Nouvelles compositions pharmaceutiques solides
à base d'alcaloides de l'ergot de seigle, permettant une libération contrôlée de ce principe
actif La présente invention a pour objet de nouvelles compositions pharmaceutiques solides à base d'alcaloïdes de l'ergot de seigle permettant une libération contrôlée du principe actif.
Les spécialistes du milieu médical s'accordent
à reconnaître que,dans certaines circonstances il est préférable d'administrer un principe actif une seule fois par jour plutôt que de l'administrer plusieurs fois par jour. Les systèmes "retard" sont appropriés pour ce type d'administration car ils permettent une libération contrôlée du principe actif, produisant ainsi un effet thérapeutique prolongé. Dans le cas d'une thérapie aux alcaloïdes de l'ergot de seigle, les systèmes utilisés habituellement pour retarder la libération du principe actif, par exemple à l'aide d'un système de matrice ou
par micro-encapsulation, entraine chaque fois une diminution de la biodisponibilité totale du principe actif.
Il est connu que les alcaloïdes de l'ergot de seigle sont stables en milieu acide, ce qui signifie qu'ils ne sont pas décomposés par les sucs gastriques
et que leur résorption se fait principalement dans le tractus intestinal. Par conséquent, on ne pouvait supposer qu'un médicament à enrobage entérique contenant un alcaloïde de l'ergot de seigle, puisse conduire à une meilleure biodisponibilité.
Or, en poursuivant ses recherches, la Demanderesse a trouvé qu'en présence d'un enrobage entérique
non seulement on observe une durée d'action nettement prolongée de l'alcaloïde de l'ergot de seigle, mais que
la biodisponibilité totale est améliorée d'une manière importante lorsque le noyau enrobé contient, en plus de l'alcaloïde de l'ergot de seigle, un éther polyalcoxyalkylènestérolique.
L'invention concerne donc plus particulièrement une composition pharmaceutique solide à enrobage entérique, sous forme de doses unitaires destinées à être administrées par voie orale, caractérisée en ce que le noyau de cette composition contient un alcaloïde de l'ergot de seigle et un éther polyalcoxyalkylènestérolique.
L'invention comprend également un procédé de préparation des nouvelles compositions pharmaceutiques spécifiées ci-dessus, caractérisé en ce qu'on enrobe avec un enrobage entérique un noyau contenant un alcaloïde

lique (désigné ci-après par éther du stérol).
Par "noyau", il faut entendre le mélange d'un alcaloïde de l'ergot de seigle avec un éther polyalcoxyalkylène du stérol, éventuellement avec des diluants, des supports et des excipients acceptables du point de vue pharmaceutique; ce noyau peut être enrobé par un enrobage entérique. Ce terme "noyau" comprend, au sens le plus large, non seulement des comprimés, des pastilles ou des granulés mais aussi des capsules, par exemple des capsules en gélatine dure ou molle contenant un liquide ou un mélange cireux d'un alcaloïde de l'ergot de seigle et d'un éther polyalcoxyalkylène du stérol, associés éventuellement à des supports et excipients acceptables du point de vue pharmaceutique. Ces capsules sont ensuite enrobées par un enrobage entérique, par exemple selon des méthodes connues.
Lorsqu'on utilise des comprimés comme noyaux, ceux-ci ont de préférence une dureté .comprise entre environ 10 et 70N.
Après avoir été enrobés par un revêtement entérique, les granulés et les pastilles peuvent être utilisés tels quels ou peuvent servir à remplir des capsules, par exemple des capsules de gélatine dure. Les compositions de l'invention se présentent donc sous forme de comprimés, de pastilles, de granulés ou de capsules.
Par alcaloïde de l'ergot de seigle, on entend les alcaloïdes naturels de l'ergot de seigle tels que l'ergotamine, l'ergocristine, l'a-ergocryptine, la pergocryptine et l'ergocornine, et leurs dérivés synthéti-ques ou semi-synthétiques tels que l'ergovaline, la dihydroergotoxine (connue également sous le nom de codergocrine) et la dihydroergotamine à l'état libre ou sous forme de sels acceptables du point de vue pharmaceutique obtenus par réaction avec des acides minéraux ou organiques tels que l'acide méthanesulfonique, maléique, tartrique ou chlorhydrique.
Les alcaloïdes de l'ergot de seigle préférés sont ceux répondant à la formule I

