Abstract

The hormone is applied in biotechnology and medicine for diagnostic and experimental purposes. The method is easily implementable and it derives a hormone of relatively high output - from 20 to 30 mkg/g of tissue, with specific activity of 11.2 ME/mg of protein and enhanced summating hormonal activity calculated on the basis of weight of hypophisic tissue. It consists of extraction of frozen and homogeneized human hypophisic glands with a buffer, centrifusion and subsequent precipitation with ammonium sulphate. The extraction is carried out with a buffer at pH from 8.4 to 8.6, while the homogen is centrifuged at low revolutions. The supernatante is subject to ionic exchange chromatography, the protein is eluated and the derived eluate is precipitated with ammonium sulphate and is centrifuged at low revolutions, after which the precipitation is consecutively solved, desalinated and concentrated via ultra-filtration. The concentrate is subject to ionic-exchange chromatography, then eluation and the eluate is treated through adsorption chromatography. The hormonal product is eluated. The derived eluate is concentrated through ultra-filtration and the concentrate is cleansed through gel-filtration.
