Par l'expression "préparations pharmaceutiques applicables par voie topique", on entend, en particulier, les préparations dans lesquelles la substance active se présente sous une forme résorbable par la peau, par exemple, conjointement . avec des substances auxiliaires et/ou les additifs habituellement utilisés pour l'application topique.

sont utilisés, par exemple, comme agents anti-inflammatoires
non stéroïdaux lors du traitement des processus inflammatoires.
En l'occurrence, les préparations correspondantes sont administrées principalement par voie orale, de même que par voie entérale ou par voie parentérale mais, lorsqu'elles sont appliquées
de la sorte, on observe des effets secondaires, principalement dans le domaine gastro-intestinal, par exemple, la formation d'ulcères sur les muqueuses des voies gastro-intestinales. Le
but du traitement de différentes formes de maladies inflammatoires, en particulier, du "rhumatisme des parties molles", consiste principalement à éviter essentiellement les effets secondaires associés à la thérapie systémique. A cet effet, il est surtout indiqué de recourir à une thérapie topique si l'on parvient à assurer le transfert de la substance active dans la région enflammée. Toutefois, une thérapie par application percutanée échoue fréquemment du fait que, lors de l'utilisation de substances actives de formule (IIIa), on ne parvient que dans une mesure insuffisante à faire pénétrer une quantité thérapeutiquement efficace de la substance active à travers la peau dans le tissu malade.
La présente invention est basée sur la constatation étonnante selon laquelle les composés de formule (I) se caractérisent par des propriétés remarquables de pénétration et de résorption percutanées.
De plus, les composés de formule I que l'on doit utiliser suivant l'invention, possèdent des propriétés anti-inflammatoires et analgésiques remarquables. Par suite des différences physicochimiques des composants, les mélanges d'isomères optiques ou les racémates obtenus peuvent être séparés en leurs constituants purs. C'est ainsi que, par exemple, des racémates d'antipodes optiques peuvent être séparés en antimères correspondants par des procédés connus en soi, par exemple, par des procédés chromatographiques, par cristallisation fractionnée,
avec des micro-organismes ou des enzymes. En outre, on peut,

ou du nombre de groupes amino substitués intervenant dans le composé correspondant de formule IIIb.
Comme sels d'addition d'acide d'aminés de formule IIIb, on utilise, par exemple, des halogénhydrates correspondants,
par exemple, des chlorhydrates.
La réaction des composés de formule IIIa avec des composés de formule IIIb a avantageusement lieu dans un solvant ou un diluant inerte, au besoin avec refroidissement et chauffage, par exemple, à une température se situant dans l'intervalle allant d'environ 0 à environ 100[deg.]C, de préférence, à la température ambiante, dans un récipient fermé et/ou sous une atmosphère d'un gaz inerte tel que, par exemple, l'azote.

L'invention concerne également les formes de réalisation du procédé selon lesquelles on prépare les substances de départ in situ, ou dans lesquelles on obtient une substance

Les préparations pharmaceutiques applicables par voie topique suivant l'invention contiennent les composés de formule
I pouvant être utilisés en pharmacie, conjointement avec une matière auxiliaire ou un additif pouvant être appliqué en pharmacie. La dose quotidienne de la substance active dépend
de l'âge et de l'état individuel, ainsi que du mode d'application.
Parmi les préparations pharmaceutiques applicables
par voie topique, il y a principalement les crèmes, les pommades et les gels, de même que les pâtes, les mousses, les teintures et les solutions contenant environ 0,5 à environ 5% de la substance active.
Les crèmes ou les lotions sont des émulsions huile-danseau contenant plus de 50% d'eau. Comme base huileuse, on utilise principalement les alcools gras, par exemple, l'alcool laurylique, l'alcool cétylique ou l'alcool stéarylique, les acides gras, par exemple, l'acide palmitique ou l'acide stéarique,
les cires liquides à solides, par exemple, le myristate d'isopropyle, la cire de suint ou la cire d'abeilles, et/ou des hydrocarbures, par exemple, la vaseline (pétrolatum) ou l'huile paraffinique. Parmi les émulsionnants, il y a les substances tensio-actives possédant principalement des propriétés hydrophiles, de même que les émulsionnants non ioniques correspondants, par exemple, les esters d'acides gras de polyalcools ou leurs produits d'addition d'oxyde d'éthylène tels que les
esters d'acides gras de polyglycérine ou les esters d'acides gras de polyoxyéthylène-sorbitanne (Tweens), de même que les éthers d'alcools gras ou les esters d'acides gras de polyoxy-éthylène, ou encore les émulsionnants ioniques correspondants tels que les sels de métaux alcalins de sulfates d'alcools gras, par exemple, le lauryl-sulfate de sodium, le cétyl-sulfate de sodium ou le stéaryl-sulfate de sodium que l'on utilise habituellement en présence d'alcools gras, par exemple, l'alcool cétylique ou l'alcool stéarylique. Les additifs ajoutés à la phase aqueuse sont, entre autres, des agents empêchant le dessèchement des crèmes, par exemple, les polyalcools tels que la glycérine, la sorbite, le propylène-glycol et/ou les polyéthylène-glycols, de même que les agents de conservation, les produits odorants, etc.
Les pommades ou les lotions sont des émulsions eaudans-huile contenant jusqu'à 70% mais, de préférence, environ
20 à environ 50% d'eau ou d'une phase aqueuse. Comme phase grasse, on emploie principalement des hydrocarbures, par exemple, les vaselines, l'huile paraffinique et/ou les paraffines dures contenant des composés hydroxy appropriés, de préférence, pour améliorer le pouvoir de fixation d'eau, notamment des alcools gras ou leurs esters, par exemple, l'alcool cétylique ou les alcools de cire de suint ou la cire de suint. Les émulsionnants sont des substances lipophiles correspondantes telles que les esters d'acides gras de sorbitanne (Spans), par exemple,

The active substance is dissolved in propylene glycol and water. We melt the constituents together
fatty, namely petrolatum, wax and fatty acid ester
sorbitan. Next, the active substance solution is emulsified in the fat phase and, if desired,
odorous products (0.1%) are added.
Similarly, an ointment containing 0.5% OR 2% of the active substance is obtained.

The active substance is dissolved in polyalkylene glycol and water. The triglyceride, the paraffinic oil and the fatty acid ester of glycerinesorbitan are melted together. Then, the aqueous phase is emulsified in the fat phase. If desired, odorous products (0.1%) are added.
Similarly, a lotion containing 0.5% of the active substance is obtained.
As described in Examples 8 to 16, ointments, creams, gels, microemulsions, lotions and solutions containing 0.5%, 2% or 5% of the compounds mentioned in Examples 1 to 7 are obtained. .
