La présente invention a pour objet de nouveaux dérivés de la vincamine, bromés en position 10, sous forme de bases ou de sels, leur procédé d'obtention et leur application à titre de principes actifs de médicaments.

Le brevet belge 868.613 auquel le présent brevet de perfectionnement se rattache, décrit quelques halogéno-10 vincamines et plus particulièrement la synthèse et les propriétés pharmacologiques de la chloro-10 déhydro-14,15 apovincamine.
De nombreux autres dérivés de la vincamine, halogènes sur le noyau indolique, dont la bromo-10 vincamine elle-même, ont été décrits notamment dans
les brevets français 77 20856, 78 17521, 78 17522,
78 17523, dans les brevets belges 823.409, 862.019
et dans le brevet allemand 2.703.920.
La Demanderesse a maintenant constaté
que le remplacement du groupe methoxycarbonyle des
dérivés halogénés de la vincamine, par un groupe alkoxycarbonyle différent, préférentiellement le
groupe éthoxycarbonyle, peut être facilement réalisé. Cette substitution conduit à des dérivés présentant comparativement des propriétés pharmacologiques plus intéressantes, en particulier au niveau de l'oxygénation cérébrale et du point de vue de la toxicité.
Le vincaminate et l'apovincaminate d'éthyle sont des composés connus dont les propriétés antianoxiques ont fait l'objet de nombreuses études.
Ils peuvent être obtenus à partir des acides correspondants en utilisant comme agent d'estérification l'éthanol ou
le bromure d'éthyle ( Arzneim. Forshung 26, 10a, p.1907,
1976) .
La présente invention concerne également un procédé de préparation des bromo-10 vincaminates et apovincaminates d'alkyle, ainsi que les intermédiaires qui caractérisent ce procédé.
Les composés de l'invention sont en effet avantageusement obtenus à partir des précurseurs des dérivés correspondants, possédant un groupe méthoxycarbonyle ( formule I dans laquelle R1= méthyle). Ces dérivés ont été décrits par la Demanderesse dans les demandes de brevet français
78 17521 et 78 17523. Les précurseurs communs de ces dérivés sont la vincadifformine et la déhydro.14,15 vincadifformine (tabersonine), composés répondant respectivement aux formules
II et III dans lesquelles X = H et R= CH3.

La première étape du procédé de l'invention consiste à transestérifier la vincadifformine I ou la tabersonine II en milieu alcool-acide. L'alcool est choisi en fonction de la nature du groupe alkoxycarbonyle que l'on veut obtenir. Il sera donc choisi parmi les alcools comportant de 2 à 6 atomes de carbone, ramifiés ou non.
Le choix de la méthode de transestérification est limité. Il a en effet été démontré que l'acide vincadifforminique est instable et se décarboxyle rapidement en dëhydro-1,2 aspidospermidine (Zsabon et Otta , Acta Chim., Budapest, 69, 87, 1971).
Les esters ainsi obtenus sont nouveaux et constituent des intermédiaires importants pour obtenir des dérivés alkoxycarbonyle autres que méthoxycarbonyle, ce dernier groupe caractérisant la plupart des alcaloides naturels de la série des aspidospermidines et des éburnamënines.
La transestérification est menée de préférence sous atmosphère inerte en chauffant entre
45 et 100 [deg.]C une solution de vincadifformine ou de tabersonine dans 5 à 20 fois son poids d'alcool strictement anhydre. La solution est saturée en acide chlorhydrique. D'autres acides peuvent toutefois
être utilisés avec succès. Après une durée qui peut varier de 12 heures à 4 jours, l'alcool est évaporé sous pression réduite. Le résidu est repris par un solvant non soluble à l'eau, tel le chloroforme ou le chlorure de méthylène, puis lavé à l'ammoniaque dilué ou toute autre solution aqueuse basique qui peut convenir.
La phase organique est séchée, par exemple par
addition de sulfate de magnésium puis filtration.
Après concentration sous vide, le produit est le plus souvent purifié par chromatographie sur colonne.
Les rendements en produit isolé se situent aux environs de 50%.
La deuxième étape du procédé conduisant aux nouveaux composés de l'invention, consiste à bromer les nouveaux esters de la vincadifformine ou de la tabersonine. La bromation est effectuée, de manière connue, en utilisant de préférence la N-bromosuccinimide. On obtient ainsi
avec de bons rendements les dérivés correspondants bromés
en position 10. Ces bromo-10 vincadifformates d'alkyle
et leurs dérivés sont également des composés nouveaux
qui sont susceptibles de devenir des intermédiaires utiles pour la synthèse de nombreux dérivés d'alcaloides.
La bromation de la vincadifformine elle-même est décrite dans la demande de brevet français 78 17521.
Le passage des bromovincadifformates d'alkyle aux vincaminates correspondants se fait de manière connue par oxydation puis réarrangement en milieu acide. Ce procédé est décrit dans le brevet belge 761.628 déposé par la Demanderesse. Le plus souvent on obtient ainsi un
un mélange de vincaminate et d'épi-16 vincaminate d'alkyle, c'est à dire des composés Dossédant le groupe hydroxyle
en position 16 respectivement au dessus ou en dessous
du plan de la molécule de formule I. Ces deux diastéréoisomères peuvent être facilement séparés par cristallisation. On utilise à cet effet le plus souvent l'acétone comme solvant de cristallisation.
Les bromo-10 apovincaminates d'alkyle sont obtenus, de manière classique, par déshydratation en milieu acide des vincaminates correspondants.
L'opération est effectuée de préférence par solubilisation de la vincamine bromée dans l'acide trifluoroacétique à température ambiante. Cette déshydratation peut également être avantageusement effectuée en utilisant conçue solvant l'acide formique pur porté à reflux pendant 1 à 6 heures.

