est administrée par la voie buaale, d'entraîner fréauemment des effets secondaires importants tels que irritations des muqueuses digestives pouvant aboutir à des gastrites simples ou hémorragiques et même à des ulcères gastro-intestinaux.
Ces inconvénients expliquent l'administration fréquente de l'Indométhacine par la voie rectale sous forme de suppositoires.
Les suppositoires traditionnels à base d'Indométhacine comportent dans la plupart des cas un support constitué d'une substance, ou d'un mélange de substances, hydrophiles telles que les polyethylèneglycols de poids moléculaires variables.
Ces supports présentent l'inconvénient d'être mal tolérés localement et d'entraîner des sensations douloureuses et des épisodes diarrhéiques notamment.
Ils sont néanmoins toujours utilisés actuellement parce qu'ils permettent une biodisponibilité satisfaisante du médicament.

s-uppositoires, telles que du beurre de cacao ou des triglycérides d'huiles végétales naturelles hydrogénées, sont bien connues pour leur bonne tolérance locale.
Cependant de telles masses présentent notamment
des propriétés d'hydrophilie insuffisantes, ce qui a pour conséquence une vitesse de libération trop lente des principes actifs.
Pour remédier à cet inconvénient, on a également proposé-d'ajouter, aux masses pour suppositoires lipophiles des agents tensio-actifs dans le but de conférer aux dites masses un certain caractère d'hydrophilie.

de qualités insuffisantes sur les plans de leurs propriétés physiques : modifications dans les vitesses de cristallisation, d'où diminution des cadences de fabrication; moindre résistance mécanique des suppositoires et vitesse de libération du principe actif encore insuffisante.
La présente invention a pour but de remédier à ces multiples inconvénients et de mettre au point un suppositoire d'Indométhacine permettant de réunir les avantages des masses hydrophiles au point de vue bonne biodisponibilité et des masses lipophiles aux points de vue bonne tolérance et facilité de fabrication.
A cet effet, suivant l'invention, le support utilisé est constitué d'une masse pour suppositoires formée par estérification directe en une seule étape du glycérol et du polyoxyethylèneglycol par des acides gras naturels.
Ces masses sont caractérisées en ce qu'elles s'avèrent douées d'une excellente tolérance rectale, d'absence de toxicité, de propriétés physiques et mécaniques convenables et d'un certain degré d'hydrophilie.
Avantageusement, lé suppositoire est constitué d'une quantité d'Indométhacine comprise par exemple entre 10 et 200 mg et d'une quantité de masse suffisante pour obtenir un suppositoire pesant, par exemple, entre 1g et 3g.
Comme masses pour suppositoires présentant les qualités requises, on citera, par exemple les masses SUPPOCIRE de type "P" des établissements GATTEFOSSE à Lyon (France).
On donne ci-après des exemples non limitatifs
de préparation de suppositoires suivant l'invention.

On peut également utiliser des masses SUPPOCIRE BP ou CP afin d'obtenir des points de fusion plus élevés.
Les suppositoires préparés suivant l'exemple 1 ont permis de mettre en évidence l'excellente biodisponibilité de l'Indométhacine.
A cet effet, on a effectué une étude croisée sur volontaires sains consistant en l'administration à 10 volontaires de deux types de suppositoires d'Indométhacine : Indocid suppositoires de Merck Sharp et Dohme et suppositoires del'exemple 1; l'ordre des administrations étant tiré au sort et les administrations étant espacées d'une semaine.
Des prises de sang ont été effectuées régulièrement pendant les 6 heures qui ont suivi l'administration des suppositoires et on a'dosé l'Indométhacine dans les échantillons sanguins ainsi recueillis.
Les résultats de ces dosages ont permi d'établir'les courbes de relations taux sanguins en fonction du temps et de mesurer les biodisponibilités relatives au moyen des surfaces sous ces courbes (SSC) ainsi que certains paramètres pharmacocinétiques résumés dans le tableau ci-dessous :