Les compositions pharmaceutiques particulièrement préférés sont celles contenant la dihydroergoto-xine, la bromocryptine et la dihydroergotamine à l'état libre ou, de préférence, sous forme de sels d'addition d'acides.
Les éthers polyalcoxyalkylène du stérol utilisés de préférence pour les compositions de l'invention sont ceux ayant une valeur HLB (balance hydrophile-lipophile) comprise entre environ 10 et 20, en particulier entre 12 et 16. Il s'agit de préférence des éthers du lanostérol, de la dihydrocholestérine et, en particulier, de la cholestérine ou de mélanges de ces éthers. Les éthers polyalcoxyalkylène du stérol préférés sont les stérols éthérifiés en moyenne par 8 à 75 unités d'alkylène-oxy, de préférence 9 à 30 unités. Le groupe hydroxy présent à la fin de la chaîne alkylène-oxy peut être acylê en totalité ou en partie, par exemple par le reste d'un acide carboxylique aliphatique tel que le reste acétyle.
Les éthers polyalcoxyalkylène du stérol particulièrement préférés sont ceux obtenus par éthérification d'un stérol avec de l'oxyde d'éthylène et/ou de l'oxyde de propylène.
Les éthers du stérol peuvent être préparés selon des méthodes connues, par éthérification du stérol avec une quantité appropriée d'époxyde, suivie éventuellement d'une acylation des groupes hydroxy ainsi obtenus. Ces éthers sont en partie disponibles dans le commerce
et sont vendus par exemple par la société Amerchol sous

nibles dans le commerce et appropriés pour les compositions de l'invention, sont ceux obtenus par alcoxylation par exemple de a) une mole de cholestérine avec environ 24 moles d'oxyde d'éthylène (Solulan� C-24) b) 1 équivalent d'alcools lanoliniques avec environ 16 <EMI ID=8.1> c) 1 équivalent d'alcools lanoliniques avec environ 25 n.

Dans les paragraphes a) à g), le mot "environ" signifie que le nombre indiqué d'équivalents d'oxyde d'éthylène ou d'oxyde de propylène est une valeur moyenne, c'est-à-dire que certains éthers du stérol comportent plus et d'autres moins de groupes éthylène-oxy ou propylèneoxy.
Les alcools lanoliniques sont également connus comme alcools gras de la laine (Handbuch der Kosmetika und Riechstoffe, 2ème éd. 1950, volume I, page 1101 [Janistyn]). Ils consistent en un mélange de cholestérine, de dihydro-cholestérine et de lanostérol.
Par enrobage ou revêtement entérique, il faut entendre tous les enrobages acceptables du point de vue pharmaceutique qui empêchent la libération du principe actif dans l'estomac et qui se décomposent suffisamment dans le tractus intestinal (par contact avec les sucs intestinaux neutres ou alcalins) pour permettre la résorption du principe actif par les parois intestinales. Divers essais in vitro permettant de déterminer si un enrobage peut être classifié ou non comme enrobage entérique, ont été publiés dans les pharmacopées de nombreux pays.
Plus particulièrement, l'expression "enrobage

dant au moins une heure, par exemple pendant 2 heures, lorsqu'ils sont au contact à 36-38[deg.] de sucs gastriques arti-ficiels tels que l'acide chlorhydrique de pH 1,2, et qui se décomposent ensuite en l'espace de 30 minutes dans les sucs intestinaux artificiels tels qu'une solution de dihydrogéno-phosphate de potassium tamponnée à pH 6,8.
L'épaisseur de l'enrobage peut être variable; elle dépend, entre autres, de la perméabilité à l'eau et aux acides et de l'effet retard désiré. D'une manière générale, on obtient des résultats satisfaisants avec un