Les composés selon l'invention forment des sels d'addition avec les acides et à partir de ces sels, on peut former les bases libres correspondantes de
manière classique en milieu alcalin.
L'invention s'étend également aux applications industrielles et notamment pharmaceutiques des produits décrits ci-dessus.
Les composés de l'invention ont été soumis à des essais pharmacologiques qui ont révélé des propriétés intéressantes notamment une activité antianoxique et une activité psychotrope. La substance de référence choisie a été la vincamine. On compare également les effets observés à ceux obtenus avec les dérivés analogues comportant ur. groupe methoxycarbonyle en position 16, composés qui ont été décrits dans les demandes de brevet français sus-mentionnées.
Toxicité aiguë
Les composés de l'invention, ainsi que la vincamine, ont été administrés par voie intragastrique et par voie intraveineuse à des souris de souche Charles

graphiquement selon la méthode de Lichfield et Wilcoxon
(J. Pharmacol. Exp. Therap. 96,99 1946). Quelques résultats sont rassemblés dans le tableau 1 ci-dessous.
Epreuve d'hypoxie hypobare chez la souris.
Des souris de même sexe, de souche Charles River, pesant environ 20 + 2 g sont réparties en trois lots de 10 animaux. Les lots 1 et 2 comportent les animaux traités, c'est à dire ayant reçu la substance
à tester ou la vincamine, plus le solvant à raison de
1 ml/ 100 g de poids corporel. Un troisième lot comporte des animaux témoins c'est à dire ayant reçu uniquement
le solvant à raison de 1 ml/100 g de poids corporel. Les composés étudiés sont administrés par voie intragastrique
30 minutes avant l'essai. Les animaux sont placés dans une atmosphère appauvrie en oxygène par réalisation d'un vide

conditions étant obtenues en 30 secondes. On mesure le temps de survie des animaux au moyen d'un chronomètre.
Les agents capables de favoriser l'oxygénation tissulaire et en particulier cérébrale, entraînent un accroissement du temps de survie et l'on peut donc exprimer par la dose efficace 50 % (DE 50 ), la dose qui augmente de 50 % le temps de survie moyen des souris placées dans les conditions décrites ci-dessus.

Nous avons constaté en outre, qu'au cinquième
de la dose létale (400 mg/kg p.o.), le composé de l'invention Ib augmente le temps de survie des animaux de 412 %. A
une dose équivalente (DL50/5) la vincamine n'augmente ce temps de survie que de 54%.
D'après les essais, les composés de l'invention possèdent donc une activité antianoxique supérieure à celle de la vincamine pour des doses égales ou inférieures à celle de vincamine. De plus, la toxicité des composés comportant un groupe éthoxycarbonyle est inférieure à celle des composés de référence comportant un groupe méthoxycarbonyle. Les toxicités et les doses efficaces des composés de l'invention correspondent donc à de meilleurs indices thérapeutiques

