La présente invention concerne des anilinométhylènemalononitriles doués d'une activité anthelminthique, la préparation de ces composés, les médicaments qui en contiennent à titre de substances actives, ainsi que leur application à la lutte contre les helminthes; plus particulièrement contre Fasciola hepatica, chez les animaux domestiques et les animaux d'élevage.

Dans le présent mémoire on entend par "helminthes" des nématodes , des cestodes et des trématodes qui vivent en parasites dans le tractus intestinal ou dans d'autres organes.
Parmi les endoparasites que l'on rencontre chez les homéothermes, les helminthes en particulier causent de graves dommages. C'est ainsi que les animaux infestés par ces parasites ont non seulement une croissance ralentie et une prodiction utile nettement diminuée mais encore ils sont parfois atteints à un degré tel qu'ils succombent. Pour éviter ou,du moins, pour diminuer les pertes de ce genre, qui peuvent prendre des proportions considérables lorsque l'attaque par les vers prend, dans les troupeaux, l'allure d'une épidémie, on s'efforce constamment de mettre au point des produits permettant de

C'est ainsi qu'on connaît déjà toute une série de substances ayant une activité anthelminthique, mais celles-ci ne donnent pas entière satisfaction, notamment parce que , lorsqu'elles sont administrées à des doses admissibles, elles n'ont pas toujours une activité suffisante ou, lorsqu'elles sont admi-

quent des effets secondaires gênants, tels que des intoxications.

se signalent par une bonne activité anthelminthique contre les nématodes, les cestodes et, surtout, les trématodes. Il convient de faire une mention spéciale pour leur activité contre les fasciolidés (Fasciola hepatica).
Les composés suivants sont particulièrement appréciés en raison de leur efficacité contre la fasciolose :
la N-(dicyano-2,2 vinyl)-N-(dichloro-2,4 benzyl) bis-trifluoro-
méthyl-3,5 aniline ;
la N-(dicyano-2,2 vinyl)-N-(dichloro-3,4 benzyl) bis-trifluoro-
méthyl-3,5 aniline ;
la N-(dicyano-2,2 vinyl)-N-(dichloro-2,6 benzyl) bis-trifluoro-
méthyl-3,5 aniline ;
la N-(dicyano-2,2 vinyl)-N-(dibromo-2,3 propyl) bis-trifluoro-
méthyl-3,5 aniline et

Les exemples suivants illustrent la présente invention.
EXEMPLE 1 :
Préparation de la N-(dicyano-2,2 vinyl)-N-(dichloro-3,4 benzyl)

Dans une solution de 45,7 g de bis-trifluorométhyl3,5 N-(dicyano-2,2 vinyl)-aniline dans 200 ml de diméthylsulfoxyde on introduit par portions, tout en agitant, 16,8 g de tert-butylate de potassium. On conserve la solution à 40[deg.]C pendant 4 heures. Tout en continuant d'agiter on y ajoute goutte à goutte 20,8 ml de chlorure de dichloro-3,4 benzyle dans
20 ml de dimêthylsulfoxyde,puis on agite le mélange réactionnel pendant 12 heures à 40[deg.]C. On verse ensuite le mélange sur
2 1 de glace et on extrait au chloroforme le produit formé. On lave à l'eau la solution chloroformique, on la sèche sur sulfate de sodium et on chasse le chloroforme par distillation. On chromatographie le produit brut sur une colonne de gel de silice en utilisant comme éluant un mélange constitué de 1 volume

L'activité anthelminthique des anilinométhylènemalononitriles de formule I est mise en évidence par les
essais décrits ci-dessous.
1. Essais sur des souris infestées par
Nematospiroides dubius.
On administre les substances actives sous la forme d'une suspension, au moyen d'une sonde oesophagienne, à des souris blanches qui sont infestées par Nematospiroides dubius. Chaque animal reçoit les substances actives une fois par jour pendant 3 jours consécutifs. Au huitième jour après le début
du traitement on sacrifie les animaux et on en fait la nécropsie.
On effectue l'évaluation, après la nécropsie des animaux d'épreuve, en comptant les nématodes qui se trouvent dans l'intestin. On utilise, comme témoins, des souris qui ont été infestées en même temps et de la même façon mais qui n'ont pas été traitées.
Les produits sont bien tolérés par les souris qui ne présentent aucun signe pathologique.
2. Essais sur des souris infestées par