Pour l'enrobage, on choisit avantageusement des polymères à haut poids moléculaire. Comme polymères appropriés,
on peut citer ceux décrits par exemple dans Hagers Handbuch der pharmazeutischen Praxis, 4ème éd. Vol. 7a, pages 739 à 742 et 776 à 778 (Springer Verlag, 1971) et dans Remington's Pharmaceutical Sciences 13ème éd.,
pages 1689 à 1691 (Mack Publ. Co., 1970) et comprenant
les dérivés des esters de cellulose, les éthers de cellulose, les résines acryliques telles que les copolymères de méthylacrylates, et les copolymères de dérivés de l'acide maléique et de l'acide phtalique.
Les films préférés sont ceux obtenus à partir d'acéto-phtalate de cellulose, de copolymères dérivés
de l'acide méthacrylique et de leurs esters, contenant
au moins 40% d'acide méthacrylique ou, en particulier,
de phtalate d'hydroxypropylméthylcellulose. Comme exemple d'acéto-phtalate de cellulose approprié, on peut citer
le produit CAP commercialisé par Eastman Kodak, Rochester N.Y., USA. Les phtalates d'hydroxypropylméthyl-cellulose appropriés sont par exemple les produits HP 50 et HP 55 commercialisés par Shinetsu, Tokyo, Japon.
Etant donné qu'un enrobage consistant essentiellement de CAP se décompose à un pH supérieur que par exemple un enrobage formé principalement de HP 50, la libération du principe actif sera plus retardée dans le premier cas.que dans le deuxième cas. De ce fait on peut,
par un choix approprié de l'enrobage, obtenir un effet retard optimal en fonction des propriétés du principe actif.
Le procédé de préparation des nouvelles compositions pharmaceutiques peut être effectué selon les méthodes habituellement utilisées pour enrober avec un revêtement entérique.
Pour préparer des comprimés, des pastilles ou
des granulés enrobés, on peut par exemple pulvériser
les noyaux avec une solution de l'enrobage entérique.
Comme exemples de solvants appropriés pour l'enrobage entérique, on peut citer les solvants organiques, par exemple un alcool tel que l'éthanol, une cétone telle que l'acétone, un hydrocarbure halogéné tel que le
chlorure de méthylène ou un mélange de tels solvants, par exemple un mélange à parts égales d'éthanol et d'acétone. Il est également avantageux d'ajouter à cette solution un plastifiant tel que le phtalate de di-n-butyle.
Avant de pulvériser la solution d'enrobage,
on chauffe les noyaux à une température comprise entre
25 et 40[deg.], par exemple au moyen d'air chaud de 40 à 70[deg.]. Pour éviter que les noyaux collent, on interrompt la pulvérisation de la solution d'enrobage de temps à autre et on réchauffe les noyaux. La pression de pulvérisation peut varier dans de larges limites; d'une manière générale, on obtient des résultats satisfaisants lorsque cette pression est comprise entre environ 1 et 1,5 bar. Toutefois, on peut également opérer sans interrompre la pulvérisation de la solution d'enrobage, par exemple par régulation automatique de la quantité à pulvériser en fonction de
la température de l'air du milieu de pulvérisation et/ou des noyaux.
Les compositions à enrobage entérique de l'invention retardent la libération du principe actif: en effet, après administration par voie orale d'une telle composition à l'être humain, on obtient une concentration maximale du principe actif dans le plasma sanguin après <EMI ID=15.1>
suite à l'administration d'une composition habituelle, par exemple un comprimé, est déjà atteint après 30 à 60 minutes.
L'effet retard des nouvelles compositions est également illustré par la détermination de la concentration du principe actif (et de ses métabolites) dans les urines: on constate une concentration maximale dans les urines, environ 6 heures après l'administration de la nouvelle composition alors que la concentration est maximale après environ 2 heures lors de l'administration d'une composition habituelle.
Par ailleurs, les compositions pharmaceutiques entraînent, 6 à 24 heures après leur administration, un niveau plasmatique supérieur à celui observé après l'administration d'une composition usuelle. Ceci a été mis en évidence par la surface sous la courbe de niveau plasmatique (AUC) et sert de mesure pour l'excellente biodisponibilité des compositions de l'invention.
Les compositions pharmaceutiques de l'invention font preuve d'un effet retard souhaité du point de vue thérapeutique, associé à une excellente biodisponibilité et, de ce fait, elles permettent un traitement à raison d'une dose unitaire unique par jour.
Les compositions de l'invention, en particulier les comprimés, peuvent être revêtues en plus par une couche externe contenant un principe actif. Outre le principe actif tel qu'un alcaloïde de l'ergot de seigle, cette couche externe peut encore comprendre des supports et/ou des excipients solubles ou dispersables dans les sucs gastriques tels que le talc, la cellulose microcris-