Les composés de l'invention, possédant à la fois
une activité antianoxique et une activité psychotrope,
peuvent être utilisés en thérapeutique pour le traitement
des troubles de la vigilance, en particulier pour lutter contre les troubles imputables à des dommages vasculaires cérébraux et à la sclérose cérébrale en gériatrie, ainsi
que pour le traitement des absences dues à des traumatismes crâniens et le traitement des états dépressifs, des affections cardiocirculatoires ou cérébrovasculaires.
Ces composés, comme la vincamine elle-même, sont susceptibles également d'être utilisés avec succès en ophtalmologie et
en O.R.L., par exemple pour le traitement des troubles cochleovestibulaires.
Pour leur application en thérapeutique, les composés
de l'invention peuvent être administrés soit par voie digestive sous forme de capsules, gélules, comprimés, dragées cachets, solution ou suspension, soit par voie parentérale sous forme de soluté stérile tamponné, préparé à l'avance
ou extemporanément, dans lesquels la substance active, à l'état de base ou salifiée, se trouve présente à raison de
0,5 mg à 150 mg de substance active par dose unitaire. La posologie quotidienne peut varier de 1 mg à 300 mg de substance active selon l'affection.
En vue de leur application thérapeutique, les composés de l'invention sont présentés sous forme de compositions pharmaceutiques contenant comme substance active au moins un composé selon l'invention, éventuellement en mélange avec d'autres substances actives et les adjuvants, diluants, véhicules ou excipients habituels en pharmacie ainsi que les colorants, édulcorants, agents de conservation,
antioxydants etc...
La préparation pharmacologique ou galénique s'ef- fectue par les méthodes classiques, essentiellement par incorporation ou mélange des substances actives dans les adjuvants utilisés.
Dans le cas où les composés sont administrés
sous forme de leurs sels d'addi ion d'acide minéral ou
organique, il convient d'utiliser des acides pharmaceutiquement admissibles qui sont bien connus en pharmacie.

Belgian patent 868,613 to which the present improvement patent relates, describes some halogeno-10 vincamines and more particularly the synthesis and the pharmacological properties of chloro-10 dehydro-14,15 apovincamine.
Numerous other derivatives of vincamine, halogenated on the indole ring, including 10-bromo vincamine itself, have been described in particular in
French patents 77 20856, 78 17521, 78 17522,
78 17523, in Belgian patents 823.409, 862.019
and in German patent 2,703,920.
The Applicant has now observed
that the replacement of the methoxycarbonyl group of
halogenated derivatives of vincamine, with a different alkoxycarbonyl group, preferably the
ethoxycarbonyl group, can be easily made. This substitution leads to derivatives exhibiting comparatively more advantageous pharmacological properties, in particular in terms of cerebral oxygenation and from the point of view of toxicity.
Ethyl vincaminate and apovincaminate are known compounds whose antianoxic properties have been studied extensively.
They can be obtained from the corresponding acids using ethanol as esterification agent or
ethyl bromide (Arzneim. Forshung 26, 10a, p.1907,
1976).
The present invention also relates to a process for the preparation of alkyl bromo-vincaminates and apovincaminates, as well as to the intermediates which characterize this process.
The compounds of the invention are in fact advantageously obtained from the precursors of the corresponding derivatives, possessing a methoxycarbonyl group (formula I in which R 1 = methyl). These derivatives have been described by the Applicant in French patent applications
78 17521 and 78 17523. The common precursors of these derivatives are vincadifformin and dehydro.14,15 vincadifformin (tabersonine), compounds corresponding respectively to the formulas
II and III in which X = H and R = CH3.

The first step of the process of the invention consists in transesterifying vincadifformin I or tabersonine II in an alcohol-acid medium. The alcohol is chosen according to the nature of the alkoxycarbonyl group that is to be obtained. It will therefore be chosen from alcohols comprising from 2 to 6 carbon atoms, branched or not.
The choice of the transesterification method is limited. It has in fact been shown that vincadifforminic acid is unstable and rapidly decarboxylates to 1,2-dehydroaspidospermidine (Zsabon and Otta, Acta Chim., Budapest, 69, 87, 1971).
The esters thus obtained are new and constitute important intermediates for obtaining alkoxycarbonyl derivatives other than methoxycarbonyl, the latter group characterizing most of the natural alkaloids of the series of aspidospermidines and eburnamënines.
The transesterification is preferably carried out under an inert atmosphere by heating between
45 and 100 [deg.] C a solution of vincadifformin or tabersonine in 5 to 20 times its weight of strictly anhydrous alcohol. The solution is saturated with hydrochloric acid. However, other acids can
be used successfully. After a period which can vary from 12 hours to 4 days, the alcohol is evaporated off under reduced pressure. The residue is taken up in a solvent which is insoluble in water, such as chloroform or methylene chloride, then washed with dilute ammonia or any other basic aqueous solution which may be suitable.
The organic phase is dried, for example by
addition of magnesium sulfate followed by filtration.
After concentration under vacuum, the product is most often purified by column chromatography.
The yields of isolated product are around 50%.
The second step of the process leading to the new compounds of the invention consists in brominating the new esters of vincadifformin or of tabersonine. The bromination is carried out, in a known manner, preferably using N-bromosuccinimide. We thus obtain
with good yields the corresponding brominated derivatives
in position 10. These alkyl bromo-10 vincadifformates
and their derivatives are also new compounds
which are likely to become useful intermediates for the synthesis of numerous alkaloid derivatives.
The bromination of vincadifformin itself is described in French patent application 78 17521.
The transition from the alkyl bromovincadifformates to the corresponding vincaminates takes place in a known manner by oxidation then rearrangement in an acidic medium. This process is described in Belgian patent 761,628 filed by the Applicant. Most often we thus obtain a
a mixture of vincaminate and epi-16 alkyl vincaminate, i.e. compounds containing the hydroxyl group
in position 16 respectively above or below
of the plane of the molecule of formula I. These two diastereomers can be easily separated by crystallization. Acetone is most often used for this purpose as the crystallization solvent.
The alkyl bromo-10 apovincaminates are obtained, in a conventional manner, by dehydration in an acidic medium of the corresponding vincaminates.
The operation is preferably carried out by dissolving the brominated vincamine in trifluoroacetic acid at room temperature. This dehydration can also be advantageously carried out using a solvent designed pure formic acid brought to reflux for 1 to 6 hours.