On peut donc conclure à l'équivalence de ces deux formes de suppositoires quant à la biodisponibilité de l'Indométhacine.
La tolérance locale supérieure des suppositoires de l'exemple 1 a également été mise
en évidence au cours d'une étude croisée en administration répétée à des patients souffrant d'affections de type inflammatoire.
Les résultats obtenus permettent de conclure à une tolérance rectale nettement meilleure chez 60% des patients et meilleure chez 30 autres pourcent, des suppositoires,de l'exemple 1 par comparaison aux suppositoires Indocid. Dans aucun des cas étudiés, la tolérance des suppositoires de l'exemple 1 n'a été jugée inférieure.
REVENDICATIONS .-
1. Suppositoire à base d'Indométhacine, caractérisé en ce qu'il contient une dose unitaire d'Indométhacine en combinaison avec une base constituée d'esters d'acides gras et possédant des propriétés hydrophiles.
2. Suppositoire suivant la revendication 1, caractérisé en ce que le dosage unitaire d'Indomêthacine se situe dans la gamme d'environ
25 à 200 mg.

is administered by the oral route, frequently causing significant side effects such as irritations of the digestive mucous membranes which can lead to simple or hemorrhagic gastritis and even to gastrointestinal ulcers.
These drawbacks explain the frequent administration of Indomethacin by the rectal route in the form of suppositories.
Traditional suppositories based on Indomethacin comprise in most cases a support consisting of a substance, or a mixture of hydrophilic substances, such as polyethylene glycols of variable molecular weights.
These supports have the disadvantage of being poorly tolerated locally and of causing painful sensations and diarrheal episodes in particular.
They are nevertheless still used today because they allow satisfactory bioavailability of the drug.

S-uppositoires, such as cocoa butter or triglycerides of hydrogenated natural vegetable oils, are well known for their good local tolerance.
However, such masses present in particular
insufficient hydrophilicity properties, which results in a too slow release rate of the active ingredients.
To remedy this drawback, it has also been proposed to add surfactants to the masses for lipophilic suppositories in order to give said masses a certain hydrophilic character.

insufficient qualities in terms of their physical properties: changes in the crystallization rates, hence a decrease in production rates; lower mechanical resistance of the suppositories and speed of release of the active ingredient still insufficient.
The object of the present invention is to remedy these multiple drawbacks and to develop an indomethacin suppository making it possible to combine the advantages of hydrophilic masses from the point of view of good bioavailability and of lipophilic masses from the point of view of good tolerance and ease of manufacture. .
To this end, according to the invention, the support used consists of a mass for suppositories formed by direct esterification in a single step of glycerol and polyoxyethylene glycol with natural fatty acids.
These masses are characterized in that they prove to be endowed with excellent rectal tolerance, absence of toxicity, suitable physical and mechanical properties and a certain degree of hydrophilicity.
Advantageously, the suppository consists of an amount of Indomethacin of for example between 10 and 200 mg and of an amount of mass sufficient to obtain a suppository weighing, for example, between 1g and 3g.
As masses for suppositories having the required qualities, mention may be made, for example, of the SUPPOCIRE masses of the "P" type from the GATTEFOSSE establishments in Lyon (France).

The suppositories prepared according to Example 1 made it possible to demonstrate the excellent bioavailability of Indomethacin.
To this end, a crossover study was carried out on healthy volunteers consisting in the administration to 10 volunteers of two types of suppositories of Indomethacin: Indocid suppositories of Merck Sharp and Dohme and suppositories of example 1; the order of the administrations being drawn by lot and the administrations being spaced one week apart.
Blood samples were taken regularly for 6 hours after administration of the suppositories and Indomethacin was dosed in the blood samples thus collected.
The results of these assays made it possible to establish the curves of blood rate relationships as a function of time and to measure the relative bioavailability by means of the surfaces under these curves (SSC) as well as certain pharmacokinetic parameters summarized in the table below:

We can therefore conclude that these two forms of suppositories are equivalent in terms of the bioavailability of Indomethacin.
The higher local tolerance of the suppositories of example 1 was also set.
highlighted in a crossover study in repeated administration to patients suffering from inflammatory type conditions.
The results obtained make it possible to conclude that rectal tolerance is significantly better in 60% of the patients and better in 30 other percent, of the suppositories, of Example 1 compared to the Indocid suppositories. In none of the cases studied, the tolerance of the suppositories of Example 1 was not considered lower.

1. Indomethacin-based suppository, characterized in that it contains a unit dose of Indomethacin in combination with a base consisting of fatty acid esters and having hydrophilic properties.