vinylpyrrolidone etc... De telles compositions peuvent être utilisées par exemple lorsqu'on désire une rapide mise en action du principe actif. Dans ce cas, la concentration élevée initiale de l'alcaloïde de l'ergot de seigle dans le sang est maintenue grâce à la libération contrôlée du principe actif provenant du noyau, dans le tractus intestinal.
Le rapport pondéral optimal dans le noyau de l'alcaloïde de l'ergot de seigle à l'éther du stérol, dépend en grande partie des propriétés physiques de l'éther du stérol mis en jeu, des adjuvants utilisés et du type et de la taille de la composition. Ainsi, la

Toutefois, on obtient généralement de bons résultats lorsque le rapport pondéral de l'alcaloïde de l'ergot de seigle à l'éther du stérol est compris entre 1:1 et 1:25, de préférence entre 1:2 et 1:8; en particulier, ce rapport est de 1:4. Le.rapport pondéral compris entre 1:2 et 1:8 est particulièrement préféré lorsque
les noyaux sont sous forme d'une solution solide de l'alcaloïde de l'ergot de seigle, notamment lorsque la polyvinylpyrrolidone est mise en jeu comme solvant solide.
Les noyaux peuvent être mélangés aux excipients habituels, par exemple des liants, des lubrifiants, des supports et des agents de désagrégation. Pour la préparation des comprimés on pourra utiliser, comme excipients, le gel de silice, le carbonate de calcium, le carbonate de sodium, le lactose, l'amidon, le talc etc.., des agents de granulation et de désagrégation tels que l'amidon, l'acide alginique etc.., des liants tels que l'amidon, la gélatine etc.., et des lubrifiants tels que l'acide stéarique et le talc.
Selon l'invention, on utilise de préférence des noyaux sous forme de solutions solides. Pour préparer de tels noyaux, on procède comme décrit ci-après.
On mélange un alcaloïde de l'ergot de seigle, un éther du stérol et un polymère acceptable du point de vue pharmaceutique, au moins partiellement soluble en milieu aqueux; ce polymère- est de préférence un polyalkylëneglycol, une polyvinylpy rrolidone, un copolymère de la vinylpyrrolidone et de l'acétate de vinyle ou un mélange de tels produits.
Les polyalkylèneglycols appropriés sont les polyéthylèneglycols et les polypropylèneglycols et leurs copolymères d'un poids moléculaire compris entre 200 et
20 000, de préférence entre 4000 et 15 000, en particulier entre 6000 et 13 000. Par polyvinylpyrrolidone, on entend