The compounds of the invention were subjected to pharmacological tests which revealed interesting properties in particular an antianoxic activity and a psychotropic activity. The reference substance chosen was vincamine. The effects observed are also compared with those obtained with analogous derivatives comprising ur. methoxycarbonyl group in position 16, compounds which have been described in the aforementioned French patent applications.

Hypobaric hypoxia test in mice.
Mice of the same sex, of the Charles River strain, weighing about 20 + 2 g are divided into three groups of 10 animals. Lots 1 and 2 include the animals treated, i.e. having received the substance
to be tested or vincamine, plus the solvent at the rate of
1 ml / 100 g of body weight. A third batch includes control animals, i.e. having received only
the solvent in an amount of 1 ml / 100 g of body weight. The compounds studied are administered intragastrically
30 minutes before the test. The animals are placed in an oxygen-depleted atmosphere by creating a vacuum

conditions being obtained in 30 seconds. The survival time of the animals is measured using a stopwatch.
Agents capable of promoting tissue and in particular cerebral oxygenation lead to an increase in survival time and we can therefore express by the effective dose 50% (ED 50), the dose which increases the survival time by 50%. using mice placed under the conditions described above.

We further found that in the fifth
of the lethal dose (400 mg / kg p.o.), the compound of the invention Ib increases the survival time of the animals by 412%. AT
an equivalent dose (LD50 / 5) of vincamine only increases this survival time by 54%.
According to the tests, the compounds of the invention therefore have an antianoxic activity greater than that of vincamine for doses equal to or less than that of vincamine. In addition, the toxicity of compounds comprising an ethoxycarbonyl group is lower than that of the reference compounds comprising a methoxycarbonyl group. The toxicities and the effective doses of the compounds of the invention therefore correspond to better therapeutic indices.

The compounds of the invention, having both
antianoxic activity and psychotropic activity,
can be used therapeutically for the treatment
vigilance disorders, in particular to combat disorders attributable to cerebrovascular damage and cerebral sclerosis in geriatrics, as well
as for the treatment of absences due to head trauma and the treatment of depressive states, cardiocirculatory or cerebrovascular disorders.
These compounds, like vincamine itself, are also likely to be used successfully in ophthalmology and

For their therapeutic application, the compounds
of the invention can be administered either by the digestive route in the form of capsules, gelatin capsules, tablets, dragees, tablets, solution or suspension, or parenterally in the form of a sterile buffered solution, prepared in advance
or extemporaneously, in which the active substance, in the basic or salified state, is present in a proportion of
0.5 mg to 150 mg of active substance per unit dose. The daily dosage may vary from 1 mg to 300 mg of active substance depending on the condition.
With a view to their therapeutic application, the compounds of the invention are presented in the form of pharmaceutical compositions containing as active substance at least one compound according to the invention, optionally in admixture with other active substances and adjuvants, diluents, vehicles or excipients usual in pharmacies as well as colorings, sweeteners, preservatives,
antioxidants etc ...
The pharmacological or galenic preparation is carried out by conventional methods, essentially by incorporation or mixing of the active substances in the adjuvants used.
In the event that the compounds are administered
in the form of their mineral acid addition salts or
organic, pharmaceutically acceptable acids which are well known in pharmacies should be used.
