On a trouvé qu'une souche de Streptomyces A 2408 isolée à partir d'un échantillon de terre d'un champ de soja de Kakegawa-shi, Shizuoka-ken, Japon, produit une enzyme qui catalyse une réaction avec le

rophosphate dans ses cellules et qui, après purificaticn,est électrophorétiquement homogène. Comme résultat, cette enzyme appartient au groupe GK, toutefois ses propriétés physico-chimiques et biochimiques sont inconnues jusqu'à présent et on l'a qualifiée de nouvelle enzyme GK.
Les propriétés taxonomiques de la souche de Streptomyces A 2408 sont les suivantes.
I. Propriétés morphologiques :
La couleur des hyphes aériens portant des spores mûres va d'un gris brunâtre à un brun grisâtre. Mycélium du substrat brun jaunâtre. Produit un pigment soluble brun jaunâtre.
Les observations sur la gélose à l'ami-don et aux substances inorganiques à 30[deg.]C pendant 10 jours sont illustrées comme suit. On observe les mêmes caractéristiques morphologiques sur de la gélose à la farine d'avoine et de la gélose glycérolée à l'asparagine.

La comparaison entre la présente souche A 2408 et Streptomyces canus ISP 5017 a montré la ressemblance en ce qui concerne les propriétés morphologiques, culturales et physiologiques.
En conséquence, la présente souche A 2408 est appelée Streptomyces canus A 2408. Cette souche a été déposée à l'Institut pour l'Industrie et la Technologie microbiennes au Japon comme collection permanente FERM-P N[deg.] 4977.

Un but de la présente invention est de fournir une nouvelle GK et un procédé pour sa production.
Un micro-organisme utilisé dans la présente invention peut être mentionné, par exemple Streptomyces illustré ci-dessus, mais ce n'est pas limitatif et on peut utiliser le présent micro-organisme producteur de GK et ses souches mutantes.

mais, de la poudre de soja, de la poudre de graines

La présente enzyme peut être isolée à partir du mycélium. On centrifuge le bouillon de culture pour recueillir le mycélium qu'on soumet à une sonication, puis à une centrifugation pour recueillir la solution surnageante. On traite cette solution par relargage en utilisant par exemple du <EMI ID=24.1> fate de sodium pour séparer les impuretés. On soumet ensuite le liquide surnageant à un relargage et on effectue une centrifugation pour obtenir le précipité contenant GK. La GK précipitée est ensuite purifiée par relargage avec Sephadex G-25 (nom commercial, Pharmacia Co.), Biogel P-2 (nom commercial, Biorad Corp.), par dialyse à travers une membrane ou au moyen de fibres creuses et traitée par échange d'ions comme en utilisant de la DEAE-cellulose.
La fraction de GK ainsi obtenue est relarguée et concentrée au moyen d'une membrane d'ultrafiltration comme l'Amicon XM-50 (nom commercial, Amicon Co.). La fraction relarguée est soumise à une chromatographie d'absorption comme en utilisant une colonne de gel de phosphate de calcium pour séparation des impuretés et en effectuant ensuite une élution à gradient au moyen d'un tampon phosphate. La fraction contenant GK est relarguée et concentrée à l'aida d'une membrane d'ultrafiltration comme l'Amicon XM-50. On soumet le concentré à une chromatographie par filtration à travers un gel en utilisant par exemple Biogel