tageusement un poids moléculaire compris entre 10 000 et
100 000, de préférence entre 11 500 et 40 000, en particulier entre 20 000 et 30 000. Les copolymères de la vinylpyrrolidone et de l'acétate de vinyle contiennent avantageusement 60% en poids de vinylpyrrolidone et 40% en poids d'acétate de vinyle; ces copolymères ont de préférence un poids moléculaire situé entre 30 000 et
100 000, en particulier entre 40 000 et 90 000.
Le cas échéant, on peut ajouter des stabilisants tels que des acides, de préférence l'acide méthanesulfonique, l'acide maléique et l'acide tartrique, pour ajuster le pH des compositions. De préférence, le pH des compositions est compris entre 4 et 6, en particulier entre 4
et 5.
Le rapport pondéral alcaloïde de l'ergot de seigle: éther du stérol: polymère acceptable du point de vue pharmaceutique, peut varier largement; toutefois, on obtient généralement de bons résultats lorsque ce rapport est de l'ordre de 1:1-10:0,1-10, de préférence de 1:2-8:
0,1-10, en particulier de 1:2-5:0,1-5.
Pour la préparation des solutions solides selon l'invention, on utilise les polymères sous forme solide. Lorsque l'un des polymères utilisés est liquide à la

d'un poids moléculaire approximatif de 200, il convient de ne pas mettre en jeu ce polymère tout seul mais d'y associer un polymère solide à la température ambiante.
Les composants indiqués ci-dessus sont dissous par agitation dans un solvant approprié, par exemple un alcool inférieur tel que le méthanol ou l'éthanol, ou le chloroforme, à une température comprise entre 30 et 70[deg.], de préférence entre 40 et 60[deg.]. On élimine ensuite le solvant par évaporation sous pression réduite à la même température. Lors de la préparation de la solution, on peut également n'ajouter qu'une partie du polymère et les éventuels ingrédients auxiliaires et ajouter le reste au cours de l'évaporation du solvant. On laisse la solution limpide ainsi obtenue se solidifier à la température ambiante (15 à 25[deg.]) puis on broie la solution solide ou la dispersion résultante en une poudre fine qu'on sèche,

toutes les traces de solvant.
On mélange ensuite la solution solide ainsi obtenue (utilisée comme noyau de la composition) avec des diluants et excipients acceptables du point de vue pharmaceutique et éventuellement une quantité supplémentaire d'éther du stérol, en procédant selon des méthodes connues; toutefois, la quantité totale d'éther du stérol présente dans le noyau doit être comprise dans les limites indiquées ci-dessus.
Les exemples suivants illustrent la présente invention sans aucunement en limiter la portée. Les températures sont toutes indiquées en degrés celsius.
Exemple 1
Dans une turbine rotative pour enrobage on chauffe à 30[deg.] pendant 10 minutes, au moyen d'air chauffé
à 50[deg.], des noyaux de comprimés composés de 3 g de dihydroergotamine, de 75 g de cholestérine éthoxylée avec environ

dispersion de gelde silice.- Au moyen d'un pistolet pulvérisateur, on pulvérise les noyaux de comprimés avec une
<EMI ID=22.1> lose (HP 50) et de 1,35 g de phtalate de di-n-butyle dans un mélange à parts égales d'éthanol et d'acétone, à une pression de pulvérisation de 1-1,5 bar; on opère selon le procédé connu d'enrobage de comprimés par pulvérisation intermittente. Les comprimés enrobés ainsi obtenus sont ensuite séchés.
Exemple 2
On procède comme décrit à l'exemple 1, mais on utilise 75 g d'alcools lanoliniques éthoxylés avec

au lieu de 75 g de cholestérine éthoxylée avec environ
24 moles d'oxyde d'éthylène.
Exemple 3
Comprimés enrobés comportant des noyaux sous forme de solution solide
Dans un ballon d'un litre à fond rond, on introduit 15 g de mêthanesulfonate de dihydroergotoxine,

(d'un poids moléculaire moyen de 25.000) et 250 ml de méthanol. On place le ballon sur un évaporateur rotatif et on le fait tourner à une température de bain de 60[deg.] jusqu'à ce que le contenu du ballon ait atteint une température de 60[deg.]. A ce moment, il se forme une solution limpide.
Tout en maintenant la température du bain à 60[deg.], on évapore le solvant sous pression réduite jusqu'à ce que le résidu ait une consistance sirupeuse. On décante le résidu dans une capsule et on le laisse se solidifier pendant 2 heures à la température ambiante.
Après avoir séché le résidu solide dans un four à 30[deg.] et sous une pression réduite d'environ 1 Torr pendant environ 12 heures, on le broie en une poudre fine que l'on sèche à nouveau.
On mélange 26,8 g de la solution solide ainsi obtenue avec les excipients suivants:

