par de ligação de peptídios de (a) ou um segmento do mesmo, fundido, quer a um domínio de ligação de DNA, quer ao seu domínio correspondente de ativação transcricional, nenhum dos quais é empregado em (a); (c) um gene luciferase operativamente associado à proteína de ativação transcricional ou parte da mesma; (d) opcionalmente, uma deleção ou mudança no DNA cromossómico da célula hospedeira para a proteína de ativação transcricional, se presente na célula hospedeira selecionada."Yeast Cell and Methods of Detecting the Interaction of a FIRST PEPTIDUS WITH A SECOND PETITUDE CONNECTION, DETERMINING THE INTERACTION OF A TEST SAMPLE WITH A FIRST AND A SECOND PEPTID PETITUS DETECTION PROTECTION OF THE INTERACTION OF TWO DIFFERENT PETIDE CONNECTION PAIRS ". This invention relates to novel modified host cells expressing heterologous fused proteins and peptide binding activity test sample screening methods wherein the modified host cell comprises: (a) a gene sequence encoding a fusion protein said fusion protein, said fusion protein comprising a first peptide of a peptide binding pair or segment of said first peptide, which joins either a DNA binding domain or its corresponding transcriptional activation domain of a transcriptional activation; (b) a gene sequence encoding a heterologous fusion protein, said fusion protein comprising a second peptide of the peptide binding pair of (a) or a segment thereof fused to either a DNA binding domain. either its corresponding transcriptional activation domain, none of which is employed in (a); (c) a luciferase gene operably associated with or part of the transcriptional activation protein; (d) optionally, a deletion or change in chromosomal DNA from the host cell to the transcriptional activation protein, if present in the selected host cell.