On administre les substances actives sous la forme d'une suspension, au moyen d'une sonde oesophagienne, à des souris blanches qui sont infestées par Nippostrongylus brasi-liensis. On utilise 5 animaux pour chaque essai. Chaque animal reçoit les substances actives une fois par jour pendant 3 jours consécutifs. La dose quotidienne par animal est de 100 mg de substance active par kg de poids corporel. Le quatrième jour après le début du traitement, on sacrifie les animaux et on en fait la nécropsie. On effectue l'évaluation, après la nécropsie des animaux d'épreuve, en comptant le nombre de parasites qui restent dans l'intestin, ce nombre étant comparé à celui que l'on obtient sur un témoin qui a été infesté en même temps et de la même façon mais qui n'a pas été traité.
3. Essais sur des souris infestées par Hymenolepis
nana.
On administre les substances actives sous la forme d'une suspension, au moyen d'une sonde oesophagienne, à des souris blanches qui sont infestées artificiellement par Hymenolepis nana. On utilise cinq animaux par essai. Chaque animal de chaque lot reçoit les substances actives une fois par jour pendant trois jours consécutifs. Le quatrième jour après le début du traitement on sacrifie les animaux et on en fait la nécropsie. On effectue l'évaluation, après la nécropsie des animaux d'épreuve, en comptant les cestodes qui se trouvent dans l'intestin et en comparant le nombre obtenu à celui que l'on obtient sur un lot d'animaux témoins qui ont été infestés en même temps et de la même façon mais qui n'ont pas été traités.
4. Essais sur des rats infestés par Fasciola
hepatica.
On infeste des rats blancs de laboratoire avec des grandes douves du foie (Fasciola hepatica)..Une fois passée la période de latence, on traite les rats par une substance

est administrée par une sonde oesophagienne. La substance active est administrée une fois par jour pendant trois jours consécutifs. On utilise trois rats infestés par essai. Deux semaines après l'administration de la substance active on sacrifie les animaux d'essai et on les examine pour rechercher les douves encore présentes.
Les résultats expérimentaux sont rassemblés dans le tableau suivant.

On utilise les produits conformes à l'invention pour combattre les helminthes parasites chez les animaux domestiques et les animaux d'élevage, tels que les bovins,
les moutons, les chèvres, les chevaux, les porcs, les chats,
les chiens et la volaille. Ils peuvent être administrés aux animaux aussi bien en une prise unique qu'en prise répétées. Les prises isolées varient, selon le type de l'animal, entre 0,5 et
100 mg par kilogramme de poids corporel. Une administration étalée dans le temps permet, dans bien des cas, d'obtenir un meilleur effet ou de diminuer les doses totales. Les substances actives, ou les mélanges qui en contiennent, peuvent également être ajoutées à la nourriture ou aux boissons. L'aliment tout préparé renferme les substances (I) en une concentration qui est de préférence comprise entre 0,005 et 0,1 % en poids.
Les produits peuvent être administrés aux animaux par la voie orale ou par la voie abomasale, sous la forme de préparations, par exemple sous la forme de solutions, d'émulsions, de suspensions (boissons), de poudres, de comprimés, de bols et de capsules.
Pour préparer les formes d'application indiquées ci-dessus on se sert par exemple de supports solides usuels, tels

phosphate de calcium, la farine de graine de coton, ou de liquides ne réagissant pas avec la substance active, tels que des huiles et d'autres solvants et diluants inoffensifs pour l'organisme animal. Lorsque les propriétés physiques et toxicologiques de solutions ou d'émulsions le permettent, on peut également injecter les substances actives aux animaux par la voie sous-cutanée.
Lorsque les produits anthelminthiques se trouvent sous la forme de concentrés destinés à être ajoutés à la nourriture, on utilise, comme matières supports, par exemple du foin, des aliments à hauts rendements, des céréales fourragères et des concentrés protéiniquas. En plus des substances actives, ces concentrés alimentaires peuvent contenir des additifs, des vitamines, des antibiotiques, des chimiothérapiques et d'autres pesticides, surtout des bactériostatiques, des fongistatiques et des coccidiostatiques, ou encore des préparations hormonales,
des substances à effet anabolisant ou d'autres composés stimulant la croissance, améliorant la qualité de la viande des animaux de boucherie ou agissant d'une autre manière favorable pour le développement de l'organisme.
On peut également les associer à d'autres anthelminthiques, ce qui permet d'étendre leur action et de l'adapter à des circonstances données. Comme anthelminthiques supplémentaires, on pourra envisager par exemple :