jusqu'à l'obtention d'un mélange homogène, puis on comprime ce mélange selon les méthodes habituelles, ce qui donne des noyaux de comprimés de 140 mg (dureté 10-32N).
On enrobe ensuite les noyaux de comprimés ainsi obtenus, en procédant comme décrit à l'exemple 1; on pulvérise une solution ayant la composition suivante:

et on poursuit l'opération jusqu'à ce que les noyaux soient enrobés d'environ 10 mg du mélange acéto-phtalate de cellulose/phtalate de di-n-butyle par noyau.
Les comprimés enrobés ainsi obtenus résistent aux sucs gastriques, étant donné que les noyaux restent intacts après le traiteront pendant une heure par des sucs gastriques artificiels de pH = 1,2. La durée de désagrégation dans les sucs intestinaux artificiels est supérieure à
1 heure à pH 5,5; à pH 6,0, elle se situe entre 23 et
28 minutes et à pH 6,8 entre 12 et 16 minutes.
Exemple 4
On procède comme décrit à l'exemple 3, mais on pulvérise des noyaux de 140 mg avec une solution de 140 g de phtalate d'hydroxypropyl-méthylcellulose (HP 50) et de 28 g de phtalate de di-n-butyle dans un mélange de 616 g d'éthanol et de 616 g d'acétone. On opère jusqu'à ce que chaque noyau soit enrobé d'environ 9 mg du mélange phtalate d'hydroxypropyl-méthylcellulose/phtalate de di-n butyle (rapport 10:2).
Exemple 5
En procédant comme décrit à l'exemple 3, on obtient une solution solide composée de 4 g de méthane-

et de 9,1 g de polyvinylpyrrolidone (poids moléculaire moyen de 2.000).
On mélange cette solution solide avec

Par compression de ce mélange, on obtient des noyaux de comprimés de 140 mg.
On pulvérise les noyaux de comprimés ainsi obtenus en procédant comme décrit à l'exemple 4, jusqu'à ce que chaque comprimé soit enrobé d'environ 10 mg ou d'environ 15 mg du mélange phtalate d'hydroxypropylméthylcellulose/phtalate de di-n-butyle.
En procédant comme décrit aux exemples ci-dessus, on obtient les comprimés spécifiés dans le tableau I ci-dessous.
Dans les cas Ib et Ic (voir tableau), le noyau des comprimés se trouve sous forme d'une solution solide.

granulés de cellulose, 104,8 g de lactose et 30 g de talc, jusqu'à obtention d'un mélange homogène. Ce mélange est comprimé en noyaux pesant chacun 140 mg.
b) On pulvérise les noyaux ainsi obtenus avec une solution composée de 140 g de phtalate d'hydroxypropylméthylcellulose et de 28 g de phtalate de di-n-butyle dans un mélange de 616 g d'éthanol et de 616 g d'acétone, jusqu'à ce que chaque noyau soit enrobé d'environ 10 mg de ce mélange. c) On prépare la couche externe de ces comprimés en mélangeant 4 g de .mésylate de dihydroergotamine avec 4 g d'une dispersion de gel de silice, 6 mg de stéarate de magnésium, 166,8 g de poudre de cellulose, 40 g de talc,
191,2 d'amidon de maïs et 348 g d'hydrogénophosphate de calcium, jusqu'à ce que le mélange soit homogène. On comprime ensuite ce mélange avec les comprimés enrobés

Les chiffres du tableau II ci-dessus illustrent clairementle bon effet-retard de la composition de l'exemple 4 ainsi que son excellente biodisponibilité. Lors des essais effectués avec une composition connue du commerce à effet retard et contenant de la dihydroergotoxine, on constate que la biodisponibilité de cette composition est nettement inférieure (60 à 70% de la biodisponibilité de la composition B).
Des essais analogues effectués avec d'autres compositions selon l'invention donnent également de bons résultats. A titre de comparaison, une composition du commerce à effet retard contenant de la dihydroergotamine possède une biodisponibilité inférieure de 30 à 40% à celle constatée avec la composition correspondante n'ayant pas d'effet retard.