Pour préparer les produits anthelminthiques qui font l'objet de l'invention on fait appel à des méthodes connues : on mélange intimement et on broie les substances actives de formule générale I avec des supports appropriés, éventuellement en ajoutant des dispersants ou des solvants inertes à l'égard des substances actives.
Les produits anthelminthiques peuvent être présentés et appliqués sous les formes suivantes :
Formulationssolides:
granulés {granulés enrobés, granulés imprégnés et granulés homogènes), concentrés dispersables dans l'eau.
Formulations liquides :
solutions, pâtes, émulsions (plus particulièrement émulsions prêtes à l'emploi).
Il est bon que la granularité des supports soit d'au plus environ 0,1 millimètre pour les agents de poudrage et poudres pour bouillies, et qu'elle soit comprise entre

Les concentrations des matières actives dans les formes de présentation solides peuvent aller de 0,5 à 80 % et, dans les préparations liquides,de 0,5 à 15 %.
On peut en outre ajouter à ces mélanges des additifs ayant pour effet de stabiliser la substance active et/ou
des substances non ionogènes, anioniques ou cationiques,
qui ont par exemple pour fonction d'améliorer la mouillabilité
(mouillants) ainsi que la dispersabilité (dispersants).
Voici, à titre d'exemple, la composition d'un mélange pulvérulent dispersable dans l'eau :
25 parties d'une substance active de formule I,
3 parties d'un mélange d'un polyoxyéthylène, d'un ester

Preparation of N- (2,2-dicyano vinyl) -N- (3,4-dichloro benzyl)

Into a solution of 45.7 g of bis-trifluoromethyl3.5 N- (2,2-dicyano-vinyl) -aniline in 200 ml of dimethylsulfoxide is introduced in portions, while stirring, 16.8 g of potassium tert-butoxide . The solution is stored at 40 [deg.] C for 4 hours. While continuing to stir, 20.8 ml of 3,4-dichlorobenzyl chloride are added dropwise thereto in
20 ml of dimethylsulfoxide, then the reaction mixture is stirred for 12 hours at 40 [deg.] C. The mixture is then poured over
2 l of ice and the product formed is extracted with chloroform. The chloroform solution is washed with water, dried over sodium sulfate and the chloroform is distilled off. The crude product is chromatographed on a column of silica gel using a mixture consisting of 1 volume as eluent.

The anthelmintic activity of anilinomethylenemalononitriles of formula I is demonstrated by the
tests described below.
1. Tests on mice infested with
Nematospiroides dubius.
The active substances are administered in the form of a suspension, by means of an esophageal probe, to white mice which are infested with Nematospiroides dubius. Each animal receives the active substances once a day for 3 consecutive days. On the eighth day after the start
of treatment animals are sacrificed and necropsy.
Post-necropsy assessment of test animals is made by counting nematodes in the intestine. Mice which were infested at the same time and in the same way but which were not treated were used as controls.
The products are well tolerated by mice which show no pathological signs.
2. Tests on mice infested with

The active substances are administered in the form of a suspension, by means of an esophageal probe, to white mice which are infested with Nippostrongylus brasi-linksis. 5 animals are used for each test. Each animal receives the active substances once a day for 3 consecutive days. The daily dose per animal is 100 mg of active substance per kg of body weight. On the fourth day after the start of treatment, the animals are sacrificed and necropsied. The evaluation is made, after necropsy of the test animals, by counting the number of parasites that remain in the intestine, this number being compared to that obtained on a control which has been infested at the same time and in the same way but which has not been treated.
3. Tests on mice infested with Hymenolepis
nana.
The active substances are administered in the form of a suspension, by means of an esophageal probe, to white mice which are artificially infested with Hymenolepis nana. Five animals are used per test. Each animal in each batch receives the active substances once a day for three consecutive days. On the fourth day after the start of treatment, the animals are sacrificed and given a necropsy. The evaluation is made, after necropsy of the test animals, by counting the cestodes which are in the intestine and comparing the number obtained with that obtained from a batch of control animals which have been infested. at the same time and in the same way but which have not been treated.
4. Tests on rats infested with fasciola
hepatica.
White laboratory rats are infested with large liver fluke (Fasciola hepatica). Once the latency period has passed, the rats are treated with a substance.