It is known that ergot alkaloids are stable in an acid environment, which means that they are not broken down by gastric juices
and that their resorption takes place mainly in the intestinal tract. Therefore, it could not be assumed that an enteric-coated drug containing a rye ergot alkaloid could lead to better bioavailability.
However, by continuing its research, the Applicant has found that in the presence of an enteric coating
not only is there a markedly prolonged duration of action of the ergot alkaloid, but that
total bioavailability is significantly improved when the coated core contains, in addition to the ergot alkaloid, a polyalkoxyalkylene esterolic ether.
The invention therefore relates more particularly to a solid pharmaceutical composition with enteric coating, in the form of unit doses intended to be administered orally, characterized in that the core of this composition contains a rye ergo alkaloid and a polyalkoxyalkylene esterolic ether .

By "nucleus" is meant the mixture of a rye ergot alkaloid with a polyalkoxyalkylene ether of sterol, possibly with diluents, carriers and excipients which are acceptable from the pharmaceutical point of view; this core can be coated with an enteric coating. This term "core" includes, in the broadest sense, not only tablets, lozenges or granules but also capsules, for example hard or soft gelatin capsules containing a liquid or a waxy mixture of an alkaloid of l 'ergot and a polyalkoxyalkylene ether of sterol, possibly combined with pharmaceutically acceptable carriers and excipients. These capsules are then coated with an enteric coating, for example according to known methods.
When tablets are used as cores, they preferably have a hardness of between about 10 and 70N.
After having been coated with an enteric coating, the granules and the pellets can be used as they are or can be used to fill capsules, for example hard gelatin capsules. The compositions of the invention are therefore in the form of tablets, lozenges, granules or capsules.

Particularly preferred pharmaceutical compositions are those containing dihydroergoto-xine, bromocryptine and dihydroergotamine in the free state or, preferably, in the form of acid addition salts.
The polyalkoxyalkylene ethers of the sterol preferably used for the compositions of the invention are those having an HLB value (hydrophilic-lipophilic balance) of between approximately 10 and 20, in particular between 12 and 16. They are preferably ethers of the lanosterol, dihydrocholesterine and, in particular, cholesterol or mixtures of these ethers. The preferred polyalkoxyalkylene ethers of the sterol are the sterols etherified on average with 8 to 75 units of alkylene oxy, preferably 9 to 30 units. The hydroxy group present at the end of the alkylene-oxy chain can be acylated in whole or in part, for example by the remainder of an aliphatic carboxylic acid such as the acetyl residue.
The particularly preferred polyalkoxyalkylene ethers of the sterol are those obtained by etherification of a sterol with ethylene oxide and / or propylene oxide.

The preferred films are those obtained from cellulose acetate phthalate, from copolymers derived
methacrylic acid and their esters, containing
at least 40% methacrylic acid or, in particular,

To prepare tablets, lozenges or

15 g of dihydroergotoxin methanesulfonate are introduced into a one liter round bottom flask,

Example 16
Clinical test
The composition of Example 4 is compared to a solid solution of dihydroergotoxin mesylate and
of tartaric acid (composition A) and to a usual composition of dihydroergotoxin mesylate (composition B).
Each person treated is given 4 mg of dihydroergotoxin mesylate.
The results are shown in Table II below.

The figures in Table II above clearly illustrate the good retardation effect of the composition of Example 4 as well as its excellent bioavailability. During the tests carried out with a known composition with a delay effect and containing dihydroergotoxin, it is found that the bioavailability of this composition is clearly lower (60 to 70% of the bioavailability of composition B).
Similar tests carried out with other compositions according to the invention also give good results. By way of comparison, a commercial composition with a delayed effect containing dihydroergotamine has a bioavailability 30 to 40% lower than that found with the corresponding composition having no retarded effect.
